Screening for Antagonistic Natural Materials Against Alternaria alternata (Alternaria alternata에 항균력이 있는 천연물 조사)

  • 79종의 식물재료의 Alternaria alternata에 대한 항균활성을 조사한 결과, 7가지 식물재료가 항균활성이 있었고, 11가지는 확실치 않았으며, 나머지 61가지는 항균활성이 나타나지 않았다. A. alternata에 항균활성이 나타난 7가지 식물재료 중 계피가 활성이 가장 높았으며, 오미지와 천궁이 중정도의 활성을 보였으며, 나머지는 약한 활성을 나타내었다. 항균 활성 성분은 계피와 천궁의 경우는 diethyl ether에 분리되었고, 오미자의 경우는 butanol에 분리되었다. 계피의 diethyl ether 추출물은 1,000 배액 이하에서 식별 가능한 항균활성이 나타났다. 계피의 성분으로 cinnam aldehyde가 가장 높은 활성을 보여, 이 물질이 계피의 주요 항균물질로 사료된다.

Elicitation of Indole-3-ylmethyl Glucosinolate Biosynthesis in Turnip Culture Cells and Their Relationship with Plant Resistance to Botrytis cinerea (잿빛곰팡이병 추출물을 이용한 순무배양세포의 Indole-3-ylmethyl glucosinolate의 생합성유도와 병원성연구)

  • Two different races of Botryris cinerea were selected by the response of plant leaves to the pathogen infection. Based on lesion size of the pathogen on the leaves, turnip showed susceptible response to 'Grape-01' race, and resistant to 'Orange' race. Turnip leaves infected with resistant pathogen race, "Orange", showed significantly higher content of indole-3-ylmethyl glucosinolate (I3M) than those infected with susceptible race, 'Grape-01'. Contents of I3M in the leaves with resistant 'Orange' race was 2.5 times as high as that in uninfected leaves, whereas I3M in the leaves infected with susceptible 'Grape-01' race showed lower content than in untreated leaves. Growth of turnip suspension cells was significantly inhibited by the treatment of MeOH extract or water extract of 'Orange' race as compared with the treatment of susceptible race, 'Grape-01'. Treatment of MeOH or water extract from 'Orange' race to turnip suspension cells, strongly inhibited cell viability up to 22.7% or 16.5%, respectively. However, plant cells treated with MeOH or water extract from resistant race, 'Orange' showed higher I3M content than that from susceptible race, 'Grape-01'. These results suggest that accumulation and degradation of I3M glucosinolate in turnip cells closely related to the resistance and susceptibility of turnip cells to Botrytis cinerea.

Antioxidant Effects of the Methanol Extracts Obtained from Aerial Part and Rhizomes of Ferns Native to Korea (자생 양치식물 지상부와 근경 메탄올 추출물의 항산화 효과)

  • Phenolic compound contents and scavenging activity on DPPH and ABTS radicals in twelve fern species were studied by analysing ultrasonification extraction of dried aerial parts and rhizomes using methanol solvent. Total polyphenol content ranged from 2.97 to 140.1 mg per 1 g dried sample and 0.80 to $49.58\;mg{\cdot}g^{-1}$ fresh sample. Highest polyphenol content was obtained with aerial part of Davallia mariesii and Polystichum lepidocaulon. Total flavonoid content of dried sample was $2.56{\sim}34.91\;mg{\cdot}g^{-1}$ and fresh sample $0.71{\sim}11.49\;mg{\cdot}g^{-1}$. Higher flavonoids were obtained with dried aerial part of Dryopteris crassirhizoma, but with fresh aerial part of P. lepidocaulon. In general, total polyphenol content was higher in rhizomes, except in case of total flavonoid. Scavenging effects on DPPH and ABTS radicals was higher with rhizome extracts compared to aerial parts. The rhizome of Polystichum polyblepharum showed highest effects on both radicals. However, the amount of fresh sample for $RC_{50}$ with consideration of water content and extraction yield. P. lepidocaulon aerial part was more advantageous than P. polyblepharum rhizome. All the fern extracts except for the Coniogramme japonica extract demonstrated superior scavenging effects on ABTS radicals, being similar activity of ascorbic acid and BHT.

