Studies on the Immunomodulating Effects of Polysaccharide Extracted from Ganoderma lucidum on Macrophage (영지버섯 다당체의 마우스 대식세포 면역증강 효과)

  • The immunomodulating effects of polysaccharide extracted from Ganoderma lucidum(PSG) on macrophage were evaluated using murine macrophage cell line ATCC TIB 71 cells or peritoneal exudate cells of BALB/c mice. The cell were incubated with various content of PSG for 24 hours at 0.5% $CO_2$ incubator under varying experimental conditions. PSG stimulated the non-specific activites of macrophage such as mitotic activity and expression of surface interleukin-2 receptors by dose-dependent pattern with statistic significance(p<0.001): however, PSG had little immunoregulatory effects on cytokines derived from peritoneal macrophages of BALB/c mice. There were no significant changes in the se-cretion of interleukin-6, interleukin-6, or tumor necrosis factors(Tn) of PSG treated cells compared to the control group. But PSG increased secretion of cytokines(IL-1 and TNF) when the cells were primed and trigged with BCG and IFN-${\gamma}$.

The Synergistic Action of the AL-Protease from Arthrobacter luteus on the Lysis of Yeast Cell Walls (Arthrobacter luteus가 생산하는 AL-Protease의 효모세포벽 용해 촉진작용)

  • The yeast cell wall lytic action of the alkaline AL-protease, which was found out of the crude Zymolyase that a kind of yeast cell wall lytic $endo-{\beta}-1$, 3-glucanase produced from Arthrobacter luteus, was investigated with the viable cells of S. sake and it's cell wall preparation. AL-protease on the lysis of the viable yeast cells showed very low activities with the alone, but the lytic activities were highly increased with the combination of AL-protease and Zymolyase. On the stepwise treatment of the viable yeast cells with AL-protease and Zymolyase, the cells were lysed highly only by the course having a treatment with Zymolyase after pretreatment with AL-protease. Thus synergistic action of AL-protease was not observed with any some commercial enzymes, known as a type of alkaline and serine protease such as AL-protease, and was also found to be affected greatly by the culture conditions and species of the yeast tested. AL-protease caused the release of some peptide and a lot of sugar from the cell wall preparation, but could not lysed the cell wall more than 66%. Whereas Zymolyase could lysed the cell walls almost completely with alone. On the basis of these results, the synergistic action of AL-protease on the lysis of S. sake cells is hypothesized that at first AL-protease bind to the yeast cell surface layer consisting of mannan and protein, and then changes their conformation to facilitate the penetration of Zymolyase from the outside to the inside framework layer constituted of alkali insoluble ${\beta}-1,\;3-glucan$.

  • In order to investigate harmful trace elements in ford additives used in food processing the contents of Copper, Lead, Cadmium, Chromium, Manganese, Nickel and Iron are determined by Atomic Absorption Spectrophotometer. These Elements are Extracted with APDC-MIBK extract method. Samples are collected from the food Industrial companies in Korea. Samples studied as shown in Table 1 are food colors(3-samples, Flavoring Agents(7-samples), Acidifiers(4-samples) Baking Powders(4-samples), Emulsifiers(2-samples), Thickeners(2-samples), Antioxidants(2-samples), Intensifiers(2-samples), Seasonings(3-samples), modifiers (10-samples) Most of the food additives contained comparatively a little small amount of harmful trace elements. However, the contents of harmful trace elements of Sodium Carbonate, Food Yellow No.5, Food Blue No.1, Food Red No.2 and Alkalies added in noodles are significantly higher than the other food additives. The obtained results are as follows ; 1) Sodium Carbonate contained Cadmium; 16.73 ug/g and Lead ; 61.55 ug/g. 2) Food Yellow No. 5 contained Cadmium: 1.67ug/g and Lead; 23.46 ug/g. 3) Food Blue No. 1 contained Cadmium; 1. 16 ug/g and Lead; 23.46 ug/g. 4) Food Red Ho. 1 contained Cadmium; 1.91 ug/g and Lead 23.08 ug./g. 5) Alkalies added in noodles contained Cadmium; 6. 11 ug/g and Lead; 53.85 ug/g.

