초석잠(Stachys sieboldii MIQ.) 줄기와 뿌리 추출물의 특성분석과 아질산염 소거능에 대한 고찰

  • This study was to search antioxidant constituents of ethyl acetate extract from Stachys siebodlii MIQ. The test of nitrite scavenging abilities were performed on the extracts of methanol, hexane, chloroform, ethyl acetate, butanol, and water, Ethyl acetate extract, The most promising one was fractionated on a silical gel column using elution solvent(chloroform:methanol:water=70:30:5 lower phase) at a flow rate 1.0ml/min. UV-VIS spectral data of each fraction showed adsorption maxima in the range of $284{\sim}330nm$ which is the characteristic range of $210{\sim}290nm$ and $300{\sim}550nm$ phenolic compounds. These results suggested that Stachys siebodlii MIQ. shows natural antioxidant activity. The nitrite scavenging abilities reached the maxium at pH 1.2 and the ethyl acetate fraction of root showed stronger scavenging ability.

Screening of electron donating ability, antibacterial activity and nitrite scavenging effect of some herbal extracts (Herb추출물의 전자공여능, 항균활성 및 아질산염 소거능 검색)

  • Six kinds of herbs(applemint, geranium, lemonbalm, rosemary, sage and thyme) were extracted with water and 95% methanol, and the electron donating ability, antibacterial activity and nitrite scavenging effect were examined. The electron donating ability of herbs ranged from 60.7 to 92.2% and the highest value was observed in geranium, followed by sage and lemonbalm. Antibacterial activity of methanol extracts was much stronger than that of water extracts and gram positive bacteria were more sensitive than gram negative bacteria. Rosemary and geranium exhibited excellent antimicrobial activities against a wide range of bacteria. Nitrite scavenging activity, which was measured at various pH conditions(1.2, 3.0, 3.6, 4.2), was the highest in all herbs at pH 1.2 and the lowest at pH 4.2, suggesting that it is pH dependent. Methanol fraction also provided higher nitrite scavenging effect than the water fraction.

  • This study was designed to analyze the election donating ability(EDA), superoxide dismutase(SOD)-like activity, nitrite scavenging ability and xanthine oxidase for food nutritional evaluation and excavation of functional materials in fruits of Elaeagnus multiflora THUNB. The electron donating ability of ethanol extracts in the 1.0 mg/ml concentration of extract was higher in extract of unripe fruits than others. The superoxide dismutase (SOD)-like activity was increased according to increasing of the extract concentration. The SOD-like activity was highest in the extract of unripe fruits among the extracts. The nitrite scavenging ability of unripe fruits ethanol extracts in 1.0 mg/ml of extraction solution in pH 1.2 was higher than extract of ripe and Ovenipe fruits. The nitrite scavenging ability was decreased according to increasing of pH. Also, inhibition of xanthine oxidase activity were higher in extracts of unripe fruits than those in extract of ripe and over-ripe fruits.

Chemical Components and Nitrite Scavenging Activity of Various Solvent Extracts from Onions (양파 추출물의 화학성분과 아질산염 소거능 효과)

  • To assess utilization of onions extract as natural food additives, ethyl acetate, ethanol and hot water extract of freeze dried onions (red, white and yellow) were exmined for their chemical component and nitrite scavenging activity. Contents of total organic acid and total free sugar were in the range of $135.4{\sim}1,2557mg%$ and $51.7{\sim}62.9%$ as dry matter bases, respectively, and then their content of three onions were higher in ethylacetate and hor water extracts than in ethanol extract. Contents of total phenol and total flavonoids were in the range of $9.3{\sim}$13.3 % and $159.8{\sim}584.1mg%$ as dry matter bases, respectively, and their content of red onion extract by three solvent were higher than those of other onion extract. Nitrite-scavenging activities (NSA) of onion extracts were increased by lowering pH and elevating onion concentrations, and their values of ethanol and hot water were about 55% in addition of 10 mg/mL of red onion extract showing that NSA of red onion was twice higher than that of the other onions. In conclusion, the result indicated that red onion extract was very effective to inhibit nitrosamine formation at low pH condition as natural nitrite scavenging agent.

