The purpose of this study is to quantitate regional neurochemical profile of regional normal adult mice brain and assess regional metabolic differences by using ex vivo $^1H$ high-resolution magic angle spinning nuclear magnetic resonance spectroscopy ($^1H$ HR-MAS NMRS). The animals were matched in sex and age. The collected brain tissue included frontal cortex, temporal cortex, thalamus, and hippocampus. Quantitative 1D spectra were acquired on 40 samples with the CPMG pulse sequence (8 kHz spectral window, TR/TE = 5500/2.2 ms, NEX = 128, scan time: 17 min 20 sec). The mass of brain tissue and $D_2O$+TSP solvent were 8~14 mg and 7~13 mg. A total of 16 metabolites were quantified as follow: Acet, NAA, NAAG, tCr, Cr, tCho, Cho, GPC + PC, mIns, Lac, GABA, Glu, Gln, Tau and Ala. As a results, Acet, Cho, NAA, NAAG and mIns were showed significantly different aspects on frontal cortex, hippocampus, temporal cortex and thalamus respectively. The present study demonstrated that absolute metabolite concentrations were significantly different among four brain regions of adult mice. Our finding might be helpful to investigate brain metabolism of neuro-disease in animal model.
Because neurochemicals are related to homeostasis and cognitive and behavioral functions in human body and because they enable the diagnosis of numerous neurodegenerative disorders, there has been increasing interest in the development of analytical platforms for neurochemical profiling in biological samples. In particular, mass spectrometry (MS)-based analytical methods combined with chromatographic separation have been widely used to profile neurochemicals in metabolic pathways. However, development of delicate sample preparation procedures and highly sensitive instrumental detection is necessary considering the trace levels and chemical instabilities of neurochemicals in biological samples. Therefore, in this review, analytical trends in MS-based metabolomics approaches to neurochemicals in multiple biological samples, such as urine, blood, CSF, and biological tissues, are discussed. This paper is expected to contribute to the development of an analytical platform to discover biomarkers that will aid diagnosis, prognosis, and treatment of neurodegenerative disorders.

많은 홀몬, 성장인자 및 신경 전달물질들은 각각에 대한 세포막의 수용체와 결합하여 Phospholipase C (PLC)를 활성화시키므로서 신호를 세포내로 전달하여 세포의 성장, 대사, 신경 흥분, 수축 및 분비 등의 생리 현상을 나타내고 있다. 이 신호 전달의 중심 효소인 PLC는 현재까지의 효소 분리, 유전자 클로닝 등의 방법으로 3가지 Class에 적어도 8종유의 등위효소(Isozyme)들이 존재하고 있는 것으로 밝혀지고 있다. 본 연구에서는 이와 같은 등위효소에 특이적인 조절 물질을 선별할 수 있는 체계를 확립하기 위하여 새로운 동위효소의 분리 및 규명과 이 동위효소들에 대한 과발현 및 발현 세포주 개발을 추진하고 있다.
Catecholamines, serotonin and their metabolites were measured in the posterior hypothalamus of urethane-anesthetized normotensive Wistar Kyoto rats (WKY) and spontaneously hypertensive rats (SHR) using brain microdialysis which is a recently developed experimental method to measure the release of neurotransmitters and their metabolites at the localized brain area in vivo. Microdialysis probe was implanted stereotaxically to the rat posterior hypothalamus and perfused by Ringer's solution. Monoamines and their metabolites were quantified by reverse phase high performance liquid chromatography with electrochemical detection. In vitro recovery test of microdialysis showed that there exist inverse relationship between the perfusion flow rate and the relative recovery of neurochemical compounds. The estimated extracellular concentration of dopamine was about 32 nM, of norepinephrine 50 nM, of epinephrine 50 nM, of serotonin 73 nM, of 3, 4-dihydroxyphenylacetic acid (DOPAC) 281 nM, of homovanillic acid (HVA) 181 nM, and of 5-hydroxyindoleacetic acid (5HIAA) 3767 nM in the hypothalamic perfusate of the normotensive rat. There was no difference in the basal level of monoamines between the SHR and the WKY. In contrast, the level of DOPAC, HVA and 5HIAA in SHR was higher than that in the WKY, This study demonstrated that the microdialysis technique should be an applicable tool for in vivo measurement of central neurochemical substances.