  • Uljin cassiterite deposit had been known to be a pegmatitic origin derived from the Wangpiri (Buncheon) granitic gneiss of Precambrian period. Lithium ore also shows the same origin and its lithium bearing mineral was ascertained to be a taeniolite. But the presence of leucocratic granites which played the role of host rocks haven't been clearly designated yet in these provinces. Even though Bonghwa and Youngweol sericite deposits situated in the vicinities of Hambaeg syncline had been known to have their host rocks as Hongjesa Granites of Precambrian period and Pegmatitic migmatite of unknown age respectively. But younger leucocratic granites are characterized by more amounts of albite and sericite (muscovite-3T type) than those of the older granitic rocks which contain plenty of biotite and chlorites. Although the younger granites show rather higher contents of alkalies such as $Na_2O$ (0.13~8.03 wt%) and $K_2O$ (1.71~6.38 wt%), but CaO (0.05~1.21 wt%) is very deficient due to the albitization and greisenization. Manisan granite, which is assumed to be Daebo granite which intruded the Gyunggi Gneiss Complex was again intruded by leucocratic granite whose microclinized part changed into kaolins. Taebaegsan region shows a wide distribution of carbonate rocks which are especially favorable to the ore depositions. And the presence of alkali granites which formed in the later magmatic evolution are well known to be worthwhile to the prospections of various rare metals and REEs resources.

  • 해양환경 중 많이 사용되는 철강재료들은 그 가혹한 부식환경에 대응하기 위하여 일반적으로 피복 도장방식법이나 음극방식법이 적용되고 있다. 여기서 음극방식법은 선박 및 해양구조물의 해중부 부식에 대해 가장 효과적인 방식법으로 알려져 있다. 한편, 이와 같이 해수 중 철강재에 음극방식을 적용할 경우, 피방식체인 그 강재 표면에 해수 중 용존된 산소의 음극환원 반응이 일어나며 국부적인 알카리 표면 조건을 형성시켜 $Mg(OH)_2$$CaCO_3$의 막을 석출시킨다. 이와같이 음극방식 중 형성된 전착물은 방식해야 될 표면적을 감소시켜 방식전류밀도를 감소시키는 효과가 있는 것으로 보고되고 있다. 이렇게 석출된 전착물은 음극표면에 부분적으로 형성되고, 여러 가지 환경 조건 등의 영향을 받아 그 피막의 형성 정도도 가늠하기 어렵기 때문에 음극방식 설계 시 그 정도에 따른 영향을 고려-반영하기가 곤란하다. 또한 이 전착물은 그 형성 메커니즘에 관한 해석이나 강도, 균일한 밀착성, 장기적인 방식효과 및 효율성 등이 아직 충분히 입증되어 있지 않은 실정에 있다. 따라서 본 연구에서는 해수 중 다양한 전착 프로세스에 의해 제작된 전착물의 기간별, 도장코팅 종류별 특성변화를 분석 및 평가하고, 전착물에 의한 희생양극 소모전류 변화 측정 분석을 통해 전착막을 균일하고 치밀하게 형성시키기 위한 최적의 조건을 찾고자 하였다. 또한 석출속도, 밀착성 및 내식특성을 향상시키기 위해 해수 중 기체를 용해시켜 제작한 막의 특성을 분석-평가하였다. 본 연구에 사용된 강 기판은 일반구조용강(KS D 3503, SS400)으로 ${\varnothing}42.7{\times}1,000mm{\times}4.0t$의 형상으로 제작하였다. 인가된 전류밀도는 1, 3 및 $5A/m^2$이고 도장 코팅 종류별 전착 석출물의 형성차이 비교 분석을 위한 실험은 선박 및 해양구조물에 많이 사용되는 Universal Epoxy 도료 2종을 선정하여 진행하였다. 또한 Steel Wire Mesh의 영향을 알아보기 위해 Mesh를 설치하여 실험을 진행하였다. 기간별-도장 종류별 외관관찰, 전착물의 두께 측정, SEM, EDS 및 XRD를 통해 막의 모폴로지, 조성원소 및 결정구조를 분석하였으며, 전착물의 내식성과 내구성을 평가하기 위해 테이핑 테스트(Taping Test) 및 전기화학적 양극분극 시험을 실시하였다. 희생양극 소모율에 대한 전착물의 영향을 확인하기 위해 외부전원을 인가하여 전착 피막을 형성시킨 강 기판에 희생양극을 연결하여 희생양극 소모효율 측정 시험을 진행하였다. 전착물의 석출량은 시간 및 전류밀도의 증가에 따라 비례하여 증가하였으며, 음극전류 인가 시 금속과 용액 계면 사이의 확산층에서 발생한 $OH^-$ 이온으로 인해 금속과 용액 계면 사이 pH가 부분적으로 증가하여 $Mg(OH)_2$ 화합물이 많이 생성되는 것으로 확인되었다. 또한 Mesh의 부착으로 평활하지 않게 형성된 미세한 굴곡구조 및 표면적 증가로 인하여 단계적으로 피복되는데 필요한 시간이 지연되면서 $CaCO_3$에 비해 $Mg(OH)_2$ 화합물이 상대적으로 증가한 것으로 사료된다. $CaCO_3$(Aragonite) 구조는 견고한 피막으로 치밀하고 화학적 친화력이 높아 우수한 밀착성을 보였으며 전착물의 영향으로 양극 전류가 감소하였고, 이로인해 방식전류 절감효과를 얻을 수 있을 것으로 기대된다.

