In vitro Plant Regeneration and Genetic Stability of the Regenerants in Wolly Grass (Imperata cylindrica 'Rubra') (홍띠(Imperata cylindrica 'Rubra') 기내식물체 재생과 재분화 식물체의 유전적 안정성)

  • 홍띠(Imperata cylindrica 'Rubra') 식물자원의 생장점 부위를 기내 배양하여 기내 식물체 재분화와 재분화식물체의 유전적 안정성을 검토하였다. 기내배양은 26±2 ℃, 25 μmol/m2/s, 14h/10h (day/night) 광조건 하의 배양실에서, MS (Murashige and Skoog, 1962) 기본배지에 생장조절물질을 첨가하여 조직절편체로부터 식물체를 유도하였다. 캘러스는 MS기본배지에 0.1 mg/L의 2,4-D와 2 mg/L의 BA를 혼용처리하여 생장점 부위로부터 유도하였다. 캘러스 증식은 MS기본배지에 0.1 mg/L의 2,4-D를 첨가한 배지에서, 이들 캘러스로부터 신초 재분화는 0.01 mg/L의 NAA 및 2 mg/L의 BA를 첨가한 배지에서 유도하였다. 다경줄기 형성(multiple shooting) 후 MS배지에서 4주 동안 배양한 재분화식물체는 멸균한 상토(버미큘라이트)를 포함한 배양병에서 7주간 배양한 다음 점차적으로 배양병 뚜껑을 개방(1/10 정도 1차 개방 1주일, 3/10 정도 2차 개방 2주일)하여 직경 6 cm의 컵포트에 이식하여 활착시켰다. 재분화식물체는 붉은색이 사라지고 녹색을 나타내었으며, 일부개체에서만 잎의 일부분만 붉은색을 나타냈다. 이는 생장점 주변조직에서 유래한 재분화체가 우세함으로써 생장점보다는 생장점 주변의 조직을 구성하는 녹색층에서 주로 식물체가 재생되는 것으로 판단된다. 이에 따라 홍띠 대조구식물체 8개체, 활착한 녹색 재분화 식물체 20개체(실내 재배중인 순화체 10개체, 2020년 6개월간 포장에서 재배중인 순화체 10개체)를 대상으로 ISSR분석을 실시하여 재분화식물체의 유전적 안정성을 검토하였다. 향후 조직학적 측면에서 신초재분화의 기원에 대한 검토가 필요하다고 사료된다.

Growth promotion and root development of Nicotiana tabacum L. by plant growth promoting fungi (PGPF) (식물 생장 촉진 진균에 의한 담배의 생장 촉진과 뿌리 발달)

  • Plant growth-promoting microorganisms promote plant growth by supplying nutrients to roots and interacting with the intrinsic factors in plants through volatile organic compounds (VOCs). In this study, we evaluated the effect of UOS, plant growth-promoting fungi (PGPF) isolated from previous study, on the growth of Nicotiana tabacum L. var Xanthi nc. Phylogenetic analysis and GC-MS were used to identify the fungal species and the VOCs emitted by the UOS, respectively. The fresh weight of UOS-treated Nicotiana tabacum L. was 3.8 and 4.2-fold higher than that of the control groups grown in vertical and I-plates, respectively. Moreover, in the UOS-treated plants, the length of the primary root was half and the number of lateral roots were twice compared to those in control plants. The UOS was identified as Phoma sp. by studying spore and mycelial morphology and using phylogenetic analysis. GC-MS revealed that the VOC emitted by the UOS was hexamethylcyclotrisiloxane (D3). These results suggest that the UOS of Phoma sp. influences plant growth and root development through D3. We expect this UOS and its VOC, D3 to be utilized in the future to increase growth and enhance yield for other plants.

  • 생장조정제 사용에 앞서 작물재배과정에서 꼭 알아둘 점이 있다면 (1) 생장조정제는 재배환경에 민감하게 작용하기 때문에 온도, 습도, 광등을 고려하여 정상생육 환경이 되도록 재배환경을 잘 맞춰줘야 약제발현 효과가 있으며 (2) 토양환경면에서 세포생리 기능이 정상적으로 분화발육 할 수 있을 때 약제 발현성을 높이기 때문에 비배관리면에 특히 유념해야 한다는 것이다.

  • 본 논문에서는 식물공장에서 사용할 수 있는 파장별 광원제어가 가능한 조명장치의 설계하고 구현하고자 한다. 식물의 생장에 필수적인 광원의 파장에 따라 생장에 다양한 영향을 미치게 된다. 따라서 본 논문에서는 식물 생장에서 필요한 다양한 파장의 빛을 공급할 수 있는 식물공장용 조명시스텡을 개발한다. 조명시스템은 마이크로 컴퓨터를 이용하여 적, 청, 녹색 3색 LED를 PWM제어하여 광량과 세기를 제어하고, 공급된 광량을 적산하여 조사된 광량을 확인할 수 있도록 구성하였다. 본 조명 시스템은 태양광 이용형 식물공장의 보조광원으로 사용 가능하며, 완전제어형 식물공장에서 사용이 가능하다.

