Analysis of Phytosterol and Fatty Acid Compositions of Grape Seeds Produced in Korea (품종별 포도씨의 식물성 스테롤 및 지방산 조성 분석)

  • In this study, phytosterol content and fatty acid composition of grape seeds from fourteen different cultivars produced in Korea were determined. Although the total phytosterol contents depended upon the cultivars, the major phytosterol was $\beta$-sitosterol (64.9$\sim$119.3 mg/100 g) in all samples. The fatty acid composition of the grape seeds were analyzed as palmitic (16:0, 6.8$\sim$16.8%), palmitoleic (16:1, 0.1$\sim$0.2%), stearic (18:0, 4.7$\sim$5.2%), oleic (18:1, 16.6$\sim$26.1%), linoleic (18:2, 16.6$\sim$20.1%), linolenic acid (18:3, 0.3$\sim$0.5%) and arachidic acid (20:0, 0.3$\sim$0.5%). Campbell early, the mainly produced and consumed cultivar in Korea, contained relatively high phytosterol content compared to other samples and high proportion of unsaturated fatty acids. Therefore, the seeds from Campbell early might be used for functional oil production and as food ingredients.

  • A mutant of Mycobacterium sp. has been isolated which is capable of degrading cholesterol and plant sterol to androst-4-ene-3, 17-dione and 9-hydroxyandrostene-3, 17-dione. Also this mutant hydroxylated the steroidal nucleus at the 9 $\alpha$ position. No ring degradation inhibitory agents are required for these processes.

Effect of Phytosterol Treatment on Plasma Lipids and Glucose in Rats (식물성스테롤 처리가 흰쥐의 혈장지질 및 혈당농도에 미치는 영향)

  • Diet effect of separated-purified phytosterol, obtained from soybean scum, in rats during 4 weeks was as follows. At this experiment, checking points were plasma lipids and glucose concentration as well as insulin level in plasma, pancreas and femur. Purity of above phytosterol was 68.3%, and composition was capesterol 21%, ${\beta}$-stigmasterol 62.3%, sitosterol 11.0% and unknowns 5.8%, respectively. Triglyceride, total cholesterol and phospholipids content was decreased as increasing of phytosterol amount($0{\sim}3%,\;w/w$). But excessive treatments more than 3% there was no any additional effect. From these experiment optimum amount of phytosterol level was around 3%(w/w). But phytosterol treating didn't show any effects on the changes of carbohydrate relative tissues like plasma glucose concentration as well as insulin level in plasma, pancreas and femur.

Comparison of lipid constituents and oxidative properties between normal and high-oleic peanuts grown in Korea (국내산 땅콩의 일반 품종과 고올레산 품종에 대한 지용성 영양성분과 산화안정성 비교)

  • Generally, peanuts are classified as high-fat foods as they possess high proportions of fatty acids. This study compared lipid constituents and properties between normal and high-oleic peanuts. Gas Chromatography-Flame Ionization Detector (GC-FID) analyses revealed that the fatty acid levels were significantly different between the normal and higholeic peanuts (p<0.05). Eight fatty acids were identified in the samples, including palmitic (C16:0), stearic (C18:0), oleic (C18:1, n9), linoleic (C18:2, n6), arachidic (C20:0), gondoic (C20:1, n9), behenic (C22:0), and lignoceric (C24:0) acids. Four tocopherol homologs were detected, and ${\alpha}$- and ${\gamma}$-tocopherols were the predominant ones. Tocopherols were rapidly decomposed during 25 day storage at $80^{\circ}C$. The main identified phytosterols were beta-sitosterol, ${\Delta}^5$-avenasterol, campesterol, and stigmasterol. Acid and peroxide values indicated that high-oleic peanuts have better oxidative stability than normal peanuts. These results can serve as the basis for the use of peanuts in the food industry.

  • The archaeological site of Nambuk-dong, Yongyu Island, Incheon which goes back to the Neolithic making it difficult to identify character of site, To get information for identifying utility and character of Open-Air Hearths within the finds, an organic residues analysis was conducted on soil and stone sample, sterol was analysis to determine the originality of archaeological sample, then percentage of fatty acid was identify the species of sample. As a result, it is assumed that No. 9, 10, 12 finds once had shellfish-related materials remain, No. 32 find seemed to have animal materials, No. 42, 43 finds were related to plant. In conclusion, it seems that open-air hearths of Yongyu Island had utility mainly related to making seafood.

Sensitivity of Colletotrichum spp. Isolated from Red-pepper to Sterol Biosynthesis Inhibiting-Fungicides and Their Field Fitness (고추에서 분리한 탄저병균의 스테롤 생합성 저해 살균제에 대한 감수성 반응과 포장 적응력)

  • Among 34 isolates of Colletotrichum spp., 20 from red pepper and 14 from apple, only one isolate 2001-45 was identified as the isolate resistant to sterol biosynthesis inhibiting fungicides. EC$_{50}$ values of the isolate 2001-45 were 153.5, 42.7, 34.0, and 17.1 $\mu\textrm{g}$/ml for myclobutanil, tebuconazole, hexaconazole and nuarimol, respectively, The resistance factor of the isolate 2001-45 against the other isolate 2001-44 to 4 above fungicides was ranged from 17 to 57. However, EC$_{50}$ value of the 2001-45 for prochloraz was 0.07 $\mu\textrm{g}$/ml, which was lower than those of the 2001-44 and the isolate JC24. For the fitness test of the 2001-45, mycelial growth, sporulation on PDA and pathogenicity on fruits were investigated. No difference in mycelial growth was found between 2001-45 and 2001-44, but great difference in sporulation. No symptom was developed by 2001-45 even by wound inoculation of pepper fruit. Therefore, this study indicated that the isolate 2001-45 was inferior to the other isolates in the fitness, although the isolate 2001-45 was highly resistant to sterol biosynthesis inhibiting fungicides.

