Plant phenomics is a technique for observing and analyzing morphological features in order to select plant varieties of excellent traits. The conventional methods is difficult to apply to the phenomics system. because the color threshold value must be manually changed according to the detection target. In this paper, we propose the convolution neural network (CNN) structure that can automatically segment plants from the background for the phenomics system. The LeafNet consists of nine convolution layers and a sigmoid activation function for determining the presence of plants. As a result of the learning using the LeafNet, we obtained a precision of 98.0% and a recall rate of 90.3% for the plant seedlings images. This confirms the applicability of the phenomics system.

본 연구는 각시톱시네고사리(Dryopteris hangchowensis Ching)의 전엽체 증식 및 포자체 형성을 위한 번식방법을 구명하고자 수행되었다. 각시톱지네고사리의 식물체를 무가온 온실에 재배하면서 포자를 채취하였으며, 수집된 포자는 기내에서 발아시켜 전엽체를 획득한 다음 계대배양하면서 실험의 재료로 사용하였다. 전엽체 대량증식을 위하여 1/4, 1/2, 1배로 조절한 MS배지와 Knop배지를 조성하였다. 이후 전엽체 300mg을 균일하게 다져 조성된 배지에 8주간 배양하였다. 연구의 결과, 전엽체의 생체중은 1/2MS와 1/4MS에서 3.4g으로 가장 많이 증가하였다. 현미경 관찰의 결과, 두 배지 모두 정상적인 전엽체의 형태로 발달하였으며, 생육도 양호하였다. 포자체 형성을 위한 적정 배양토의 혼합조건을 확인하고자 원예상토, 피트모스, 펄라이트 및 마사토의 혼합비율을 5종류로 달리하여 조성하였다. 준비된 배양토를 사각분($7.5{\times}7.5{\times}7.5cm$)에 충진 후 기내 배양한 전엽체 1g과 증류수를 함께 10초간 분쇄하여 토양표면에 균일하게 분주한 다음 12주간 재배하였다. 연구의 결과, 원예상토, 피트모스, 펄라이트를 1:1:1(v:v:v) 비율로 혼합한 토양에서 포트 당 223.7개로 가장 많은 포자체가 형성되었다. 또한 형성된 포자체의 생육을 조사한 결과, 상기 토양에서 생체중, 엽수, 엽장, 엽폭, 근수, 근장 및 SPAD value 등의 생육수치가 우수하였다. 따라서 각시톱시네고사리의 전엽체 증식에 적합한 배지는 경제성과 생육수준을 고려하여 1/4MS 배지가 적합하며, 포자체 대량생산을 위해서는 원예상토, 피트모스, 펄라이트를 1:1:1(v:v:v)로 혼합한 토양이 적합하다고 판단되었다.
To investigate the function of chloroplast small HSP, transgenic tobacco plants (Nicotiana tabacum L. cv. Samsun) that constitutively overexpress the chloroplast small HSP (NtHSP21) from N. tabacum cv. Petit Havana SR1 were generated. Five homozygous lines of transformants showing different constitutive expression levels of the NtHSP21 were selected. To determine whether constitutive overexpression of NtHSP21 protein affects thermotolerance, wild-type and transformants were grown in Petri dishes, heat-stressed at 52$^{\circ}C$ for 45 min, and then incubated in normal growth condition. When heat-stressed wild-type plantlets were incubated at $25^{\circ}C$, leaf color gradually became white and all trio plantlets finally died within a week. As for the transformants, however, more than 70% of them remained green and survived under the conditions in which all the wild-type plants were dying. It was also found that the levels of NtHSP21 were correlated with the degree of thermotolerance. These results suggest that the NtHSP21 protein in transformants is responsible for the increase in thermotolerance.

Cotyledonary abnormalities were observed in secondary somatic embryos which were developed from calli cultured on MS medium with various concentrations of 2,4-D. In MS medium containing hormone-free or $0.1mg/{\ell}$ 2,4-D, the frequency of normal embryo formation with two cotyledons were above 57%. According to concentration of 2,4-D increment the frequency of normal embryos were decreased. In MS medium containing $4mg/{\ell}$ 2,4-D, the frequency of normal embryo formation was just 10%. The rate of germination was as follows; 37.7% of somatic embryos had one cotyledon, 85% two cotyledons, 38% three cotyledons, 35% four cotyledons, 25% five cotyledons and 29% trumpet-like cotyledons. About 80% of the embryos with two cotyledons were converted into normal plants, but one, three or four cotyledons were only 6.8~10%. The five or trumpet-like embryos were not developed into normal plants.
