Plant Regeneration and Protein Analysis from Cadmium Resistant Callus of Tobacco (Nicotiana tabacum cv. BY4) (담배 (Nicotiana tabacum cv. BY4)카드뮴 저항성 캘러스로부터 식물체 재생과 단백질 분석)

  • Calli were induced from diploid and haploid tobacco after 4 weeks and maintained on MS medium with combination of 2.0 mg/L 2,4-D,0.1 mg/L BAP and 2.0 mg/L kinetin. Suspension cells were screened through 65 $\mu$m-nylon mesh and 100 $\mu$m-mesh, then they were smeared on selection medium combined with cadmium and PFP by using the low melting agarose of 0.8%. After 30days smeared cultures of the medium the cell was treated with 500 $\mu$M and 1000 $\mu$M to select the resistant cell line were selected. Plant regeneration was induced from the selected cell lines on medium with 0.5, 1.5, 2.0 mg/L BAP and on media with combination of auxin and BAP under 500 $\mu$M and 1000 $\mu$M cadmium. At this time, plant regeneration was achived on cadmium free medium. In case of haploid, occurred from the cell line which is selected in medium with cadmium and PFP. In case of diploid regeneration occurred is in the medium with cadmium alone. The plantlet regenerated from cadmium resistant calli grew well in cadmium 500 $\mu$M. Protein pattern of leaf, root, stem of regenerated plants was analyzed. The quantum was 6.5188 ug/mg.fr.wt in the leaf of plant, 5.3611 ug/mg.fr.wt in the stem, 3.0213 ug/mg.fr.wt in the root. On the other hand, 5.9652 ug/mg.fr.wt. in the leaf of control, 3.5974 ug/mg.fr.wt in the stem of the control, 4.3766 ug/mg.fr.wt. in the root of the control. The one dimension bends regenerated from cadmium resistant calli resistant to cadmium in leaf were 49 involving 198.7KD etc. Disappeared were 4 involving 160.5KD etc, The protein bends were combinized were 3 involving 83.4KD etc. The bends resistant to cadmium stress in stem were 41 involving 4.3KD etc. Disappeared were 5 involving 114.8KD etc. The protein bends combinized were 6 involving 128.7KD etc. The bends which had the resistance to cadmium stress in root is 27 in volving 166,9KD etc. The bends which disappeared were 198.7KD etc. There were 5 involving 83.4KD etc.

  • 눈개승마 (Aruncus dioicus)는 장미과(Rosaceae) 눈개승마속(Aruncus)에 속하는 다년생 초본식물로 울릉도에서 자생하는 잎줄기 채소이다. 독특한 향기가 특징이며, 인삼맛, 두릅맛, 고기맛이 나는 산채로 세가지 맛이 난다고 하여 예로부터 삼나물로도 불러왔다. 눈개승마에는 caffeic acid, 1-O-Caffeoyl-β-D-glucopyranose 등의 항산화 성분이 다량 함유되어 있으며, 전초는 해독, 편도선염에 효과가 있어 약용으로 쓰이고 어린순은 탄수화물과 무기물 함량이 풍부하다. 비지는 대두를 이용하여 두부를 제조하는 과정에서 발생되는 것으로서 대두로부터 수용성 단백질이 추출되었지만 나머지 약 23%의 단백질, 57%의 식이섬유와 이소플라본 등의 유용한 성분을 다량 포함하고 있다. 또한 비지의 단백질은 다른 식품에는 부족한 황 함유 아미노산과 리신의 함량이 비교적 많고, 두부에 거의 없는 식이섬유가 다량 함유된 고 식이섬유 소재이다. 그리하여, 이러한 콩비지를 고부가가치 식품 소재로 활용하기 위해 건조 미세분말 비지를 제조하고, 이를 산채와의 혼합 가공을 통한 기능증진 효과를 연구하였다. 일반적으로 산채는 쓴맛 제거와 보관성 증진을 위해 데침공정을 주로 하나 산채를 물에 데칠 경우 산채의 유용성분이 대부분 손실된다. 눈개승마의 경우 유용성분이 17.25%, 그 중 폴리페놀 성분이 약 90% 손실된다. 본 연구에서는 기존의 데침공정 대신 찜공정을 이용하여 산채의 유용성분을 보존하고 콩비지를 첨가하여 산채 유용성분의 생이용성 증진 효과를 연구하였다. 눈개승마를 콩비지와 혼합하여 고온고압 가공함으로써 산채의 쓴맛을 줄이고 보관성을 높이는 동시에 눈개승마 폴리페놀의 단량체화를 통해 눈개승마 단독가공에 비하여, 단량체 비율이 약 100~200배 증가하는 것을 확인하였다. 본 연구를 통하여 콩비지의 고부가가치 소재로의 가능성을 확인하였을 뿐만 아니라 산채의 다이어트·건강기능식품 소재로의 산업화에도 기여할 수 있을 것으로 기대된다.

  • Plant pathogenic fungi are the most diverse and drastic causal agents of crop diseases threatening stable food production all over the world. Plant have evolved efficient innate immune system to scout and counterattack fungal invasion and pathogenic fungi also developed virulence system to nullify plant resistance machinery or signaling pathways and to propagate and dominate within their niche. A growing body of evidences suggests that post translational modifications (PTMs) and selective/nonselective degradations of proteins involved in virulence expression of plant pathogenic fungi and plant defense machinery should play pivotal roles during the compatible and incompatible interactions. This review elucidates recent investigations about the effects of PTMs and protein degradations on host defense and fungal pathogens' invasions.

