Anti-oxidative Effects of Extracts of Korean Herbal Materials (한방재료 열수 추출물의 항산화 효과)

  • This study was carried out to analyze antioxidative effects of extracts of Korean herbal materials, Diospyros kaki, Teucerium veronicoides and Zanthoxylum schinifolium. Total phenol compound content of Diospyros kaki extacts was higher than those of other extracts. The electron donating ability was $70\%$ in 250 ppm of D. kaki extracts, but was $70\%$ in 1,000 ppm of T. veronicoides and Z. schinifolium extracts. The superoxide dismutase (SOD)-like activity of T. veronicoides and Z. schinzfolium extract was 25, $30\%$ in 1,000 ppm of concentration, respectively, but those of D. kaki extracts was $99\%$. The nitrate scavenging ability of D. kaki extracts was $99\%$ in pH 1.2 and 1,000 ppm of extract solution, but those of T. veronicoides and Z. schinifolium extracts was 53, $72\%$, respectively. The nitrate scavenging ability of D. kaki, Z. schinifolium and T. veronicoides extract was 93, 55, and $40\%$ in pH 3.0 and 1,000 ppm of extract solution, respectively. The nitrate scavenging ability of extracts were $10\~20\%$ in pH 6.0. The nitrate scavenging ability of extracts was decreased according to increase of pH. The nitrate scavenging ability of D. kaki extracts was higher than those of T. veronicoides and Z. schinifolium extracts.

The DPPH and Nitrite Scavenging Activity of Flower Tea by Year of Manufacture in Magnolia denudata (백목련 꽃차 제조연도에 따른 유리산소 소거능(DPPH)와 아질산염 소거능 특성)

  • 색과 향과 맛 및 기능성까지 뛰어난 대표적인 꽃차가 백목련이나 기능성에 관한 연구는 아직 매우 미미하다. 본 연구에서는 유리산소 소거능(DPPH)와 아질산염 소거능(Nitrite Scavenging Activity)이 백목련 꽃차가 제조된 2017년부터 2021년까지의 연도에 따라서 차이가 있는 지를 알아보고자 수행하였으며 그 결과는 다음과 같았다. 1. DPPH는 측정농도(0.5g/100ml=500ppm)에서 제조연도가 2017년 것은 87.8±0.0, 2018년 것은 87.5±0.1, 2019년 것은 85.6±0.6, 2020년 것은 88.5±0.2, 2021년 것은 86.1±0.1으로 2020년에 제조한 것이 가장 높은 함량을 보였다. RC50이란 소거능력이 50%를 환원시킬 수 있는 환원율을 의미하며 낮을수록 적은 양으로 환원이 가능하여 함량이 높다고 할 수 있으며 RC50값은 2017년이 140,2, 2018년이 165.0, 2019년이 178.6, 2020년이 112.2, 2021년이 151.4으로 2020년이 가장 뛰어난 활성을 보였다. 2. 아질산염 소거능은 측정농도(0.5g/100ml=500ppm)에서 제조연도가 2017년 것은 31.8±1.9, 2018년 것은 21.1±4.6, 2019년 것은 18.2±1.9, 2020년 것은 31.0±1.4, 2021년 것은 19.3±5.7의 수치를 보여 2017년과 2020년이 높은 활성을 보였다.

Depletion of Nitrite by Lactic Acid Bacteria Isolated from Commercial Kimchi (시판 김치 분리 젖산균에 의한 아질산염 소거)

  • This study confirmed depletion efficiency of nitrite when incubate lactic acid bacteria was isolated from commercial Chinese cabbage pickles (Baechu kimchi), pickled ponytail radishes (Chongkak kimchi), radish cube kimchi (Kaktugi) and pickled Wakegi (Pa kimchi) at temperatures of $15^{\circ}C$ and $25^{\circ}C$. At $15^{\circ}C$, lactic acid bacteria isolated from commercial kimchi depleted nitrite actively except Leuconostoc mesenteroides subsp. mesenteroides and Leuconostoc paramesenteroides. In particular, Leuconostoc mesenteroides subsp. dextrinicum and Leuconostoc lactis depleted nitrite by very actively without period adaptation in nitrite. Lactobacillus sake, Lactobacillus plantarum, Lactobacillus casei subsp. pseudoplantarum, and Lactobacillus coryniformis subsp. torquens depleted nitrite very actively after 1 day. L. mesenteroides subsp. mesenteroides depleted nitrite relatively actively after 2 days pass. In contrast, L. paramesenteroides displayed very low nitrite depletion ratio compared to other species. At $25^{\circ}C$, all lactic acid bacteria isolated from commercial kimchi did not need adapting period in nitrite, and depleted nitrite very actively except L. paramesenteroides. Also, all lactic acid bacteria except L. mesenteroides subsp. mesenteroides and L. paramesenteroides nitrite of more than 90% after 1 day, and L. mesenteroides subsp. mesenteroides depleted nitrite of more than 90% after 2 days. However, because L. paramesenteroides was not active even at $25^{\circ}C$, nitrite depletion efficiency was very low compared to other species. On the other hand, the same species of Lactobacilli and Leuconostocs except L. mesenteroides subsp. mesenteroides and L. paramesenteroides of other kimchi origin at $15^{\circ}C$ as well as $25^{\circ}C$ by vitality depleted nitrite very actively without statistically significant difference (p<0.05).

