Convergence study on anti-caries effect of chlorhexidine and essential oils (클로르헥시딘과 에센셜 오일의 항우식 효과에 대한 융합 연구)

  • The aim of this study was to evaluate the anti-caries effect of the essential oils and chlorhexidine mouth rinse. 42 subjects were randomized allocated to the chlorhexidine (n=22) and essential oils group (n=20). Subjects were instructed to rinsing each mouthwash by 15 ml once a day during the 7 days. The CRT and Cariview scores were recorded at baseline and 4 weeks after treatment. Data was analysed by using PASW 18.0 program with independent t-test and paired t-test. After 1 month, Cariview score was shown reducing compare to baseline. However, there were no significant differences between the groups. High risk level of MS and LB was decreased to 9~15% after treatment in two groups. Both of the chlorhexidine and essential oils mouth wash showed an antiplaque effect. Although chlorhexidine continues to be the "gold standard" in terms of antiplaque effect, essential oils could be considered a reliable alternative antimicrobial agent.

  • Oleanolic acid (OA) and ursolic acid (UA) are triterpenoid compound present in many plants. This study examined the antimicrobial activity of OA and UA against Streptococcus downei. The antimicrobial activity was evaluated by the minimal inhibitory concentration (MIC) and time kill curves. The MIC values of OA and UA for S. downei isolated from the Korean population were $8{\mu}g/ml$. OA and UA had a bactericidal effect on S. downei ATCC $33748^T$ above $2{\times}MIC$, $16{\mu}g/ml$ and $8{\mu}g/ml$, respectively. The results suggest that OA and UA can be used in the development of oral hygiene products for the prevention of dental caries.

  • Biochemical characteristic of Streptococcus iniae and Streptococcus parauberis that are pathogens of streptococcosis of cultured flounder, Paralichthys olivaceus, in Jeju area was examined. The result of experiments on the grow according to temperatures showed that only S. parauberis grew at 10℃, the result of hemolysis test showed that only S. iniae bacteria showed β hemolysis. Only S. parauberis were positive in VP test and HIP test, both bacteria used α-D-glucose, D-mannose, D-psicose, D-trehalose, pyruvatic acid methyl ester, and glycerol as substrates. L-lactic acid was used only S. iniae bacteria, and β-methyl-D-glucosid was used only by S. parauberis. S. iniae exhibited acute infection patten, differently S. parauberis exhibited chronic infection patten in pathogenic test.

  • Group B Streptococcal sepsis and/or meningitis is one of the most serious and common diseases in the neonatal period with high mortality and frequent complications. We have experienced a case of late onset type group B streptococcal sepsis and meningitis with a complication of subclavian vein catheterization catheterization. This 29-day-old male neonate was admitted to intensive care unit with the presentation of fever and septic shock. He was born with Cesarean delivery at 36 weeks and 3 days of gestational age. He showed multiple episodes of seizure after admission and group B streptococcus was isolated from blood. CSF profiles 10 days after admission showed the features of bacterial meningitis without organism isolated. Diffuse cerebral infarction was detected on brain CT 24 days after admission. In the 13th hospital day, the complication of subclavian vein catheterization occurred; Guide wire was cut during insertion and the distal portion of it(2.5cm) was retained in the left subclavian vein. We removed the retained guide wire with goose-neck snare catheter via right femoral vein. This case was presented with a brief review of the literatures.

  • Bacteria have been regarded as a one of major etiologic factors in root canal infections. In endodontic treatment the effective removal of pathogenic microorganisms in the root canal is the key to successful outcome. Bacterial cell wall components may play an important role in the development of pulpal and periapical disease. The purpose of this study was to evaluate the effect of sonic extracts of Streptococcus spp. on cultured L929 cells and to characterize cell wall protein profiles of Streptococcus spp. Streptococcus spp. were isolated from infected root canals and identified with Vitek Systems(Biomeriux, USA). Five streptococci, namely S. sanguis, S. mitis, S uberis, S. mutans (ATCC 10449) and S. faecalis (ATCC 19433) weere enriched in brain heart infusion broth. Cell pellets were sonicated and cell wall extracts were dialyzed and membrane filtered. Prepared cell wall proteins were applied to cultured L929 cell. The cell reaction were evaluated by monitoring DNA synthesis, cell numbers and the change of cell morphology. The total cell wall protein profiles of microorganisms were characterized by sodium dodecyl sulfate polyacrylamide-gel eledruphoresis(SDS-PAGE). DNA synthesis of L929 cells were reduced by the increasing concentration of sonic extracts. DNA synthesis was significantly suppressed in more than $50{\mu}g$/ml of sonic extract conentration in five streptococci. S. nutans (ATCC 10449) showed stronger suppression on DNA synthesis than remaining four streptococci, which had the similar effect on DNA synthesis. Analysis of DNA synthesis measured by [$^3H$]-thymidine uptake was more sensitvie method than cell counting. Sonic extracts affected the microscopic findings of L929 cells. The protein profiles indicated that all five strains shared two major proteins with molecular masses of 70.8 and 57.5 kD respectively. S. uberis and S. mutans shared common minor proteins of which molecular weights were 147.9 and 112.2 kD respectively. However some minor proteins were unique for S. mitis, S. uberis and S. faecalis.

