The aim of this study is to compare the anti-inflammatory and immunological activity of different parts (bone, meat, and rind) of Yeonsan Ogye (YO). In order to evaluate cytotoxicity, MTT assay was performed. We investigated the production of nitric oxide (NO) and pro-inflammatory cytokines, such as IL-$1{\beta}$, IL-6, and TNF-${\alpha}$, in LPS-induced RAW264.7 cells. All parts of the YO showed no toxicity at concentrations of 1, 10, and $100{\mu}g/m{\ell}$. Rooster's bone, hen's bone, and rind decreased the production of NO. And rooster's bone, meat, and hen's bone also attenuated TNF-${\alpha}$ production in LPS-induced RAW 264.7 cells. In addition, all parts of the YO decreased IL-$1{\beta}$ and IL-6 production in LPS-induced RAW264.7 cells, whereas they all increased IL-$1{\beta}$, IL-6 and TNF-${\alpha}$ production in normal RAW264.7 cells. Rooster exhibited higher immune activation and inhibitory activity on inflammation than a hen, and among different parts of the YO, bone showed the highest activity. Our results demonstrated and compared the anti-inflammatory and immunological activity of different parts of the YO. These results suggest that YO may be developed as a raw material for new health supplement food and medicine to attenuate various symptoms related to inflammation and immunity.
Objective : The aim of this study is to investigate anti-arthritis activity using a korea's natural monument No. 265 designate 'Yeonsan-Ogye'. In this study, research by using extracts from different concentration of the Yeonsan-Ogye through an MIA-induced arthritis animal model was being conducted in vivo and scientifically verifying the efficacy of medicinal food. Methods : Yeonsan-Ogye was administered 500 mg/kg/day, 1000 mg/kg/day, 2000 mg/kg/day to SD-Rat, where arthritis was induced by monosodium iodoacetate (MIA) at $60mg/m{\ell}$. MMP-9, COMP, CTXII, calcitonin and glycosaminoglycan level in serum were measured by ELISA. The changes of relative hind paw weight bearing ratio by Incapacitance Test Meter and The cartilage of meniscus volume was examined and 3-D high-resolution reconstructions of the cartilage of meniscus were obtained using a Micro-CT system. Also, the histopathological analysis of knee was observed by H&E and safranin-O staining. Results : Production of MMP-9, COMP (all groups) and CTXII (500, 1000 mg/kg/day) level in serum was decreased, respectively, in comparison with control. The other way, production of calcitonin (500, 1000 mg/kg/day) and glycosaminoglycan (all groups) level in serum, Hind paw weight bearing ratio (all groups) was increased, espectively, in comparison with control. The cartilage of patella volume in micro-CT increased significantly. In addition, all groups showed a increase in the cartilage volume and proteoglycan. Conclusion : The results for Yeonsan-Ogye showed significant antiarthritis activity in serum and the cartilage. Therefore, it is thought to be that Yeonsan Ogye can be utilized as a variety of new korea medicie and health foods against arthritis diseases.

Ogye rooster sperm chromatin status can be detected using well established sperm assays. In this paper, a simple and fast method to monitor rooster sperm chromatin status could be employed in field for assessment of chicken sperm quality. Using standard bright field microscope, Diff-Quik stains can be reproducibly, easily and routinely monitored with simple staining. The presence of abnormal chromatin staining of rooster sperm was determined by darker stain in head. In the fresh semen, the viabilities of three tested Ogye spermatozoa were 93.53%, 82.42% and 90.63% and normal chromatin rates were 87.96%, 74.25% and 85.10% respectively. However, after freezing, the rates of viability of thawed semen were reduced to 69.58%, 61.98% and 72.20% and normal chromatin rate also reduced to 58.91%, 48.49% and 63.34%. A significant correlation between live sperm and normal sperm nuclei was 0.875 in fresh semen and 0.513 in frozen semen. After incubation of sperm at $37^{\circ}C$ for 5min, the rates of viability, chromatin normality and sperm head activity were shown as $90.63{\pm}1.28%$, $82.44{\pm}8.09%$ and $66.68{\pm}10.29%$ in fresh semen. However, the rates of thawed semen were reduced to $67.92{\pm}7.55%$, $56.92{\pm}12.15%$ and 47.32{\pm}5.02%, respectively. The relationship between chromatin normality and sperm head movements in fresh and thawed semen were 0.564 and 0.540, respectively. With these results, the chicken sperm normality could be assessed by the Diff-Quik staining that could be used for chromatin status of sperm head and activated morphology of live spermatozoa, as a simple and rapid staining method.
