Fabrication of Poly(L-lactide) Scaffold Membranes through Phase Inversion with Pure and Mixed Solvents (순수용매와 혼합용매를 이용한 상전이를 통한 Poly(L-lactide) 스캐폴드 막의 제조)

  • This paper reports a fabrication of poly(L-lactic acid) (PLLA) scaffold membranes through phase separation process using pure and mixed solvents. Chloroform and 1,4-dioxane were used as pure solvents and mixed solvents were obtained by mixing the pure solvents together. Morphologies, mechanical properties and mass transfer characteristics of the scaffold membranes were investigated through SEM, stress-strain test and glucose diffusion test. Scaffold membranes from the solution with pure chloroform showed solid-wall pore structure. In contrast, nano-fibrous membranes were fabricated from the solution with pure 1,4-dioxane. In case of mixed solvents, the scaffold membranes showed various structures with changing composition of the solvents. When 1,4-dioxane content was lower than 20 wt% in the solvent, scaffold membrane showed solid-wall pore structure. When the content was 20 wt%, scaffold membranes with macropores with the maximum size of $100{\mu}m$ was obtained. In the concentration range of 1,4-dioxane over 25 wt%, the scaffold membranes showed nano-fibrous structures. In this range, the fibers showed different diameters with changing composition of the solvent. The minimum fiber diameter was about $15{\mu}m$, when 1,4-dioxane composition was 80 wt%. These results indicate that the composition of the solvent showed a significant effect on the structure of scaffold membrane.

Optimization for Chia Seed Antioxidative Activity of Solvent Extraction Using the Response Surface Methodology (반응표면 분석법을 이용한 치아씨 항산화 활성 추출의 최적화 조건)

  • The purpose of this study was to determine the optimum conditions of solvent extraction type and solvent concentration (60, 70, 80%, v/v), extraction time (30, 80, 130 mins) and extraction temperature (10, 15, $20^{\circ}C$) in order to increase the antioxidant activity of the chia seed. The total polyphenol content and DPPH radical scavenging activity was measured by using response surface methodology (RSM) to establish the optimal conditions. Using ethanol and methanol extractions at extraction concentrations of 60%, and time and temperature of 130 mins and $20^{\circ}C$, the maximum total polyphenol content was 871.00 mg% ($R^2=0.9507$) and 557.70 mg% ($R^2=0.9784$) for ethanol and methanol extraction respectively. Using the same extraction conditions, the maximum level of DPPH radical scavenging activity was 72.14% ($R^2=0.9675$) and 52.79% ($R^2=0.9524$) for ethanol and methanol extraction respectively. The results indicate that ethanol extracts showed a higher antioxidant activity than methanol extracts. The ethanol extraction conditions of response surface analysis (RSA) were affected more by ethanol concentration than by extraction time or temperature. In contrast, the methanol extraction conditions of response surface analysis (RSA) were affected more by extraction time. Based on the RSM, the optimum ethanol extraction conditions were the following: extraction concentration, 63%: time, 100 mins: and temperature, $18^{\circ}C$. The optimum methanol extraction conditions were the following: extraction concentration, 65%; time, 120 mins; and temperature, $16^{\circ}C$.

  • 효소는 반응을 효율적으로 촉진시킬 뿐만 아니라 기질 및 입체특이성을 갖고 있어 광학활성 화합물을 합성하는데 매우 유용한 촉매이다. 효소 반응은 일반적으로 수용액에서 진해오디지만, 유기 용매를 포함하는 용액에서도 가능하다. 대부분의 유기 화학 반응이 유기 용매에서 진행된다는 점을 생각할 때, 유기 용매에서 효소 반응을 이용한 유기합성이 갈수록 중요해지고 있다. 본 글에서는 유기 용매에서 사용가능한 대표적인 효소인 지질 가수분해 효소(lipase)를 이용한 유기합성에 대해서 간략히 기술하고자 한다.

Solvent-tolerant Lipases and Their Potential Uses (유기용매 내성 리파아제와 그 이용가능성)

  • This review described solvent-tolerant lipases and their potential industrial, biotechnological and environmental impacts. Although organic solvent-tolerant lipase was first reported in organic solvent-tolerant bacterium, many organic solvent-tolerant lipases are in not only solvent-tolerant bacteria but also solvent-intolerant bacterial and fungal strains, such as the well-known Bacillus, Pseudomonas, Streptomyces and Aspergillus strains. As these lipases are not easily inactivated in organic solvents, there is no need to immobilize them in order to prevent an enzyme inactivation by solvents. Therefore, the solvent-tolerant lipases have the potential to be used in many biotechnological and biotransformation processes. With the solvent-tolerant lipases, a large number insoluble substrates become soluble, various chemical reactions that are initially impossible in water systems become practical, synthesis reactions (instead of hydrolysis) are possible, side reactions caused by water are suppressed, and the possibility of chemoselective, regioselective and enantioselective transformations in solvent and non-aqueous systems is increased. Furthermore, the recovery and reuse of enzymes is possible without immobilization, and the stabilities of the lipases improve in solvent and non-aqueous systems. Therefore, lipases with organic-solvent tolerances have attracted much attention in regards to applying them as biocatalysts to biotransformation processes using solvent and non-aqueous systems.

