  • This study was conducted to evaluate the potential of indigenous vesicular-arbuscular mycorrhizal fungi(VAMF) in the rhizosphere soil of horticultural crops grown under greenhouse and open-field condition, in the southern area of Kores. Soil samples collected from the rhizosphere of some sellected horticultural crops, such as cucumber, hot pepper, lettuca, tomato and eggplant grown under greenhouse or open-field condition. All tested crops are considered as mycorrhizal plants. The infection rate of horticultural crops investigated ranged from 38% to 70%, hot pepper and eggplant grown under greenhouse condition showed the highest infection being 66.0% and 70.0%, respectively. Spore densities were from 4.8 to 20.0g-1 on dried soil basis. Spore densities of VAMF in the rhizosphere soils under greenhouse condition were higher than that of open-field conditions. The highest distribution of spores in diameter ranged from $75{\mu}m$ to $106{\mu}m$ in the rhizosphere soil of lettuce, cucumber and tomato while those in hot pepper and eggplant ranged from $75{\mu}m$ to $250{\mu}m$. Glomus sp.-type spores predominated in the slightly acid soil(pH 6.3), while Acaulospora sp.-type spores greatly predominated in the very strongly acid field(pH 4.9).

  • Composition of fungal communities in three microhabitats such as soil, rhizosphere and rhizoplane were studied to understand the root environment of healthy and diseased plants in Phytophthora non-infested and infested fields, respectively. Samples were collected from the tomato- and red pepper-growing greenhouses in Kyungsang-Nam Province on April, 1999. Twenty-five species were isolated from each vegetation field using the dilution plate technique. There were a greater variety of species in infested fields than non-infested and in soils than in both rhizospheres and rhizoplanes. The number of species isolated were varied amongst the different microhabitats. A Trichoderma species was isolated only from non-infested fields.

  • Chemical characteristics and their interrelationships of 156 soils included by 74 sandy loam and 82 loam soils collected from plastic film house in Chungbuk area were investigated from 1998 to 2001. Seventeen chemical properties including pH, organic matter (OM), electrical conductivity (EC), inorganic nitrogen, available phosphorus, exchangeable cations, CEC, etc., were analyzed by correlation, standardized partial regression coefficient, and principal factor analysis. Standardized partial regression coefficients of chemical properties were estimated to determine the degree of contribution of EC and OM contents in soils. Principal factor analysis was applied to classify the studied chemical properties into different groups having similar chemical properties. The pH of experimental soils ranged from 4.24 to 7.14 and 4.95 to 7.35 for loam and sandy loam soils, respectively. The EC of soils varied from 0.93 to $15.65dS\;m^{-1}$ for loam and $0.91{\sim}22.30dS\;m^{-1}$ for sandy loam soils, respectively with significant differences among them. The EC measured by 1:5 $H_2O$ dilution method and saturation method were significantly related with 8.163 and 8.599 as the slopes of regression equation for loam and sandy loam soils, respectively. These slopes more than 8.0 in this regression equation was higher than the slope of 5.0 that is estimated from dilution coefficient suggesting that EC measured by 1:5 dilution method might be erratic. The standardized partial regression coefficient of different chemical properties for the estimation of EC was in the order of $NO_3{^-}$ > $Cl^-$ > OM > exchangeable Mg for loam soils and $NO_3{^-}$ > exchangeable Mg > $Cl^-$ for sandy loam soils. Contribution order of the chemical properties based on standardized partial regression coefficient differed 1:5 dilution method and saturation method, indicating that different chemical compounds might be present in the extract solutions of these two methods. Consequently the measurement of EC by saturation method was thought be still better for estimation of chemical property because accuracy of EC measurement by 1:5 dilution method can't be improved by any specific coefficient for adjustment of EC. Regardless of differences in soil textures and extraction methods, correlation coefficients between EC and the other chemical properties were routinely in the order of $NO_3{^-}$ > $Cl^-$ > degree of base saturation > exchangeable Mg > exchangeable Ca > $SO{_4}^{2-}$. The principal factor analysis revealed four factor groups of the chemical properties studied. The groups for sandy loam were as follows; ; 1. salt components, 2. soil reaction components, 3. fixed and adsorption components, 4. CEC components. The groupings of loam soils were similar to sandy loam except that exchangeable Na substituted the CEC of sandy loam.

  • Yield response of tomato to nitrogen and potassium fertilizer and the balance of the two elements were determined in salt-accumulated greenhouse soil to improve the efficiency of fertilizer appiication. The response of tomato yield to nitrogen and potassium fertilizer application was not significant. The current parameters such as OM and $K/{\sqrt{Ca+Mg}}$ that were used to determine the level of nitrogen and potassium fertilizer in open field were not suitable in salt-accumulated greenhouse soil condition. The temporal and spatial distribution of $NO_3{^-}-N$showed the same pattern to those of $Cl^-$ ion that is non-reactive with soil, while the content of Ex. K was extraordinarily high in soil after harvesting of tomato, which had experienced relatively dry condition during harvesting time. The loss of $NO_3{^-}-N$ and Ex. K out of 28cm below the soil surface was 2~5 and 1.5~3.5 times greater than the amount of nitrogen and potassium uptake by the plant.

