A Study on the Antimicrobial Effect of Ginkgo biloba Leaves Extracts according to Concentrations of Ethanol for staphylococcus aureus (포도상구균에 대만 에탄올 농도별 은행잎 추출물의 항균효과에 관한 연구)

  • A optimal condition for the Ginkgo biloba extraction in ethanol and water binary solvent system has been proposed based on concentration of bilobalide and ginkgolide known as having a antimicrobial components in the range 5% to 70% ethanol in water at $80^{\circ}C$. Concentration of bilobalide as a single component of Ginkgo biloba leaves extract is the highest at the 60% ethanol and ginkgolide A and B is highest at 50% ethanol. Antimicrobial effect of Ginkgo biloba leaves extracts on the S. aureus was also examined by disc diffusion test and optical density test. In case of the disc diffusion test, the clean zone diameter was increased from 0.95 cm to 1.70 cm as ethanol concentration increased from 5 to 70%. However, over the 40% of ethanol concentration the antimicrobial effect was almost flat. Based on these results, we propose that the 40% of ethanol and 60% water solvent is most desirable for Ginkgo biloba extract considering vapor pressure problem in concentrating process after extraction. We introduced SEM and TEM to figure out the morphological change on the surface and inside body of S. aureus when Ginkgo biloba leaves extract was treated. After mixed with Ginkgo biloba leaves extract blast like blebs appeared on the surface of S. aureus cells and cell wall was not observed. From the these results, it seems that the Ginkgo biloba leaves extract including bilobalide and ginkgolide A, B prevent cell wall synthesis.

Effects of Main Constituents of W/O Emulsion on Removal of Acetic Acid in a Simulated Hemicellulosic Hydrolysate (W/O 에멀젼의 주요 구성 성분들이 모사 헤미셀룰로오스 가수분해액에 있는 초산의 제거에 미치는 영향)

  • Acetic acid is the most abundant and serious ethanol fermentation inhibitor in dilute-acid hemicellulosic hydrolysates. A mixture of xylose, acetic acid and sulfuric acid was chosen as a simulated hemicellulosic hydrolysate so as to find an optimal separation system to selectively remove acetic acid from the hydrolysates. In order to attain the purpose, emulsion liquid membrane was applied to removal of acetic acid from the simulated hemicellulosic hydrolysate. The effects of main constituents of water-in-oil (W/O) emulsion, such as amine extractant type, surfactant composition, additive type, and type and concentration of stripping agent, on extraction of acetic acid, xylose, and sulfuric acid in the simulated hemicellulosic hydrolysate were investigated. Under specific experimental conditions, degree of extraction of acetic acid was higher than 95% while loss of xylose was insignificant, which means that the current emulsion liquid membrane can be an economically feasible process.

  • An investigation was carried out to study the effects of extraction, heat treatment, and storage temperature on functional ingredients of onion. Extracts of onion paste and freeze dried onion were prepared by treatments with several solvents including hot water, methanol, and ethanol, and then contents of functional ingredients such as total phenol, flavonoid, quercetin, and thiosulfinate were determined. The highest levels of functional ingredients were observed in samples extracted using water at 100$^{\circ}C$ or ethanol. The skin extract contained 60-fold more quercetin than onion paste extract, whereas no thiosulfinate was detected in onion skin extract. Heat treatment of onion extracts at 60, 80, 105, and 121$^{\circ}C$ did not effect on the levels of all functional ingredients tested. During four-week storage, the levels of total phenol, flavonoid, and quercetin were not significantly changed regardless of storage temperature. However, the total thiosulfinate was rapidly reduced as the storage temperature increased.

Effect of Fermented (Oenanthe stolonifera DC) Extract on the Activity of Enzymes Related to Liver Function of Alcohol-administered Rats and Mice (밭미나리 발효액이 알콜투여 흰쥐의 간기능관련효소활성에 미치는 영향)

