Effects of 3-dimensional Co-culture of Human Endometrial Cells Decidualized with Progesterone and TGF-${\beta}1$ on the Development of Mouse 2-cell Embryos In Vitro (Progesterone과 TGF-${\beta}1$에 의해 탈락막화가 유도된 인간 자궁내막세포의 삼차원 공배양이 2-세포기 생쥐배아의 체외발달에 미치는 영향)

  • Objective: This study was carried out to investigate the effects of 3-dimensional co-culture of human endometrial cells decidualized with progesterone and TGF-${\beta}1$ on the development of 2-cell mouse embryos. Methods: Stromal and epithelial cells isolated from human endometrial tissue were immunostained for cytokeratin and vimentin. Expression of TGF-${\beta}1$, its receptor-1, -2, integrin-${\beta}3$ and prolactin in mono or co-culture according to three different hormone conditions was investigated by RT-PCR. Differential staining was used to investigate the number of ICM and trophectoderm of hatched mouse blastocysts in different three conditions. Results: The immunohistochemical study was positive for cytokeratin or vimentin and confirmed that epithelial and stromal cells were isolated from endometrial tissue successfully. In co-culture, TGF-${\beta}1$, its receptor-1, integrin-${\beta}3$ and prolactin except TGF-${\beta}1$-r2 were expressed in progesterone dominant condition. The hatching and attaching rate were higher in the co-culture with decidualized cells (p<0.05). However, we observed that lots of the incomplete hatched blactocysts attached on non-decidualized cells. The ICM number of hatched mouse blastocysts was higher in co-culture with decidualized and non decidualized cells than media only culture (p<0.05). The trophectoderm number of hatched blastocyst was higher in the co-culture with decidualized cells than non-decidualized cells or media only culture (p<0.05). Conclusion: The administration of progesterone, estrogen and TGF-$\beta$ could induce decidualization of stromal and epithelial cells isolated from human endometrial tissue using 3-dimensional co-culture, and the decidualization of human endometrial cells could increase the hatching and attaching rate of 2-cell mouse embryos.

Changes of the Preoperative and Postoperative Nutritional Statuses in Patients with Gastric Cancer and Assessment of the Nutritional Factors That Are Correlated with Short-Term Postoperative Complications (위암 환자에서 수술 전후의 영양지표의 변화와 수술 후 단기간 합병증과의 연관성)

  • Purpose: The aim of this study was to evaluate the preoperative and postoperative nutritional statuses of patients with gastric cancer and to investigate the nutritional factors that are correlated with perioperative complications. Materials and Methods: From January 2008 to Jun 2008, 669 patients who underwent curative gastrectomy were enrolled in a retrospective study. To evaluate the changes of their nutritional status preoperatively and postoperatively, we measured the total lymphocyte count, the serum albumin, the body weight change and the BMI. The nutritional factors correlated with short-term postoperative complications were analyzed. Results: The total lymphocyte count and serum albumin decreased from the first preoperative day to the $5^{th}$ day after operation, but they tended to increase and approach the normal range 6 months after operation. The only factor correlated with the short-term postoperative complications (defined as the ones that occurred for 30 days) was the serum albumin checked on the $5^{th}$ day after operation. Conclusion: Low serum albumin on the $5^{th}$ day after operation was correlated with postoperative short-term complications. Serum albumin can be the preoperative statistical parameter that can predict the occurrence of postoperative complications.

Iron Deficiency Anemia and Vitamin D Deficiency in Breastfed Infants (모유수유아에서의 철결핍 빈혈과 비타민 D 결핍)

  • Purpose: Iron deficiency anemia (IDA) is one of the most common nutritional problems, despite a recent improvement of nutritional status of infants and children. We assessed the risk factors for IDA in infants and vitamin D deficiency and IDA by nutrition analysis. Methods: We analyzed blood tests and evaluated 103 children with IDA and 123 children without IDA, 6-36 months of age, who were cared for in our hospital between March 2006 and July 2010. Nutritional analysis using Canpro was performed among breastfed infants 6~12 months of age who had been diagnosed with IDA and had detailed diet histories. Results: Breastfed infants accounted for 87.4% and 40.7% of the IDA and comparison groups, respectively. The IDA and comparison groups began weaning food at 6.4${\pm}$1.8 and 5.9${\pm}$1.3 months, respectively. In the IDA and comparison groups, 46.4% and 53.5% began to adapt to weaning food within 4 weeks, respectively. The most common reason for hospital care of the IDA group was respiratory symptoms constituting 36.2%. Only 18.6% visited the hospital for palloror anemia. The Canpro analysis, performed on 11 infants with IDA, showed that iron and vitamin D were <40% and 30% of recommended intakes, respectively. Conclusion: Weaning food should be started 4~6 months of age in breastfed infants. In infants at high risk for IDA and vitamin D deficiency, screening tests should be recommended. The high-risk infants may require iron, vitamin D fortified formula, or oral supplements.

