Effects of Organic Matter Application on Soil Microbial Community in a Newly Reclaimed Soil (신규 유기농경지 토양의 유기물 공급이 토양 미생물군집에 미치는 영향)

  • soil microbial activities and diversities in a newly reclaimed soil. Soil chemical properties, population of microbe, microbial biomass, and properties of microbial community were investigated under 4 different treatment (animal manure compost+green manure, chemical fertilizer, and without fertilizer). The experiment was conducted for 3 years from 2012 to 2014. The most of chemical properties in the animal manure compost+green manure treatment were increased continually compare to chemical fertilizer and without fertilizer. The population of bacteria and fungi were higher in the animal manure compost+green manure treatment, however, there was no difference on actinomyces. Soil microbial biomass C content was higher in the animal manure compost+green manure treatment than in chemical fertilizer and without fertilizer. Biolog examination showed that catabolic diversities of bacterial communities were higher in the treatment of animal manure compost+green manure. It was showed that principle component analysis of the Biolog data differentiated the organic matter amended soils from NPK and control. These results indicated that application of animal manure compost+green manure had a beneficial effect on soil microbial properties.

  • Cystocin, a derivative of Puromycin, is a new material derived from Streptomyces sp GCA0001, new strain of Actinomycetes spiecies. This compound has outstanding biological activities in anti-bacteria, anti-tumor and anti-virus than former Puromycin compounds. And it is chosen by natural selection processing through extraction, isolation and purification from , so it may replace old Puromycins in most applications.

  • Epoxide hydrolase activities of various microbial cells were analyzed by colorimetric assay based on alkylation of epoxides with 4-(p-nitrobenzyl)pyridine (NBP). The epoxide hydrolase activity was determined by measuring the decrease of color intensity at 560 nm due to the decrease of styrene oxide substrate by epoxide hydrolase-catalyzed hydrolysis reaction. The experimental conditions of NBP colorimetric assay were optimized for the efficient measurement of epoxide hydrolase activities from various microbial cells.

Effects of Animal Manure Compost, Tillage Method and Crop System on Soil Properties in Newly Organic Corn Cultivation Field (신규 유기농 옥수수 재배 시 가축분 퇴비, 경운방법 및 작부체계가 토양 환경에 미치는 영향)

  • This study was conducted to investigate the effects of organic farmland soil and nutrient management on soil properties depending on organic (animal manure compost and green manure [hairy vetch]) and chemical fertilization, tillage and no-tillage, and crop rotation (corn-wheat, corn-.hairy vetch). It was found that the application of organic matter such as animal manure compost and hairy vetch, increased the soil organic matter content, the soil microbial density and microbial biomass C content as compared with the chemical fertilizer treatment. It was also confirmed that the functional diversity of soil microbial community was increased. As a result of the comparison with the crop rotation and single cropping, the soil chemistry showed no significant difference between the treatments, but the corn-wheat and corn-hairy vetch rotation treatments tended to have higher microbial biomass C content and shannon's diversity index than the single cropping. Soil chemical properties of tillage and no-tillage treatments showed no significant difference between treatments. There was no statistically significant difference in substrate utilization of soil microbial community between tillage and no-tillage treatment. Correlation analysis between soil chemical properties and soil microbial activity revealed that soil organic matter content and exchangeable potassium content were positively correlated, with statistical significance, with substrate utilization, and substrate richness. To conclude, organic fertilization had positive effects on the short-term improvement of soil chemical properties and diversity of microbial communities.

  • AF-011A is a novel lipopeptide with potent antifungal activity isolated from Pseudomonas cepacia AF6008 deposited as KFCC 10759. The compound was isolated from the fermentation broth by extraction with 50% isopropyl alcohol. Purification was effected by chromatography on Diaion HP-20, Alumina and C18 followed by HPTLC on silica gel. These techniques affored two closelt related compounds. AF-011A1 ans AF-011A2. The molecular weights of AF-011A1/A2 were determined by fast atom bombardment mass spectrometry(A1 m/z 1,215 : A2 m/z 1,199).

