A Study on the properties of mushroom compost produced by different methods for the development of a self-propelled compost turner (자주식(自走式)퇴비교반기 개발을 위한 제조방법별 양송이 퇴비배지의 특성에 관한 연구)

  • A self-propelled turner was developed to produce mushroom compost at low costs and high efficiency, and the uniformity of compost produced by an excavator, compost mixer, and self-propelled turner was compared. The material of the compost was mixed with rice straw and chicken manure at a dry weight ratio of 3:1. After the final turning, internal temperature distribution, water content of the compost pile, ash ratio, and uniformity of the compost pile were examined. After the compost was completed, the water content was $69.9{\pm}0.54%$, $72.1{\pm}0.15%$, and $74.5{\pm}0.82%$, respectively. The length of rice straw was $47.5{\pm}15.4cm$, $24.9{\pm}10.1cm$, and $31.0{\pm}10.6cm$, respectively. The ash content in the dry weight of each compost was $25.0{\pm}6.2%$, $33.6{\pm}4.2%$, and $28.2{\pm}1.1%$, respectively. The deviation in the length of rice straw was affected by the linear velocity of the spinner in the compost mixer and the self-propelled turner, which were 21.5 m/sec and 9 m/sec, respectively. As a result, the most uniform mushroom compost was produced by the self-propelled turner.

Characteristics and Distribution of Microorganisms in a Rice Straw Compost for Cultivation of Button Mushrooms (Agaricus bisporus) (양송이 재배에서 볏짚 배지의 발효 단계별 관여 미생물의 분포양상 및 특성)

  • In this study, we analyzed the densities and taxonomic characteristics of various microorganisms that play important roles in Agaricus bisporus culture medium composting, and examined changes in the levels of decomposition-related enzymes secreted by these microorganisms. Various microorganisms such as thermophilic bacteria, actinomycetes, fluorescent Pseudomonas spp., and filamentous bacteria are closely associated with culture medium composts of Agaricus bisporus. The population densities of microorganisms change, and harmful bacteria disappear during thermophilic composting. Psychrobacter sp., Pseudomonas sp., Bacillus sp., and Pseudoxanthomonas sp. accounted for the highest proportion of bacteria in the culture media during outdoor composting, whereas Bacillus sp. and Psychrobacillus sp. were dominant after pasteurization. Cellulose and hemicellulose enzymes of the microorganisms were important at an early stage of rice straw composting and after decomposition of carbon sources, respectively. Microorganisms that secreted these enzymes were present in the second and third turning stage of composting.

Selection of Agaricus bitorquis hybrid strains based on RAPD analysis (RAPD분석을 통한 여름양송이(Agaricus bitorquis)의 교잡주 선발)

  • Agaricus bitorquis is an edible white mushroom of the genus that is cultivated at high temperature($25{\pm}1^{\circ}C$) unlike A. bisporus is cultivate at $16{\pm}2^{\circ}C$. Unlike Agaricus bisporus, an edible white A. bitorquis mushroom is cultivated at high temperature ($25{\pm}1^{\circ}C$). Most farmers cultivate this mushroom for a long cultivation period in Korea. For this reason, we made heterokayons to develop a new cultivar that generate fruitbodies for short cultivation period. Over one hundred SSIs(single spores isolates) were collected from selected A. bitorquis ASI1151 and ASI1349 strains. Seventy-three SSIs were germinated on CDA(compost dextrose agar) media after 20 days (minimum) or 83 days (maximum) incubation under different media condition. The mycelial growth rate of germinated SSIs was different. 9 homokaryons in ASI 1151 and 11 homokaryons in ASI 1349 from SSIs were selected by OPN-02 primer in RAPD analaysis. Also this primer was used to select heterokaryon that cross among each homokaryon with compatible locus. Therefore 44 compatible matings were confirmed of 99 crossed lines.

Good Agricultural Practices (GAP) analysis of hazardous materials in button mushroom (Agaricus bisporus) (GAP 양송이버섯 재배를 위한 위해물질 분석)

  • Good Agricultural Practices (GAP) is the world's leading quality certification for food safety. Since its introduction in Korea in 2006, its importance has been increasing every year. In particular, food safety issues are becoming increasingly important in society, and food safety is directly linked to health. The core of GAP certification is the traceability of the production, distribution, and consumption of hazardous materials, including pesticide residues, heavy metals, and microbes. In the present study, pesticides and heavy metals in button mushroom (Agaricus bisporus) and associated cultivation materials were analyzed. Tricyclozole (0.0144 ppm), flubendiamide (0.147 ppm), and trifloxystrobin (0.0340 ppm) were detected in rice straw and wheat straw, and carbendazim (0.0142 ppm) was detected in mixed wheat straw and rice straw medium. Lead and cadmium were detected at levels higher than the standard level in rice straw and mixed medium. However, lead and cadmium were not detected in mushrooms, and levels of arsenic and mercury were below the safety limit. Therefore, it was confirmed that the residual pesticides and heavy metals are safely managed in the investigated mushroom species. The results of the present study suggest that if these materials are adequately managed in the surroundings during cultivation, all hazardous materials can be managed during mushroom production.