This study was conducted to establish the method for preserving PGCs that enables long-term storage in liquid nitrogen for developmental engineering or preservation of species. The purpose of this study is to clarify the effects of freeze-thaw treatment on viability of PGCs in chickens. PGCs were collected separately from a germinal gonad of an early embryo of 5.5~6 day (stage 28) of Isa brown, Korean Oge (KO), White Leghorn and Commercial breeds. PGCs separated from a germinal gonad of an early embryo of 5.5~6 day (stage 28) are suspended in a freezing medium containing a freezing and protecting agents (e.g. dimethyl sulfoxide (DMSO), ethylene glycol (EG) and glycerol). The PGCs were then purified using magnetic activated cell sorting (MACS) method. The viability of PGCs after thawing was $87.4{\pm}0.4%$ and $89.4{\pm}0.2%$ with the 10% EG treatments with no significant difference between the Isa brown and Commercial breeds. The viability of PGCs after freeze- thawing was significantly higher for Isa brown ($87.4{\pm}0.4%$) and Commercial breeds ($89.4{\pm}0.2%$) than Korean Oge (KO) ($77.6{\pm}1.1%$) and White Leghorn ($76.2{\pm}0.9%$)(p<0.05) using 10% EG cryoprotectant. This study established a method for pre- serving chicken PGCs that enables systematic storage and labeling of cryopreserved PGCs in liquid ($LN_2$) at agermplasm repository and ease of entry into a data base. In the future, the importance for this new technology is that poultry lines can be conserved while work is being conducted on improving the production of germline chimeras.

In this study, viability, growth performance and egg production performance were investigated to determine the productive characteristics of 12 Korean domestic chicken varieties which have been collected and conserved for over 25 years in National Institute of Animal Science, Rural Development Administration, Korea. The 1,134 hens were tested and their production traits including survival rate, body weight, age at first egg laying, hen-day and hen-housed egg production, egg weight, and egg quality were measured. Survival rate was the highest in Korean Rhode-D and Korean Native Chicken (KNC) White and the lowest in Korean Cornish Brown (92.2% and 54.3%, respectively). Body weights from 4 to 50 wks were consistently high in the order of Korean Cornish, Korean Rhode, KNC, Korean Ogye, and Korean Leghorn breeds. There was no significant difference in weight between varieties within a breed. Age at first egg laying was the shortest in Korean Leghorn, while it was the longest in Korean Cornish. The hen-day egg production from $1^{st}$ egg to 57 wks was the highest in Korean Leghorn-K, and the lowest in Korean Cornish Brown. Egg weight was the heaviest in Korean Leghorn-F and the lightest in KNC White. The Haugh unit was the highest in Korean Rhode-C and the lowest in Korean Ogye. Taken together, these results suggest that it is desirable that the Korean Cornish variety is improved as the Korean meat breed because of its excellent growth ability, the Korean Leghorn variety is improved as the Korean layer breed because of its good laying performance, and the Korean Rhode and KNC varieties are improved as strong viable breeds because of their good survival rate.
Biological materials, such as stuffed specimens, can release various acids or volatiles. There has been no research carried out on the emission characteristics of organic compounds generated from the preservatives used in taxidermy specimens or associated manufacturing materials and methods. Therefore, in order to identify the organic compounds generated from taxidermy specimens, a degradation experiment was conducted on specimens for each material and for storage specimens. To produce Ogye chicken specimens, naphthalene and borax were used as preservatives, and planer sawdust, newspaper, and polystyrene foam were used as the core body materials. The deterioration experiment was conducted for 2 weeks in a high-temperature environment(50℃) and a high-humidity environment (95%), with an Ogye chicken specimen (year 2015) kept in an animal storage facility. Results indicated that the concentration of organic compounds generated by the specimen in the high-temperature environment tended to be greater than that in the high-humidity environment. The preservatives benzene, toluene, xylene, and p-dichlorobenzene were detected in the specimens using naphthalene, confirming that naphthalene is a major organic compound release factor, and the specimens that used sawdust, newspaper, and polystyrene foam also exhibited organic compounds. This appears to have been due to degradation of the material. In addition, ammonia was detected in the specimens for each material due to decay. In particular, the specimens using borax at high temperature were subject to approximately 9 times higher rates of ammonia-related deterioration than the specimens using naphthalene. These results can be considered to result from the prevention of biological damage through insecticidal effects by accelerating the sublimation of naphthalene in a high-temperature environment. Naphthalene is a potentially carcinogenic substance, and when used as a preservative, proper use management is required. Taxidermy specimens can release various organic compounds depending on the manufacturing techniques used, so a systematic preservation management plan is required that depends on conditions such as the applicable manufacturing materials and preservatives.