  • The potency of adventitious root initiation was investigated in relation with germination days and ike end of culture days healed with root induction medium on the initiation of adventitious roots from explants of soybean (Glycine max L.) cotyledon. Also, the changes in the pattern of protein profiles related with adventitious mot initiation have been examined by two-dimensional polyauyl-amide gel elecoophoresis during the germination and the initiation of adventitious roots. The potency of adventitious root initiation from germinated cotyledons was high after 4 days, and adventitious roots were initiated from after 4 days and very high after 6 days healed with mot induction medium There was a reproducible quantitative change in 34 specific protein spots during germination and adventitious root initiation. A necessary basic protein for adventitious root initiation with rotative molecular weight of 27 kD was induced during the germination. And three basic protein groups with relative molecular weight 22 kD, 23 kD and 33 kD, and two acidic protein groups with relative molecular weight 27kD and 29 kD were induced during the initiation of adventitious roots.

Genetic variation of 7S and 11S globulins in soybean seed (콩 종실 단백질의 유전변이)

  • 7S and 11S globulins are two major storage proteins in soybean seed. For improving the quality of soybean seed protein, an increase of 11S/7S ratio would be a desirable objective because 11S globulin contains much more sulfur-containing amino acids than 7S globulin. In this study, six soybean varieties grown at three locations were used for genetic variation analysis of 7S and 11S globulins. It was possible to screen the soybean genotypes having aberrant subunit compositions of the two globulins by a sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE). So, heritabilities, genotypic and phenotypic correlations among eight globulin fraction contents of soybean seeds were estimated. The mean value of 7S and 11S globulin fraction contents were 38.9% and 61.1%, respectively, and the ratio of 7S to 11S globulin ranged from 0.58 to 0.74. The high heritability value was found in $\beta$ subunits but the values of acidic and basic subunits were relatively low. Genotypic correlations were higher than the corresponding phenotypic correlations in most of globulin subunit contents. $\beta$ subunits was negatively correlated with $\alpha$ and $\alpha$' subunits among 7S fractions, while no significant correlation between $\alpha$ and $\alpha$' subunits could be found In case of 11S fractions, acidic and basic subunits exhibited no genotypic but negative phenotypic correlation.

  • 머드는 알루미늄, 철, 마그네슘 등의 미네랄이 풍부하여 원적외선 방사율이 높고, 미량 포함된 단백질과 비타민은 피부의 세포조직을 강화하고 탄력성을 주며, 항균, 소취 등의 기능을 보유하고 있다. 이러한 유익한 기능을 가진 머드를 매일 사용하는 면타올에 적용하려 하나 무기 염재인 머드는 섬유와 친화성이 없어 일반 염료처럼 고농도로 섬유에 염색되지 못하고 있다. 본 연구에서는 섬유의 촉감에 영향을 미치지 않으면서 머드의 염색농도를 높이기 위해, 4종의 고착제를 이용한 염색법을 검토하였다. 먼저 식물성 단백질액으로 면타올을 전처리하고 나노입자의 머드액를 사용하여 염착시킨 경우가 가장 진하게 염색되며, 염색견뢰도 측정 결과 세탁과 땀에 모두 우수한 것으로 나타났다.

  • 계사내 공기중의 암모니아 농도 감소 목적으로 식물 기원 소화율 촉진제(phytogenic digestibility enhancer) 사용이 가능하다. 이 소화율 촉진제는 사료 설계 명세서 내 특정 성분(조단백질)을 감소시키고 영양소 이용성을 개선하여 동물생산성을 향상시키기 때문이다. 조단백질 함량이 낮은 사료 급여는 가금에게 적정 요구량 이상 흡수된 단백질(excess protein)의 뇨산(尿酸 : uric acid) 전환에 에너지를 소비하지 않아서 열역학적 이점(熱力學的 利點 : energetic advantage)이 있고, 이 에너지는 유익한 성장에 사용이 가능하도록 존재한다. 더욱이 동물 사료 중 조 단백질 1%는 약 3% 더 많은 물의 섭치가 필요하고, 그 중 80%는 다시 배설되므로 단백질 함량 적정화 사료들(protein optimized diets)은 음수 요구량을 감소시킨다. 이러한 단백질 적정화 사료는 동물 사육업자들에게 관심의 대상으로 명확한 경제적 이득이 발생한다.

  • Panax ginseng C.A. Meyer is a well-known Korean traditional medicine. Until now, even though major research of ginseng has been focused on the pharmacological effect, clinical application and chemical analysis of extracted secondary metabolite for several years, the physiology and gene functions of ginseng were not well known. In this research, we have developed the protein extraction methods of ginseng root and hairy root for proteome analysis in order to elucidate the gene(s) function of ginseng. Using the liquid nitrogen (equation omitted) TCA method as protein extraction method, about 660 protein spots were detected on the 2-DE gel of hairy root. Additionally, comparative analysis result of 2-DEs of ginseng root (equation omitted) hairy root suggested that proteomes of same organism could be changeable according to the culture condition, growth stages and other stimulus.

  • The effect of defatted and dehydrated beef muscle on the physical properties of thermoplastically extruded defatted soybean, cottonseed, peanut and sunflower seed flours were investigated. To minimize the adverse effect of meat fat and to increase the mixing efficiency of the meat with defatted oilseed flours at a given moisture level, beef muscle was extracted with chloroform-ethanol(2 : 1) at 2$0^{\circ}C$ and air dried. The variety of oilseed flours used had greater effects on color, expansion, bulk density, water absorption rate and textural strength of the extrudate than did the added level of defatted, dehydrated beef muscle (0 to 20% on a dry weight basis).
