Evaluation of Susceptibility of Spoladea recurvalis (Lepidoptera: Crambidae) to Five Insecticides in Spinach (시금치 해충 흰띠명나방(Spoladea recurvalis)의 살충제 감수성)

  • The susceptibilities of five insecticides to the larvae of Spoladea recurvalis, a major pest of spinach, were evaluated. Lufenuron EC, chromafenozide EC, chlorantraniliprole WP, tebufenozide WP, and pyridalyl EW showed 90% or more insecticidal activity at dilution concentrations of 2 (12.5 ppm), 4 (12.5 ppm), 8 (2.5 ppm), 4 (20.0 ppm), and 8 (12.5 ppm), respectively. At their recommended concentrations, Chromafenozide EC, chlorantraniliprole WP, tebufenozide WP, and pyridalyl EW showed very high insecticidal activity of 98.3%, 100%, 95.0%, and 100%, respectively, after 72 h on S. recurvalis larvae exposed after 7 days of treatment. Field tests at two spinach greenhouses in Hwaseong and Yeoncheon showed a high control effect of over 90% after 7 days of treatment in both areas. In addition, the insecticides can be used as exclusive agents for S. recurvalis that occurs in spinach, as there is no phytotoxicity even at double doses.

Relationship of Somatic Cell Count, Physical, Chemical and Enzymatic Properties to the Bacterial Standard Plate Count in Different Breeds of Dairy Goats

  • The objective of the present study was to investigate the accuracy of mastitis diagnostic indicators for different dairy goat breeds. Biweekly milk samples were collected from individual half mammary gland of seven Saanen and seven Alpine dairy goats in the period of 40 to 120 days in milk. With threshold value set at 2.8 and 3.1 for Alpine and Saanen dairy goats, respectively, log (SPC) offered good sensitivity (0.89, 0.93), specificity (0.88, 0.95), positive predictive value (0.75, 0.85) and negative predictive value (0.95, 0.98) as a mastitis diagnostic tool. The correlations of log (SPC) with milk yield, log (SCC), ALP, LDH, $Na^{+}$, $K^{+}$ and EC were significant in Saanen dairy goats (p<0.05), with the highest correlation coefficient (0.653) existing between log (SPC) and log (SCC). The correlations of log (SPC) with milk yield, milk fat, milk protein, log (SCC), $Na^{+}$, $K^{+}$, EC were significant in Alpine dairy goats (p<0.05), with the highest correlation coefficient (0.416) existing between log (SPC) and log (SCC). There were different best-fit regression equations with different multiple diagnostic indicators for Saanen and Alpine dairy goats. In conclusion, different breeds of dairy goats may have to adapt different mastitis diagnostic parameters for a better diagnosis.

Flavobacterium jocheonensis sp. nov., Isolated from Marine Green Alga Ulva pertusa

  • A bacterial strain, labeled $UR11^T$, was isolated from green alga Ulva pertusa collected from Jeju Island, Korea. $UR11^T$ was identified as a gram-negative, rod-shaped, motile by gliding and aerobic bacterial strain with yellow colonies on R2A plates. The strain $UR11^T$ grew over at a temperature range of $10^{\circ}C$ to $30^{\circ}C$ (optimally at $25^{\circ}C$), a pH range of 6.0-11 (optimally at pH 7.0) and a Nacl range of 0.5-5% Nacl (w/v). Phylogenetic analysis based on 16S rRNA gene sequences revealed that strain $UR11^T$ was a member of the genus Flavobacterium. Strain $UR11^T$ shared close similarity with F. jejuensis $EC11^T$ (98.0%) F. jumunjinense $HME7102^T$ (96.1%), F. haoranii $LQY-7^T$ (95.3%), F. dongtanense $LW30^T$ (95.1%), and F. ahnfeltiae 10Alg $130^T$(94.9%). The major fatty acids (>5%) were $iso-C_{15:0}$ (33.9%), $iso-C_{15:1}$ G (12.4%), $iso-C_{17:0}$ 3-OH (9.0%), $isoC_{16:0}$ (7.0%) and $iso-C_{15:0}$ 3-OH (6.3%). The major polar lipids were phosphatidylethanolamine, seven unknown aminolipids, two unknown aminopolarlipids and two unknown lipids. DNA-DNA hybridization value was 58% at strain $UR11^T$ with F. jejuensis $EC11^T$. Based on phenotypic, chemotaxonomic and phylogenetic evidence, strain $UR11^T$ represents a novel species of the genus Flavobacterium, for which the name Flavobacterium jocheonensis sp. nov. is proposed. The type strain is Flavobacterium jocheonensis is $UR11^T$ (=KCTC $52377^T$ =JCM $31512^T$).