Physiological Activity of Five Kinds of Medicinal Plant Extracts with Various Solvents and Their Composites (약용식물 5종의 용매별 추출물 및 조성물의 생리활성)

  • This study was performed to investigate the antioxidative activity of solvent (water, 50% ethanol, and 100% ethanol) extracts from five kinds of medicinal herbs Cutellaria baicalensis Georgi; SB, Paeonia lactiflora Pall.; PA, Salvia miltiorrhiza Bunge; SM, Phellinus linteus; PH, Morus alba L.; MA). The total content of phenolic compounds was highest in the 50% ethanol extract from PH (280.05 mg/g), the 100% ethanol extract from PH (308.88 mg/g), and the water extract from SM (80.27 mg/g). The total content of flavonoids was highest in the 50% ethanol extract from SB (62.71 mg/ml), the 100% ethanol extract from SB (64.59 mg/ml), and the water extract from SM (35.85 mg/ml). ACE inhibitory activity only occurred in the water extracts, and it was highest in the water extract from SB (45.33%). Cholesterol adsorption activity was higher in the SB and PA extracts than in the other extracts. In water extracts, SM showed the highest antioxidative activity. Among the 50% and 100% ethanol extracts, DPPH radical scavenging activity and FRAP were highest in the PH extract, and ABTS radical scavenging activity was significantly higher in the PA extracts. Seven types of compositions were prepared with different mixing ratios of 0.2 to 2.0 from relatively high-activity medicinal herbs, such as PH, SM and PA. The total phenolic and flavonoid compound contents of the compositions were 50.53-61.96 and 16.91-33.81 mg/ml, respectively. Cholesterol adsorption activity was 46.27-70.03%.

Screening for Herbicidal Medicinal Plants against Digitaria Sanguinalis and Taraxacum Platycarpum in Turf (잔디밭 잡초 바랭이와 민들레 방제를 위한 살초활성 약용식물의 탐색)

  • Crabgrass (Digitaria sanguinalis L.), Dandelion (Taraxacum platycarpum H. DAHLST.), Annual bluegrass (Poa annua L.), Gree kyllinga (Kyllinga brevifolia var.), Red clover (Trifolium pratense L.), Annual lespedeza (Kummerowia striata (Thunb.)Schindl.), Mugwort (Artemisia capillaris Thunb.), Horseweed (Erigeron canadensis L.), Field horsetail (Equisetum arvense L.) are major weeds in turf. In this study, 23 medicinal plant extracts were tested for herbicidal activities against crabgrass and dandelion in the course of major weed in turf. In pot experiment, we implemented pre-emergence application used medicinal plant extracts showed herbicidal activity in petridish experiment. In effect, inhibition rates of germination were 61.0% at Curcuma longa extract and 62.5% at Cnidium officinale extract in crabgrass pot experiment, and 77.8% at Ailanthus altissima extract in dandelion pot experiment.

  • 전 세계 고등 식물자원은 약 350,000종, 한반도 식물자원은 약 4.500~5,000종 이라고 알려져 있습니다. 2018년 8월부터 국내에도 나고야의정서 발효되어 국내 산학연 연구자들의 해외생물자원 활용을 통한 연구개발에 장벽이 높아졌습니다. 저희 한국생명공학연구원에서는 지난 2001 ~ 2010년 동안 자생식물이용기술개발사업단을 운영하여 국내 유용식물자원을 확보하여 추출물 또는 건조시료 형태로 연구자들에게 분양을 해오고 있습니다. 또한 해외생물소재센터에서는 2006년부터 현재까지 해외 36개 국가와 국제공동연구협약을 체결 하여 ABS 규정에 따라 약 35,000종의 식물자원을 확보해 오고 있습니다. 또한 해외 4개 국가(중국, 베트남, 인도네시아, 코스타리카)에 거점센터를 구축하고 박사급 연구자를 파견하여 열대식물 자원전문가로 양성해오고 있습니다. 4개 거점센터에서는 국내 바이오 기업들의 산업화 지원을 위하여 야생종의 대량재배 시스템 구축 및 원료 표준화 작업을 지원해오고 있습니다. 현재까지 산학연 연구자들에게 누적 3,000,000점(1점 : 20mg)이상의 해외추출물을 분야하여 식물자원을 활용한 연구개발을 지원해 오고 있습니다.