  • It has long been recognized that dropwort contains specific funtional subtances for protecting human liver and preventing and curing its diseases, and thus it has been widely utilized in traditional folk remedy. In the present study. fresh of fermented extract of dropwort shoots grown on dryland and fresh extract of those grown on flooded fie1d were fed to the rats suffering from acute, subacute or chronical toxication induced by alcohol administration, and their affects were investigated. Administration of alcohol to rats and mice for 2 days at 5ml of 30% EtOH/kg/day raised total cholesterol and total glyceride which were, however, great1y supressed when alcohol was administered to the laboratory animals previously fed on fresh or fermented extract of dryland dropwort, or fresh extract of flooded field-grown dropwort for 20 days, without significant differences among the extracts. The levels of alanine aminotransferase and aspartate aminotransferase which were raised by alcohol adminstration were also lowered by feeding dropwort extract, among which that of fermented dryland-grown one was more effective than the other two. Chronic alcohol toxication was induced to rats by administering 10% alcohol for 10 months and fermented dropwort extract or tap water was fed to the rats for 5 days. The rats fed on fermented dropwort extract were lower in total cholesterol by 40% and in tota1 glyceride by 60% than the control. Alanine aminotransferase and aspartate aminotransferase in the rats fed on fermented dropwort extract were decreased by 87.2% and 91.7%, respectively, compared to the control, and the rats recovered almost to normal. Activity of alkaline phosphatase, catalase, superoxide dismutase or glutathione peroxidase changed greatly by alcohol administration in the rats suffering from chronic as well as acute toxication. The extract of fermented dry land dropwort significantly lowed the activity of those enzymes, especially, alkaline phosphatase and superoxide dismutase. The present results suggesting the possible medicinal effect of fermented dropwort extract to liver diseases.

  • The chemical attractants for sea eels, Astroconger myriaster, were identified by using fish traps in their natural habitat around namhae island, southern part of Korea. The ethyl alcohol extracts of anchovy, Engraulis japonica, which was a very good baits for sea eels showed higher attraction activities than residuals of it. Because sea eels showed high attraction activities to the free amino acid or nucleotides of synthetic anchovy extract, the succeeding experiment was conducted to identify the major chemical attractants among 9 free amino acid and 2 nucleotides by omission test. L-Glutamic acid, L-glycine, taurine, L-methionine or IMP showed highest attraction activities among 11 chemical compounds. But sea eels showed significantly weaker attraction activities to the synthethic extracts and single component than to the natural extract.

A Study on Antioxidant Activity of Ethanol Extract from Rumex crispus and Metal Adsorptivity of it's Root (소리쟁이 에탄올 추출물의 항산화 활성 및 뿌리의 중금속 흡착능에 관한 연구)

  • Antioxidant activities such as reducing power, electron donating ability(EDA) and oxidation stability of Rumex crispus extracts with ethanol were examined, and adsorption of Pb(II) and Cd(II) with dried Rumex crispus powder in aqueous solution was experimented. Extraction yields based on the soluble solid contents ranged 1.9% to 3.0%, and were inversely proportional to solvent ratio but proportional to extraction temperature. Reducing power and EDA were also increased with extraction temperature but decreased with solvent ratio. These were corresponded to 86.4% and 91.0% of ascorbic acid reducing power and EDA, respectively, on 20 folds of solvent ratio and $75^{\circ}C$ Oxidation stability of Rumex crispus extracts on the corn seed oil was 86~91.3% of butylated hydroxytoluene. Pb(II) adsorptivity of Rumex crispus was higher than Cd(II), and adsorption rate of Pb(II) was 88% in 20 ppm concentration and 60min.

Preparation of Functional Healthy Drinks by Ethanol Extracts from Defatted Safflower Seed Cake (탈지 홍화씨박 에탄올추출물 함유 기능성 건강음료의 제조)