Study on Decline of Trees by Acid Rainfall (산성 강우에 의한 수목의 쇠퇴현상(I) - 음이온을 중심으로 -)

  • This study was carried out to analyze pH, Electrical conductivity(EC), cation and anion on rainfall and throughfall raining at industrial area(Onsan and Nongso of Ulsan city), suburban area(Myeongsek-myeon of Jinju city) and rural area(Haman and Changyoung-gun), and to investigate $SO_2\;and\;NO_2$ concentration in the air of industrial area and rural area, and to survey degree of decline of Pinus thunbergii growing at each area. The rainfall pH was higher than the throughfall pH at the all surveyed areas. The rainfall pH and throughfall pH was $4.76{\sim}5.15\;and\;4.64{\sim}4.98,\;4.95{\sim}5.30\;and\;4.87{\sim}5.13,\;5.29{\sim}5.76\;and\;5.12{\sim}5.73$ at the industrial area, suburban area and rural area, respectively. The EC of rainfall and throughfall was $13.64{\sim}17.22\;and\;41.60{\sim}103.59,\;4.18{\sim}8.65\;and\;17.02{\sim}23.89,\;4.09{\sim}6.65\;and\;13.75{\sim}21.44$ at the industrial area, suburban area and rural area, respectively. Generally EC at the industrial area was higher than those of suburban area and rural area. In the correlation analysis among Degree of decline on Pinus thunbergii and ingredients of rainfall, it showed that there was a negative correlation between Degree of decline and pH. There was a positive correlation between pH and $Cl^-$ respectively.

Recovery of Cyclodextrin Glucanotransferase by Adsorption to Starch (전분흡착에 의한 Cyclodextrin Glucanotransferase의 회수)

  • Cyclodextrin glucanotransferase (EC 2.4.1.19 : 1,4-$alpha$-glucan 4-$alpha$-D-(1,4-glucano) transferase, cyclizing; CGTase) was recovered by starch adsorption. The adsorption and desorption of CGTase to starch was studied as a function of pH, temperature, and starch type. The optimal pH, temperature, and starch for adsorption were, 8.0, $4^{circ}C$, and 1% (w/v) corn starch, respectively, per 205 U/mL enzyme activity in the presence of 25% (w/v) ammonium sulfate. The maximum adsorption ratio was 95%. On the other hand, the optimal pH, temperature, and starch type for desorption were 8.0 (tris-buffer), $50^{circ}C$, and oxidized starch, respectively. The maximum desorption ratio was 98% by tris-buffer solution at pH 8.0. The efficiency of adsorption and desorption were affected slightly by the removal of cells from the fermentation broth.

Development of Method for Possibility Assessment on Organic Resources for Using Raw Material of Compost (유기성자원의 퇴비원료로 활용 가능성 평가방법 개발)

  • This study was conducted to find a system for screening organic resources with 16 species, 62 samples which were selected to randomizing point from city, province and industrial areas in the whole country. Content of organic matters were $65.3%{\sim}98.0%$ in all samples so that they were largely over than 60%, raw material regulation of compost. Concentrations of total nitrogen and total phosphorus were $0.7{\sim}4.8%\;and\;0.8{\sim}5.0$, they could look for effect of the nitrogen and phosphorus supply as a raw material of compost. In case of 8 elements concentrations of heavy metal, they were too high to use as raw materials of compost which were over to regulation limit in Cu, Cr, Ni, and As from fiber industry, Ni from food company and leather industry, and the others are adapt to limit levels. HEM contents fro the highest to $113mg\;kg^{-1}$ from liber industry and PAHs content were the highest to $3,462ug\;kg^{-1}$ from paper-mill manufacture. Distribution of PAHs concentiations were naphthalene>phenanthrene>pyrene>fluoroanthene>acenaphthene. $Microtox{(R)}\;EC_{50}$ values for bioassay were pharmaceutical company>paper-mill manufacture>industrial area sewage sludge>fiber industry>urban sewage sludge>metropolitan sewage sludge. HEM between Zn, Cu, and Ni was significant at the 99% and between Cd was significant at the 95%, Microtox between Hg and BEM significant at the 95%.