Antioxidative and Nitrite Scavenging Activity of Pine Needle and Green Tea Extracts (솔잎 및 녹차추출물의 항산화성 및 아질산염 소거작용)

  • The natural sources extracted from green tea and pine needle were utilized to investigate the effects of extracts on free radical reaction, lipid oxidation and nitrite scavenging ability. The degree of lipid oxidation is very sensitive to kinds of oil emulsion reacted with iron sources and oxygen species. The antioxidants of extracts from green tea and pine needle were different depending on concentration of extracts, which were a lower TBARS value in 0.3% extracts concentration, compared to 0.1% extracts concentration. And also, the binding ability on iron sources was superior in hot water extracts, but oxygen scavenging ability was the lowest TBARS values in ethanol extracts. The hydroxyl radical scavenging ability of green tea and pine needle extracts had a little low TBARS value in 0.1% and 0.3% extracts concentration in deoxyribose. The ethanol extracts of pine needle were higher than those of hot water extracts on the basis of Fe$\_$2+/ ion content. The ascorbic acid content of green tea showed 14.3 mg/100g in hot water extracts and 16.7 mg/100 g in ethanol extracts. Electron donating ability of extracts showed difference depending on extracts concentration, which were higher in ethanol extracts than those of hot water extracts. The nitrite scavenging effects were tended to be different depending on pH value, however they were decreased overall as pH value was increased. Especially, the nitrite scavenging ability of 0.3% extracts from green tea and pine needle were the mosts effective in pH 1.2 and pH 3.0, which were showed 95% nitrite scavenging ability.

Nitrite scavenging and lipid peroxidation inhibitory effects of solvent fractions from Phragmites communis rhizome extract (갈대(Phragmites communis) 뿌리 추출물로부터 얻어진 용매 분획물의 아질산염 소거 및 지질과산화 억제 효과)

  • Phragmites communis rhizome (Phragmitis rhizoma) was extracted with 70% ethanol, and then the extract was fractionated sequentially using n-hexane, chloroform, and ethyl acetate as solvents. Among the solvent fractions of Phragmitis rhizoma extract, the polyphenol content in ethyl acetate fraction was the highest. The chloroform and ethyl acetate fractions possessed a good nitrite scavenging activity. In addition, each solvent fraction showed an effective lipid peroxidation inhibitory ability at a concentration of 10 mg/mL.

Screening of Lactic Acid Bacteria as a Starter Culture in Fermented Sausage (발효소시지 제조에 적합한 스타터 선발)

  • The aim of this study was to select the most suitable starter cultures for production of fermented sausages. A total of 27 strains isolated from Korean fermented foods and natural substances were characterized with respect to their physicochemical properties in a fluid (submerged) model system modified according to the special conditions of fermented sausages. Three of these strains were pre-selected for testing as potential cultures based on their ability to grow fast and initiate rapid acidification. The selected strains were identified by API and partial sequence analysis of 16S rRNA. The results exhibited sequence similarity to known sequences of Staphylococcus warneri, Staphylococcus epidermidis and Lactobacillus plantarum. Among them, relatively good growth properties and nitrite reduction activities were detected for S. epidermidis and L. plantarum and low pH values and high total acidities were observed in the model system fermented with these isolates compared with reference strains.

In Vitro antioxidant effect of ethanol extract from Pennisetum purpureum (Napier grass (Penninsetum purpureum) 에탄올 추출물의 in vitro 항산화 효과)

  • In vitro antioxidant effect of 50% ethanol extract from Napier grass (Penninsetum purpureum) was investigated. The yield and polyphenol content of the Napier grass extract were $6.3{\pm}0.35%$ and $79.6{\pm}3.65{\mu}g$ gallic acid equivalents/mg-extract, respectively. Antioxidant ability of the Napier grass extract such as fee radical and cation radical scavenging activities, reducing power, nitrite scavenging activity, and lipid peroxidation inhibitory activity proportionally increased as concentration of the extract increased. $EC_{50}$ values of the Napier grass extract for free radical scavenging, cation radical scavenging, reducing power, and nitrite scavenging were 1,930.0, 350.0, 840.0, and $1,470.0{\mu}g/mL$, respectively. In the presence of $85.0{\mu}g/mL$ of the Napier grass extract, lipid peroxidation was potently inhibited by 74.6%.

