Mode of Occurrences and Depositional Conditions of Arsenopyrite from the Yeonhwa 1 Mine, Korea (연화 제1광산에서의 유비철석의 산상과 배태 조건)

  • The chemical composition of the arsenopyrite Ib adjoining“triple mutual contact”arsenopyrite + pyrite + hexagonal pyrrhotite may serve as a useful geothermometer in Stage II. In this study it corresponds to temperature T=33$0^{\circ}C$ and f( $S_2$)=10$^{-9.5}$ atm. And the pyrite-hexagonal pyrrhotite buffer curve indicates the probable range of the two variables; T= 315∼345$^{\circ}C$, and f( $S_2$)=10$^{-1}$0.5/∼10$^{-9}$ atm. The present antimony-bearing arsenopyrite (arsenopyrite Ic) is characterized by relatively high content of antimony, ranging from 4.95 to 8.91 percent Sb by weight and excess of iron and deficiency of anions are evident. Such a high antimonian arsenopyrite has never been known within single grain. But being the high content of antimony as in the arsenopyrite Ic, it does not serve as a geothermometer. The results of microprobe analyses for four pairs of asenopyrite and sphalerite in Stage III indicate the temperature range from 310 to 34$0^{\circ}C$, and sulphur fugacity range from 10$^{-10}$ ∼10$^{-9}$ atm. These values seem to correspond with those inferred from the Fe-As-S system.m..

Pood Component Characteristics of Cuttle Bone as a Mineral Source (무기질 소재로서 갑오징어갑의 성분 특성)

  • This study was conducted to get a knowledge on food components of cuttle bone (CB) as a food resource. The yields and ash contents of CB were about $7.5\%$ on whole cuttle fish and about $90\%$ on dry basis, respectively, The contents of heavy metal might not invoke health risk in using food resource, The major mineral of CB was calcium as about $22\%$ in content. The yields, proximate compositions, heavy metal and mineral contents were not significantly different between domestic and imported CB. Judging from X-ray diffraction pattern, most of calcium in CB was present as a form of calcium carbonate (CC), and scanning electron micrograph showed irregular form. Buffering capacity of CB showed strongly at pH 7, and its pattern was the same as shown in that of CC, And CB solution showed a very high degree in turbidity comparing to that of CC solution. The solubility of CB was superior to that of CC, but inferior to those of calcium powders on the market, It was concluded from above results that CB could be effectively utilized as a calcium source.

Development and Application of the Assessment Method of No Net Loss of Greenness for Urban Ecosystem Health Improvement (도시생태계 건전성 증진을 위한 녹지총량 평가법 개발과 적용)

  • This study defined and classified no-net-loss-of-greenness (NNLG) based on the law, and then assessed the NNLG index by metropolitan cities and provinces in Korea after estimating NNLG evaluation indicators for the introduction of NNLG for health improvement of urban ecosystems. The results are as follows. First, NNLG was the comprehensive meaning that was included in the greenbelt and park greenbelt and the green area which was defined by the Act on Urban Parks, Greenbelts, etc. and the National Land Planning and Utilization Act respectively. Second, NNLG was classified as a park greenbelt which was included urban parks and greenbelts such as buffer greenbelts, scenic greenbelts, and connecting greenbelts, green areas which was included in green conservation areas, green production areas, green natural areas, and green coverage which is included forests, grasslands, and wetlands that were occupied by vegetation such as trees, shrubs, and plants. Third, NNLG index by cities and provinces was assessed based on the estimation of NNLG evaluation indicators, which included parks and greenbelt areas per capita, green areas per capita, green coverage per capita, ratio of parks and greenbelts, ratio of green areas, and ratio of green coverage. As a result, Sejong city got the highest point of NNLG index and Seoul and Daegu got lowest points of NNLG index among metropolitan cities in Korea. Chungbuk got the highest point of NNLG index and Kyonggi and Jeju got lowest points of NNLG index among provinces in Korea.

