Effects of Meju Manufacturing Periods on the Fermentation Characteristics of Kanjang, Korean Traditional Soy Sauce (메주의 제조기간에 따른 재래간장의 발효특성)

  • This study was carried out to clarify the microorganisms which participated in the fermentation of kanjang. The changes in the viable cell counts of total aerobic bacteria, lactic acid bacteria and yeasts for raw soybean, soybean during cooking, meju during cultivation, and kanjang mash during maturing were investigated along with the changes in components during those periods. Lactic acid bacteria that were found to be $6{\times}10^2\;CFU/g$ in raw soybean were disappeared after cooking process, but total aerobic bacteria were diminished from $1.9{\times}10^6\;CFU/g$ to $10^2\;CFU/g$. Aerobic bacteria of inner and outer parts of meju increased to more than $10^9\;CFU/g$. The higher viable cell counts of lactic acid bacteria in the inner parts of meju were observed than those in outer ones. On the contrary, significantly higher viable cell counts of yeasts in the outer parts of meju were found. Total nitrogen content and color density of kanjang increased by using meju with extended cultivation periods. No significant differences were observed in microbial counts between kanjang mash with aeration and non-aeration during kanajng mash maturing.

Two-step High Temperature Pretreatment Process for Bioethanol Production from Rape Stems (유채대의 이단 고온 처리에 의한 알콜 발효용 당화물 생산)

  • Two-step pretreatment process was investigated to efficiently hydrolyzed rape stems for obtaining fermentable sugars. The process was consisted of two consecutive steps as $200^{\circ}C$ and 15 MPa and $374^{\circ}C$ and 24 MPa with the flow rate of 5 mL/min. Under this condition, 5.5 (g/L) of glucose and 25.6 (g/L) of xylose were obtained from rape stems, showing 18% of glucose yield based on 25% cellulose in the rape stems. It was also found that this process could generate less amounts of toxic residues, such as HMF (Hydroxy- Methyl-Furfural) and other fulfural components during hydrolysis process. It could reaction maintain relatively high ethanol production yield as 90% of theoretical conversion yield from glucose. Therefore, this pretreatment process could be applied to hydrolyze other cellulosic and marine resources such as woods, stem and algae for bioethanol production.

홍국주의 제조와 품질특성에 관한 연구

  • 최근 홍국색소는 천연계착색료로서 뿐만 아니라 발효식품, 발효조미료등에도 이용되어 기호에 맞는 새로운 홍국 개발도 적극적으로 진행되고 있어 인지도가 높아지고 있다. 혈청콜레스테롤저하작용, 혈압강하작용, 혈당강하작용, 항산화작용등 여러가지 약리효능을 지닌 홍국균을 이용해 재래적 방법에 근거하여 홍국주를 제조하여 그 제조공정을 확립하고 품질특성을 확인하였다. Monascus purpureus를 액체배양법에 의해서 홍국코지를 제조하고 코지의 역가는 $\alpha$-amylase와 $\beta$-amylase 활성은 400unit와 4.44unit를 나타났으며, acid protease 활성은 80unit, neutral protease 활성은 91.4unit로 나타났다. 홍국코지에 증자된 쌀과 물을 혼합하여 발효경과를 측정하고, 발효가 끝난 홍국주의 DPPH법에 의한 전자공여능, 아질산염 소거능, Total phenolic compound함량, ACE저해작용등을 측정하였다. 홍국주의 발효경과 분석은 알콜도수는 2차 담금 후 2일째-13.2도, 4일-13.5, 6일째-13.5, 8일째-13.5, 10일째-13.4로 나타났다. 환원당은 2일째-23.26, 4일-20.68, 6일째-21.74, 8일째-19.93, 10일째-14.23 $^{\circ}$Brix로 점점 감소한다는 것을 볼 수 있으며, 색도에서 적색을 나타태는 Hunter value a값은 2일째 -14.72, 4일-14.97, 6일째-15.70, 8일째-16.43, 10일째-17.29로 시간이 지날수록 붉은 색소가 증가한다는 것을 알 수 있었다. pH는 2일째-4.52, 4일-4.02, 6일째-4.11, 8일째-13.11, 10일째-13.18로 pH가 약간 낮아진 것을 볼 수 있다. 산가는 2일째-8.4, 4일-6.7, 6일째-7.0, 8일째-7.5, 10일째-11.5로 pH가 산성화 될 수록 산가는 증가하는 것을 볼 수 있다. 홍국주의 전자 공여능에 의한 항산화력은 25.6%, 아질산염 소거능은 27.6%, Total phenolic compound 함량은 12.34mg%, ACE저해작용은 38%의 항산화력을 나타냈다.