FS11052, a novel microbial metabolite from Streptomyces spp. was identified as a small molecular substance and shown inhibition activities for the release of neurotransmitter from rat hippocampal neuron and PC12 cells. FS11052 is an inhibitor of tritiated norepinephrine ($[^3H]-NE$) release in high $K^+$ buffer solution containing ionomycin, indicating that FS11052 inhibits neurotransmitter release after the influx of $Ca^{2+}$ ions. When examined the effect of FS11052 on glucuronidase release from guinea pig neutrophils, FS11052 inhibited glucuronidase release: when treated with $5{\mu}g/ml$ of FS11052, which was not induced cellular cytotoxicity. The fact that the glucuronidase release in neutrophil and norepinephrine release in neuron was inhibited suggests the similarity in the locations and the mechanisms of FS11052 action targets. When treated with $5{\mu}g/ml$ of FS11052, $[^3H]-NE$ release and neurite extension for both rat hippocampal neurons and PC12 cells were prevented. These observations of FS11052 functioning as an inhibitor of neurotransmitter release suggest that FS11052 has an important role in synaptic transmission in neuron.
We provide the reader with a brief introduction to the neurobiology of neuropeptides. Several comprehensive reviews of the distribution and neurochemical, neurophysiological, neuropharmacological and behavioral effects of the major neuropeptides have recently appeared. In reviews of the large number of neuropeptides in brain and their occurance in brain regions thought to be involved in the pathogenesis of major psychiatric disorders, investigators have sought to determine whether alternations in neuropeptide systems are associated with schizophrenia, mood disorders, anxiety disorders, alcoholism and neurodegenerative disease. There is no longer any doubt that neuropeptide-containing neurons are altered in several neuropsychiatric disorders. One of the factors that has hindered neuropeptide research to a considerable extent is the lack of pharmacological agents that specifically alter the synaptic availability of neuropeptides. With the exception of naloxone and naltrexone, the opiate-receptor antagonists, there are few available neuropeptide- receptor antagonists. Two independent classes of neuropeptide-receptor antagonists has been expected to be clinically useful. Naltrexone, a potent ${\mu}$-receptor antagonist, has been used successfully to reduce the need for alcohol consumption. And cholecycstokinin antagonists are now in development as a new class of anxiolytics, which would be expected to be free from tolerance and physical dependence and lack of sedation. In this review, we deal with these two kinds of neuropeptide system, the opioid system and cholesystokinins in the brain. The role of opioid systems in the reinforcement after alcohol consumtion and that of cholesystokinins in the pathogenesis of anxiety will be discussed briefly. As we know, the future for neuropeptides in psychiatry remains bright indeed.

Hematoxylin is the main component of Hematoxylon campechianum which has been utilized in the southern provinces of Korea as a folk remedy for diabetic complications. In the present study, to investigate the hypoglycemic mechanism of hematoxylin, the 2-deoxyglucose uptake and phospholipid metabolism were examined in sciatic nerves from three groups of rats : normal control, diabetic control, diabetic hematoxylin-treated group. Hematoxylin significantly reduced blood glucose levels in diabetic control rats. On a wet weight basis, the nerves from diabetic rats showed a 20% decrease in total phospholipid from that of controls and a relative decrease in phosphatidylinositide. Hematoxylin treatment increased the incorporation rate of 2-[3H] myo-inositol into total phosphoinositids in diabetic rat. The effectiveness were more potent in higher dose hematoxylin-treated rats than lower dose hematoxylin-treated rats. These results suggest that hematoxylin increases glucose transport and lipid metabolism by partially normalizing concerned with myo-inositol metabolism in diabetic rat. Therefore we propose that hematoxylin can be a promising candidate for diabetes medication.

포유 동물의 중추 신경계에서 주 억제성 신경전달물질로 알려진 4-aminobutyrate(GABA) 대사에 관여하는 효소인 GABA transaminase (GhBA-T)와 Succinic semialdehyde reductase(SSR)를 각각 여러 가지 크로마토그래픽 방법을 사용하여 순수 분리 정제하였다 GABA-T는 subunit 분자량 50kDa을 가진 dimer이며 화학 변형(chemical modification), 흡광, 형광스펙트럼 연구에 의하면 cysteinyl잔기가 활성 부위에 존재하고 있음을 보여주었다 SSR은 2-mercaptoethanol이 존재하든 안하든 o-pthalaldehyde(OPA)에 의하여 효소 활성도가 상실되었으며 흡광 및 형광스펙트럼에 의하면 OPA가 효소의 활성 부위의 lysyl잔기와 반응하여 cysteinyl잔기와 함께 intermolecular cross-linking을 이루는 것을 알 수 있었다. 한편 SSR은 몇가지 anticonvulsant drug에 의하여 부분적인 저해 효과를 나타내었다.