Comparative analysis of nitrite scavenging activity and anti-inflammation effects in the fruiting bodies of medicinal mushrooms (약용버섯류 자실체의 아질산염 소거능 및 항염증 효능 분석)

  • This study was carried out to compare the anti-inflammation effects of various fruiting body of Ganoderma species and Cordyceps militaris, Phelinus linteus extracts. We concentrated Ganoderma species and other medicinal mushrooms by extracting with ethanol. And We made it $100{\mu}g/ml$ concentration. As a result of nitrite scavenging activity, in the contrast to the positive control; Ascorbic acid was 25%, ASI 7080 of Ganoderma species was disappeared up to around 40%. And in the contrast to Ascorbic acid was 55%, ASI 7002 was 78.5% that was the highest anti-inflammation effect in the result of "No assay test". The Cordyceps militaris showed 75% and Hericium erinaceus showed 59.7% of anti-inflammation effect. As a result of the fungus yield control test of $TNF-{\alpha}$ through ELISA method to ASI 7002 of Ganoderma species that showed the highest anti-inflammation, it was reduced as same as LPS non-treatment. We extracted RNA from ASI 7002 Ganoderma species 10, 50, $100{\mu}g/ml$ concentration and LPS $10{\mu}g/ml$ of Raw 264.7 cell. And we tested the expression of iNOS, COX-2 and TNF-a that are kinds of inflammation gene after synthesizing RNA with cDNA. Finally we could find that iNOS, COX-2 and TNF-a were all controlled expression in the result of above experiment.

Antioxidative Activity and Nitrite Scavenging Ability of Methanol Extract from Salvia plebeia R. Br. (배암차즈기 메탄올 추출물의 항산화 활성 및 아질산염 소거능)

  • Efficacy of antioxidative activity and nitrite scavenging ability of methanol extract from Salvia plebeia was investigated. Electron-donating ability of extract at RC$_{50}$ was 51.10 ${\mu}$g/ml. After addition of 0.92 mg/ml extract, autooxidation of pyrogallol decreased to 62.11% by superoxide dismutase-like activity. In antioxidative activity of extract against linoleic acid during incubation times of 1,2, and 4 days at 40$^{\circ}$, lipid peroxidation values significantly decreased by 72.86%, 77.18%, and 78.32% with addition of 0.2 mg/ml, respectively. Total phenolic content was determined as gallic acid equivalents (GAE) and values revealed as 202.35${\pm}$ 1.26 GAE ${\mu}$g/ml of extract. Nitrite scavenging ability showed the most remark-able effect at pH 1.2, exhibited to 44.29% by addition of 0.2 mg/ml. These results suggest that methanol extract from Salvia plebeia can be used as bioactive and functional materials.

Chemical Components in Stalks and Leaves of Sasa borealis Makino and Antioxidative and Antimicrobial Activities of Extracts (조릿대 줄기와 잎의 화학성분과 그 추출물의 항산화 및 항균활성)

  • This study investigated the cremical components of 1he s1aks anI leaves of Sasa borealis Makino, and the antioxidative activities, nitrite scavenging levels, and antimicrobial activities of extracts of the stalks and leaves. The moisture contents of stalks and leaves were 59.87%(w/w) and 28.79%(w/w), the crude protein levels 2.09%(w/w) and 6.33%(w/w), the crude fat concentrations 1.21%(w/w) and 3.43%(w/w), and the ash levels 0.99%(w/w) and 3.76%(w/w), respectively. The major mineral components were K, Na, Mg and Mn; and the K contents of stalks and leaves were the highest amongst the minerals tested, at 350.27 mg% and 639.60 mg%, respectively. The principal organic acids of stalks and leaves were acetic acid, citric acid and succinic acid, and the organic acid content of leaves was higher than that of stalks. The antioxidative activity of stalk extracts was higher than that of leaves, and the antioxidative activities of ethanol extracts were higher than those of hot water extracts. The 0.5%(v/v) ethanol stalk extracts showed stronger antioxidative effects than did the 0.02%(w/v) butylated hydroxyanisole. All extracts showed nitrite scavenging activities, and hot water extracts from stalk showed the highest activities. Hot water extracts showed antimicrobial activities against all bacterial strains tested, and ethanol extracts from leaf showed strong antimicrobial activities against most bacteria, except Listeria monocytogenes and Salmonella typhymurium. The antimicrobial activities of most stalk and leaf extracts were higher than those obtained using 1.0%(w/v) sorbic acid.