Immature flower buds of 'Desio' carnation were cultured on MS agar medium supplemented with 1 ㎎/L 2,L-D. Embryogenic calli were formed from 5-10% of the buds less than 20 ㎜ in length, but only non-embryogenic calli were produced from explants of shoot apex leaf, internode, and flowere buds larger than 20 ㎜. The same method was applied to 16 cultivars of cut Sower carnation and embryogenic calli were obtained in 7 cultivars. Several embryogenic callus lines were selected and maintained through subcultures over 120 weeks without loss of embryogenic competence. The embryogenic cultures were also proliferated rapidly in liquid agitation cultures using MS medium supplemented with 1mg/L 2,4-D. Numerous embryos were formed on the periphery of the cell aggregates upon transfer to auxin-free MS agar medium. Plantlets were transplanted in potting soil and grown to bloom in six months.
The development of selectable markers for transformation has been a major factor in the successful genetic manipulation of plant. We established a new selectable marker system for tobacco transformation using chimeric adenosine deaminase (ADA) gene, which confers resistance to cytotoxic adenosine analogues, 9-$\beta$-D-arabinofuranosyl adenine(Ara-A) and cordycepin. The transformants with the chimeric ADA gene in tobacco grew in the presence of normally lethal level of cytotoxic adenosine analogues, 100 $\mu$M Ara-A and 50 $\mu$M cordycepin. We successfully distinguished transformed shoot from non-transformed shoot on the same selectable media with cytotoxic adenosine analogues. In this selectable media, we were able to select seeds with/ without ADA gene from transgenic tobacco seeds. Theses results show that the mammalian ADA gene may serve as a new selectable marker for tobacco transformation.
For the germination and differentiation of immature embryos obtained by artificial crossing between tetraploid grape 'Fujiminori' (Vitis vinifera ${\times}$ V. labruscana) and triploid 'Summer Black' (V. labruscana ${\times}$ V. vinifera), were incubated in vitro using MS medium supplemented with $GA_3$ or coconut water (CW) at various concentrations. The percentage of embryo formation of 'Fujiminori' ${\times}$ 'Summer Black' was 64.3%. Embryo germination percentage was higher than 95% in all the $GA_3$ treatments at the concentrations of 0.01, 0.05, 0.25, and $1.25mg{\cdot}L^{-1}$. However, only 15.8-31.6% of the germinated embryos successfully developed into normal plantlets. At higher concentration of $GA_3$, the plantlets developed infirm hypocotyls with over elongated and less enlarged structure. Among the treatments of CW at the concentrations of 5, 10, 15, and 20% (v/v), 10% and 15% were more effective and plantlet achievement percentage were 68.4 and 66.7%, respectively. The addition of 10% CW was most effective to obtain plantlets with optimal shoot length, node and root numbers. 15% CW was suitable to obtain plantlets with longer roots. Accordingly, the embryo culture using the MS medium supplemented with 10-15% CW was observed to be more efficient for germinating and growing the immature embryos produced from artificial crossing between tetraploid grape 'Fujiminori' and triploid 'Summer Black'.
Chinese foxglove (Rehmannia glutinosa) is receiving much attention as one of the principal medicinal crops and the crude drug damand expands rapidly. This study was conducted to obtain the basic breeding information of Chinese foxglove. Meristem culture was employed for virus free seedling production and miropropagation. Both Jiwhang 1 and domestic local were severely infected by potexvirus and TMV. Several growth regulators were used for the virus free stock production from Jiwhang 1 and Danyang local. Shoot formation and callus induction from the meristem culture seemed to be influenced by the content of various kinds of plant growth regulators. Kinetin supplement was the most effective on shoot formation and NAA addition was good on callus induction among the treatments. The acquired virus free stocks were confirmed using transmission electron microscope and indicate plants.
Antifungal bacterium, Bacillus subtilis YS1 was isolated from Yusong hot spring showed broad antifungal spectrum against 12 kinds of plant pathogenic fungi and Candida albicans, animal pathogen. From the gamma($Co^{60}$) radiation sensitivity test, $D_10$ value was 2.08 kGy and it survived above 20 kGy of radiation dose. Several mutants were induced by gamma radiation. Among them, YS1-1009 mutant showed resistance against tebuconazole of herbicide, increased activity against Botryoshaeria dothidea and ligninase activity. YS67 mutant was antifungal deficient auxotrophic mutants(trp-pro-or arg-ura-). From this results, it suggested that gamma irradiation could be useful method for mutant induction.