  • Experiments of alcohol fermentation of the yeast,K. fragi1is CBS 1555 were performed to obtain the following results. In these experiments, the initial concentrations of sugar which was composed of inulin and fructose as weight ratio of one to one were 30, 50, 75, 100 and 150g/l and the initial densities of the microorganism were less than 0.5g/l, 10g/1 to 15g/1, and 50g/l. The functional relationship among specific growthrates, sugar concentrations, and alcohol concentrations could be expressed by Aiba-shoda equation and the specific growth rate represented the trend that decreased with increase in the initial concentration of the microorganism. Also, $\mu$max and Ks of Monod's equation could be expressed as the function of initial cell concentration like the following equations. $\mu$max=0.8-0.008X Ks=0.54X+8 In the region that sugar, alcohol and cell concentrations were 10g/1 to 120g/l, 0g/l to 60g/l and 0.5g/l to 50g/l respectively, the differences between the experimental values and the calculated ones for specific growth rate approached to 40% with respect to experimental values at the worst cases, but in most cases, those were distributed in the range of less than 20%.

  • This study was conducted to improve the Prunus mume vinegar production. The most suitable concentration of the Prunus mume juice was 6%. Static fermentation was a more suitable process for acetic acid fermentation of the Prunus mume vinegar than shaking fermentation. Major components of the organic acids were acetic, citric, tartaric and malic acid at 4.2, 1.2, 0.3, and 0.1%, respectively. Also, major components of the free sugars were glucose and fructose, and 80.96 mg% of asparagine was included in the Prunus mume vinegar as a main free amino acid. Alcohol components of the Prunus mume vinegar were n-propyl alcohol, iso-butyl alcohol, n-butyl alcohol, iso-amyl alcohol, and n-amyl alcohol.

The Changes of Microflora During the Fermentation of Takju and Yakju (약.탁주 발효과정 중 미생물 균총의 변화)

  • Korean traditional rice wine Takju and Yakju were manufactured using 2-step-brewing method. To investigate microflora involved in fermentation step, number of microorganisms, pH, titratable acidity, and alcohol contents of Takju and Yakju were measured. In Takju and Yakju, although not significantly, $1.1{\times}10^{8}$ and $2.0{\times}10^{6}\;CFU/mL$ lactic acid bacteria at initial stage of second fermentation decreased to $8.3{\times}10^{6}\;and\;1.0{\times}10^{4}\;CFU/mL$ at the end of second fermentation, respectively. For Takju, micrococci and yeast occupied 80 and 20% at initial stage of second fermentation, whereas bacteria and yeast occupied 35 and 65% at the end of second fermentation, respectively. Yeast occupied 88% throughout the second fermentation of Yakju. The main yeast isolated from both Takju and Yakju was identified as Saccharomyces cerevisiae using API 20C AUX kit. The yeast strain Candida magnoliae was also detected during fermentation of Takju and Yakju.

Analysis of Volatile Components in Korean Rice Wine by the Addition of Yulmoo (탁주 제조 시 율무 첨가에 따른 휘발성 성분의 분석)

  • In order to use nutraceutical components in Yulmoo (Coix lachryma-jobi L. var. ma-yuen Stapf), the acceptability of Yulmoo for Takju (Korean rice wine) was examined in terms of the production of volatile components. When Yulmoo was added to the Takju made by commercial koji as a substitute of starch at 0 (rice 100%), 15, 30, 60, and 100%, the ethanol content slightly increased to 13.6, 15.2, 15.2, 14.1, and 13.8%, the Production of isobutanol and iso-imyl alcohol increased as the amount of Yulmoo increased. While the use of Yulmoo to the Takju as the nuruk (Korean traditional koji), inoculated with Aspergillus kawachii (AK) and Rhizopu japonicus (RJ), decreased the contents of n-propanol, iso-butanol, and iso-amyl alcohol as well as the contents of ethanol from 14.3% to 11.2 [AK], 7.5 [RJ], and 10.4% [AK + RJ]. The Yulmoo nuruk in Takju increased the production of acetaldehyde and ethyl acetate indicating the negative effect in the organoleptic evaluation. It was determined that the addition of Yulmoo as a starch source or as nuruk has a critical effect on the production of fusel oils and other volatile compounds in Takju fermentation.