  • Functional healthy drinks were processed with freeze dried powders of ethanol extract from of defatted safflower (Carthamus tinctorious L.) seed cake and some useful components of the drinks were investigated. Yield of freeze dried powder was the highest as 8.42% when it extracted with 60% ethanol (60% EFDP). Each drink contained 0.02% of freeze dried powder and ranged 10.6∼13.8% of soluble solid, 2.90∼3.68 of pH, 0.10∼0.83% of titratable acidity. ‘L’ value of drink-I (DSD-I) was the highest as 94.82$\pm$2.45, ‘b’ and ‘a’ value of drink-V (DSD-V) was highest as 27.15-2.65 and 28.67$\pm$2.69, respectively. Major free sugars of drink were 6015.3∼7918.2 mg% of glucose and 1511.4∼2091.0 mg% of sucrose. The content of citric acid was the highest as 179.2∼981.3 mg%. The content of total phenol in 60% EFDP was 99.17 mg% and that of drink-II(DSD-II) and DSD-V was 307.84 mg% and 224.06 mg%, respectively. Total flavonoid was contained as 50.29 mg% in 80% ethanol extract (80% EFDP) and 125.20 mg% in DSD-V. N-[2-(5-hydroxy-1H-indol-3-yl) ethyl] ferulamide (serotonin-I) was determined as high as 18.81 ppm in 80% EFDP and ranged 2.42∼2.89 ppm in drinks. N-[2-(5-hydroxy-lH-indol-3yl)ethyl]-p-coumaramide (serotonin-II) was determined as 30.17 ppm in 80% EFDP and ranged 3.79∼4.59 ppm in drinks. Acacetin, flavonoid compound were 9.83 ppm in amyloglucosidase hydrosis + 60% ethanol extract (A + 60% EFDP) and ranged 0.98∼1.26 ppm in drinks. Electron donating ability (EDA, %) was measured and compared with 100 ppm BHA as chemical antioxidant. EDA was 93.97$\pm$2.21% in A+60% EFDP, 94.79$\pm$2.26% in DSD-I, 94.69$\pm$1.37% in DSD-II, and 93.83$\pm$1.49% in BHA. DSD-II added with hot water extract solution from Korean ginseng and safflower yellow pigment recorded the highest sensory score.

Effect of Pine Needle and Green Tea Extracts on the Survival of Pathogenic Bacteria (솔잎과 녹차 추출물이 식중독세균의 생존에 미치는 영향)

  • The sensitivity of various pathogenic bacteria(Listeria monocytogenes, Staphylococcus aureus, Aeromonas hydrophila, Escherichia coli O157:H7 and Salmonella typhimurium) to the pine needle and green tea extracts was tested. Water extract of pine needle(PNW), 70% ethanol extract of pine needle(PNE), water extract of green tea(GTW) and 70% ethanol extract of green tea(GTE) were prepared for the test of antibacterial activty. Tryptic soy broth(TSB) containing 0∼2%(w/v) of pine needle and green tea extracts were inoculated with 10$\^$5/∼10$\^$6/ cells/ml of each bacterium and incubated at 35$\^{C}$ for 24 hours. The standard plate count method was used to measure the inhibitory effect of the extracts. Minimum inhibitory concentration(MIC) and minimum bactericidal concentration(MBC) were derived from the survival curves of pathogenic bacteria. Antibacterial activities of the pine needle and green tea extracts were compared with that of sodium benzoate, a preservative, by clear zone test. L. monocytogenes, S. aureus and A. hydrophila were completely inhibited at 0.4∼1.6% level while E. coli and S. typhimurium were very resistant to the pine needle extracts. Green tea extracts completely inhibited all strains tested at 0.2∼1.0% level and bactercidal to all strains except L. monocytogenes at 0.5∼2.0% level. Antibacterial activities of pine needle and green tea extracts were stronger than that of sodium benzoate. The order of antibacterial activities of pine needle and green tea extracts to the pathogenic bacteria was GTE > GTW > PNE > PNW. This result suggests that green tea extracts can be used as an effective natural antibacterial agent in food.

  • The interest in and development of healthy foods and nutraceuticals have increased because of the trend for a health-oriented society. Cinnamon is used as a food ingredient as well as a herbal medicine because of its functional properties. In this study, flavoring compounds and antioxidative activities of cinnamon extracts were investigated with different extraction solvents and extraction methods. The contents of flavoring compounds such as coumarin, cinnamic acid, cinnamaldehyde, and cinnamyl alcohol were investigated. The contents of courmarin, cinnamic acid, and cinnamylaldehyde in 70% ethanol extract were higher than those in hot water and subcritical water extracts. The contents of courmarin, cinnamic acid, and cinnamaldehyde in subcritical water extract were higher than those in hot water extract, whereas the content of cinnamyl alcohol was lower. DPPH scavenging activity increased with increasing concentration of the extracts, and the 70% ethanol extract showed the highest antioxidant activity. The ascorbic acid content of the 70% ethanol extract was largest in the antioxidative activity measurement by FRAP analysis. The ascorbic acid contents of the hot water and subcritical water extracts were similar.
