Lipopolysaccharide로 활성화된 마우스 대식세포에서 애엽(艾葉) 물추출물의 면역활성 연구 (Effect of Water Extract from Artemisiae Argi Folium on Mouse Macrophage Stimulated by LPS)

  • Objectives : The purpose of this study is to investigate the effect of Water Extract from Artemisiae Argi Folium (WAAF) on mouse macrophage Raw 264.7 cells stimulated by lipopolysaccharide (LPS). Methods : Cell viabilities were measured by MTT assay. And the intracellular productions of hydrogen peroxide (H2O2) were measured by dihydrorhodamine 123 assay. TNF-$\alpha$ and IL-6 production from Raw 264.7 were measured by ELISA method. Results : The results of the experiment are as follows. 1. WAAF significantly increased the cell viability compared to the control group (treated with LPS only) at the concentrations of 10, 50, 100, 200, 400 ug/mL. 2. WAAF significantly increased the intracellular production of H2O2 compared to the control group at the concentrations of 50, 100, 200 ug/mL. 3. WAAF significantly decreased the production of TNF-$\alpha$ compared to the control group at the concentrations of 100, 200 ug/mL. 4. WAAF significantly decreased the production of IL-6 compared to the control group at the concentrations of 50, 100, 200 ug/mL. Conclusions : WAAF could be supposed to have the immune-modulating activity related with the macrophage's immunoactivity.

강화산 애엽(艾葉)의 보관기간에 따른 정유성분의 변화 (Analysis of the content of essential oils from Ssajuari and Sajabalssuk according to storage period)

  • Objective : The content of essential oils from Ssajuari and Sajabalssuk was analyzed based on storage period. A total of eight samples old Ssajuari (harvested in 2000, 2001, 2002, 2003, and 2004), fresh Ssajuari (harvested in 2004), old Sajabalssuk (harvested in 2002), and fresh Sajabalssuk (harvested in 2004) was analyzed. Methods : The analysis of the content of essential oils from Ssajuari and Sajabalssuk was conducted by GC/MS. Results : The main essential oils of Ssajuari were ${\beta}$-thujone and 1.8-cineole. The ${\alpha},{\beta}$-thujone and 1,8-cineole were increased but the caryophyllene oxide was decreased. in accordance with storage period. The main essential oils of fresh Sajabalssuk were trans-${\beta}$-caryophyllene, terpinen-4-ol. The main essential oils of old Sajabalssuk was ${\beta}$-thujone and 1,8-cineole like Ssajuari. But 1,8-cineole was higher than that of Ssajuari samlpes. Conclusion : All samples had trans-sabinene hydrate, 1,8-cineole, terpinen-4-ol and caryophyllene oxide. According to stored year, ${\beta}$-thujone, ${\alpha}$-thujone were increased but terpinen-4-ol, caryophyllene oxide were decreased.

에탄올-유발 위점막손상에 대한 애엽추출물 (DA-9601)의 방어효과 및 기전에 관한 연구 (Studies on Protective Effect of DA-9601\ulcorner an Artimisiae Herba Extract, against Ethanol-induced Gastric Mucosal Damage and its Mechanism)

  • Protective effect of DA-9601, an extract of Artemisia Herb, against ethanol-induced gastric mucosal injury was evaluated in rats. In the prophylactic study, DA-9601 exhibited total protection (99.4%) against absolute ethanol-induced gastropathy, And the protective effect of DA-9601 lasted up to 2 hours, which was longer than those of other contemporary mucoprotectants. In the treatment study, DA-9601 significantly facilitated the healing of 70% ethanol-induced mucosal damage, which was superior to cetraxate, a commonly used anti-ulcer drug. The mechanisms of mucoprotection of DA-9601 were also assessed. DA-9601 increased the release of prostaglandin E$_2$ from murine neutrophils in a dose-dependent manner in vitro. The cytoprotective effect of DA-9601 against ethanol-induced mucosal damage was significantly diminished by the concommitant injection of N$\omega$-nitro-L-arginine methyl ester (L-NAME, 5 mg/kg, i.v.), a non-specific nitric oxide (NO) synthase inhibitor, while it was not affected by preinjection of indomethacin (5 mg/kg, s.c.), a prostaglandins-depletor. And it was found that DA-9601 significantly enhanced adaptive cytoprotective action of 10% ethanol against absolute ethanol (56.9$\pm$6.5 vs 23.0$\pm$3.3 mm$^2$, p<0.05, mean$\pm$SEM), though its exact underlying mechanism remains to be clarified. The present fin[lings demonstrate that DA-9601 exerts gastroprotecticv actions for the stomach against ethanol through several different underlying mechanisms, in which prostanglandins and NO are involved. In conclusion, the results obtained suggest that DA-9601 can be useful both in prevention and treatment of ethanol-induced gastric damage.

