반응표면 분석법을 이용한 계란 난황내 레시틴 분리 최적조건 연구

  • 난황으로부터 인지질을 회수하기 위하여 전처리, 표면반응분석기법을 실시한 결과 인지질 95%이상 PC함량 55%의 제품을 생산할 수 있는 공정을 확립하였다. 난황 인지질 분리조건(액난황 100g 기준)은 알코올 농도 91.38%, 알코올 첨가량 1: 7.3, 균질온도 $29.22^{\circ}C$, 균질시간 5분으로 한 후 원심분리한 다음 여과후 진공농축하고 아세톤 100ml로 1회 세척하고 나서 여과지로 여과하고 $100^{\circ}C$, 40분 건조하거나 $40^{\circ}C$ 진공건조기에서 overnight 정도 건조과정을 휘발시키는 것이 바람직한 것으로 생각된다.

States of Am in Aqueous Solution (수용액중의 Am의 상태)

  • The state of Am in the concentration of 10$^{-5}$M has been studied in the pH range of 5 to 10 by filtration and centrifugation method. By the experiments, we could estimate possible Am hydrolysis products and solubility constants. If the solubility of Am (OH)$_3$ estimated by Baes and Mesmer is increased about a factor of 10, i.e. changing logk=-l8.7 to logk=-17 5 it was found that the calculated curve of Am concentration versus pH agreed completely with experimental values.

Studies on Carrier-Free Silver-111 with Membrane Filters (막여과지에 의한 무단체 Ag-111에 관한 연구)

  • The state of carrier-free silver-111 has been studied by applying filtration method. The studies involved that the effects of pH and concentration of silver-111 in aqueous solution have been determined with membrane filters. The present studies revealed that the retainment of silver-111 on membrane filters followed Freundlish adsorption isotherm, and the adsorbed state of silver-111 was present in the form of AgOH. Also it was supposed that the formation of the non-adsorbed hydroxide of$Ag(OH)_{2}-$ may prohibit the existance of AgOH at higher pH, and it seems to be valid that the carrier-free silver-111 in aqueous solution exists in$Ag^+$state.

Effects of Filtration or Centrifugation on the Oxidative Stabilities of Sesame Oil (여과 및 원심 분리가 참기름의 산화 안정성에 미치는 영향)

  • Effects of filtration and centrifugation on the oxidative stabilities of sesame oils during storage at $70^{\circ}C$ were studied by combination of determining peroxide values and conjugated dienoic acid values of oils and measuring the hexanal formation using headspace gas chromatography. Crude sesame oil from roasted seeds contained more free fatty acids, conjugated dienes, and metals (Fe, Cu, Mg and Zn); on the other hand, higher contents of moisture and ${\gamma}-tocopherol$ were found in the filtered or centrifuged oil. Only filtered oil contained more peroxides than the crude oil in spite of the color advantage of the highest L and b values among three oils. All the oils showed the tendency of increasing in total color difference during storage at $70^{\circ}C$, fatty acid compositions were relatively constant except for decreasing tendency of linoleic acid in filtered oil. No significant difference at 1% in the oxidative stabilities was observed between centrifuged oil and crude oil with higher susceptibility to the oxidation in the filtered oil. Centrifuged sesame oil was the best in the aspect of both oxidative stability and the oil color.

Shelf-life Extension of Acer mono Sap Using Ultra Filtration (한외여과법을 이용한 고로쇠 수액의 저장성 향상)

  • This study was investigated the effect of the ultra filtration (UF) on extending shelf-life and increasing quality of Acer mono sap. To evaluate the quality changes of Acer mono sap before and after UF, crude ash, crude protein, minerals, free sugar, organic acid, pH, total acidity, browning index, turbidity and total microbial number were measured during storage periods at -1, 4 and $10^{\circ}C$. After UF, the ash, minerals, organic acid, total acidity, browning index and turbidity of Acer mono sap were slightly decreased, while the pH was slightly increased, and total microbial number was not detected. After UF, the pH, total acidity, browning index, turbidity and total microbial number of Acer mono sap were very stable during storage periods at -1 and $4^{\circ}C$. During the storage at $10^{\circ}C$, turbidity and total microbial number was slightly increased, while the pH of before UF was decreased and the total acidity, browning, turbidity and total microbial number was increased during storage periods. As the results of this study indicate application of UF on Acer mono sap could extend the shelf-life and quality without loss of natural minerals and useful components.