Electrochemical Degradation of Phenol by Using Reticulated Vitreous Carbon Immobilized Horseradish Peroxidase (Horseradish Peroxidase가 고정화된 다공성 탄소 전극을 이용한 페놀의 전기화학적 분해)

  • Horseradish peroxidase, had the phenol degradation rate of 95% in aqueous phase, was covalently immobilized on the surface of reticulated vitreous carbon(RVC) and the degradation of phenol was performed with in situ generated $H_2O_2$-immobilized HRP complex in an electrochemical reactor. The incorporation of carboxylic group on the RVC surface was confirmed by FT/IR spectrometry and 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide hydrochloride(EDC) was used for peptide bonds between the carboxylic groups on the RVC surface and amine groups from HRP. The optimal conditions of in situ $H_2O_2$ generation such as concentration($10{\sim}200$ mM) and pH($5.0{\sim}8.0$) of electrolyte, supply of $O_2(10{\sim}50$ mL/min) and applied voltage($-0.2{\sim}-0.8$ volt, vs. Ag/AgCl) from potentiostat/galvanostat were determined by concentration of hydrogen peroxide and current efficiency. It was observed that the RVC immobilized HRP was stable maintaining 89% of the initial activity during 4 weeks. The phenol degradation rate of 86% was attained under the optimal condition of in situ $H_2O_2$ generation.

  • A new method for immobilization of glucose oxidate by the aerobic gamma radiation of synthetic monomers was developed. The radiocopolymerization was conducted aerobically at -70 to -8$0^{\circ}C$ with the mixture of several polyfunctional esters, acrylates and native enzyme. The retained activity of immobilized glucose oxidase was about 50 to 55% when a NK 23G ester, acrylamide-bis and water mixture (1:1:2) in cold toluene treated with 450 krad of gam-ma radiation. The radiation dose did not influence significantly to the enzyme activity. The solvents used to prepare the beads of glucose oxidase and monomers were toluene, n-hexane, petoleum ether and chloroform. 0.05M tris-gycerol (pH 7.0) was a more suitable bugger solution for immobilizing the enzyme than was 0.02M phosphate. Immobilization of glucose oxidase shifted the optimum pH for its reaction from 6.0 to 6.5. The pH profile for the immobilized enzyme showed a broad range of optimum activity while the native enzyme gave a sharp pick for its optimum pH value. The immobilized enzyme reaction temperature was at the range of 30~4$0^{\circ}C$.

Microfluidic System for the Measurement of Cupric Ion Concentration using Bilayer Lipid Membrane on Silver Surface (은 표면의 이중층 지질막에 의한 구리 이온 농도 측정용 마이크로플루이딕 시스템)

  • A microfluidic system has been developed using biomaterial for the measurement of cupric ion concentration. The cell-membrane-mimicking bilayer lipid membrane(BLM)-coated silver electrode was used for the sensing of cupric ion concentration. The silver-supported BLM could increase its stability. A silver-supported bilayer lipid membrane(s-BLM) was easily obtained using its self-assembling characteristics by immersing silver wire into lipid(phosphatidylcholine; PC) solution and then dipping into aqueous KCl solution. These s-BLMs were used to determine the relationship between $Cu^{2+}$ concentration and current crossing s-BLM. Their relationship showed high linearity and reproducibility. The calibration curve was constructed to express the relationship between $Cu^{2+}$ concentration and current in the $Cu^{2+}$ concentration range of 10 and $130{\mu}M$. This calibration curve was used to measure $Cu^{2+}$ concentration in an unknown sample. Microfluidic system with s-BLM was made of PDMS(polydimethyl siloxane) using typical soft photolithography and molding technique. This integrated system has various functions such as activation of the silver surface without cutting silver wire, coating of BLM on silver surface, injection of KCl buffer solution, injection of $Cu^{2+}$ sample and measurement of $Cu^{2+}$ concentration in the sample.