Studies on the Allelopathy of some Poaceae Plants (수종 화본과식물의 Allelopathy에 관한 연구)

  • This investigation includes the sick soil phenomenon caused by the self-poisoning of Setaria italica, Sorghum nervosum, Zea mays and Miscanthus sinensis among Poaceae. It elucidates whether the poison is directly excreted from the root or the secondary product resulting from the decomposition in the soil; the effect of Miscantus sinensis on the germination and growth of other plants, and the effect of Zea mays grown between furrows to shade Angelica gigas on its growth. The results obtained are as follows; Supplied with the leakage water from the pots, in which the same plants as the test ones were grown, in anticipation of the poison to be directly excreted from their roots, Sataria itlaica and Zea mays exhibited the growth inhibition more than 30%, whereas Sorghum nervosum and Miscanthus sisnensis were not effected in growth at all. When cultivated in the soils mixed with the roots of the some plants as the test ones, in anticipation of the poison to be the secondary product resulting from the decomposition in soil, Setaria italica and Zea mays showd growth inhibition of more than 50%, which is greater than that of the case of the leakage water, and Miscanthus sinensis exhibited no inhibition either, whereas Sorghum nervosum in the 50% plot showed heavy growth inhibition of more than 80% to the case of the leakage water. The common or uncommon plants found easily in the group of Misscanthus scinenis were not affected by the extracts of the steam and leaves of Miscanthus sinensis in germination and growth. Supplied with the leakage water from the pots in which Miscanthus sinensis was grown, among Lespedeza crytobotrya, Oenothera odorata, Raphanus sativus val'. acarlthiformis, Zoysia japonica, Patrinia scabiosaefolia. which are easily found in the group of Miscanthus sinensis, only Patrinia scabiosaefolia was slightly inhibited in growth in the 100% plot, whereas the others did not show any inhibition at all. Mean while, Amaranthus patulus. Solanum nigrum, Capsella bursa-pastoris val'. triangularis, Alopecurus amurensis, Chenopodium album val'. centrorubrum, which could not be found in the group of Miscanthus sinensis, were all distinctly inhibited. In the experiment on the effect of Zea mays on the growth of Angelica gigas, its growth was severely inhibited by one-half to two thirds with the increased concentration in both the cases of growing in the mixture of the soil and the powdered root of Zea mays and being supplied with the leakage water from the pot in which Zea mays was grown.

Effects of Simulated Acid Rain on the Shoot Growth and Internal Tissue of Arabidopsis thaliana (애기장대의 shoot 생장과 내부조직에 미치는 인공산성비의 영향)

  • This experiment was carried out to investigate the effects of simulated acid rain (SAR) in the shoot growth and internal structure of Arabidopsis thaliana. In the shoot growth, the plants treated with SAR (pH 3.0) for 15 days showed no morphological change compared to the control plants. Some change was observed in the internal structure of the stems: the epidermis and cortex tissues of the stems treated with SAR were partly damaged. The plants treated with SAR showed no noticeable difference compared to the control plants, but morphological changes were observed in the leaf. The leaves of the plants treated with SAR showed many white necrotic spots on the part of upper epidermis. A light microscopic examination of the leaves with necrotic spots showed that the upper epidermis was severely compressed with the damaged cuticle layer and the mesophyll cells were also damaged and compressed. However, noticeable structural change of vascular bundle cells was not observed.

Effects of Plant Growth Regulators on the Growth and Flowering of Pot Pelargornium hortorum (분식(盆植) 제라니움의 생육(生育) 및 개화(開花)에 미치는 식물생장조절제(植物生長調節濟)의 효과)

  • This study was carried out to investigate the effects of plant growth regulators on the growth and development of pot 'Pinto Rose' and 'Pinto Scarlet' geraniums. Plant height was increased by $GA_3$ treatments in both 'Pinto Rose' and 'Pinto Scarlet' geranium. However, the number of branches was not affected by $GA_3$ and daminozide treatments. The combined treatments of $GA_3$ and daminozide delayed the days to first flowering in 'Pinto Scarlet' geranium. Also, in 'Pinto Rose', $GA_3$ or the combination of $GA_3$ and daminozide treatments delayed the first flowering time. 'Pinto Scarlet' geranium was more sensitive to growth regulator treatments than 'Pinto Rose'. The number of flower stalks and branches were increased by uniconazole in 'Pinto Scarlet', while the flower stalk length and canopy diameter were severely decreased by uniconazole treatments in both cultivars. The days to first flowering tended to be delayed by all growth retardant treatments in geranium.