Determination of Representative Renal Depth for Accurate Attenuation Correction in Measurement of Glomerular Filtration Rate in Transplanted Kidney (이식 신의 사구체 여과율 측정에서 정확한 감쇄 보정을 위한 신장 깊이 대표값 설정)

  • Purpose: To measure reliable glomerular filtration rate by using the representative values of transplanted renal depths, which are measured with ultrasonography. Materials and Methods: We included 54 patients (26 men, 28 women), with having both renal scintigraphy and ultrasonography after renal transplantation. We measured GFR with Gates' method using the renal depth measured by ultrasonography, and median and mean ones in each patient. We compared GFR derived from ultrasonography-measured renal depth with GFR derived from median and mean renal depths. The correlation coefficients were obtained among GFR derived from ultrasonography-measured renal depths, median and mean renal depths under linear regression analysis. We determined whether GFR derived from median or mean renal depth could substitute GFR derived from ultrasonography-measured renal depth with Bland-Altman method. We analyze the expected errors of the GFR using representative renal depth in terms of age, sex, weight, height, creatinine value, and body surface. Results: The transplanted renal depths range from 3.20 cm to 5.96 cm. The mean value and standard deviation of renal depths measured by ultrasonography are $4.09{\pm}0.65cm$ in men, and $4.24{\pm}0.78cm$ in women. The median value of renal depths measured by ultrasonography is 4.36 cm in men and 4.14 cm in women. The GFR derived from median renal depth is more consistent with GFR derived from ultrasonography-measured renal depth than GFR derived from mean renal depth. Differences of GFR derived from median and ultrasonography-measured renal depth are not significantly different in the groups classified with creatinine value, age, sex, height, weight and body surface. Conclusion: When median value is adapted as a representative renal depth, we could obtain reliable GFR in transplanted kidney simply.

Detection of Denitrifying Bacteria in Groundwater by PCR (PCR을 이용한 지하수 내의 탈질화 세균의 검출)

  • Groundwater samples were collected at 6 sites in Seoul area. DNA extraction from the sample was performed by the boiling method. Samples were boiled with guanidinium thyocyanate and phenol-chloroform. One set of primer was designed for amplification of 16S rDNA. For detection of denitrifying bacteria in groundwater sample, the author used primer sets consensus regions in gene sequences encoding the two forms of nitrite reductase (NIR), a key enzyme in the denitrification pathway. Two sets of PCR primer were designed to amplify $cd_1$-and Cu-nir. We confirmed the existence of denitrifying bacteria in 3 sites using $cd_1$-nir primer and in 4 sites using Cu-nir primer.

Optimization of Extraction and Clarification Condition for Preparation of Liquid Extract Tea from Artificially Cultivated Phellinus linteus (인공재배 상황버섯 액상추출차 제조를 위한 최적추출 및 청징화 조건)

  • Optimum extraction condition and clarification process were investigated to manufacture liquid extract tea using Phellinus linteus. Carbohydrates content in the raw Phellinus linteus was 80.9%. Major minerals were K, Ca, Fe and Na, but vitamin B$_1$, B$_2$, and C were not detected. The best extraction condition was 5% raw material at 10$0^{\circ}C$ within 3 hr. To clarify the extract, three methods of filtration with depth filter pad, centrifugation and addition of several filter aids were studied. Filtration with depth filter pad and centrifugation were highly effective on the clarification and the changes in the turbidity for 7 weeks at 4$0^{\circ}C$ were not occurred. The optimum clarification condition was centrifugation above 6,000 rpm or filtration with 8 ${\mu}{\textrm}{m}$ depth filter pad.

Experimental Evaluation Method of Mass Transfer Coefficient on Biotrickling Filtration for Air Pollution Control (대기오염제어를 위한 생물살수여과법에서 물질전달계수 실험평가방법에 관한 연구)

  • Biological treatment is promising alternative to conventional air pollution control method. Bioreactors for air pollution control have found most of their success in the treatment of dilute and high flow waste air streams containing volatile organic compounds and odor. The studies of mass transfer in biotrickling filters for air pollution control were of importance in order to control and optimize the purification process. The objectives of this study were to develop the experimental methodologies to evaluate the mass transfer coefficients of gas/liquid(trickling liquid), gas/solid(biomass) and liquid/solid in three phase biotrickling filtration. Also, this study characterized the influence factors on mass transfer such as dynamic holdup volume, gas/liquid flow rate ratio, biomass weight in reactor and recirculation rate of trickling medium for each phase of biotrickling filter.