애엽(艾葉)의 미토콘드리아 보호 효과 (Cytoprotective Effects of Artemisia princeps Extract through Inhibition of Mitochondrial Dysfunction)

  • Objectives : Artemisia princeps is used as moxa in moxibustion and traditional herbal medicine. And its extracts or compounds is known to have an efficacy of antioxidant, anti-diabete, anti-cancer, anti-inflammation and neuroprotection. This study was performed to investigate the cytoprotective effect of Artemisia princeps extract (APE) against arachidonic acid (AA)+iron-induced oxidative stress on HepG2 cell. Methods : The effects of APE on cell viability has been assessed using MTT assay. And flow cytometric analysis was performed to estimate APE's effects on mitochondrial function. To investigate its underlying mechanism, related protein was analysed by using immunoblot analysis. Results : Treatment of APE increased relative cell viability, prevented a decline of B-cell lymphoma-extra large (Bcl-xL) and cleavage of poly(ADP-ribose) polymerase (PARP) and procaspase-3, and also protected mitochondrial membrane permeability (MMP) against oxidative stress induced by AA+iron. In addition, APE treatment increased phosphorylation of AMP-activated protein kinase (AMPK) exerts a cytoprotective effect. Conclusions : This results demonstrate that APE has an ability to activation of AMPK which protects cells from AA+iron-induced oxidative stress and restores MMP.

전탕 시간에 따른 애엽의 성분패턴 비교연구 (The Comparative Study on Compositional Pattern Analysis of Decoction of Extracted Artemisia argyi by Different Extraction Time)

  • Objectives: This study was conducted to find out the optimal extraction time for Artemisia argyi. Methods: The compositional pattern was compared with HPLC (High Performance Liquid Chromatography) and GC (Gas-Chromatography) by decocting Artemisia argyi 10, 60, 120 minutes respectively. Results: With longer extraction time, the contents of reference compounds were extracted 1.1 times more when 3,4-dicaffeoylquinic acid was extracted for 60 minutes than when extracted for 10 minutes in HPLC test, but the contents were reduced when extracted for 120 minutes compared to 60 minutes extraction time. 3,4-di-O-caffeoylquinic acid, 3,5-di-O-caffeoylquinic acid, 4,5-di-O-caffeoylquinic acid, jaceosidin, and eupatilin showed the largest yield rate when extracted for 10 minutes, and it decreased as time passed. The contents of chlorogenic acid, 3,5-dicaffeoylquinic acid, 4,5-dicaffeoylquinic acid, jaceosidin, scoparone, and eupatilin were detected only in 10 minutes extraction but not in 60 or 120 minutes extraction according to GC test. Conclusions: The results show that extraction time could affect the physicochemical characteristic or composition of Artemisia argy extracted. Thus, short extraction time could be useful for decoction of Artemisia argyi.

애엽(艾葉) 추출(抽出) 약침(藥鍼)이 허혈성(虛血性) 손상(損傷)에 미치는 효과(效果) (Effects of FOLIUM ARTEMISIAE ARGYI' Herbal Acupuncture on the Expression of nNOS protein Following Transient Forebrain Ischemic Injury in Rats)