A Study on the Solvent Fraction for Extraction of Bioactive Substances in Sea Cucumber (해삼의 생리활성물질 추출 용매분획에 따른 연구)

  • In this study, a study was attempted to confirm the yield of the solvent fraction for extracting physiologically active substances of polyphenols and flavonoids in sea cucumber. In the previously reported cases, the 50% ethanol extraction solvent fraction was confirmed as a result of high yield of sea cucumber antioxidants. The determination of the amount of antioxidants extracted from sea cucumber was confirmed as a result of the extraction solvent fraction applied to the extraction. In addition, the necessity of extraction of the solvent fraction was obtained through a previous study that 'there is a significant difference in the content of sea cucumber physiologically active antioxidants extracted depending on the extraction solvent.' The results of extracting high antioxidants from the phenolic substances contained in the sea cucumber extract of the 50% ethanol extraction solvent fraction were demonstrated. In the study case of extraction solvent fraction, the ethyl acetate solvent fraction, which yielded the lowest, yielded a higher phenol content than other solvent fractions, and the antioxidant effect was confirmed. Accordingly, it was confirmed that the effect on the high yield of the antioxidant extract through the yield change according to the application of the extraction solvent fraction was performed. Therefore, it was verified as an optimized sea cucumber physiologically active substance extraction solvent fraction of 50% ethanol solvent.

  • Solvolysis of tert-butyl halides and 1-adamantyl tosylate in methanol-acetone mixtures and solvolysis of tert-butyl bromide in methanol-chloroform mixtures have been studied. Solvent ionizing power of methanol-acetone mixtures were calculated by the Grunwald-Winstein equation and discussed the Y value variation caused by substrate changes. Y values based on 1-adamantyl tosylate is superion to others since it varies in wide range for methanol-acetone mixtures. It was found that the order of electrophilic assistance to leaving group is OTs > Cl > Br > I.

Solvation Number and Selective Solvation of Ions in Water-Methanol Systems (물-메탄올 混合溶媒中에서 이온들의 溶媒和數와 選擇的 溶媒和)

  • The limiting equivalent ionic conductances of Li$^+$, Na$^+$, K$^+$, Cl$^-$, and Br$^-$ ions were obtained with TATB[Tetraphenyl Arsonium Tetraphenyl Borate] method from limiting equivalent conductances of LiCl, NaCl, KCl and KBr measured in water-methanol systems by conductometric method. With those values and viscosity of water-methanol systems, the Stokes radii of corresponding ions have been extracted. The latter values corrected by calibration method of Nightingale and supplemented by the crystallographic radii were utilized to calculate the volume of solvation-shell that surrounds ions. The reasonable solvation numbers (h$_{H_2O}$ + h$_0$) were estimated by comparing the values obtained by the various suggested methods. The selective properties of ion solvation were also discussed.

  • Membrane formation mechanism by the phase inversion has been investigated in order to control the membrane morphology. It was found that finger-like structures were obtained when the heat of mixing between the solvent and non-solvent is exothermic, while sponge-like structures were obtained when they are endothermic in polystyrenes as well as in polyimides (Torlon and polyimide made frorn 3,3'4,4'-benzophenontetracarboxylic diahmydride and 1,4-phenylene diarnine) membranes. This concept was applied to control the morphology of polysulfone membranes simply by adding a solvent additive by adjusting the heat of mixing. A crude, but simple, relationship between the heat of mixing and the exchange rate of the solvent with non-solvent has been also developed.

  • In this work, the solvent permeation and separation performance of organic solvent nanofiltration (OSN) membranes were evaluated. Particularly, the PuraMem (PM) series developed for nonpolar solvents were analyzed and tested in dead-end filtration system. PM membranes exhibited higher permeance for nonpolar solvents compared to polar solvents, and their rejection data did not follow conventional trends with respect to solute size. The data showed that simple solution-diffusion model is not suitable to describe the OSN membrane behavior, and a better solvent-solute-membrane interaction parameter must be developed.

  • In order to further elucidate the process of electron solvation in liquids, the medium effect, as the difference between the free energy of $H^+$ in aqueous and non-aqueous states (${\Delta}{\Delta}G_0$), of THF-water mixtures has been investigated. (${\Delta}{\Delta}G_0$) were determined by electromotive force masurements of the cell Pt$H_2Q$, Q, HCI, THF, $H_2O$|KC1 | $Hg_2Cl_2$|Hg(Pt), where $H_2Q$ and Q are hydroquinone and quinone respectively. The effect of dielectric constant on the difference of free energy and the absorption energy of solvated electrons have been studied. For the consideration of these effect the polymerization of water in THF has been studied. Near infrared spectrum of O-H stetching energy has been used to measure the extent of water aggregates. The expermental results indicate that at least in some composition of binary mixtures the electrons or other ions are solvated preferentially with one component of solvents.