A Study on the Effect of Electrolyte Additives on Zn Electrode with Pb3O4 in Zn-AgO Secondary Battery System (Zn-AgO 이차 전지에서 Pb3O4가 첨가된 아연 전극에 미치는 전해질 첨가제의 영향에 관한 연구)

  • Zn electrode was widely used as an anode material in alkaline battery systems in highly concentrated KOH electrolyte, however it was well known that its cycle life is significantly shortened by growth of dendrite due to the high dissolution of $Zn(OH)_2$ and rapid electrochemical reaction. In this study when by the additives such as $Ca(OH)_2$, Citrate, tartrate and Gluconate were added to $40\%$ KOH electrolyte at solution temperature of $25^{\circ}C$ and the amount of $5wt\%\;Pb_3O_4$ was mixed to Zn electrode and then the effect of $Pb_3O_4$ and additives on the electrochemical behavior of Zn electrode was investigated by Potentiodynamic Polarization Curves, Cyclic Voltammetry, Accelerated Life Cycle lest, and SEM image analyses. The addition of $Pb_3O_4$ reduced the corrosion rate of Zn electrode. The corrosion potential of Zn electrode with $Pb_3O_4$ was higher or lower than that of pure Zn electrode however was not influenced practically to the open circuit voltage. And the addition of 4 type additives had an important role in improving both cycle life in accelerated cycle life test and corrosion resistance. Furthermore the additive of Tartrate indicated comparatively a good effect to corrosion resistance as well as charging-discharging property Improvement among those four type additives.

Characteristics of Whey Protein (WPC-30) Hydrolysate from Cheese Whey (치즈유청으로부터 제조한 유청단백질 가수분해물의 특성에 관한 연구)

  • Whey protein concentrate (WPC) is widely used to increase the nutritional and functional properties of food. In this study, the physiochemical and functionality of WPC-30 hydrolysates were examined to evaluate the possibility of application in the food industry. The WPC-30 was manufactured using ultrafiltration and spray-drying, and then hydrolyzed with proteolytic enzyme including alcalase, flavourzyme, nuetrase and protamex. Enzymatic hydrolysis had a significant influence on the physicochemical properties as evident from the increased foaming capacity, solubility. Alcalase caused highest protein hydrolysis (3.26%) and the bitterness. Foaming capacity was largest in WPC-30 hydrolysate treated with flavourzyme. Protein solubility at various levels of pH was highest in protamex-treated WPC-30 hydrolysate. However, the solubility of WPC-30 hydrolysates was significantly improved in alkaline condition than in acidic and neutral conditions. The study revealed that spray dried enzyme modified WPC can be used in various functional food.

