In recent years, significant importance has been given to chitooligosaccharides (COS) due to its potent notable biological applications. COS can be derived from chitosan which is commonly produced by partially hydrolyzed products from crustacean shells. In order to produce COS, there are several approaches including chemical and enzymatic methods which are the two most common choices. In this regard, several new methods were intended to be promoted which use the enzymatic hydrolysis with a lower cost and desired properties. Hence, the dual reactor system has gained more attention than other newly developed technologies. Enzymatic hydrolysis derived COS possesses important biological activities such as anticancer, antioxidant, anti-hypersentive, anti-dementia (Altzheimer's disease), anti-diabeties, anti-allergy, anti-inflammatory, etc. Results strongly suggest that properties of COS can be potential materials for nutraceutical, pharmaceutical, and cosmeceutical product development.
This study was conducted to produce oligosaccharides from buckwheat hull by using commercial enzymes. Yields of oligosaccharides obtained by enzymatic hydrolysis of the cellulose and hemicellulose fractions were 132.37 and 393.04 g/kg, respectively. Xylose, glucose, fructose, xylobiose, xylotriose, cellobiose, and cellotriose were detected in the hydrolysate produced from buckwheat hull. Antioxidant activity of oligosaccharide from cellulose fraction (OSC) reduced with increasing hydrolysis time; however, the antioxidant activity of oligosaccharide from hemicellulose fraction (OSF) increased as the hydrolysis time was prolonged. OSF and OSC showed higher increase in viable counts compared to the control. As a result, oligosaccharides produced from buckwheat hull by enzymatic hydrolysis showed antioxidant activity and prebiotic effects. It is suggested that utilization of oligosaccharides produced from buckwheat hull as functional food materials may be improved when hydrolysis time and conditions are controlled for this purpose.
Quality characteristics of strawberry jams made of 2 different strawberry cultivars, Bogyo-joseng and Suhong, with 25, 50, 75, 100% fructo-oligosaccharide in place of sucrose were evaluated. Moisture content of Suhong jam was higher than Bogyo-joseng jam. pH of Bogyo-joseng and Suhong jam was not different. The lightness, redness and yellow-ness of Bogyo-joseoung jam were higher than those of Suhong jam. Spreadmeter value of jams decreased with addition of fructo-oligosaccharide. The sweetness of Suhong jam was higher than Bogyo-joseng. Reducing sugar content of Suhong jam was higher than Bogyo-joseng jam. More of fructo-oligosaccharide added, reducing sugar content of jam increased. Residual anthocyanin in Bogyo-joseng strawberry jam was decreased by addition of fructo-oligosaccharide. However, the residual anthocyanin content of Suhong strawberry jams with fructo-oligosaccharide was lower than that of sugar-only control jam with an exception of 50% fructo-oli-gosaccharide jam. The 50% fructo-oligosaccharide jam had a higher residual anthocyanin content than sugar-only jam. As the addition rate of fructo-oligosaccharide increased, springiness and cohesiveness of jam increased, while resilience decreased. In sensory evaluation, Bogyo-joseng strawberry jams obtained highest score with 50% fructo-oligosaccharide, while Suhong strawberry jam was favored most with 25% fructo-oligosaccharide.
Chitosan is the deacetylated product of chitin. Chitosan and its derivatives have many properties that make them attractive for a wide variety of health applications. This study was conducted to investigate change of oligosaccharide content and antiviral effect on rotavirus of chitooligosaccharide and water soluble chitosan after heat treatment. The quantitative analysis of oligosaccharide using colorimetry showed that oligosaccharide contents in water soluble chitosan and chitooligosaccharide were decreased from 62.67% to 60.45% and from 59.48% to 54.31%, respectively, after heating. The inhibitory effect of chitosan derivatives on MA-104 cell infected with human rotavirus(HRV) measured using AEC staining method. The inhibition level of 0.125% water-soluble chitosan against cell infection by human rotavirus was 91.98 3.09% in HRV S2 and was 89.92 1.68% in HRV Wa. But, chitooligosaccharide had not shown inhibitory effect against cell infection by HRV. It considered that most oligosaccharide of chitooligosaccharides consist of oligomer of lower polymerization degree. Heat treatment of water soluble chitosan and chitooligosaccharide did not influence their antiviral effects on rotavirus.