  • Objectives : Acupuncture has been used to prevent and treat the cerebrovascular accident, such as a stroke, and many studies of acupuncture and moxibustion concerning to the stroke have been undertaken in the human and various animals. Also, herbal acupuncture, namely aqua acupuncture has been applied and developed to various diseases including the cerebrovascular accident. FOLIUM ARTEMISIAE ARGYI is the dry leaf of Artemisia argyi Levl. et Vant. collected in summer before the plant blooms and used to moxibustion and has been recommended for use as an analgesic and hemostatic. In this study, effects of FOLIUM ARTEMISIAE ARGYI (艾葉)' herbal acupuncture on the $LR_3$, namely Taechung on neuroprotection after the transient forebrain ischemia were investigated in Sprague-Dawely rats. Methods : Expressions of neuronal nitric oxide synthase (nNOS) protein in the hippocampus and cortex were observed at 2 hrs after transient forebrain ischemia by immunohistochemistry. Results : Expression of nNOS protein was increased in the hippocampus and cortex at 2 hrs after transient forebrain ischemia. However, pretreatment with FOLIUM ARTEMISIAE ARGYI' herbal acupuncture on $LR_3$ significantly decreased expression of nNOS protein protein compared to ischemia group. These features were observed in the motor cortex and the hippocampus. Conclusions : These results suggest that pretreatment with FOLIUM ARTEMISIAE ARGYI' herbal acupuncture on $LR_3$ inhibits the expression of nNOS protein induced by transient forebrain ischemia and may modulate excitatory toxicity of neuron related to neuronal cell death.

Acetaminophen 유도 간 손상에 대한 조릿대 애엽 추출물의 보호 효과 (Protective Effects of Sasa borealis Bamboo Browse Extract on Acetaminophen-induced Liver Damage in Mouse Model)

  • Acetaminophen (N-acety1-p-aminophenol, paracetamol) is widely used as an over-the-counter analgesic and antipyretic drug. Intake of a over dose of acetaminophen may result in severe hepatic necrosis. In this study, we investigated the liver damage in mice using single dose (300 mg/kg) of acetaminophen and the possible protective effects of administration (50-200 mg/kg body weight) of SB-Ex on acetaminophen-induced liver damage in mice. The alanine aminotransferase (ALT), and aspartate aminotransferase (AST) activities were determined in the plasma of mice. The effect of SB-Ex on lipid peroxidation product thiobarbituric reacting substances (TBARS) and some antioxidant enzymes superoxide dismutase (SOD), catalase, d-aminolevulinate dehydratase (${\sigma}$-ALA-D) activities, and gluthathione peroxidase (GPx), were also evaluated in the mouse liver homogenate. Acetaminophen caused liver damage as evident by statistically significant increased in plasma activities of AST and ALT. There were general statistically significant losses in the activities of SOD, catalase, ${\sigma}$-ALA-D, and GPx and an increase in TBARS in the liver of acetaminophen-treated group compared with the control group. However, SB-Ex was able to counteract these effects. These results suggest that SB-Ex can act as hepatoprotectives against acetaminophen toxicity and is a good candidate for further evaluation as an effective chemotherapeutic agent.

애엽(艾葉) 발효 추출물의 면역활성에 관한 연구 (Studies on the Immuno Modulating Acitivity of Fermented Artemisiae Argyi Folium Extract)

  • Objectives : This research aimed to study the cytotoxicity and immuno modulating activity of fermented Artemisia argyi Lev. et Vant.(Compositae). Methods : Effect of fermented Artemisiae Argyi Folium extracts, which were fermented by Sacchromyces cerevisiae STV89(AFS), on cell viability, generation of ROS within cells, generation of NO and the level of cytokines($TNF-{\alpha}$ and IL-6) was measured using mouse macrophage RAW 264.7 cell. Results : 1. Result of MTT assay conducted to verify the cytotoxicity of fermented Artemisiae argyi folium extract illustrated that, when fermented Artemisiae argyi folium extract was processed for each concentration, there was no excessive induction of cytoxicity in the RAW 264.7 cell. 2. Fermented Artemisiae Argyi Folium extract increased the generation of H2O2 within RAW 264.7 cell as well as significantly increased inhibition of generation of H2O2 in macrophage induced by LPS. 3. Fermented Artemisiae Argyi Folium extract inhibited generation of NO in RAW 264.7 cell, and significantly inhibited increase in generation of NO of macrophage induced by LPS. 4. Fermented Artemisiae Argyi Folium extract, AFS has significantly reduced the increase in the generation of $TNF-{\alpha}$ above 10 ${\mu}g/mL$. 5. Fermented Artemisiae Argyi Folium extract, AFS has significantly reduced the increase in generation of IL-6 above 50 ${\mu}g/mL$. Conclusions : AFS fermented extract produced from Artemisiae Argyi Flium, have increased generation of ROS and reduced generation of NO in RAW 264.7 cell without excessively inducing cytotoxicity of RAW 264.7 cell. In addition, they displayed significant immuno modulating activities including inhibition of generation of $TNF-{\alpha}$ and IL-6 in macrophage, induced by LPS.