Bleaching of cold-pressed rapeseed oil using activated clay (산성백토를 이용한 저온압착 유채유의 탈색 평가)

  • Acid-activated clays (SUPER-DC, DC-A3, and P1) are used for the bleaching of cold-pressed rapeseed oil. In this study we tested the bleaching performance of cold-pressed rapeseed oil according to the different reaction time (20, 40, 60, 80 min) and temperature (40, 80, $120^{\circ}C$). Oil color (lightness, redness, yellowness), pigments (chlorophyll A and carotenoid content) and quality properties (fatty acid composition, tocopherols (${\alpha}$, ${\beta}$, ${\gamma}$, ${\delta}$), and plant sterols (${\beta}$-sitosterol, campesterol, stigmasterol) content) were analyzed. The results showed that bleaching of cold-pressed rapeseed oil with 2% acid-activated clays at $40^{\circ}C$ for 20 min, brightness (L) increased, but redness (a) and yellowness (b) decreased. Bleaching of cold-pressed rapeseed oil with 2% DC-SUPER at $40^{\circ}C$ removed chlorophyll A and carotenoids pigments significantly. In addition, about 50% of total tocopherol content in cold-pressed rapeseed oil was reduced by bleaching. Originally total tocopherol content was 46.62mg/100g in cold-pressed rapeseed oil. But after bleaching, total tocopherol content was 12.67mg/100g (20 min bleaching), 15.31mg/100g (40 min bleaching), and 13.56mg/100g (60 min bleaching). However plant sterols content in cold-pressed rapeseed oil remained unchanged by bleaching. Overall, acid-activated clays were useful for the bleaching of pigmented rapeseed oil.

Effect of stigmasterol in liposome bilayer on the stabilization of encapsulated ascorbic acid (리포솜 이중층의 stigmasterol이 포집된 ascorbic acid의 안정성에 미치는 영향)

  • The effect of stigmasterol (SS), a phytosterol, in the liposome bilayer on the stability of encapsulated ascorbic acid (ASA) was evaluated. Liposomes, consisting of phosphatidylcholine (PC) and SS, and ASA were encapsulated by the dehydration/rehydration method. The average particle size of the liposome increased with increasing SS content. SS significantly increased the stability of encapsulated ASA. For example, ASA remaining in the liposomes of 100:0, 90:10, and 70:30 (PC:SS, w/w) ratios was 34.12%, 49.88%, and 58.58%, respectively, after storage for 8 days at 4℃, while only 7.66% ASA remained in the buffer under the same conditions. These results indicated that SS in liposomes increased the stability of encapsulated ASA.

Development of Natural Antioxidants Stable at Frying Temperatures (고온에서 안정한 천연 항상화제 개발)

  • The addition of antioxidants to fats and oils is one of the most effective ways to prevent oxidation of lipids. The popularity of natural antioxidants has increased because of the possible toxicity of synthetic antioxidants. Common natural antioxidants, tocopherols, retard oxidation at ambient temperatures, but they are ineffective at retarding oxidation at frying temperatures. The need for the development of novel natural antioxidants which are effective at frying temperatures is obvious. Sterols present in vegetable oils and certain herbal plant extracts have been reported to have antioxidant properties. Some sterols have been shown to retard thermal changes at frying temperatures. All the sterol effective at preventing oxidation at frying temperatures have an ethylidene group in their side chain. These effects can be explained by the hypothesis that sterols with a structure that allows them to react with lipid free radicals to form relatively stable free radicals are effective as antioxidants. Organic Residues Analysis of Oil Bottle of Goryeo Dynasty Excavated from the Soejeoul Site, Geumneung-dong, Chungju (충주 금릉동 쇠저울유적 출토 고려시대 유병의 유기물 분석)

  • Organic residues are substances derived from diverse natural sources. Recent scientific analysis of organic residues has yielded important information in restoring the lifestyles of ancient peoples. In this study, the organic material contained within the celadon oil bottle of the Goryeo dynasty, excavated from the Soejoul site in Geumneung-dong, Chungju, was analyzed using Fourier-transform infrared spectroscopy (FT-IR) and gas chromatograph-mass spectrometer (GC-MS). The results showed that the organic materials in the bottle were plant-derived oils. In particular, polyunsaturated fatty acids and phytosterols were detected using GC-MS analysis. Sesamin components were also identified. Sesamin, which is a characteristic component of sesame seeds, is a lignan and an antioxidant. As the organic residues in the oil bottle were derived from sesame seeds, it is presumed that sesame oil was stored in the bottle.
