This study aimed to investigate the impact of protease treatments on the yield of rice starch (RST) from frozen rice flours, and to compare the physicochemical properties of RST by alkaline steeping (control) and enzymatic isolation (E-RST) methods. Although the yield of E-RST, prepared according to conditions designed by the modified 23 complete factorial design, was lower than the control, the opposite trends were observed in its purity. E-RST (RST1, isolated for 8 h at 15℃ with 0.5% protease; RST2, isolated for 24 h at 15℃ with 1.5% protease; RST3, isolated for 24 h at 15℃ with 0.5% protease) with the yields above 50% were selected. Amylose contents did not significantly differ for the control and RST2. Relative to the control, solubilities were higher for all E-RST, but swelling power did not significantly differ for E-RST except for RST1. Although all E-RST revealed higher gelatinization temperatures than the control, the reversed trends were found in the gelatinization enthalpy. The pasting viscosities of all E-RST were lower than those of the control. Consequently, the enzymatic isolation method using protease would be a more time-saving and eco-friendly way of preparing RST than the alkaline steeping method, even though its characteristics are different.
Stevia (Stevia rebaudiana Bertoni) is a perennial herb widely distributed in the mountainous area of Paraguay. It belongs to the family Compositae and contains 6 to 12 percent stevioside in the leaves. Stevioside is a glucoside having similar sweetening character to surgar and the degree of sweetness is approximately 300 times of sugar. Since Korea does not produce any sugar crops, and the synthetic sweetenings are potentially hazardous for health, it is rather urgent to develop an economical new sweetener. Consequently, the current experiments are conducted to establish cultural practices of stevia, a new sweetening herbs, introduced into Korea in 1973 and the results are summarized as followings: 1. Days from transplanting of cuttings to the flower bud formation of 6 stevia lines were similar among daylengths of 8, 10 and 12 hours, but it was much greater at daylengths of 14 or 24 hour and varietal differences were noticable. All lines were photosensitive, but a line, 77013, was the most sensitive and 77067 and Suweon 2 were less sensitive to daylength. 2. Critical daylength of all lines seemed to be approximately 12 hours. Growth of plants was severely retarded at daylengths less than 12 hours. 3. Cutting were responded to short daylength before rooting. Number of days from transplanting to flower bud formation of 40-day old cuttings in the nursery bed was 20 days and it was delayed as duration of nursery were shorter. 4. Number of days from emergence to flower bud formation was shortest at short day treatment from 20 days after emergence. It was became longer as initiation of short day treatment was earlier or later than 20 days. 5. Plant height, number of branches, and top dry weight of stevia were reduced as cutting date was delayed from March 20 to May 20. The highest yield of dry leaf was obtained at nursery duration of 40-50 days in march 20 cutting, 30-40 days in April 20 cutting, and 30 days in May 20 cutting. 6. An asymptotic relationship was observed between plant population and leaf dry weight. Yield of dry leaf increased rapidly as plant population increased from 5,000 to 10,000 plants/10a with a reduced increasing rate from 10,000 to 20,000 plants/l0a, and levelled off at the plant population higher than 20,000 plants/l0a. 7. Stevia was adaptable in Suweon, Chengju, Mokpo and Jeju and drought was one of the main factors reducing yield of dry leaf. Yield of dry leaf was reduced significantly (approximately 30%) at June 20 transplanting compared to optimum transplanting. 8. Yield of dry leaf was higher in a vinyl house compared to unprotected control at long daylength or natural daylength except at short day treatment at March 20. Higher temperature ill a vinyl house does not have benefital effects at April 20 transplanting. 9. The highest content of stevioside was noted at the upper leaves of the plant but the lowest was measured at the plant parts of 20cm above ground. Leaf dry weight and stevioside yield was mainly contributed by the plant parts of 60 to 120cm above ground but the varietal differences were also significant. 10. Delayed harvest by the time of flower bud formation increased leaf dry weight remarkably. However, there were insignificant changes of yield as harvests were made at any time after flower bud formation. Content of stevioside was highest at the time of flower bud formation and earlier or later harvest than this time was low in its content. The optimum harvesting time determined by leaf dry weight and stevioside content was the periods from flower bud formation to right before flowering that would be the period from September 10 to September 15 in Suweon area. 11. Stevioside and rebaudioside content in the leaves of Stevia varieties were ranged from 5.4% to 14.3% and 1.5% to 8.3% respectively. However, no definit relationships between stevioside and rebaudioside were observed in these particular experiments.

