Extraction and Purification of Polysaccharides from Phellinus linteus Mycelia (상황버섯의 균사체로부터 다당류의 추출과 정제)

  • To maximize the obtained polysaccharides from Phellinus linteus mycelia, effects of extraction and purification conditions including dialysis time (8, 16, 24, 48 h) on an amount of the polysaccharides were investigated. As extraction temperature ($50{\sim}95^{\circ}C$), ratio of solvent volume to the dry weight of mycelia (10, 15, 20, 25 ml/g), extraction time(2, 4, 6, 8 h), final concentration of ethanol (70, 75, 80, 85%), and aging time (1, 4, 8, 16 h) increased, an amount of the polysaccharides was increased. An increase in precipitation time up to 24 h increased an amount of the polysaccharides but a further increase in precipitation time after 24 h did not changed largely an amount of the polysaccharides. Three precipitation solvents (ethanol, methanol, acetone) were tested. An amount of the polysaccharides was increased in order to acetone, ethanol, and methanol. On the optimal extraction condition, extraction temperature, ratio of water volume to the dry weight of mycelia, and extraction time were $95^{\circ}C$, 25 ml/g, and 8 h, respectively.

  • 디지털 TV 서비스의 시작으로 시청자의 효율적인 방송 컨텐츠 이용의 필요성이 제시되었고, 이를 위한 사용자 맞춤형 서비스와 방송 컨텐츠의 특징 추출 및 표현 방법 등, 지능형 TV에 대한 연구가 수행되고 있다. 지능형 TV의 방송 컨텐츠의 비디오 요약과 비디오 하이라이트 등의 제작을 위해 MPEG-7 비주얼 기술자를 이용하여 비주얼 특성을 추출할 수 있다. 특히 비디오의 컬러 특징을 추출하는 MPEG-7 컬러 기술자를 이용한 특징 추출 위해서는 압축된 비디오를 복호하고 비디오의 프레임을 획득하여 특징을 추출하게 되지만, 이 과정은 비주얼 특징 추출 시간을 증가시켜 압축된 방송용 컨텐츠에 MPEG-7 컬러 기술자의 적용을 어렵게 한다. 본 논문은 압축된 비디오의 컬러 특성 추출 속도의 향상을 위해 비디오 압축 영역에서 MPEG-7 컬러 기술자를 적용할 수 있는 방법을 제안하였다. 압축된 비디오에서 DC 프레임을 추출하여 컬러 특징 추출 시간이 감소하였고 검색 율의 변화는 크지 않았다. 본 논문에서 제안한 방식은 방송용으로 압축된 비디오의 비주얼 특징을 고속으로 추출할 수 있는 방법으로의 활용이 가능할 것이다.

Quality characteristics of Sikhye prepared by different ratio of Seomaeyaksuk (Artemisia argyi H) extract (섬애약쑥 추출물의 첨가 비율에 따른 식혜의 품질특성)

  • This study was conducted to increase the functionality of Korean traditional beverage, Sikhye. The Sikhye were prepared by supplementation up to 20% of Seomaeyaksuk extract (SE) and saccharification for five hours (h). During the saccharification, turbidity of SE-added Sikhye (SES) was the highest at two hours of saccharification and then lowered gradually. L vlaue of SES did not show significant difference feduring saccharification, and both a and b value of Sikhye were increased as saccharification time passed and SE addition ratio was increased. The pH before saccharification was not significantly different by SE addition ratio but it was increased to 5.68-5.73 at 5 h saccharification. Soluble solid, total sugar and reducing sugar contents were enhanced as saccharification time and SE addition ratio were increased. The total phenolic compounds contents and DPPH radical scavenging activity showed a similar trend by saccharification time and SE addition. In sensory test, the evaluations of 5% SES and control were not significant different, but 20% SES group showed significant lower evaluation than other groups because strong color and mugwort flavor. From all of these results, physicochemical characteristics and antioxidant activity of SES was improved by SE addition but sensory characteristics was lower. Acceptable addition ratio of SE in Sikhye is determined to be within 15%.