Effects of Foreign Plant Extracts on Cell Growth and Biofilm Formation of Streptococcus Mutans (해외 자생식물추출물이 Streptococcus mutans의 세포 성장 및 생물막 형성에 미치는 영향)

  • Chemically synthesized compounds are widely used in oral hygiene products. However, excessively long-term use of these chemicals can cause undesirable side effects such as bacterial tolerance, allergy, and tooth discoloration. To solve these issues, significant effort is put into the search for natural antibacterial agents. The aim of this study was to assess the extracts of foreign native plants that inhibit the growth and biofilm formation of Streptococcus mutans. Among the 300 foreign plant extracts used in this study, Chesneya nubigena (D. Don) Ali extract had the highest antimicrobial activity relatively against S. mutans with a clear zone of 9 mm when compared to others. This plant extract also showed anti-biofilm activity and bacteriostatic effect (minimal bactericidal concentration [MBC], 1.5 mg/ml). In addition, the plant extracts of 19 species decreased the ability of S. mutans to form biofilm at least a 6-fold in proportion to the tested concentrations. Of particular note, C. nubigena (D. Don) Ali extract was found to inhibit biofilm formation at the lowest concentration tested effectively. Therefore, our results reveal that C. nubigena (D. Don) Ali extract is a potential candidate for the development of antimicrobial substitutes, which might be effective for caries control as well, as demonstrated by its inhibitory effect on the persistence and pathogenesis of S. mutans.

  • The methanol extracts from 30 species of oriental medicinal plants belonging to 24 families were tested for their lhicidal and antifeeding activit~es against diamondback moth (Plutello xylostella L) and tobacco cutworn (Spodoptera litura F.) by a leaf-dipping method at a concentration of 5, 000 ppm. The methanol extract from Copti chinensis only showed a potent larvicidal activity against P xylostello. Strong antifeeding activity against P. xylostello was observed from the extmds from Platycodon grandiflorurn, Codonopsis pilosula, Asomm sieboldii, Rhus chinensis and Uthospermum erythrorhizon And a potent antifeeding activity against S. liturn was obtained from Akebia quinata and Equlsetum hyemale extracts. A significant antifeeding activity against both species was obtained from R chinensis and C. chinensis extracts.

Antioxidative Activities and Inhibitory Effects on Tyrosinase and Elastase by Water Extracts of Allium sp. (Allium 속 식물 물 추출물의 항산화 활성과 Tyrosinase 및 Elastase 저해 효과)

  • Three different Allium plants, including wild garlic (Allium monanthum Max; AMM), onion (Allium cepa L.; ACL) and green onion (Allium fistulosum L; AFL), have been widely used as spicy food in daily life. This study was attempted to investigate the properties of antioxidant, whitening and anti-wrinkle activities in water extracts in Allium plants. The antioxidant, whitening and anti-Wrinkle activities were evaluated by performing DPPH and ABTS radical scavenging activities, inhibitory effects on tyrosinase, and elastase, respectively. The water extract (10 mg/mL) of onion, wild garlic and green onion exhibited DPPH radical scavenging activities of 28.46%, 28.45%, and 15.91%, respectively. The inhibitory effects of tyrosinase activities by wild garlic, green onion, and onion were increased by heating the extracts at $90^{\circ}C$. Additionally, the wild garlic extract showed higher elastase inhibitory activity than those of other plant extracts. These results suggest that water extracts of Allium plants may be useful as potential sources of beauty foods due to higher antioxidant, anti-melanin and anti-wrinkle activities.