Optimization for Alcohol Fermentation by Kluyveromyces marxianus using Jerusalem Artichoke Powder (돼지감자 분말을 이용한 Kluyveromyces marxianus의 알콜올 발효)

  • In order to produce alcohol for the alternative energy from dried powder of Jerusalem artichoke was investigated with Kluyveromyces marxianus UCD(FST)55-82, which was reported to assimilate inulin. The optimal condition for the production of ethanol by K. marxianus was elucidated to be incubation temperature of $30^{\circ}C$, initial pH 5.44, agitation of 100 rpm, 1,000 ml of medium in a 2.5l-vessel, anaerobic state, and inoculation of 2.5%(v/v). Addition of antifoam A concentrate(si1icon polymer) of 0.01% and urea of 0.1% increased the concentration of ethanol effectively. The optimized condition showed ethanol concentration of 6.8%(v/v) in Jerusalem artichoke liquid medium, production yield of 91.91% and productivity of 2.71 g/l/hr during the first day and 0.71g ethanol/l/hr during four days of incubation.

Fermentation of wheat bran through lactic acid bacteria: Changes in flavor components and free amino acids and potential applications in baking (밀기울의 유산균 발효: 향기성분 및 유리아미노산 변화를 통한 제빵 소재로서의 가능성)

  • The aim of this study was to enhance the use of wheat bran in lactic acid bacteria (LAB) fermentation. LAB fermentation of wheat bran and the flavor components and amino acids of fermentation products were analyzed using gas chromatography-mass spectrometry (GC/MS) and high-performance liquid chromatography (HPLC). The results showed that total flavor components increased by 93% and 73% in the animal-based LAB mixture (T2) and plant-based LAB mixture (T3), respectively, after fermentation. Among these components, 2,3-butanedione (diacetyl), known for its buttery flavor, was detected at concentrations of 18.44 ng/g (T2) and 16.95 ng/g (T3). Levels of hexanal and nonanal, which causes off-flavor components in wheat bran, dramatically decreased after T2 fermentation; similarly, levels of total free amino acids decreased by 37.6% (T2) and 36.7% (T3) after fermentation. This may explain why some components were bound to volatile compounds during LAB fermentation. These results suggest that LAB-fermented wheat bran is a potential value-added food material.

The Chemical Characteristics of Korean Red Wine with Different Grape Varieties (포도품종을 달리한 적포도주의 이화학적 성분변화 (I))

  • Five kinds of red wine from three different grape varieties, Gerbong (G), Campbell (C), Moru (M), Gerbong+Moru (70:30, GM) and Gerbong+Campbell (70:30, GC), were prepared. Their chemical and microbiological changes were monitored during alcohol fermentation and aging. The changes of pH, brix and total acidity were $3.2{\sim}3.6,\;17.9{\sim}6.0$ and $2.4{\sim}4.6\;g/L$, respectively. The viable cell numbers of initial, stationary and death phases were $6.0{\times}10^6,\;1.0{\times}10^8$ and $7.0{\times}10^5\;cfu/mL$ during alcohol fermentation. The sugar fermentability, glucose and fructose contents were greatly decreased less than 0.2 g/L, and the final contents of ethanol and sulfur dioxide were $11.4{\sim}12.3%$ and $40{\sim}62\;mg/L$, respectively. The conversion ratios of malic acid to lactic acid were 23% (G), 67% (M), 28% (C), 33% (GM) and 39% (GC). The chemical characteristics of five red wine were significantly different in pH, total acidity, sulfur dioxide and lactic acid contents (p<0.05).