한약재 보관중 발생하는 해충류 (Insect Pests Occurring in Storage Medicinal Plants)

  • 국내 유통 중인 한약재 대부분을 수입에 의존하고 있으나, 관리시설이 빈약하여 해충들에게 쉽게 노출될 수 있는 환경에 놓여 있다. 이 연구는 총 67과 134속 158종의 저장 한약재들을 대상으로 이들에서 발생하고 있는 저장물해충들을 조사하였다. 그 결과, Cassia 속의 육계, Juniperus 속의 자단향 그리고 Artemisia 속 애엽을 비롯한 44종의 한약재들에서는 곤충의 발생을 확인할 수 없었다. 한약재들에서 나타난 해충들의 발생빈도는 쌀머리대장 (A. advena) 12.3%, 권연벌레 (L. serricorne) 11.5%, 머리대장가는납작벌레 (O. surinamensis) 10.3%, 거짓쌀도둑거저리 (T. castaneum) 9.4%, 인삼벌레 (S. paniceum) 8.4% 그리고 화랑곡나방 (P. interpunctella) 7.9% 순이었다. 또한 이들 곤충들은 저장 중인 한약재들 중에서도 뿌리 또는 뿌리줄기 형태의 한약재들을 가장 선호한 것으로 보였다. 이 연구는 천연물 연구자들에게 곤충들이 발생하지 않았던 한약재들에 대한 기본 정보를 제공해줌으로써 향후 이들이 살충활성 또는 훈증활성물질을 탐색하는데 있어 이들 한약재들을 우선적으로 검토할 수 있는 유용한 자료로서 활용될 수 있을 것이다.

애엽(艾葉)의 RBL-2H3 비만세포 탈과립, cytokine 생산 및 $Fc{\varepsilon}RI$ 유전자 발현에 미치는 영향 (Inhibitory Effect of Artemisiae Asiaticae Herba on Degranulation, Production of Cytokine, and $Fc{\varepsilon}RI$ Expression in RBL-2H3 Cells)

  • Artemisiae Asiaticae Herba (AAH) has been used to remedy of symptoms such as bleeding, dysmenorrhea, eczema and itchy skin in Oriental Medicine. In this study, we investigated the protective effect of AAH on allergic response. The effect of AAH was analyzed by ELISA and RT-PCR in RBL-2H3 cells. We investigated cell viability, ${\beta}$-hexosaminidase and histamine as markers of degranulation, production of IL-4 and TNF-${\alpha}$, and gene expression of HDC2, cytokines and $Fc{\varepsilon}RI$ ${\alpha}{\beta}{\gamma}$ subunit. We found that AAH suppressed ${\beta}$-hexosaminidase and histamine release, the production of IL-4 and TNF-${\alpha}$ in RBL-2H3 by the anti-DNP IgE plus DNP-HSA stimulation. AAH also significantly decreased cytokine mRNA expressions, such as IL-$1{\beta}$, IL-2, IL-3, IL-4, IL-5, IL-6, IL-12, IL-13, TNF-${\alpha}$, and GM-CSF, and increased cytokine mRNA expressions of IL-10 in RBL-2H3. In addition, AAH suppressed mRNA expression of $Fc{\varepsilon}RI$ ${\alpha}{\beta}{\gamma}$ subunit on cell surface. Our results indicate that AAH protects against allergic response and exerts an anti-inflammatory effect through the inhibition of degranulation and production of cytokines and expression of $Fc{\varepsilon}RI$ ${\alpha}{\beta}{\gamma}$ subunit.
