Screening of α-Amylase, α-Glucosidase and Lipase Inhibitory Activity with Gangwon-do Wild Plants Extracts (강원도 자생 산채 추출물의 α-Amylase, α-Glucosidase, Lipase 효소 저해활성 탐색)

  • We investigated ${\alpha}$-amylase, ${\alpha}$-glucosidase and lipase inhibitory activity of extracts collected from wild plants in Gangwon-do. 90 wild plants were collected and their water and ethanol extracts were obtained. Results of measuring ${\alpha}$-amylase inhibitory activity indicated more than 80% of activity inhibition in 10 mg/mL concentration for ethanol extracts of three plants and water extracts of two plants. For ${\alpha}$-glucosidase inhibitory activity, ethanol extracts of thirteen plants and water extracts of three plants showed more than 80% of activity inhibition in 10 mg/mL concentration. In the experiment of inhibiting lipase activity, ethanol extracts of seven plants and water extracts of one plants showed above 80% of activity inhibition in 10 mg/mL concentration. These results suggest that the selected extracts could be potentially used as a resource of bioactive materials for health functional foods.

Antioxidant Activities of Medicinal Plant Extracts (약용식물 추출물의 항산화 효과)

  • The antioxidant activities of ethanolic extracts obtained from medicinal plants (Scutellaria baicalensis Georgi, Acanthopanax sessiliflorum Seeman, Pueraria lobata Ohwi, Portulaca oleracea Linne, Crataegus pinnatifida Bunge var. typica Schneider, Euonymus alatus Apterus, Hovenia dulcis Thunberg, Prunus yedoensis Matsumura, Albizzia julibrissin Durazz., Chrysanthemum indicum Linne) were evaluated for total phenolic content, total flavonoid content, DPPH radicals, nitrites, $Superoxide^-$ radicals, $ABTS^+$ radical scavenging activity, and reducing power. Antioxidant capacities were the highest in Prunus yedoensis Matsumura for DPPH radical scavenging activity ($IC_{50}$ $5.39{\mu}g/mL$), reducing power (2.72, $A_{700}$), and nitrite scavenging activity ($IC_{50}$ $167.94{\mu}g/mL$). Hovenia dulcis Thunberg and Acanthopanax sessiliflorum Seeman were effective for their nitrite scavenging activities (over 90% at 1 mg/mL). The $Superoxide^-$ radical scavenging activity of Prunus yedoensis Matsumura ($IC_{50}$ $43.39{\mu}g/mL$) was stronger than tannic acid ($IC_{50}$ $46.51{\mu}g/mL$). Five samples (Prunus yedoensis Matsumura, Acanthopanax sessiliflorum Seeman, Hovenia dulcis Thunberg, Crataegus pinnatifida Bunge var. typica Schneider, Scutellaria baicalensis Georgi) were effective for their $ABTS^+$ radical scavenging activity (more than 90% at 0.5 mg/mL). These results suggest that the ethanolic extracts of Prunus yedoensis Matsumura could be used as a functional ingredient in food products.

Anti-oxidant and Anti-inflammatory Activities of Beet (Beta vulgaris) Sprout Extract (비티 새싹 추출물의 항산화 및 항염효과)

  • 본 연구에서는 비트 새싹 추출물의 항산화 및 항염증 효능을 평가하기 위해 3가지 품종의 레드 비트(메를린, 우단, 디트로이트다크레드) 새싹을 60% 에탄올로 추출한 후 free radical 소거활성, 총 폴리페놀 함량을 측정하였다. 또한, LPS로 염증반응이 유도된 대식세포인 RAW264.7 세포에서 nitric oxide(NO) 생성억제 효과를 알아보았다. 메를린, 우단, 디트로이트다크레드 새싹 추출물의 총 폴리페놀 함량을 querceitn을 표준물질로 사용하여 측정한 결과 2.84 mg Que/g, 2.61 mg Que/g, 2.71 mg Que/g으로 나타났고 DPPH 라디칼 소거능은 농도 의존적으로 증가하였으며, 200 ㎍/ml일 때, 55%, 72%, 63%로 나타났다. 또한, DPPH 라디칼 소거능이 가장 높은 메를린 새싹 추출물은 LPS로 염증반응이 유도된 RAW264.7 세포에서 농도 의존적으로 iNOS와 COX-2를 억제하는 것을 확인하였다. 이러한 결과를 바탕으로 메를린 비트 새싹 추출물이 항산화 및 항염증제의 천연물 소재로 활용이 가능할 것으로 생각된다.