First, the Agrimonia pilosa Ledeb. was extracted with methanol at room temperatures, and fractionation of the methanol extracts from Agrimonia pilosa Ledeb. was carried out by using petroleum ether, chloroform, and ethyl acetate, and methanol respectively. The antimicrobial activity of the Agrimonia pilosa Ledeb. extracts was determined using a paper disc method against food-borne pathogens and food spoilage bacteria. The petroleum ether extracts of Agrimonia pilosa Ledeb. showed the highest antimicrobial activity against Pseudomonas aeruginosa. The synergistic effect has been found in combined extracts of Agrimonia pilosa Ledeb. and Perillae folium as compared to each extracts alone. Finally, the growth inhibition curve was determined using ethyl acetate extracts of Agrimonia pilosa Ledeb. against Bacillus Cereus and Salmonella Enteritidis. The petroleum ether extract of Agrimonia pilosa Ledeb. showed strong antimicrobial activity against Bacillus Cereus at the concentration of 4,000 ppm. The 4,000 ppm of petroleum ether extract from Agrimonia pilosa Ledeb. retarded the growth of Bacillus Cereus more than 24 hours and Salmonella Enteritidis up to 36 hours. The petroleum ether extracts of Agrimonia pilosa Ledeb. has been shown the antimicrobial effect against Bacillus Cereus and Salmonella Enteritidis. (Korean J Nutrition 38(2): 112~116, 2005)

This study was performed to investigate the antimicrobial effect of the Hedyotis diffusa extracts against food-borne pathogens. First, the Hedyotis diffusa was extracted with methanol at room temperature and the fractionation of the methanol extracts was carried out by using petroleum ether, chloroform, and ethyl acetate, and methanol respectively. The antimicrobial activity of the Hedyotis diffusa extracts was determined by using a paper disc method against food-borne pathogens and food spoilage bacteria. The methanol extracts of Hedyotis diffusa showed the highest antimicrobial activity against Staphylococcus aureus and Shigella flexneri. Synergistic effect in inhibition was observed when Hedyotis diffusa extract was mixed Sophora subprostrata extract as compared to each extracts alone. Finally, the growth inhibition curves were determined by using ethyl acetate extracts of Hedyotis diffusa against Staphylococcus aureus and Salmonella Typhimurium. The methanol extract of Hedyotis diffusa had strong antimicrobial activity against Staphylococcus aureus at the concentration of 4,000 ppm. At this concentration, the growth of Staphylococcus aureus was retarded more than 36 hours and up to 24 hours for Salmonella Typhimurium. In conclusion, the methanol extracts of Hedyotis diffusa inhibit efficiently Staphylococcus aureus and Salmonella Typhimurium.
This study was performed to investigate the antimicrobial activities of the Plagiorhegama dubium extracts against food-borne pathogens. The methanol extract was partitioned into petroleum ether, chloroform, ethyl acetate, and methanol portions. The antimicrobial activity of the P. dubium extracts was determined using a paper disc method against food-borne pathogens and food spoilage bacteria. The methanol extracts of P. dubium showed the highest antimicrobial activity against Staphylococcus aureus and Escherichia coli. The synergistic effect has been found in combined extracts of P. dubium and Hedyotis diffsa as compared to each extracts alone. Finally, the growth inhibition curve was determined using methanol extracts of P. dubium against S. aureus and E. coli. The methanol extract of P. dubium showed strong antimicrobial activity against S. aureus at the concentration of 4,000 ppm. The 4,000 ppm of ethyl acetate portion from P. dubium retarded the growth of S. aureus more than 36 hours and E. coli up to 60 hours. The methanol extracts of P. dubium has been shown the antimicrobial activity against S. aureus and E. coli.