  • This study was carried out to enhance the stability and activity of polygalacturonase (PGase) purified from Kluyveromyces marxianus IFO 0288. Gums such as xanthan gum, guar gum, and locust bean gum were capable of increasing the catalytic stability and activity of the PGase. At $30^{\circ}C$, the rate constants for the inactivation of the PGase with xanthan gum, guar gum, and locust bean gum were estimated to be $0.0003min^{-1}$, below $0.0001min^{-1},\;and\;0.0001min^{-1}$ respectively, whereas control was estimated to be $0.0082min^{-1}$. The yield of the maceration reaction catalyzed by the PGase for the production of carrot single cells increased by 13% in the presence of guar gum, where the relative enzyme activity supplemented with guar gum was two-fold greater than that of the PGase alone.

Lytic Action of Egg White Lysozyme Isolated from Ogol Fowl on Staphylococcus aureus Phage Type 29 (Staphylococcus aureus Phage Type 29에 대한 오골계 난백 Lysozyme의 용균성)

  • This experiment was carried out to investigate the bacteriolytic action of the egg white lysozyme isolated from Korean native Ogol fowl and to obtain the data for utilization of the enzyme as a food preservative. Staphyococcus aureus phage type 29 and Bacillus subtilis ATCC 6633 among the microorganisms tested were lyzed by the treatment with 0.05% lysozyme, but Staphylococcus aureus phage type 57 in addition to E. coli etc. was found to be a lysozyme- insensitive species. The lysis of S. aureus phage type 29 was maximized when incubated in nutrient broth (pH 7.0) at $37^{\circ}C$ for 24 hours and suspended it to absorbance 0.6 at 540nm in 0.05M sodium acetate but fer (pH 4.5) and then treated it with the 0.05% lysozyme for 30 min. at $30^{\circ}C$. It was found that the effect of 0.05% lysozyme in combination with 1% glycine on the growth inhibition of S. aurecus phage type 29 increased more 50% than that in the absence of glycine, but not effect with other any additeves and metal ions tested.

  • Nitrifier consortium entrapped in Ca and Ba-alginate beads were packed into two reactors and studied for removing ammonia-nitrogen in aquaculture system. The ammonia-nitrogen concentration of the influent was continually kept about 2 ppm. At the hydraulic residence time of 0.6 hours, ammonia-nitrogen removal amount of two reactors was about 52.6 and 51.0 g $NH_3-N/m^3/day$, respectively. The ability of adjusting to an impulsive leading which was happened according to variations of HRT was better at Ba-alginate reactor, but its discrepancy was not so large. At the respect of removing ammonium-nitrogen, two reactors showed the similar ability of treating recirculating water.

Composting of Livestock Manure Blending Humic Acid Powder and Influences on Growth of Lettuce by Its Application (부식산분말 처리에 따른 가축분의 퇴비화 특성 및 시비효과)

  • Humic acid was used soil amendment or functional fertilizer in Korean agriculture, and its cation exchangeable capacity was high enough to increase soil buffering from plant toxicant. This study was conducted to evaluate effects of humic acid powder (HA) on composting of livestock manure (LM) and of its application on growth of lettuce. Treatments were designed as follows; livestock manure compost (LM+sawdust+bark+castor meal; Control), control+0.1% HA (0.1% HA), control+0.5% HA (0.5% HA), control+1.0% HA (1.0% HA), control+3.0% HA (3.0% HA), and control+5.0% HA (5.0% HA). The changes of temperature, water content, organic matter content, total nitrogen and ratio of organic matter and nitrogen in HA treatments were similar to those of control. Although pH of 3.0% HA and 5.0% HA blending with HA and LM was lower than those of others, it unaffected by HA blending during composting. Humic acid content of HA treatments was increased by 1.7~4.4 folds than that of control. As compared with odor index, 3.0% HA and 5% HA were decreased than control for composting time. Application of 3.0% HA increased the dry weight of lettuce by about 7% than that of control.