Lactobacillus bulgaricus and Kluyveromyces lactis were inoculated to Jangyeob and Jinpum soy milks together after the addition of different amounts of lactose to increase the contents of oligosaccharides, which were compared with single cultured samples. The contents of stachyose, raffinose, sucrose, and glucose of samples without lactose decreased by single culture method, but the oligosaccharides decreased less than in single cultured samples containing of lactose. The oligosaccharides of single cultured samples were equal or decreased compared with soy milks. While those of mixed cultured Jangyeob and Jinpum samples containing 2% lactose for 24 hr incubation increased 125.0% and 118.1%, respectively and those of samples for 36 hr incubation increased 127.0% and 141.0%, respectively, those of mixed cultured samples containing 4% lactose for 24 hr incubation increased 112.5% and 123.0%, respectively and those of samples for 36 hr incubation increased 120% and 135.9%, respectively. Therefore, the oligosaccharides in samples containing 2% lactose were slightly more than in samples containing 4% lactose. Among the cultured methods, oligosaccharides were produced in the largest amounts by the mixed culture for 36 hr. The addition of lactose in soy milks for soy yogurts was effective in the formation of oligosaccharides since the galactose, produced by the hydrolysis of lactose, was thought to be combined with sucrose by the action of ${\beta}-galactosidase$ in yeast.
The quality characteristics of Korean rice cakes (Karedduk) with a mixture of fructooligosaccharide (95%) and AA (acetylated adipate distarch) added, after 2 and 24 hours of storage at $5^{\circ}C$, were investigated. A central composite design was used for arrangement of treatment. Different levels of fructooligosaccharide (95%), 0, 3, 6, 9, and 12%, were added to dry rice flour. In addition, different levels of AA, 0, 0.3, 0.6, 0.9, and 1.2%, were added to the same dry rice flour. The texture properties analysis using a Texture Analyzer revealed that the springiness, cohesiveness, chewiness, gumminess, adhesiveness, and hardness were significantly different. The effect of retarding retrogradation of Korean rice cakes (Karedduk) with added mixtures of fructooligosaccharide (95%) and AA showed an increasing trend as the amount of fructooligosaccharide (95%) and AA increased. Overall, the instrumental texture properties were highly correlated with the sensory characteristics. These results suggest that adding a mixture of 6% fructooligosaccharide (95%) and 1.2% AA to Korean rice cakes (Karedduk) is effective for retarding retrogradation.
Effects of the addition of three commercial oligosaccharide syrups into a Korean rice cake (Karedduk) on the textural characteristics and retrogradation of the rice cake were examined during the storage for 5 days at $25^{\circ}C$ and $4^{\circ}C$. Each syrups contained maltose (M75), isomaltose and panose (HL), or maltotetraose (G4) as major sugars. The increment (rates) in gumminess, hardness and chewiness during the storage were significantly reduced by replacing rice flour (up to 10%) with the oligosaccharides. The retardation in the textural changes by the oilgosaccharides was more significant when the rice cake was stored at $25^{\circ}C$ than at $4^{\circ}C$. Among the three types, HL exhibited most effective in retarding the textural changes. Thermograms by a differential scanning calorimeter (DSC) showed that the oligosaccharide increased the onset temperatures and enthalpy for the starch melting, but the recrystallinity measured from the enthalpy ratio before and after the storage was significantly reduced by the presence of the oligosaccharide. Especially with 5% HL, the recrystallinity was significantly low (72.7%) compared to rice cake without HL (88.1%). Therefore, HL had great efficiency in retarding starch retrogradation as well as textural changes of the rice cake during the storage.

The study was conducted to the effect of inclusion of chitosan-oligosaccharide in drinking water on the blood component profile, immunity and antioxidative enzyme of broiler chickens. A total of 28,000 broiler chickens (Arbor Acre) at 1 days of age were fed the commercial diet until 35 days of age, the treatment divided into two treatments, normal control group and chitosan-oligosaccharide in drinking water group. In concentration of glucose, treatment was significantly decreased than control (P<0.05), but not statistically different on the Triglyceride, Total cholesterol, HDL-cholesterol and LDL-cholesterol in the Blood component profile of broiler chicks. The concentration of glutamic oxalacetic transaminase (GOT) was statistically different but the concentration of glutamic pyruvate transaminase (GPT) was significantly decreased in treatment fed chitosan-oligosaccharide than control (P<0.05). Immunoglobullins in the blood, concentration of IgG was not significantly different among control and treatment but concentration of IgM was significantly increased in fed chitosan-oligosaccharide than control (P<0.05). Antioxidant and super oxide dismutase (SOD) was not different among control and treatment and concentration of catalase and glutathione peroxidase were significantly increased in fed chitosan-oligosaccharide than control (P<0.05).
Agar, one of the most abundant marine products has not been utilized extensively because of low level of processing technology in Korea. This research was carried out to improve the utilization of agar and consequent increase in profit. Antibacterial activity of agarooligosaccharides were evaluated against bacteria causing putrefaction and flood poisoning. Addition of 0.4% agarooligosaccharides showed antibacterial activity toward Staphylococcus aureus and Escherichia coli O157:H7; furthermore, autoclave treatment of agarooligosaccharides solution enhanced the antibacterial activity. Agarooligosaccharides showed high stability against the pH change. Addition of amino acid(alanine, lysine, glycine, phenylalanine) in agarooligosaccharides solution enhanced antibacterial activity in E. coli O157:H7, Streptococcus mutans and Staphylococcus aureus.