Production of Mushroom Mycelium (Agaricus campestris) in Shaking Culture (진탕배양법(振?培養法)에 의한 양송이 균사체(菌絲體)의 생산(生産))

  • Conditions for submerged culture of Agaricus campestris var. bisporus and the chemical composition of its mycelium were investigated. In shaking culture with TGY basal medium at $27{\sim}30^{\circ}C$, pH tended to increase upon culture period, mycelial growth was the highest on 12th day, with relatively high nitrogen content of 7% and sugar in the medium disappeared almost at the end of culture period. As a nitrogen source, ammonium phosphate (dibasic) gave relatively high mycelial yield and the addition of yeast extract gave rise to better results. As a carbon source, glucose was the best, fructose, maltose, lactose and sucrose gave the same results, and soluble starch was utilized slightly. Mushroom mycelium contained 48% of protein, 8 free amino acids including arginine, histidine, lysine, isoleucine, leucine, phenylalanine, proline, tyrosine and its protein consisted of most essential amino acids, with relatively high contents of lysine and threonine. Therefore, mushroom mycelium deserves to be a high quality protein food.

Cultural conditions and morphological characteristics of Portobella, Agaricus brunescens (큰양송이 Portobella 균주의 배양조건 및 형태적 특성)

  • The compost dextrose(CD) medium was selected a an optimum liquid culture media for Portobella(Agaricus brunescens). The optimal conditions for the mycelial growth of Agaricus brunescens were 25 and pH 7.0. Sucrose and soluble starch were favorable to mycelial growth. Malt extract and ammonium nitrate appeared to be more favorable than the other kinds of nitrogen source. The optimum C/N ratio as about 10.When the collected Portobella trains were compared with growth characteristics, KME 49007 appeared the highest of $44.6kg/3.3m^2$.

  • This study was conducted to select the promising antifungal microoganisms for biological control of wet bubble, Mycogone perniciosa. Ninty one isolates of fungi, 342 isolates of bacteria, and 556 actinomycetes were isolated from mushroom composts and soils, were subjected to primary screening test on agar medium base for their antimicrobial spectra. Among them, 12 bacteria and 71 actinomycetes were selected. Among the antibiotic producing microoganisms, 5 cultures were selected on the basis their antibiotic activities on casing soil with Benlate nontolerence M. perniciosa. Finally, AJ-117, AJ-136 and AK-139 were selected as microoganism with antifungal activity against two strains of M. perniciosa.

Antagonistic Effects of the Bacterium Alcaligenes sp. HC12 on Browning Disease Caused by Pseudomonas agarici (버섯 세균성회색무늬병균(Pseudomonas agarici)에 대한 Alcaligenes sp. HC12의 항균활성)

  • A gram-negative bacterium was isolated from spent substrates of Agaricus bisporus and showed significant antagonistic activity against Pseudomonas agarici. The bacterium was identified as Alcaligenes sp. based on cultural, biochemical, physiological characteristics and a 16S rRNA sequence analysis. The isolate is saprophytic, but not parasitic or pathogenic on cultivated mushroom, whereas it showed strong inhibitory effects against P. agarici cells in vitro. The control efficacy of Alcaligenes sp. HC12 against brown blotch of P. agarici was up to 63% on Agaricus bisporus. The suppressive bacterium may be useful for the development of biocontrol systems.

Antagonistic Effect of Bacillus safensis HC42 on Brown Blotch Mushroom Disease Caused by Pseudomonas agarici (버섯 세균성회색무늬병균 (Pseudomonas agarici) 에 대한 항균활성을 가지는 Bacillus safensis HC42)

  • A gram-positive bacterium was isolated from the spent substrate of Agaricus bisporus that showed a marked antagonistic activity against Pseudomonas agarici. It was identified as Bacillus safensis HC42 based on its cultural, biochemical, and physiological characteristics, and 16S rRNA sequence. B. safensis HC42 was saprophytic, but not parasitic or pathogenic, in cultivated mushrooms and showed strong inhibition of P. agarici in vitro. Moreover, it showed a control efficacy of 66 % against browning disease caused by P. agarici in Agaricus bisporus. Therefore, B. safensis HC42 may be useful in the future for the development of a biocontrol system.

Study of the composting method using wheat straw on Agaricus bisporus cultivation (밀짚을 이용한 양송이 퇴비제조방법에 관한 연구)

  • The effect of wheat straw mix was tested to develop rice straw replacement media. The chemical contents were analysed after late fermentation. Nitrogen content and C/N ratio of the 20% wheat straw mixing medium were 2.2% and 16.7 and the only rice straw using medium were 2.0% and 17.9, respectively. When the 20% wheat straw mixing treatment was compared to the only rice straw medium, pinhead initiation period was shortened and the yield was increased as much as 30%. Therefore the 20% wheat straw mixing medium is expected to replace the rice straw medium.