Antioxidant Activity of Major Cultivars Prunus mume in Korea (국내 주요 재배 매실 품종의 항산화 활성)

  • This study examined the total phenolic and flavonoid compounds, and the antioxi dant activity of major cultivars of Prunus mume in Korea. The total phenolic content ranged from 64.13 to $93.43mg{\cdot}100g^{-1}$, with the content in 'Osuku' being higher than that in the other cultivars; a difference in the content among cultivars was observed. The total flavonoid content was $6.16{\sim}18.57mg{\cdot}100g^{-1}$, showing a significant difference, with the highest content being noted in 'Gojirou'. The DPPH radical scavenging activity was measured and was relatively high for Nanko, Kamakami, Natsuka, Dana, and Osuku cultivars. When ABTS radical scavenging activity was measured, all the cultivars showed a scavenging activity over 80%, with the scavenging activity of 'Osuku' being the highest. With respect to nitrite scavenging ability, all cultivars exhibited a high scavenging activity of over 50%, with the highest activity observed for 'Nanko' (76.03%), 'Gojirou' (70.56%), and 'Yourou' (70.32%). Based on the study results, it is considered that Prunus mume can be a useful resource because of various substances with functional physiological activities.

Antioxidant Activity and Nitrite Scavenging Ability of Olive Leaf (Olea europaea L.) Fractions (올리브 잎 분획물의 항산화기능과 아질산염 소거능력 평가)

  • In this study, the antioxidant activities and nitrite scavenging abilities of olive leaf fractions acquired from plants cultivated in Australia (Olea europaea L. var. Picual) and Spain (Olea europaea L. var. Hojiblanca) were evaluated. Oleuropein was found to be the major phenolic compound in the leaves, with the butanol fractions presenting the highest contents. Antioxidant activity was evaluated in terms of superoxide dismutase (SOD)-like activity, 1,1-diphenyl-2-picryl hydroxyl (DPPH) radical scavenging activity, and the inhibitory effect on the auto-oxidation rate of linoleic acid. The SOD-like activities of the olive leaf extracts ranged from 0 to 36.8%. DPPH radical scavenging activity was highest in the ethanol extract of the Australian cultivated olive leaves. Finally, the chloroform fractions of the extracts showed inhibitory effects on the auto-oxidation rate of linoleic acid as well as nitrite scavenging ability.

Change of Antioxidant Activities in Preparing Freeze Dried Wild Vegetable Block for the Long-term Storage (장기저장을 위해 제조한 동결건조 산채 블록의 항산화 활성 변화)

  • The antioxidant activities of water extracts from wild vegetables such as Ligularia fischeri (GC), Capsicum annuum L. (GCY), Aster scaber (CNM), Petasites japonicus S. et Z. Max (MYD), Ipomoea batatas L. (Lam) (GGM) were evaluated and compared with water extracts from freeze dried block. The antioxidant properties of water extracts from wild vegetables and their freeze dried block were evaluated using different antioxidant tests; 2.2-diphenyl-1-picryl-hydrazyl (DPPH) radical scavenging, hydroxyl radical scavenging and nitrite scavenging activities. The water extracts from wild vegetables were found to have a higher total phenolic content than water extracts from freeze dried block. Total phenolic contents of water extracts from GC, GCY, CNM, MYD, and GGM were $471.66{\pm}3.52\;{\mu}g/mg,\;141.33{\pm}2.51\;{\mu}g/mg,\;177.33{\pm}2.88\;{\mu}g/mg,\;238.66{\pm}9.50\;{\mu}g/mg\;and\;122.67{\pm}3.51\;{\mu}g/mg$, respectively. At the concentrations of 1000 ppm, water extracts from GC, GCY, CNM, and GGM showed higher activities than water extracts from their freeze dried block on DPPH radical scavenger activity. The activity of water extracts from CNM, GC, GCY, MYD, and GGM were 90.9%, 89.9%, 76.6%, 71.1%, and 57.4%, respectively. When 10000 ppm of GC, GCY, CNM, MYD, and GGM water extracts tested for hydroxyl radical scavenging activity, activities were increased by 38.8%, 33.4%, 35.9%, 34.3%, and 33.8%, respectively and a similar effect was found with water extracts from GCY, CNM, and GGM freeze dried block at 10000 ppm concentration. However, the water extracts from GC and MYD was slightly more effective than freeze dried block extracts. The water extracts from wild vegetables and their freeze dried block had effective DPPH radical scavenger activity and hydroxyl radical scavenging activity at all tested concentrations. Nitrite scavenging activity of GC water extract significantly increased in a dose-dependent manner and the extract had higher nitrite scavenging activity than extracts freeze dried block extracts. We found that freeze dried block maintained antioxidant activities of the wild vegetables.
