Occurrence of Bacterial Soft Rot of Melon Caused by Erwinia carotovora subsp. carotovora (Erwinia carotovora subsp. carotovora에 의한 메론의 세균성무름병 발생)

  • Water-soaked spots or lesions were observed on fruits or stems of melon plants at house-melon farms in Andong, Korea. The symptoms developed to soft rot of the fruits of wilting of the whole plants. The causal organism isolated from the water-soaked le-sions was identified as Erwinia carotovora subsp. carotovora based on the morphological and physiological characteristics. The causal bacterium was susceptible to not only two kinds of medical antibiotics but also two kinds of agrochemicals tested. Since the bacterial soft rot is a first described bacterial disease in melon in Korea, we propose to name the disease as "bacterial soft rot of melon".

Suppression of Bacterial Soft Rot on Chinese Cabbage by Calcium Fertilizer Treatment (칼슘비료 처리에 의한 배추 무름병 발생 억제)

  • Bacterial soft rot by Erwinia carotovora subsp. carotovora is one of the diseases causing the biggest damages in Chinese cabbage cultivation. This study was conducted to evaluate on suppressive effect of calcium fertilizer to bacterial soft rot of Chinese cabbage. Seven calcium fertilizers were selected for evaluation. And screening was conducted to select effective agents for controlling bacterial soft rot. When applied by the nursery test condition using mineral oil inoculation method with Chinese cabbage, calcium hydroxide had more suppressive efficacy than any other calcium fertilizer, While nitrogen fertilizer was induced the disease, calcium hydroxide was suppressed soft rot disease in field test as well as seedling test. Treatment of calcium+nitrogen fertilizer as well as calcium only showed a significant control effect in the field experiment with Chinese cabbage 'Sanchon' in 2003.

미국에 땅콩 줄무늬 바이러스병 발생

  • 땅콩 줄무늬 바이러스(peanut stripe virus, PSV)는 땅콩의 가장 피해가 큰 병 중 하나이며, 기타 중요한 두과(대두, cowpeas, arrowhead, crimson clovers)작물에도 피해를 입힌다. 최근에 이 바이러스는 중공에서 수입한 땅콩종자 유전자원(germ plasm)에 감염되어 있다는 사실을 발견하고 미국의 식물병리학자들이 세밀한 조사시험연구를 한 결과 미국에도 이미 이 바이러스가 발생되었다고 보고하였다. 그 결과 미국 동식물검역국(Animal and Plant, Health Inspection Service)은 최근에 중공으로부터 종자용을 포함한 땅콩(Arachis species)수입을 금지하기에 이르는 등 식물검역상 중요한 문제로 대두하고 있다.

  • Organic farming is necessary to sustainable agriculture, preserve biodiversity and continued growth the sector in agriculture. In organic farming, reduced usage of chemical agents that adversely affect human health and environment, employing amino acids and oil cake fertilizer, plant extracts, and microbial agents are used to provide safe agricultural products to consumers. To investigation microbiome structure, we proceeded on the pepper plant with difference fertilizers and treatments in organic agriculture for three years. The microbial communities were analyzed by the next generation sequencing approach. Difference soil microbiota communities were discovered base on organic fertilizer agents. Occurrences of virus and anthracnose diseases had a low incidence in conventional farming, whereas bacteria wilt disease had a low incidence in microbial agents treated plots. Microbe agents, which applied in soil, were detected in the microbial community and the funding suggested the applied microbes successfully colonized in the organic farming environment.

  • The medicinal plants of 33 species with symptoms of mosaic, necrosis, line pattern, malformation (fine leaf), stripe, and ring spot infected by cucumber mosaic virus spontaneously, were collected in Korea and Japan. Among them, 2 mosaic diseases of portulaca oleracea L. and Celocia argentea L., 2 necrotic mosaic diseases of Aristolochia debilis Sieb. and Tetragonia exponsa Murr., 6 mottle diseases of Cassia torosa Cav., Bupleurum falcantum L., Angelica acutioloba Kitagawa, A. keiskei Koidz., Peucedanum japonicum Thunb., and Foeniculum vulgare Mill., and ringspot disease of Basella rubra L., were named newly in these studies.