Effects of Deodorizing Plant Products on the Odor and Fermentation Characteristics of Kimchi (소취식물이 김치 냄새특성 및 발효성에 미치는 영향)

  • The use of deodorizing plant products such as persimmon leaves, pine needles and fennel seeds to mask the characteristic odor of kimchi has been proposed. Therefore, in this study, the effect of deodorizing plants on kimchi fermentation and the profile of volatile flavor compounds was investigated. During sensory evaluation tests of sour and fermentation odors of kimchi, plant extract-added kimchi showed higher sensory scores than chopped plant-added kimchi. Additionally, kimchi containing pine needle extracts (0.3-0.6%) and fennel seed extracts (0.05-0.1%) showed higher sensory scores than untreated kimchi. In addition, pine needle extract and fennel seed extract had a significant influence on kimchi fermentation characteristics such as pH, total acidity and the concentration of lactic acid bacteria. The volatile flavor compounds of kimchi and deodorizing plant extracts were also analyzed using GC/MS and the correlation of volatile flavor compounds in kimchi, plant extracts and plant-added kimchi was examined. The results revealed that the sour and fermentation odors of kimchi were reduced using deodorizing plant extracts by changing the kimchi fermentation characteristics.

The Antioxidative Effects of the Water-Soluble Extracts of Plants Used as Tea Materials (다류원료 식물류 물 추출물의 항산화 효과)

  • The water-soluble extracts of plants used as tea materials were investigated for their antioxidant activities and active components(total phenolics, ascorbate and selenium). Antioxidant activities of the plant extracts were determined by measuring the changes in both peroxide values(POV method) during storage(36 day, $55^{\circ}C$) and conductivity of soybean oil at $110^{\circ}C$(Rancimat method). Soybean oil without any additive was used as a control. Soybean oil treated with 0.02% BHT was used as a positive control. The test samples were prepared by mixing the plant extracts with soybean oil in 0.02% concentration by weight. The water-soluble extracts of lycii fructus(23 d), oolong tea(23 d), orange peel(23 d), citron(22 d), and apricot(22 d) showed longer induction periods, compared to control(21 d) and BHT(21 d) by POV method. Also water-soluble extracts of oolong tea(12 h), instant coffee(11 h), citron(10 h), cinnamomi cortex(10 h), schizandrae fructus(10 h), lycii fructus(10 h) and apricot(10 h) demonstrated longer induction periods, compared to control(8 h) and BHT(8 h) by Rancimat method. The contents of total phenolic compounds were observed to be high in water-soluble extracts of oolong tea, green tea, black tea, coffee, cinnamomi cortex, and cassiae semen. Ascorbate contents were found to be high in coffee, eucommiae cortex, black tea, ganoderma, cinnamomi cortex, persimmon leaf, chicory, green tea and oolong tea extracts. The selenium contents were not detected in all the extracts. The antioxidative effects of some plant extacts were seemed to be the combined effects of various antioxidant components such as phenolics and ascorbate.

  • This study was to evaluate the efficiency of different solvent systems to extract active ingredients from different parts of medicinal plants used as oriental medicinal herb in the cosmetic industry. We tested efficacies related to the antioxidative effects and UV absorption of herbal extracts as complex of active ingredients, not each single ingredient. When extracting medicinal plant which is used effective medicine, we should consider the collecting conditions like collecting time, place, part and extracting conditions like solvent, temperature, time, and etc. Among them, extraction solvent and collecting part are very important factors for the efficacy. As increased interest in medicinal plants, more intensive studies on collecting conditions and extracting processes were needed for the developments in the herbal ingredient industry. We evaluated the efficiency of different solvent systems to extract active ingredients from different parts of plants (Terminalia chebula, Syzygium aromaticum, Paeonia lactiflora, Morus alba, Scutellaria baicalensis) widely used as medicine and food. As results, we found that proper condition can make better data and decrease the required quantity.