Nutrition Knowledge, Dietary Habits and Nutrition Attitudes of Elementary and Middle School Teachers in Masan City (마산시 초.충 교사들의 영양지식, 식생활 습관 및 영양태도)

  • This study was carried to investigate the nutrition knowledge, dietary habits and nutrition attitude of 227 teachers of elementary and middle school in Masan city. The survey was conducted by a self-administered questionnaire. The results showed that the subjects had a high level of perceived nutrition knowledge (85.2%), but the accuracy of the knowledge was 76.2%, and percentage of correct answers was 65.5%. The average nutrition knowledge score was 13.1 out of possible 20 points, the average score of dietary habits was 76.2 and nutrition attitudes score was 70.8 out of 100 points. Most of the subjects are belonged to the 'good'group in terms of nutrition knowledge, dietary habits and nutrition attitudes level. Female teachers scored significantly higher on nutrition knowledge and nutrition attitudes than male teachers did (p<0.001). And elementary school teachers scored significantly higher on dietary habits than noddle school teachers (p<0.01). There was a highly significant correlations between nutrition knowledge and nutrition attitude (r =0.3, p<0.001); dietary habit and nutrition attitude (r : 0.4, p<0.001). But the correlation between nutrition knowledge and dietary habits was not significant.

  • 노인에게서 두드러지게 나타나고 있는 저근육형 비만은 근육감소를 동반한 체지방의 증가로 신체상의 뚜렷한 체성분의 변화를 야기 시킨다. 이때 골감소증을 동반하여 신체기능의 감소 및 골절장애 그리고 대사성 관련 질환의 위험도가 올라가는 것으로 보고되고 있다. 노화로 인한 체성분의 변화는 단순한 저근육형일 경우와 비만일 때 보다 급격히 증가된 복부내장 지방조직에서 분비되는 염증성 사이토카인, C-반응성 단백질(CRP), 인터루킨(IL)-6, IL-8 및 종양 괴사 인자(TNF-${\alpha}$)들이 단백질 대사를 저해하여 근육량의 감소를 더욱 촉진시키며, 염증관련 대사질환의 유병률에 중요한 요인이다. 본 연구에서는 DNA 메틸화가 당뇨병, 심혈관질환, 암과 같은 만성염증성 질환에 관계하고 있다는 최근 연구 결과를 기초로 하여 항염증 영양소와 생리활성을 갖는 식품인자들의 충분한 섭취가 염증조절에 중요하게 기여할 것으로 생각되며, 또한 염증성 질환의 주요 표식자인 DNA 메틸화와 히스톤 변형을 유발하는 효소의 활성 또는 비 암호화된 RNA의 발현을 조절함으로써 근육량 증가와 체지방 감소에 중요한 역할을 하는 것을 살펴보았다. 따라서 최근 새롭게 인식되는 후생유전학적 연구의 중심에 있는 항염증 영양소의 효과와 체성분 변화와의 긍정적 관계를 중심으로 저근육형 비만의 예방 및 인구고령화에 건강한 노화를 위한 효과적인 방법을 제시하였다.

Associations between Clinical Characteristics and Plasma BDNF Levels of Panic Disorder (공황장애의 임상적 특성과 Brain-Derived Neurotrophic Factor 농도와의 관계)

  • Objective : Brain-derived neurotrophic factor (BDNF) is implicated in the pathophysiology of several neuropsychiatric disorders. However, there are few studies on BDNF of panic disorder. In this study, we investigated plasma BDNF levels in patients with panic disorder, and evaluated whether there are associations between clinical characteristics of panic disorder and plasma BDNF levels. Methods : We included 110 patients with panic disorder and 110 health controls in the current study. Plasma BDNF levels were measured by the enzyme-linked immunosorbent assay (ELISA). Plasma BDNF level differences were evaluated according to the clinical characteristics, such as duration of illness, recent stressful life event, agoraphobia, and insomnia. Results : The mean plasma BDNF levels of patients with panic disorder were significantly lower, as compared with those of controls (192.50 pg/mL vs. 693.75 pg/mL, t=8.838, p<0.001). The mean plasma BDNF levels of patients who had recent stressful life events were significantly higher, as compared with those who did not ($269.79{\pm}358.96pg/mL$ vs. $136.94{\pm}187.06pg/mL$, t=-2.525, p=0.013). Conclusion : These results suggested that BDNF plays a potential role in the pathophysiology of panic disorder.

