Antioxidative Effects of Solvent Fractions from Nandina domestica Fruits (남천(Nandina domestica) 열매 분획 추출물의 항산화 효과)

  • This study was carried out to compare the antioxidative effects of various Nandina domestica fruits extracts. Organic fractions, including n-hexane, chloroform, ethyl acetate, and butanol fractions, were obtained from the water and ethanol extracts of N. domestica fruits. The phenolic compound content of the EE fraction was 922.22 mg/g. The flavonoid compound content was highest in the EB fraction (282.49 mg/g). The electron-donating ability was highest (93%) in the WA and EH fractions at 0.1 mg/mL. The SOD-like activity was showed the highest in the EA fraction (56.36%), and EH and EC both showed higher than 50% activity. The nitrite-scavenging ability of the EC fraction at 1.0 mg/mL under pH 1.2 conditions was 82.03%. The xanthine oxidase inhibitory activities of all the fractions, except WE, were higher than 90% at 0.5 mg/mL. The effect of tyrosinase inhibition was highest in the WE fraction (46.75%). These results show that the N. domestica fruits fractions contained high levels of polyphenol and flavonoid compounds, along with excellent antioxidative effects. This suggests that N. domestica fruits can be used as a functional material.

Antihypertensive Effects of Casein Hydrolysate in Spontaneously Hypertensive Rats (자연발증고혈압쥐에서 카제인 가수분해물의 혈압강하효과)

  • The aim of this study was to determine if a low-molecular weight casein hydrolysate has an anti- hypertensive effect in spontaneously hypertensive rats (SHR). Prior to the in vivo experiment, the casein hydrolysate was confirmed to be resistant to gastrointestinal digestion by confirming the retention of its potency as an inhibitor of angiotensin I-concerting enzyme after incubation with pepsin, trypsin, or chymotrypsin. The in vivo anti-hypertensive effect of the hydrolysate was determined by the tail cuff method. Following an oral administration of the hydrolysate solution, the systolic blood pressure (SBP) decreased by 12.9% (-28.9mmHg; P<0.05) at 3 h after the administration at a dose of 500mg/kg body weight. When the hydrolysate was administered as an emulsion with 30% egg yolk, its anti-hypertensive effect was even more greater at the same dose(-30.8mmHg or -15.9%; P<0.01). In a 50-day long-term trial where the casein hydrolysate was administered once a day, the SBP-lowering effect of the hydrolysate was apparent (P<0.05) from day 35 through the end. Moreover, organ weights and plasma glutamate oxaloacetate transaminase and glutamate pyruvate transaminase activities of the administered SHR were not significantly different from those of controls at the end of the long-term trial.

Study on the Anti-oxidant and Anti-inflammatory Activities of Sarcocarp and Calyx of Persimmon (Cheongdo Bansi) (청도 반시의 과육과 감꼭지(시체: 枾滯)의 항산화 및 항염증 활성 연구)

  • Biological activities of sarcocarp and calyx of persimmon (Cheongdo Bansi) were investigated. The electron donating ability of the calyx extract was 91% at a 500 ${\mu}g/mL$ level, which was higher than that of the sarcocarp extract (40%). The superoxide dismutase (SOD)-like activity of calyx extracts was about 22% at a 1,000 ${\mu}g/mL$ level. During the entire experimental period, there was no cytotoxicity found from sarcocarp and calyx up to 50 ${\mu}g/mL$. Our results indicated that sarcocarp and calyx significantly inhibited NO production and iNOS and COX-2 expression accompanied by an attenuation of interleukin (IL)-$1{\beta}$, IL-6, and tumer necrosis factor (TNF)-${\alpha}$ formation in human keratinocyte. These results suggest that sarcocarp and calyx of persimmon may have significant effect on inflammatory factors and can be used as potential anti-inflammatory agents.

EYE IRRITATION TEST OF LENS WASHING AGENTS CLlWELLTM ON NEW ZEALAND WHITE RABBITS (렌즈 보존액 CLlWELLTM에 대한 안점막자극성시험)