  • 중국(中國) 장홍화(藏紅花)는 해발 1500m~3000m에 자생하는 고원식물이다. 장홍화는 여성질환예방과 두뇌의 신경안정, 호르몬분비 촉진, 학습증진과 기억력향상에 의한 알츠하이머 치료의 효과가 있는 것으로 알려져 있으며, 함유되어 있는 크로신은 암세포를 파괴시키어 종양세포를 줄이는 작용을 하며, 카로티노이드 성분이 항산화 활성과 면역을 증진시키어 항암효과를 가져다 준다. 또한 다량의 칼륨을 함유하고 있어서 나트륨배출에 효과적이어서 혈압을 떨어뜨리는 작용을 한다. 또한 신경전달물질의 활동을 증진시키어서 스트레스 해소와 치매 및 인지력을 높여준다고도 알려져 있다. 본 실험에서는 장홍화(藏紅花)추출액의 총 폴리페놀 함량과 항산화 물질 활성을 조사했던 바, 장홍화(藏紅花) 메탄올과 에탄올추출액에서 항산화 반응이 매우 양호하였다. 또한 총 폴리페놀 함량도 에탄올 추출물과 메탄올 추출물 처리구에서도 비슷한 결과를 나타내었다. 이러한 결과들로 미루어 볼 때 장홍화(藏紅花)의 추출액은 건강음료와 화장품의 기능성원료로도 이용이 가능할 것으로 추측되었다.

A Study on Pectin Extraction from Apple Cell Wall by Enzyme (효소에 의한 사과 세포벽 펙틴 추출)

  • Two methods were used to extract pectin from apple cell wall by enzyme(Hemicellulase) and by acid. Hemicellulase was used to extract high functional pectin with higher degree of polymerization. The yield of weight of pectin by hemicellulase treatment was slightly higher than that by acid treatment. The optimal condition for extraction was accomplished by providing 1.5 grams of Hemicellulase at 38$^{\circ}C$ for 24 hours. The comparison of the pectin purity and the yield of extracts by the portion of galacturonic acid between two methods showed that the purity of pectin extracted by enzymatic method was lower than that by acid treatment.

Screening of Natural Antimicrobial Edible Plant Extract for Dooboo, Fish paste, Makkoli Spoilage Microorganism (식용 식물로부터 얻은 추출물의 두부, 어묵, 막걸리 변질균에 대한 항균성 검색)

  • To extend the self-life of Dooboo (tofu), fish paste and Makkoli (Korean traditional rice wine) for which severe heat treatment are not expectable, main putrefactive microorganisms were isolated from each product and tested their growth inhibition by ethanol and water extracts of several edible plants. The ethanol extract of Phellodandron amurense Ruprs had the strong effect of growth inhibiting to all three isolates from Dooboo and Makkoli, and Eugenia caryophyllus, Pinus rigia Mill, Bletilla striata (Thunb) Reichb. Fill and Paeonia albiflora Pall were also same effect to isolates from fish paste. The ethanol extract was more effective inhibiting than water extract in all test microorganisms. The evident inhibition level of each extract was 2000 ppm of Pa ethanol for fish paste and 1000 ppm for Makkoli.

Effect of Ainsliaea acerifolia Extract on Anti-acetylcholinesterase and Peroxynitrite Scavenging and Analysis of Caffeoylquinic Acids (단풍취 추출물의 Acetylcholinesterase 저해 및 Peroxynitrite 소거에 대한 활성과 Caffeoylquinic Acid 성분의 분석)

  • Anti-acetylcholinesterase (anti-AChE) activity and peroxynitrite scavenging activity of Ainsliaea acerifolia (Compositae) were studied to find its anti-Alzheimer's activity. The $IC_{50}$ was $73.4{\mu}g/mL$ in AChE assay and $8.60{\mu}g/mL$ in peroxynitrite assay. Caffeoylquinic acids that have been reported to have anti-Alzheimer's activity were analyzed in the leaf- and stem extract by HPLC. Caffeoylquinic acids occupied 25.1% of the leaf extract which was higher than 8.1% of the stem extract. Particularly, the content of 3,5-dicaffeoylquinic acid (145.6 mg/g) was highest of the tested caffeoylquinic acids. In addition, the $IC_{50}$ values of 3,5-dicaffeoylquinic acid were shown to be $3.79{\mu}g/mL$ in peroxynitrite assay and $69.19{\mu}g/mL$ in anti-AChE assay.