  • In order to develop new antitumor agents from fermentation broths, we used cccDNA breakage assay abd sulforhodamine B assay for prescreening. As a result, it was shown that sample reach 3.3% when using cccDNA breakage assay. In sulforhodamine B assay, we obtained 4 acive fraction against A549 (a cell line of human lung carcinoma) and SK-OV-3 (a cell line of human adenocarcinoma). These results suggest that these assay would be a promising method for antitumor prescreening from microbial sources.

  • AF-001A is a novel antifungal cyclic glyco-peptise isolated from Pseudomonas cepacia AF6008. AF-001A is a mixture of AF-011A1 and AF-001A2. Each compound contains glycine(1), serine(2), asparagine(1), 2,4-diaminobutyric acid(1), beta-hydroxytyrosine(1), xylose(1) and a methylene chain amino acid(1). Additionally, A1 contains one beta-hydroxyasparagine that is replaced with as asparagine in A2. AF-011A showed high in vitro antifungal activity against various animal and plant pathogenic fungi and caused no irritation on the skin of rabbits.

  • In the fed-batch fermentation of penicillin specific uptake rates of carbon source and ammonia nitrogen, and specific production rate of penicillin as the most important process variables were evaluated over the fermentation course and their effects on the productivity studied. As the results, glucose and lactose each as a major carbon source showed the following values, respectively ; the specific uptake rates of 47-93 mg hexose per gm-DCW per he and 37-44 mg hexose per gm-DCW per hr, the specific uptake rates of 4.6-6.8 mg $NH_3-N$ per gm-DCW per hr, and 1.2 mg $NH_3-N$ per gm-DCW per he and the specific production rates of 32-42 arbitrary unit per gm-DCW per hr and 46-50 arbitrary unit per gm-DCW per hr. The productivity of penicillin could be improved by controlling the feed rates of glucose and ammonia nitrogen to meet the uptake rates.

Development of Self-Repairing Smart Concrete Using Micro-Biologically Induced Calcite Precipitation (미생물의 방해석 석출 작용을 이용한 자기보수 스마트 콘크리트 개발에 관한 연구)

  • This paper presents a study on the development of next generation smart concrete in an eco-friendly manner using micro-biologically induced calcite precipitation (MICP) via microbial biomineralization. It seems that currently, the reformation and functional improvement of concrete using MICP can be achieved using Sporosarcina pasteurii, which is a representative microorganism that produces calcite precipitation. Based on previous studies on MICP the biochemical tests and crystallinity evaluation of cement using sporoasrcina pasteurii and four additional micro-organisms from the concrete structures as identified by 16S rDNA sequence analysis were conducted. Also by applying the Sporosarcina pasteurii and separated four effective micro-organisms from the concrete structures to mortar, the compressive strength improvement by varying curing conditions, repair of crack were examined, and plans for future study were suggested. The effect of the application of effective micro-organisms can lead to the development of a new material that will contribute to resolution of environmental problems and facilitate repair work, and this can also serve as a new research theme in the future. In addition, the importance of this study is to use micro-organism, which is found common in concrete structures, this new microbial is not only environmentally safe but also persists in the natural environment for an extended period of time. Therefore, it seems to have a great potential to became a new environmentally low-burdened functional material.

Isolation and characterization of an Enterococcus faecalis bacteriophage (Enterococcus faecalis 특이적 박테리오파지의 분리와 특성규명)

  • Enterococcus faecalis is a Gram-positive and facultative anaerobic bacterium that causes many hospital-acquired infections. Novel E. faecalis specific bacteriophage (phage) ECP3 that had been isolated from thirty-four environmental samples and characterized phenotypically and genotypically. ECP3 phage belongs to the family Myoviridae with contractile tail and lysed E. faecalis specifically but other bacteria including Enterococcus faecium. The genome was double-stranded linear DNA and its size was 145,518 bp comprising of 220 open reading frames. The GC content was 35.9%. The genome sequence showed 97% identity in 90% coverage region with Myoviridae phage PhiEF24C. ECP3 is the first E. faecalis-specific Myoviridae phage isolated in Korea which can be a promising antimicrobial agent against E. faecalis infections.