  • The eye irritant test of lens washing agent, CLIWELL$^{TM}$ was conducted using Draize methods according to KFDA Guidelines. In addition, to test the potential toxicity of test articles, the ratio of inflammatory cells and non-inflammatory epitheloid cells was also observed using smear cytology methods against ocular discharge. At sacrifice, the histopathological changes on Cornea, Iris, Retina and Sclera were also observed in all animals. Slight irritancy of cornea and conjunctiva was observed at 1, 2 and 3 days after dropping in non-washing group. The MIOI of these points are detected as 4.50, 1.67 and 0.67, respectively. In washing group, slight irritancy of cornea and conjunctiva were observed at 1 and 2 days after dropping with MIOI as 0.67, respectively. Therefore, CLIWELL$^{TM}$ was considered as non-irritating materials because the MIOI is detected below 5.00 throughout the whole experimental periods in both washing and non-washing groups and the IAOI was also detected as 4.50. Except for significant increase of the inflammatory cell ratios in ocular discharge at 1 day after dropping of non-washing group compared to that non-treated intact eyes, no meaningful changes on smear cytology of ocular discharges are observed in this study. In addition, no abnormal histopathological changes on the cornea, iris, retina and sclera were also detected in CLIWELL$^{TM}$ dropping group compared to that of non-treated intact eyes.

Antioxidant and Whitening Effects of the Fermentation of Barley Seeds (Hordeum vulgare L.) Using Lactic Acid Bacteria (유산균을 이용한 보리의 발효를 통한 항산화 및 미백 효과)

  • Barley (Hordeum vulgare L.), of the Poaceae/Gramineae family, is a common grain in the surrounding area. It has been used in Ancient Egyptian medicine and it has been used worldwide for many years as food and as an ingredient in beer. Barley has been reported to have anti-inflammatory, anti -carcinogenic and anti-diabetic effects. So far, a lot of research has been done on barley but the effects of fermented barley seeds with lactic acid bacteria have not been studied largely. In this study, we investigated the effects of ethanol-extracted barley seeds after their fermentation with lactic acid bacteria. The biological activities of fermented barley seeds with lactic acid bacteria and non-fermented barley seeds were analyzed for total polyphenol content, total flavonoid content, DPPH radical scavenging, superoxide dismutase-like activity and tyrosinase inhibition. These results showed that fermented barley seeds with lactic acid bacteria have more advanced anti-oxidant and whitening properties than non-fermented barley seeds. Hence, we suggest that fermenting barley seeds with lactic acid bacteria can be an impressive material in the food and cosmetic industries.

Eye Irritation Test of Lens Washing Agents SOLOCARETM on New Zealand White Rabbits (렌즈 보존액 SOLOCARETM에 대한 안점막자극성시험)

In addition, to test the potential toxicity of test articles the ratio of inflammatory cells and non-inflammatory epitheloid cells were also observed using smear cytology methods against ocular discharge. At sacrifice, the histopathological changes on Cornea, Iris, Retina and Sclera were also observed in all animals. Slight (1~2 degrees) irritancy of cornea and conjunctiva were observed at 1, 2 and 3 days after dropping in non-washing group. The MIOI of these points are detected as 4.33, 3.33 and 2.00, respectively. In washing group, slight irritancy of cornea and conjunctiva were restricted to 1 and 2 days after dropping with MIOI as 3.00 and 1.33, respectively. Therefore, $SOLOCARE^{TM}$ was also considered as non-irritating materials because the MIOI is detected below 5.00 throughout the whole experimental periods in both washing and non-washing groups and the IAOI is also detected as 4.33 (1 day after dropping). No meaningful changes on smear cytology of ocular discharges are observed in this study compared to that of non-treated intact eyes. In addition, no abnormal histopathological changes on the cornea, iris, retina and sclera were also detected in $SOLOCARE^{TM}$ dropping group compared to that of non-treated intact eyes.

Flavor Compounds of Domestic Meju and Doenjang (재래식 메주 및 된장의 향기성분)

  • Volatile components of domestic Meju and Doenjang were extracted by simultaneous steam distillation extraction, and analyzed by GC-MS. Sixty-four kinds of compounds were identified from neutral fraction. The contents of pentanal, hexanal and 1-octen-3-ol were high in cooked soybean while those of 3-methylbutanal and 1-butanol were high in Meju. In the case of Doenjang, so many compounds including acetic acid, ethylester were identified which was not appeared in Meju. The main compounds in Meju were 3-methyl-1-butanol, 2-furancarboxyaldehyde, 1-octen-3-ol, benzeneacetaldehyde, methyloctadecadienoate and methyloctadecenoate. Of the eleven compounds identified from basic fraction, the contents of 2,6-dime-thylpyrazine, trimethylpyrazine and tetramethylpyrazine were high in Meju and Doenjang. Nine kinds of compounds were identified from phenolic fraction and appeared that 4-vinylphenol and p-ethylguaiacol were major compounds in Meju and Doenjang. Fifteen kinds of volatile compounds were contained in acidic fraction. Only four acidic compounds were identified in cooked soybean and Meju, but in Doenjang ten compounds were identified which did not appeared in other samples. Among them pentadecanoic acid was major compound.