This study was conducted to establish the BMPs (Best Management Practices) for preventing pollutant loadings from paddy rice field applied livestock liquid manure from 2008 through 2011. Cultivated paddy rice fields (Gyeonggi province, Korea) were treated with SCB (Slurry composting and bio-filtration process) liquid fertilizer. The BMPs for paddy rice field developed in this study includes: 1) the controlling a drainage water gate in paddy rice field from right after SCB liquid fertilizer application to 3 weeks after rice transplanting; 2) livestock liquid fertilizer application to paddy rice soils in 20 days before rice transplanting to encourage the utilization of liquid fertilizer; 3) preservation of surface water depth to 5 cm in a paddy field right after SCB liquid fertilizer applied to minimize a water pollution and enhance the utilization of liquid fertilizer; and 4) blocking a water gate at least for 2 days to inactivate E. coli survival. The findings of this study will provide useful and practical guideline to applicators of agricultural soil in deciding appropriate handling and time frames for preventing pollution of water quality for sustainable agriculture.
The avian embryos have been used as a good model to study embryonic development. Due to its unique development in the eggshell, avian embryos can be cultured and hatch in the surrogate eggshell system. In this study, we examined the viability, normal development and hatchability of Korean Oge (KO) chicken embryos in White Leghorn (WL) surrogate eggshells. Donor KO embryos at 3-day and 4-day-old were transferred into recipient WL eggshells, incubated for further 18 days at $37.5^{\circ}C$ with 70% of humidity until hatching. The viability of 3-day-old KO embryos at 7, 14 and 21 day in surrogate eggshell were 70.0%, 43.8% and 23.1%, respectively. In contrast, the viability of 4-day-old KO embryos at 7, 14 and 21 day in surrogate eggshells were 87.1%, 55.6% and 36.0%, respectively. The hatchability of KO embryos transferred into surrogate eggshells at 3-day-old was 23.1%, whereas embryos transferred at 4-day-old was 36.0%. Furthermore, the development of all viable embryos from 3-day group and 4-day group were normal. Our results suggested that culture of KO embryos in WL surrogate eggshells is highly possible, and transfer of donor embryos at 4-day-old may yield higher percentage of hatchability. This study may provide potential knowledge for the conservation of wild and endangered birds through surrogate system.
We compared the degrees of vitality of 12 Korean native chicken breeds, such as Jaeraejong, Korean Rhode Island Red (Rhode), Korean White Leghorn, Korean Cornish (Cornish), and Korean Ogye breeds. The survival rate and telomere length were measured as vital markers. Telomere length was analyzed via quantitative fluorescence in situ hybridization method using the lymphocytes of 466 chickens. We found that the telomere length decreased linearly with increasing chronological age in all chicken breeds. Telomere length and telomere shortening rates (TSR) were significantly different among the chicken breeds after 20 weeks of age (P<0.01). Rhode had the longest telomere length and the lowest TSR, whereas Cornish had the shortest telomere length and the highest TSR. In terms of TSR, the telomere length of 50-week-old chickens was half of that of 8-week-old chickens. There was also a significant difference in survival rates among the breeds. Both Rhode and Korean Ogye had the highest survival rates, while Cornish had the lowest. There was a significant positive correlation between survival rate and telomere length, and telomere length in old age showed a higher correlation with survival rate than that in young age. Therefore, it is considered that TSR is more closely related to survival rate than the telomere length. Based on the telomere dynamics and survival rates of 12 Korean native chicken breeds, it was concluded that the Rhode breed and Cornish breed had the highest and lowest vitality, respectively.

Chickens are a species of vertebrate with varying colors. Various colors of chickens must be classified to find color-related genes. In the past, color scoring was performed based on human visual observation. Therefore, chicken colors have not been measured with precise standards. In order to solve this problem, a computer vision approach was used in this study. Image quantization based on k-means clustering for all pixels of RGB values can objectively distinguish inherited colors that are expressed in various ways. This study was also conducted to determine whether plumage color differences exist in the reciprocal cross lines between two breeds: black Yeonsan Ogye (YO) and White Leghorn (WL). Line B is a crossbred line between YO males and WL females while Line L is a reciprocal crossbred line between WL males and YO females. One male and ten females were selected for each F1 line, and full-sib mating was conducted to generate 883 F2 birds. The results indicate that the distribution of light and dark colors of k-means clustering converged to 7:3. Additionally, the color of Line B was lighter than that of Line L (P<0.01). This study suggests that the genes underlying plumage colors can be identified using quantification values from the computer vision approach described in this study.