  • To find a quick test procedure for soil nitrate concentration, two methods, i.e. "Nitracheck 404" reflectometer (Eijkelkamp, Netherlands) with nitrate test strip and a portable colorimeter "Hanna Ion Specific meter(USA) with a color development reagent, were tested for twenty soils with different nitrate contents ranged between from $10mg\;kg^{-1}$ to $340mg\;kg^{-1}$. The standard deviation, coefficient of variability, and recovery from these quick test procedures were compared with those measured by conventional Kjeldahl distillation method and nitrate ion electrode method. The nitrate concentration measured by portable colorimeter method was higher in soils with low concentration and lower in soils with high concentration than those measured by conventional methods. However, concentrations measured by test strip reflectometer method was showed the similar coefficient of variability and recovery as those by conventional methods for all soil samples. From the experimental results in this study, the test strip reflectometer method was thought to be recommendable one revealed the satisfied results for accuracy, quickness, and simplicity for field test of soil nitrate concentration.

  • This study was conducted to evaluate the microbiological safety of leafy vegetables (perilla leaf and lettuce) in relation to cultivation methods. A total of 2,304 samples were collected from plants, harvesting tools and soil mulching film during the production and harvest stages from organic- and conventional- farms. From the samples, sanitary indicator microorganisms (total aerobic bacteria, coliforms, E. coli., Environmental Listeria, and yeast and mold) and pathogenic microorganisms (S. aureus, B. cereus, Salmonella spp., Clostridium spp., and L. monocytogenes) were analyzed. In the production stage of leafy vegetables, the sanitary indicator microorganisms was not detected regardless of cultivation method or it was detected to be less than $3.4\;Log\;CFU/100cm^2$. B. cereus and S. aureus were found to be 0.22~1.55 Log CFU/g in perilla leaf and lettuce produced by organic farms, and S. aureus was not detected and B. cereus was found to be 0.42~2.19 Log CFU/g in conventional farms. There were no significant differences between two cultivation methods. In the harvesting tools and soil mulching film, the contamination levels of sanitary indicator microorganisms and pathogenic microorganisms was low regardless of the cultivation method. However, there was a positive correlation ($R^2=0.4526$) in that the higher the microbial contamination level in the harvesting tool, the higher the microbial contamination on the surface of the plant. In addition, sanitary indicator microorganisms and pathogenic microorganisms were not detected or low in soil mulching during the production of organic leafy vegetables. As a result of this study, microbial hygiene control by soil mulching and harvesting tools was more important than difference of cultivation method in production of leafy vegetables.

  • To determine the economic threshold level of Meloidogyne incognita on cucumber in greenhouse conditions, cucumber seedlings(Cucumis sativa L. cv. 'Super Manchon') grafted on 'Jangsushintozoa'(Cucurbita maxima x Cu. moschata) were planted in wooden boxes($30{\times}40{\times}15cm,\;L{\times}W{\times}D$) under a plastic house in August 01 and harvested from Sep. 01-Oct. 30, 2006. The initial nematode population densities(Pi) in the wooden boxes were adjusted to 0, 10, 30, 100, 300, and 600 second-stage juveniles(J2)/100 $cm^3$ soil. The relationship of total fruit yield to Pi level could be adequately described by a linear regression equation, $Y=0.82-0.04{\cdot}Log_{10}$(Pi+1). Initial nematode densities(Pi) before planting in excess of 5 J2/1,000 $cm^3$ soil caused in total yields loss that is equivalent to the costs of granular nematicide application; Pi level in excess of 25 J2/1,000 $cm^3$ soil caused in total yields loss that exceed the costs for application of fumigants at current control costs. We propose two different management strategies depending on nematode densities, (i) application of non-fumigant granular nematicides for M. incognita Pi level of 5 J2/1,000 $cm^3$ soil and (ii) fumigant treatment with Pi level over 25 J2/1,000 $cm^3$ soil. Soil samples to determine population density of Meloidogyne spp. for advisory purposes should be taken several months before planting time, which allows a period of time enough for implementing management procedures.

  • To find diagnosing method of nitrogen status in tomato plant for determining optimum application rate of side dress, chlorophyll reading values were measured by portable chlorophyll meter(SPAD 502, Minolta), and compared with nitrogen supplying capability of soils. Regression between dry weight, amount of nitrogen uptake, and chlorophyll reading at stalk positions of tomato grown on the condition of no fertilization were evaluated For 6 green house soils with different nitrate concentrations ranged from $55mg\;kg^{-1}$ to $306mg\;kg^{-1}$. The chlorophyll reading of tomato leave was significantly correlated with amount of nitrogen per unit area of leave suggesting that chlorophyll content is useful for nitrogen diagnosis of tomato plant. The chlorophyll reading showed peak at the 15th leaf of stalk position on the 45th days after transplanting and this suggested that below or near the 15th leaf and before or near the 45th days after transplanting is the critical stalk position and time for diagnosing nitrogen status of tomato by chlorophyll test. The chlorophyll reading at the 14th leaf on the 40th days after transplanting was significantly correlated with soil nitrate status, dry weight and amount of nitrogen uptake by tomato grown with no fertilization. From the above correlation, the chlorophyll reading value of 57.1 at the 14th leaf of tomato was estimated as the critical level for maximum dry weight and amount of nitrogen uptake by tomato grown with no fertilization. Consequently, chlorophyll reading of tomato leaves measured by portable chlorophyll meter was thought to be available as a rapid plant test for predicting the nitrogen supplying capability of green house soils.