Antioxidant Activity and Cytotoxicity Effect of Extracts from Taraxacum mongolicum H. (민들레 추출물의 항산화 활성 및 세포독성 효과)

  • This study was investigated antioxidant and anticancer activity of water, methanol extract from upper and root part of Taraxacum mongolicum H. Total phenolic compound contents of methanol and water extracts from upper part were $51.95{\pm}0.18$ mg/g and $48.16{\pm}0.89$ mg/g respectively, and total flavonoid compound contents were estimated as $20.57{\pm}1.12$ mg/g in methanol extract and $6.55{\pm}1.20$ mg/g in water extract. $EC_{50}$ values for DPPH radical scavenging activity of methanol and water extract from upper part were $138.47{\pm}3.78{\mu}g/mL$ and $204.38{\pm}5.32{\mu}g/mL$, and methanol and water extracts from root part were as $512{\pm}8.11{\mu}g/mL$ in methanol extract and $1315.05{\pm}11.98{\mu}g/mL$ in water extract. Reducing power and hydroxyl radical $({\bullet}OH)$ scavenging activity estimated that methanol extract of each part were higher than water extracts. The cell viability showed that the methanol extract from upper part had a cytotoxicity in the growth of colon carcinoma cell (44.58%). Both water extract $(51.97{\pm}11.43%)$ from upper part and methanol $(53.46{\pm}19.77%)$, water $(52.79{\pm}13.53%)$ extracts from root part had quite higher cytotoxicity than that of methanol extract $(88.25{\pm}2.02%)$ from upper part. Based on the results, It was suggested that the methanol extract of Taraxacum mongolicum H. were potential materials for use as functional food and medicine.

An Analysis of Growth Conditions of old Trees in Yangdong Villages (양동마을의 노거수 생육실태 분석)

  • The purpose of this study is to provide the basic data by analyzing and analysing the Growth Conditions of old Trees in Yangdong village. This study investigated about the conducted on tree information, soil information, and tree health. The result are as follows. The trees information in Yangdong Village consists of Juniperus chinensis, Salix chaenomeloides, Salix pseudolasiogyne, Celtis sinensis, Zelkova serrata, Gleditsia japonica, and Gleditsia sinensis trees, The range of height was 4.0~17.0m, and the diameter was 0.51~1.34m, Juniperus chinensis trees of No.17 was most large. In the results of soil analysis, there showed that acidity was pH4.1~6.3, hardness of 5~48mm, organic matter content of 21.2~29.1g/kg, electrical conductivity(EC) of 0.34~1.76dS/m, available P2O5 of 79.8~451.6mg/kg, exchangeable K of 0.22~1.71cmol+/kg, exchangeable Ca of 4.98~7.44cmol+/kg, exchangeable Mg of 0.67~2.19cmol+/kg, exchangeable Na of 0.19~1.04cmol+/kg and cation exchange capacity(C.E.C) of 7.23~13.02cmol+/kg. As a result, the highest number of tree health levels is 8 of 11trees of Celtis sinensis, 2 of 7trees of Zelkova serrata, and 3 of Gleditsia sinensis, and 13 of 30 trees of health levels, The Older trees with high infection, spoil and hollowed part were the remaining trees except for the healthy part. Relatively, more than half of the number of targets is infected, decay, and the hollowed site, and it is necessary to perform surgery on the damaged area. In addition, preservation and protection measures should be implemented by supplying root nutrients for trees, controlling nutrients in the body to prevent secondary and tertiary damages that cause the infection site to metastasize to the health site, In order to continually monitor the trees, measures to improve the location environment and management of the trees should be sought.