Water-insoluble, Whey Protein-based Microcapsules for Controlled Core Release Application (유청단백질을 이용한 미세캡슐의 응용)

  • Microcapsules consisting of natural, biodegradable polymers for controlled and/or sustained core release applications are needed. Physicochemical properties of whey proteins suggest that they may be suitable wall materials in developing such microcapsules. The objectives of the research were to develop water-insoluble, whey protein-based microcapsules containing a model water-soluble drug using a chemical cross-linking agent, glutaraldehyde, and to investigate core release from these capsules at simulated physiological conditions. A model water soluble drug, theophylline, was suspended in whey protein isolate (WPI) solution. The suspension was dispersed in a mixture of dichloromethane and hexane containing 1% biomedical polyurethane. Protein matrices were cross-linked with 7.5-30 ml of glutaraldehyde-saturated toluene (GAST) for 1-3 hr. Microcapsules were harvested, washed, dried and analyzed for core retention, microstructure, and core release in enzyme-free simulated gastric fluid (SGF) and simulated intestinal fluid (SIF) at 37$^{\circ}C$, A method consisting of double emulsification and heat gelation was also developed to prepare water-insoluble, whey protein-based microcapsules containing anhydrous milkfat (AMF) as a model apolar core. AMF was emulsified into WPI solution (15-30%, pH 4.5-7.2) at a proportion of 25-50% (w/w, on dry basis). The oil-in-water emulsion was then added and dispersed into corn oil (50 $^{\circ}C$)to form an O/W/O double emulsion and then heated at 85$^{\circ}C$ for 20 min for gelation of whey protein wall matrix. Effects of emulsion composition and pH on core retention, microstructure, and water-solubility of microcapsules were determined. Overall results suggest that whey proteins can be used in developing microcapsules for controlled and sustained core release applications.

Problems and Solutions of Zymography Techniques (자이모그라피 기술의 문제점과 해결)

  • Enzymes are widely used in industrial applications such as detergents, food, feed production, pharmaceuticals and medical applications and major contributors to clean industrial products and processes. To screen, identify, and characterize the enzymes the zymography techniques are routinely used. The zymography technique is a simple, sensitive, and quantifiable technique that is widely used to detect functional enzymes following electrophoretic separation in sodium dodecyl sulfate (SDS)-polyacrylamide gels. The method is a versatile two-stage technique involving protein separation by electrophoresis followed by the detection of enzyme activity in polyacrylamide gels under non-reducing conditions. It is based on SDS-polyacrylamide gel (PAG) copolymerization with substrates, which are degraded by the hydrolytic enzymes restored in enzyme reaction buffer after the electrophoretic separation. Any kind of biological sample can be applied and analyzed on zymography, including culture supernatants of microbes, plants extracts, blood, tissue culture fluids, enzymes in foods extracts and metaproteome. The advantage of zymography is that it is possible to directly detect the protein with activity on the electrophoretic gel as well as confirm the activity at the nanogram level. Thus, this zymography technology can be applied in various fields. However, these advantages are rather disadvantageous and can often lead to experimental errors. In this review, the advantages, disadvantages, and problem solving of zymography technique are described.

Antibiotics Resistance for Isolated Pathogenic Bacteria from Manufacturing Establishment of Sandwich Products (샌드위치 제조사업장에서 분리된 병원성세균에 대한 항생제 내성 검사)

  • The Collected 70 samples from 5 sandwich shops were analysed for the pathogenic bacteria such as Escherichia coli O157:H7, Salmonella spp., Listeria monocytogenes, and Staphylococcus aureus. As a result of Listeria monocytogenes and Saphylococcus aureus were detected in 1 sample, 11 samples, respectively. However, Escherichia coli O157:H7 and Salmonella spp. were not detected in anywhere. The antibiotics test of isolated bacteria was pelformed by the disk diffusion method from NCCLS. The resistance rate of Listeria monocytogenes isolates was confirmed 38.5% to 10 species such as Am, B, P, and Va for antibiotics of 26 species. MRSA was determinated 4 strains in S. aureus isolates. The resistance pattern of Staphylococcus aureus isolates were confirmed 36.4% to P Am OX B K E CXM, 18.2% to P Am B K E CXM B, 9.1% to P Am B K, 27.3% to P Am B, and 9.1% to Te B Nb. Therefore, continuous surveillance and monitoring for antibiotic resistance strains are demanded for prevention of increases in multiple antibiotic resistance strains.