Change of Solubilization Characteristics of Rice Straw by Different Pre-treatments (전처리 방법에 따른 볏짚의 가용화 특성 변화)

  • In order to find a feasibility of applying an agricultural biomass to the anaerobic digestion system, the effects of pre-treatment of rice straw was investigated by different sizes,temperatures, ultrasonic frequencies, and with/without NaOH treatment on the change of soluble organic matter, which is generated most dominantly in Korea. It was observed that SCOD(soluble chemical oxygen demand) as the index for the decomposition of rice straw increased with the smaller particle size, higher reaction temperature and alkali treatment. With treatment of 5% NaOH, it was shown that the highest concentrations of SCOD were observed at 9,000 mg $L^{-1}$ and 6,000mg $L^{-1}$ at $35^{\circ}C$ and $55^{\circ}C$, respectively. Agitating with ultrasonic irradiation could be enhanced SCOD of rice straw less than 3 cm with 40 kHz of ultrasound. Optimal RPM in this study was at 150 rpm regardless of reaction temperatures.

Isolation and Purification of Fibrinolytic Enzyme of Edible Mushroom, Sarcodon aspratus(Berk.)S. Ito (능이버섯으로부터 Fibrin 분해활성이 있는 단백질의 분리 및 정제)

  • To isolate and purify fibrinolytic active substance from Sarcodon aspratus(N $H_4$)$_2$S $O_4$ precipitation, DE52 anion exchange column chromatography, Sephacryl-S 200gel filtration chromatography and Mono S cation FPLC were carried out and the characterizations of the purified enzyme were investigated. The bound active fraction on DE52 anion exchange column chromatography were eluted with 0.2 M NaCI and the fibrionlytic enzyme was purified after following Sephacryl-S200 gel fitration chromatography and Mono S cation EPLC. The specific activity of purified enzyme was 55.2 U/mg protein and increased 11.3 fold comparing crude extract and the yield was 49.5%. 12% SDS-PAGE electrophoresis and gel filtration chromatography revealed that Sarcodon aspratus fibrionloytic enzyme was highly purified and had 29.300 Da molecular weight. Enzyme activity of the purified fibrinolytic enzyme from Sarcodon aspratus was increased on higher pH and was stable until pH 10.5. On temperature dependent stability, the enzyme activity was decrease sharply but remained 25% relative activity on 8$0^{\circ}C$. This enzyme activity was inhibited by heavy metal ion, C $U^{2+}$ and $Co^{3+}$ with 68% and 38%, respectively. And also, the enzyme activity was inhibited with $Ca^{2+}$ chelator EDTA and serine protease inhibitor PMSF. These results from this study suggested that the fibrinolycit enzyme from Sarcodon aspratus is a serine protease and the enzyme activity was increased by $Ca^{2+}$ or $Mg^{2+}$ ion.n.ion.n.

Development of a continuous electrolytic system with an ion exchange membrane for pH-control with only one discharge of electrolytic solution and its characteristics (단일 전해액 배출만을 가지는 pH조절용 연속식 이온 교환막 전해 시스템의 개발과 그 특성)

  • In order to produce only a pH-controlled solution without discharging any unwanted solution, this work has developed a continuous electrolytic system with a pH-adjustment reservoir being placed before an ion exchange membrane-equipped electrolyzer, where as a target solution was fed into the pH-adjustment reservoir, some portion of the solution in the pH-adjustment reservoir was circulated through the cathodic or anodic chamber of the electrolyzer depending on the type of the ion exchange membrane used, and some other portion of the solution in the pH-adjustment reservoir was discharged from the electrolytic system through the other counter chamber with its pH being controlled. The internal circulation of the pH-adjustment reservoir solution through the anodic chamber in the case of using a cation exchange membrane and that through the cathodic chamber in the case of using an anion exchange membrane could make the solution discharged from the other counter chamber effectively acidic and basic, respectively. The phenomena of the pH being controlled in the system could be explained by the electro-migration of the ion species in the solution through the ion exchange membrane under a cell potential difference between anode and cathode and its consequently-occurring non-charge equilibriums and electrolytic water- split reactions in the anodic and cathodic chambers.