Functional Properties of Medicinal Plant Extracts (한방식재료 추출물의 기능성)

  • The purpose of this study was to investigate the functional properties of medicinal plant extracts. Four kinds of medicinal plants, Dioscorea batatas(DB), Armeniacae Semen(AS) Crataegus pinnatifida Bunge(CP) and Ponciri Fractus(PF), were extracted with water and 70% ethanol and the extracts were tested for their electron donating ability(EDA), nitrite scavenging ability(NSA) and inhibitory effects on cancer cells(MDA cell and A549 cell)growth. EDA at 100-1,000 ppm of water extract ranged from 3% to 14%, 14% to 36%, 29% to 72% and 14% to 43%, and that of ethanol extract ranged from 9% to 62%, 27% to 59%, 33% to 89% and 14% to 44%, in DB, AS, CP and PF, respectively. NSA of extracts measured at various pH(1.2, 3.0, 4.2, 6.0) showed the highest ability in all extracts at pH 1.2 and decreased with increasing pH. The highest NSA of water extracts of 1,000 ppm at pH 1.2 was 6%, 31%, 55% and 44% and that of ethanol extract was15%, 32%, 69% and 52%, in DB, AS, CP and PF, respectively Inhibition ratio of water and ethanol extracts on MDA cell growth was 24% and 17%, 51% and 93%, 46% and 69%, and 48% and 47%, while that on A549 cell was 18% and 9%, 6% and 3%, 7% and 3%, and 43% and 11%, at 1,000 ppm, in DB, AS, CP and PF, respectively.

Studies on Substitution Effect of Chitosan against Sodium Nitrite in Pork Sausage (돈육 소시지에 첨가한 키토산의 아질산염 대체 효과에 관한 연구)

  • Sodium nitrite which is added in processing of meat process product to develope color and to keep bacteria from growing, produces toxic substance after reacting, bring about deterioration by oxidation and toxic substance. So natural material is needed to substitute this sodium nitrite for. Chitosan which is made of chitin by processing of deacetylase, has various function of antibiosis and antimutation. We studied about the substitution effect of chitosan against sodium nitrite in pork sausage. As a result, of storing the sausage, antimicrobial effect of sodium nitrite was detected by 0.35% of chitosan(M.W. 30 kDa). This chitosan had same color developing effect even though addition content of sodium nitrite reduced until 15 ppm which is less than 1/10 of standard level. And chitosan decreased fast a residual nitrite. This result shows that chitosan inhibited a formation of nitrosamine.

Physiochemical Properties, and Antioxidative and Alcohol-metabolizing Enzyme Activities of Nectarine Vinegar (천도복숭아 식초의 이화학적 특성과 항산화 및 알코올 대사 효소 활성)

  • This study investigated the physiochemical properties, antioxidative, nitrite-scavenging, and alcohol metabolism enzyme activities of nectarine vinegar prepared by a traditional fermentation method. The pH of nectarine vinegar was 3.70, the sugar content was $8.87^{\circ}Brix$, and the total acidity was 6.29%. Among organic acids detected, acetic acid was highest at 32.42 mg/ml, followed by lactic acid, malic acid, and succinic acid. Total phenol content of the nectarine vinegar was $121.84{\mu}g$ tannic acid equivalents (TAE)/100 ml. The antioxidative effects of muskmelon vinegar were measured using 1,1-Diphenyl2-picrylhydrazy (DPPH) radical-scavenging activity and superoxide dismutase (SOD) assay. DPPH of nectarine vinegar was increased in a dose-dependent manner, which was 84.47% at 40% concentration. SOD activity was increased in a dose-dependent manner, which was 89.06% at 60% concentration. Nitric scavenging activities of nectarine vinegar were 94.17%, 76.91%, and 20.21% at pH values 1.2, 3.0, and 6.0 at 100% concentration, respectively. The effects of nectarine vinegar on alcohol-metabolism were determined by measuring the generation of reduced nicotinamide adenine dinucleotide (NADH) by alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase (ALDH). ADH and ALDH activities of nectarine vinegar were increased in a dose-dependent manner, which were 153.61% and 178.20 % at 60% concentration, respectively. These results suggest that nectarine vinegar has great potential as a resource for high quality functional health beverages.