Effect of Lentinula edodes water extracts and Lentinan on proliferation of myosatellite cell of Bos taurus Hanwoo (표고 자실체 물 추출물과 베타글루칸이 한우 근육위성세포 증식에 미치는 영향)

  • Lentinula edodes is one of the most produced mushrooms in the world. In this study, the effects of L. edodes water extracts and lentinan, a beta-glucan from this mushroom, on the proliferation of Bos taurus Hanwoo myosatellite cells were studied. The betaglucan content of the L. edodes water extract was approximately 15.20% at 85 ℃ for 4 h, 13.64% at 100 ℃ for 4 h, 9.48% at 40 ℃ for 8 h and 8.21% at room temperature for 24 h. L. edodes water extract was added to the culture of Hanwoo myosatellite cells. The expression of the MyoD gene increased in the addition of the extract at 40 ℃ for 8 h and 100 ℃ for 4 h, and the expression of the Myogenin gene increased in the addition of the extract at 40 ℃ for 8 h, but proliferation and activity did not increase compared to no addition. However, the addition of lentinan to the culture of Hanwoo myosatellite cells increased the expression of Myogenin gene related to muscle formation increased and the proliferation and viability of the cells. This study proved that the components of L. edodes can affect the proliferation of Hanwoo myosatellite cells, and further research will help develop the mushroom industry and cultured meat industry in the future.

Detection and Extraction Condition of Physiological Functional Compounds from Bran of Heugjinju rice (Oryza sativa L.) (흑진주벼 미강으로부터 생리기능성 물질의 탐색 및 추출조건)

  • Physiological functionalities in various extracts of Heugjinju rice bran were determined and its optimal extraction condition were also investigated Angiotensin-converting enzyme(ACE) inhibitory activity, fibrinolytic activity and tyrosinase inhibitory activity were higher in water extracts than those of 80% ethanol and methanol, hexane. Electron donating abilities were 97.8% in hexane extract and 83% in water extracts. ACE inhibitor was maximally extracted from Heugjinju rice bran when it was treated with 20 times of distilled water for 12 h at 20 $\^{C}$. Fibrinolytic compound was also maximally extracted by treatment of 10 times of distilled water for 18 h at 20 $\^{C}$. Electron donating compound and tyrosinase inhibitor were maximally extracted by treatment of 20 times of hexane and 10 times of distilled water at 20 $\^{C}$ for 18 h, respectively

Evaluation on Extractability of Heavy Metals in Mine Tailings of Disused Metal Mines with Concentrations and Kinds of Soil Washing Solutions (토양세척용매의 종류 및 농도에 따른 폐금속광산 폐기물내 중금속의 추출특성)

  • The objectives of this study were to assess extraction kinetics of heavy metals with extraction times and to assess extraction efficiencies of heavy metals with concentrations and kinds of washing solutions. Target materials were obtained from disused metal mines. Washing solutions were water, HCl(0.1, 0.3, 1.0 N), EDTA(0.01, 0.05, 0.1 M), and sodium dodecyl sulfate(SDS, 0.1. 0.5, 1.0%). Extraction efficiencies of heavy metals by water and SDS were below 1%, and extraction efficiencies of Zn and Cd were higher than those of Pb and Cu. As results, water and SDS were not effective in extracting heavy metals from mine tailings as washing solution, but extraction efficiencies of Pb and Cu with SDS solution increased as extraction time increased. Extraction kinetics of heavy metals with HCl and EDTA were faster than those with water and SDS. The majority of heavy metals were extracted within 6 hours, and extraction kinetics was almost independent of the solution concentration. Extraction kinetics of heavy metals after 6 hours was slow, but extraction kinetics was dependent on the solution concentration. Also, as concentrations of HCl and EDTA solution were stronger, heavy metals were extracted rapidly and extraction efficiencies were increased. The extraction efficiency was high in order of Cd>Pb>Zn>Cu in using 1.0 N HCl, and Pb>Cd>Zn>Cu in using 0.1 M EDTA. Consequently, extraction effectiveness was highest for Pb in using HCl, and for Pb and Cd in using EDTA with concentration increase. Extraction time of over 6 hours was not effective in extracting heavy metals.