본 연구에서는 생체 뇌 국소부위의 세포외액을 대상으로 실험할 수 있는 뇌 미세투석법을 이용하여 후시상하부에서 유리되는 monoamine의 세포외액 농도에 대한 전신 혈압변화의 영향을 관찰함으로써, 심혈관 조절에 대한 후시상하부의 monoamine성 뉴론의 생리적 역할을 규명하고자 하였다. 마취한 흰쥐의 머리를 뇌정위 고정장치에 고정시키고 미세투석관 (microdialysis probe)을 후시상하부에 위치시킨 후 링거액으로 관류하였다. 미세투석액내에 존재하는 monoamine성 신경 전달물질들과 그 대사체들의 정량분석은 고속액체크로마토그라피와 전기화학검출기를 이용하여 실시하였다. 전신 혈압을 40분 동안 약 40mmHg 상승 혹은 30mmHg 감소시키기 위하여 L-phenylephrine hydrochloride (800ng/100 g/min) 혹은 nitroprusside dihydrate (500 ng/100 g/min)를 대퇴정맥을 통하여 주사하였다. 후시상하부로 부터 20분 간격으로 얻은 투석액에서 신정화학물질들의 농도는 미세투석관 삽입후 2시간에 안정되었다. 관류액의 $K^{+}$ 농도를 90 mM로 증가시켰을 때 후시상하부의 투석액에서 norepinephrine (NE) 과 serotonin (5-HT)의 농도는 각각 기준치의 176.5 $\pm$ 14.8%, 149.1 $\pm$ 2.3%로 증가하였다. Phenylephrine (i.v.)으로 유발된 전신 혈압상승에 의하여 NE과 5-HT의 양은 각각 기준치의 79.3 $\pm$ 4.4%, 61.4 $\pm$ 10.3%로 유의하게 감소하였다. 또한 nitroprusside (i.v.)에 의하여 전신혈압이 감소하였을 때 투석액내 5-HT의 양은 기준치의 195.0 $\pm$ 23.0% 로 유의하게 증가하였다. 후시상하부의 투석액내 monoamine성 대사체들의 경우에는 전신 혈압의 변화에 대한 유의한 반응을 나타내지 않았다. 이상을 종합하면 전신 혈압이 증가되었을 때는 말초 압수용체의 흥분에서 기시한 신경충동이 후시상하부에 전달되어 신경말단에서 5-HT 과 NE 의 유리가 감소되고, 5-HT의 경우에는 전신 혈압이 감소되었을때 그 반대 현상이 일어난다는 가설을 제시할 수 있다. 본 연구의 결과는 생체상태에서 혈압변화에 대한 후시상하부의 monoamine성 조절양상을 규명한 것으로서 특히 후시상하부의 serotonin성 신경계를 통한 심혈관 중추조절의 가능성을 처음으로 제시한 것이다.
한국산 도롱뇽 (Hynobius leechi BOULENGER) 발생배의 질소화합물의 대사를 연구하고져 발생단계에 따른 단백질구성(아미노) 산의 촉진(아미노) 산의 정성적 분석과 질소 배설물(ammonia, urea, uric acid, creatine and/or creatinine, total nitrogen)의 정량분석을 미수정란, 란할배, 포배, 양배, 신경배 및 부화직전의 배에 대하여 행하였다. 1. 발생단계에 따른 발생배내 단백질구성 (아미노산)은 전 단계를 통하여 일양하게 18종씩 검출되어TRh 유리 (아미노산) 은 17 내지 23 종 검출되었으며, 전체적으로 유리(아미노산) 분포상에 큰 변화가 없었다. 2. 발생단계에 따른 질소배설물 분석의 결과는 다음과같다. Ammonia 와 urea 의 배내 함량은 미수정란에서부터 양배까지 점차 감소하여 양배 이후 부화직전배까지 급격히 증가한다. Uric acid의 함량은 전 단계를 통해 미소하였으며 creating and/or creatininie 은 본 실험에서는 검출되지 않았다. 3. 발생단계에 따른 발생배내 ammonia 함량에 대한 sodium azide의 영향은 농도에 따라 이원적인 특이성을 가지고 있었다.