This study was performed to investigate the antimicrobial effect of the Indigofera kirilowii extracts against food-borne pathogens. The Indigofera kirilowii was extracted with methanol at room temperature, and fractionation of the methanol extract from Indigofera kirilowii was carried out by using petroleum ether, chloroform, ethyl acetate, and methanol, respectively. The antimicrobial activity of the Indigofera kirilowii extracts was determined using a paper disc method against food-borne pathogens and food spoilage bacteria. The ethyl acetate extract of Indigo/era kirilowii showed the highest antimicrobial activity against Staphylococcus aureus and Shigella dysenteriae. The water extract of Indigofera kirilowii showed relatively low antimicrobial activity against microorganisms used in this experiment. The synergistic effect has been found in combined extracts of Indigofera kirilowii and Pulsatilla koreana as compared to each extract alone. The growth inhibition curve was determined using ethyl acetate extracts of Indigofera kirilowii against S. aureus and S. dysenteriae. The ethyl acetate extract of Indigofera kirilowii showed strong antimicrobial activity against S. aureus at the concentration of 4,000 ppm. The 4,000 ppm of ethyl acetate extract from Indigofera kirilowii retarded the growth of S. aureus more than 24 hours and S. dysenteriae up to 48 hours. This study showed the possibility of using ethyl acetate extract of Indigofera kirilowii as a material of food preservative.

The ethyl acetate extracts of Cutellaria baicalensis George showed the highest antimicrobial activity against Staphylococcus aureus and Shigella dysenteriae. The synergistic effect has been found in combined extracts of Cutellaria baicalensis George and Portulaca oleracea as compared to each extracts alone. Finally, the growth inhibition curve was determined using ethyl acetate extracts of Cutellaria baicalensis George against Staphylococcus aureus and Shigella dysenteriae. The ethyl acetate extract of Cutellaria baicalensis George showed strong antimicrobial activity against Staphylococcus aureus at the concentration of 4,000 ppm. The 4,000 ppm of ethyl acetate extract from Cutellaria baicalensis George retarded the growth of S. aureus more than 24 hours and Shigella dysenteriae up to 36 hours. The ethyl acetate extracts of Citrus unshiu Markovich showed the highest antimicrobial activity against Bacillus cereus and Shigella sonnei. A synergistic effect was found in combined extracts of Citrus unshiu Markovich and Hedyotis diffusa Willd as compared to each extract alone. Finally, the growth inhibition curve was determined using ethyl acetate extracts of Citrus unshiu Markovich against Bacillus cereus and Shigella sonnei. The ethyl acetate extract of Citrus unshiu Markovich showed strong antimicrobial activity against Bacillus cereus at the concentration of 5,000 ppm. The 5,000 ppm of ethyl acetate extract from Citrus unshiu Markovich retarded the growth of Bacillus cereus more than 24 hours and Shigella sonnei up to 24 hours. The ethyl acetate extracts of Citrus unshiu Markovich have shown an antimicrobial effect against Bacillus cereus and Shigella sonnei.
To investigate the antimicrobial effect of the Patrinia scabiosaefolia extracts against food-borne pathogens, we extracted the P. scabiosaefolia with methanol at room temperature and the fractionation of the methanol extracts was carried out by using petroleum ether, chloroform, ethyl acetate, and methanol, respectively. The antimicrobial activity of the P. scabiosaefolia extracts was determined by using a paper disc method against food-borne pathogens and food spoilage bacteria. The ethyl acetate extracts of P. scabiosaefolia showed the highest antimicrobial activity against Escherichia coli and Shigella sonnei. Synergistic effect in inhibition was observed when P. scabiosaefolia extract was mixed Forsythiae fructus extract as compared to each extracts alone. Finally, the growth inhibition curves were determined by using ethyl acetate extracts of P. scabiosaefolia against Staphylococcus epidermidis and Shigella sonnei. The ethyl acetate extract of P. scabiosaefolia had strong antimicrobial activity against S. sonnei at the concentration of 4,000 ppm. At this concentration, the growth of S. Sonnei was retarded more than 72 hours and up to 48 hours for S. epidermidis. These results suggest that the ethyl acetate extracts of P. scabiosaefolia can be used for the efficient material against the growth of S. epidermidis and S. sonnei.