The efficacy and influence on growth of olive flounder Paralichthys olivaceus vaccinated against Edwardsiella tarda and Streptococcus iniae (넙치, Paralichthys olivaceus에 투여된 Edwardsiella tarda와 Streptococcus iniae에 대한 혼합백신의 예방효과와 성장에 미치는 영향)

  • This study was performed to verify the efficacy and safety of vaccine mixed formalin killed Edwardsiella tarda and formalin killed Streptococcus iniae in olive flounder Paralichthys olivaceus. Sera were obtained from each group at 2, 4, 6 and 8 weeks after vaccination and agglutination titers to E. tarda and S. iniae were determined using the microtiter method. Three weeks after vaccination, the fish challenged by intraperitoneal injection of E. tarda or S. iniae and immunization resulted in a relative percentage survival (RPS) of above 81.0-92.8%. Neither the groups vaccinated nor control groups resulted in significantly reduced weight gain 140 days post-vaccination, but differences between the groups vaccinated and control groups were found at the early days post-vaccination. This results suggest that it is possible to prevent edwardsiellosis and streptococosis after a vaccination without affecting growth of olive flounder.

  • Between January 1986 and June 1994, 23 patients underwent surgical intervention for infective endocarditis at Wonkwang University Hospital. There were 13 male and 5 female, ran ing in age from 13 to 67(mean 43.7 $\pm$ 5.8) years. 21 had native valve endocarditis and 2 had prosthetic valve endocardits. The most common causative organism was streptococci in 9 patients, and the others were staphylococci in 7 patients and unknown in 7 patients. The infection was in the mitral valve in 10 patients, the aortic valve in 6 patients, the aortic and mitral In 5 patients, 2 in tricuspid. Surgical indications for surgery were intractable congestive heart failure, giant growing vegetation and severe valvular insufficiency. There were 3 perioperative death (13%) and no late death. The main causes of death were cerebral embolism in 2 due to recurrent endocarditis and low cardiac output in 1. By NYHA functional classification, all patients were in class III or IV preoperatively, and all patients were improved postoperatively with HYHA functional classification in class I or II. In conclussion, early operative intervention is life-salving in patients with persistent or progres ive congestive heart failure, irrespective of the activity of the infectious process or the duration of antibiotic therapy.

  • Beta-hemolytic streptococci (BHS) are frequently isolated pathogens in clinical microbiology laboratories. Among them, Group C, G and F streptococci infrequently cause respiratory infections, but they often invade other sites. Patients with underlying diseases are prone to be infected by the organisms and some of them can be fatal. Therefore, combination tests of serological and biochemical tests are needed to correct the identification of non-A, B streptococci because to various species belong to this species. The aim of this study was to investigate the isolation rates of strains and distribution of serogroup C, G and F streptococci at Wonju Hospital during the period of 2003-2004. 133 clinical isolates of group C, G and F streptococci were examined. Of them, 13 (9.8%) were group C, 41 (31.8%) were group F, 79 (59.4%) were group G. The prevalent isolation of the strains by serogroup were: group C, Streptococcus constellatus (38.5%); group F, Streptococcus anginosus (56.1%), Streptococcus constellatus (26.9%); group G, Streptococcus anginosus (54.4%), Streptococcus dysgalatiae subsp. equisimilis (26.5%). Overall, the most frequently isolated BHS was Streptococcus anginosus (51.2%).

Use of 16S-23S rRNA Intergenic Spacer Region for identification in the fish pathogenic Streptococcus iniae (16S-23S rRNA Intergenic Spacer Region을 이용한 어류 병원성Streptococcus iniae의 분자생물학적 동정)

  • This study was performed for the identification of Streptococcus sp. from cultured flounders (Paralichthys olivaceus) showing streptococcosis in the Jeju island. We isolated 10 strains of Streptococcus iniae from the cultured olive flounders with streptococcosis. Isolated strains were identified in S. iniae since they have formed the expected band through performing PCR assay using specific primers, Sin-1 (5'-CTAGAGTACACATGTACT(AGCT)AAG-3') and Sin-2 (5'-GGATTTTCCACTCCCATTAC-3'). In addition to 16S-23S rRNA intergenic spacers (ISR), operon structure of isolated strains showed that all strains had three 16S-23S rRNA ISR band patterns. The 16S-23S rRNA ISR sequence of isolated strains showed 96% sequence identity with S. iniae (GenBank accession number AF 048773). This paper is the first report that S. iniae is associated with streptococcosis of Olive flounder in Korea.

  • This examination was carried out to evaluate the sanitary quality of slices of raw fish being served in the restaurants. Twenty five kinds of slices of raw fish collected from various restaurants in Busan Korea were examined during the period from March to May in 1971. As the evaluation factors of sanitary quality, the contents of sanitary indicative bacteria such as coliform group, fecal coliform, feral streptococci and enterococci and plate counts were determined. The results obtained are as in below: 1. The numbers of fecal streptococci and enterococci MPN were much greater than those of coliform group and fecal coliform. 2. The median value of coliform group MPN was 3,300 per 100 grams of the sample examined and those of enterococci was 5,400. The median value of plate counts was $1.8\times10^5$ per gram. 3. Fifty-two percent of the samples examined were exceeded fecal coliform MPN 930 per 100 grams. 4. As a sanitary indicative bacteria fecal coliform MPN was more reasonable than enterococci 5. The grade of restaurants was not correlated with the bacterial quality of the foods served. 6. No correlation between the numbers of sanitary indicative bacteria ana plate counts was observed.