  • SWAT 모형을 사용하여 수문 및 환경인자들의 예측에 있어서 수문응답단위(Hydrological Response Unit, HRU) 수를 적절하게 결정하는 것은 매우 중요하다. SWAT 모형에서는 수문응답단위라는 계산단위를 통하여 모형의 입력매개변수를 생성하고 모의를 수행하기 때문이다. 본 연구에서는 SWAT모형에서 하천유역의 적절한 HRU 수를 결정하기 위하여 대상유역을 낙동강의 제 1지류이자 국제수문개발계획(International Hydrologic Project, IHP)의 국내 대표유역 중 하나인 위천유역으로 선정하였으며, 토지이용과 토양의 면적비를 조정함으로써 HRU 수를 여러 단계로 적용하여 유출량, 유사량 및 영양염류를 모의하였다. SWAT 모형의 적용을 위하여 위천 유역의 $DEM(30m{\times}30m)$, 토지이용도(1:25,000), 토양도(1:25,000) 등의 GIS 자료와 강우량 및 기상자료를 이용하였다. 본 논문은 위천 유역에 대한 적절한 HRU 수의 기준을 제시하였으며, 이와 같은 분석결과를 이용하여 모형의 입력자료 구축시간을 단축할 수 있어 차후 모형의 적용시에 모형의 적용 효율을 높일 수 있을 것으로 판단된다.

Effect of oral health status on risk factors for obstructive sleep apnea in middle-aged Koreans: the Korea National Health and Nutrition Examination Survey (2019) (한국 중장년층의 구강 건강 상태가 폐쇄성 수면 무호흡증 위험인자에 미치는 영향: 국민건강영양조사를 바탕으로(2019년도))

  • Objectives: This study aimed to investigate the effect of the oral health status on risk factors for obstructive sleep apnea (OSA) using data from the 2019 National Health and Nutrition Examination Survey. Methods: Of a total of 2,422 persons, 1,295 and 1,127 were categorized into the control group (CG) and OSA risk group (OSARG), respectively. The effect of the oral health status on OSA risk factors was presented in Model 1 by performing a complex sample linear regression analysis. Results: Our findings showed that OSA risk factors decreased by 0.075 points when there were no speaking problems with demographic characteristics adjusted. In addition, when systemic diseases were adjusted for, OSA risk factors decreased to 0.074 points (p<0.05). Conclusions: Therefore, in order to reduce oral problems that affect OSA risk factors, dentists and dental hygienists should seek accurate recognition of OSA and effective oral care methods.

Effects of Extract Mixture (Yg-1) of Anti-Inflammatory Herbs on LPS-Induced Acute Inflammation in Macrophages and Rats (급성염증성 발열 모델에서의 항염증성 약재 혼합 추출물(YG-1)의 효과)

  • Traditional herbs, such as Lonicera japonica, Arctii Fructus, and Scutellariae Radix have been used as traditional drug due to their anti-inflammatory and anti-oxidant activities. The aim of this study was to investigate the anti-inflammatory effects of extract mixture (YG-1) in a model of lipopolysaccharide (LPS)-induced acute inflammation in both macrophage (RAW 264.7) cells and Sprague-Dawley rats. YG-1 did not show specific cellular toxicity in RAW 264.7 cells until a concentration of $100{\mu}g/mL$. YG-1 reduced various markers related to inflammation such as IL-$1{\beta}$, COX-2, and iNOS caused by LPS in RAW 264.7 cells. Consistent with these results, YG-1 exerted significant anti-inflammatory effects in an acute inflammation rat model. Acute fever and high concentration of IL-$1{\beta}$ in serum induced by LPS were significantly reduced by YG-1. These results were similar to flubiprofen, a commercial anti-inflammatory and anti-febrile drug. Therefore, these results indicate that YG-1 has beneficial effects on LPS-induced acute inflammation and suggest that YG-1 can serve as an effective anti-inflammatory and anti-febrile drug.

Inhibitory Effects of Flavonoids on Growth of HT-29 Human Colon Cancer Cells (Flavonoid의 HT-29 대장암세포 증식 억제 효과)

  • This study was performed to elucidate the anti-proliferative and apoptotic mechanism of flavonoids in HT-29 human colon cancer cells. We investigated the anti-proliferative activity of flavonoids in HT-29 human colon cancer cells via cell viability assay (MTT assay), caspase-3 activity, RT-PCR, and western blotting. We cultured HT-29 cells in the presence of various flavonoids (apigenin, rutin, naringenin, and myricetin) at a concentration of $100{\mu}M$. In the MTT assay, naringenin showed the strongest effect on cell viability in HT-29 colon cancer cells. Caspase-3 activity, a marker of apoptosis, significantly increased upon naringenin treatment. For RT-PCR, myricetin significantly increased Bax protein levels, naringenin increased p53 protein levels, and rutin reduced expression of the anti-apoptotic protein Bcl-2. Western blotting of HT-29 colon cancer cells showed that myricetin increased cleaved caspase-3 protein levels, naringenin significantly increased poly (ADP-ribose) polymerase protein levels, and rutin increased E-cadherin protein levels. These results indicate that flavonoid exerts anticancer effects on human colon HT-29 cells through a caspase-dependent apoptotic pathway.