Establishment of Microwave-Assisted Extraction Conditions for Antioxidative Extracts from Pleurotus eryngii (새송이버섯의 항산화성 추출물 제조를 위한 마이크로웨이브 추출조건 설정)

  • Through the application of a microwave-assisted extraction, the aim of study was to investigate the extraction conditions for the total yield, electron donating ability, total polyphenol contents and nitrite scavenging effect from Pleurotus eryngii. This experiment was conducted under the following extraction conditions: the microwave power (60, 90, 120 W), sample to solvent ratio (1:50, 1:20, 1:10), extraction time (1, 5, 10, 15 mins), and extractant like water, $50\%$ EtOH and $99\%$ EtOH. During the increase of microwave power ranging from 60 to 120 W, the maximized effective components were obtained when the microwave power was set at 60 W. By decreasing the ratio down to 1:10 (g/mL), the extraction of effective components but not total yield was the highest at 1:10 (g/mL). As going up to 15 min, the maximum extraction of effective components was achieved at 1 min for the extraction time and was little affected at over 5 min. In conclusion, the most appropriate conditions for the extraction of effective compounds from Pleurotus eryngii were as follows: the microwave power at 60 W, 1:10 of the sample to solvent ratio, extraction time at 1 min and water as the extractant.

Optimization of Extraction Method for the Quantitative Analysis of Gallic Acid from Cornus officinallis (산수유로부터 gallic acid 추출 및 HPLC에 의한 정량분석)

  • Cornus fructus has been used as a tonic, astringent, and haemostatic agent in Korea, China, and Japan. In this study, the fruit of Cornus officinalis was treated with different osmotic pressures, pH values, heat, and ethanol percentages in order to establish optimum extraction conditions for gallic acid, an example of a hydrolyzable tannin. The extract was analyzed by HPLC and LC-MS/MS to identify the gallic acid. As a result, the highest extraction rate of gallic acid (1.57 mg/g) occurred when the Cornus fructus was extracted with 100% ethanol for 1 hr at $80^{\circ}C$. Also, when it was treated with 70% ethanol for 24 and 48 hr, contents of gallic acid were 1.35 and 1.50 mg/g, respectively.

Antioxidative Effects and Characteristics of Methanol Extracts from Perilla Oils Roasted for Different Time (볶음시간에 따른 들기름 메탄올 추출물의 항산화 효과와 특성)

  • This study was carried out to investigate the oxidative stability of oils from peril1a seeds roasted at $190^{\circ}C$ for $0{\sim}50$ min. The oxidative stability of perilla oils increased as the roasting time increased. Oxidative stability of perilla oils extracted methanol extracts significantly decreased. When 1.0%(w/w) methanol and hexane fractions prepared from methanol extracts added to the unroasted perilla oils, methanol fractions showed strong antioxidative effects, but hexane fractions showed weak effects. As the roasting time increased, the browning intensity, fluorescence and electron donating ability of methanol extracts, methanol and hexane fractions increased, and those were closely related with antioxidative effects.

Optimization of Extraction Conditions for Dried Jujube by Response Surface Methodology (반응표면분석에 의한 건대추의 추출조건 최적화)

  • Extraction characteristics of dried jujube and functional properties of corresponding extract were monitored by response surface methodology. Maximum extraction yield of 53.69% was obtained at extraction temperature of $50.35^{\circ}C$, extraction time of 16.69 hr, and ethanol concentration of 72.88%. At extraction temperature, extraction time, and ethanol concentration of $45.80^{\circ}C$, 15.47 hr, and 73.12%, respectively, maximum cyclic adenosine monophosphate content was 8.20 mg/100 g. Maximum total polyphenol content was 18.85 mg/g at extraction temperature, extraction time, and ethanol concentration of $64.91^{\circ}C$, 20.84 hr, and 66.91%, respectively. Maximum total flavonoid content was 0.48 mg/g at extraction temperature, extraction time, and ethanol concentration of $57.36^{\circ}C$, 15.14 hr, and 71.08%, respectively. $IC_{50}$ value of electron donating ability showed maximum level of 32.34 mg/mL at extraction temperature of $48.46^{\circ}C$, extraction time of 19.25 hr, and ethanol concentration of 65.36%. Maximum ascorbic acid equivalent antioxidant capacity was 3.58 mg ascorbic acid equivalent per gram sample at extraction temperature, extraction time, and ethanol concentration of $56.09^{\circ}C$, 21.86 hr, and 65.36%, respectively.