  • Septoria brown spot caused by Septoria glycines Hemmi is one of the serious fungal diseases in soybean. Since little has been known about the disease progress in the field, the present study was conducted to determine the factors affecting the disease progress in the soybean plant. Disease severity and pattern of the progress of the Septoria brown spot were different with varieties. Susceptibility of soybean plants increased with increase of plant age and leaf maturity in order from the primary leaf to the newly expanded leaf. It seemed to be related with conidial germination on the leaves. Germination and germtube elongation were more inhibited by the diffusates obtained from upper leaves than those from lower leaves and they were higher in a susceptible variety than in a moderately resistant one.

Stem Blight of Brunfelsia Caused by Fusarium oxysporum (Fusarium oxysporum에 의한 브룬펠지아 줄기마름병)

  • Stem blight of brunfelsia (Brunfelsia calycina) caused by Fusarium oxysporum was found in greenhouse around Sungnam area, Kyunggi province, Korea in September 2001. The initial infection appeared as a slight wilting of the foliage, turned yellow from the lower leaves. The yellowing leaves were falled, resulting in blight of stem and eventual death of the entire plant. The vascular tissue of a diseased plants became dark brown and browning of the vascular system was a characteristic of the disease and the pith remained healthy, Isolates obtained from the lesions of the diseased plant were identified as F. oxysporum, based on the morphological characteristics of conidia. Symptom by artificial inoculation was same to the symptom of naturally infected plants. This is the first report demonstrating the stem blight on a brunfelsia caused by F. oxysporum in Korea, and we proposed to name this disease "stem blight of brunfelsia".

Disease Management in Road Trees and Pepper Plants by Foliar Application of Bacillus spp. (Bacillus spp. 엽면살포에 의한 가로수 및 고추의 병 방제)

  • Out of plant-associated bacteria, certain plant growth-promoting bacteria (PGPB) have been reported to increase plant growth and productivity and to elicit induced resistance against plant pathogens. In this study, our objective was to broaden the range of applications of leaf-colonizing PGPB for foliar parts of road tress and pepper. Total 1,056 isolates of endospore-forming bacteria from tree phylloplanes were collected and evaluated for the enzymatic activities including protease, lipase, and chitinase and antifungal capacities against two fungal pathogens, Colletotrichum graminicola and Botrytis cinerea. Fourteen isolates classified as members of the bacilli group displayed the capacity to colonize pepper leaves after spraying inoculation. Three strains, 5B6, 8D4, and 8G12, and the mixtures were employed to evaluate growth promotion, yield increase and defence responses under field condition. Additionally, foliar application of bacterial preparation was applied to the road tress in Yuseong, Daejeon, South Korea, resulted in increase of chlorophyll contents and leaf thickness, compared with non-treated control. The foliar application of microbial preparation reduced brown shot-hole disease of Prunus serrulata L. and advanced leaf abscission in Ginkgo biloba L. Collectively, our results suggest that leaf-colonizing bacteria provide potential microbial agents to increase the performance of woody plants such as tree and pepper through spray application.

Development of DNA markers linked to resistant gene to Psmodiophora brassicae Woronin in Chinese cabbage (배추무사마귀병 저항성 유전자와 연관된 DNA 마커개발)

  • 배추무사마귀병 저항성 유전 양식을 증명하기 위해서 CR계 F1에서 유래된 F2 세대를 포장시험과 유묘 검정을 실시하였다. F$_2$ 세대의 7 집단은 단인자우성으로 3:1의 분리비를 보였고, 5 집단은 중복 유전자가 관여하는 9:7의 유전 분리비를 보였다. 배추무사마귀병 저항성 유전자와 연관된 DNA 마커를 개발하기 위하여 CR-Saerona F$_2$ 집단을 배추무사마귀병 발병포장에서 재배하여 저항성 평가를 하였다. 220개의 임의의 프라이머를 이용하여 BSA-RAPD (Bulked segregant analysis-Randomly amplified polymorphic DNA)를 수행하였지만 CR-Saerona F2 집단에서 배추무사마귀병 저항성 유전자와 꼭 들어맞는 DNA 마커는 발견되지 않았다. 300개의 임의의 프라이머를 이용하여 CR-Saerona에서 유래된 F$_2$ 세대를 QTL 분석하였다. 저항성 정도는 발병지수에 따라 조사되었고 QTL 분석을 위해 one-way ANOVA 테스트를 하였다. 통계분석 결과 두 프라이머(K16-1, L2-2)가 저항성과의 상관관계를 보여 주었으나 유의성은 인정되지 않았다.
