Transcriptional Response of Major Antioxidant Enzyme Genes to Heat Stress in Mud Loach (Misgurnus mizolepis) (고온 스트레스에 대한 미꾸라지(Misgurnus mizolepis) 항산화 효소 유전자들의 발현 특징)

  • Expression of major antioxidant enzyme (AOE) including Cu/Zn superoxide dismutase (Cu/Zn-SOD), catalase (CAT), glutathione-S-transferase (GST) and 3 glutathione peroxidase isotypes (GPXs) at mRNA levels during heat stress was examined in mud loach (Misgurnus mizolepis) liver. Based on the semi-quantitative RT-PCR, real-time RT-PCR and/or northern dot blot hybridization, the antioxidant enzyme genes were generally up-regulated during elevation of water temperature from $23^{\circ}C$ up to $32^{\circ}C$. GPXs and SOD displayed the most significant elevation of mRNA levels (up to 3 and 2 folds, respectively) while CAT showed the steady-state expression irrespective of thermal conditions. GST represented the relatively moderate response (1.3-fold increase) in its transcription to thermal stress. The transcriptional activation of AOE genes was not significant at the treatment temperature lower than $29^{\circ}C$. Increased mRNA levels of GPX (extracellular form) and SOD genes in the fish exposed to $32^{\circ}C$ was readily detectable 1 day after exposure to heat stress.

The coping and defense features of the North Korean Defectors in the Rorschach test (로샤 검사에 나타난 북한이탈주민의 대처와 방어)

  • Coping and defense are similar psychological mechanisms to reduce stress response, but they are regarded as distinct from each other. Defense involved distortions of reality, but coping was used for strategies associated with accepting the true nature of the situation. The purpose of the present study was to investigate the coping and defense features of the North Korean Defectors in the Rorschach test. The subjects were then categorized into four groups based on their birth place and gender: Male North Korean defectors( N=16), female North Korean defectors(N=28), male South Koreans(N=17), and female South Koreans(N=25). All subjects were individually administered the Rorschach test, and protocols were scored according to the Exner(2003)'s Rorschach Comprehensive systems. Coping or defense-related Rorschach variables were selected for analysis: EA, CDI, Adj D, EB style, Lamda, Zd, X-%, Xu%, W:Dd:D, P, reaction time, and other behavior characteristics during the testing. Compared with other groups, female North Korean defectors were more likely to show poor coping resources(low EA), adjustment difficulties(Adj D), and distortions of reality(high X-%). They also demonstrated more 'don't know' responses and more delayed responses than other groups. These findings suggest that poor coping resources make female North Korean defectors more vulnerable to stress, and psychological intervention such as social skills training may help them settle successfully in south Korea. Limitations of the current study and implications for future research were discussed.

Changes of Drought Tolerance and Photosynthetic Characteristics of Populus davidiana Dode According to PEG Concentration (PEG농도에 따른 사시나무의 내건성과 광합성 특성의 변화)

  • We investigated changes in photosynthetic characteristics of P. davidiana in order to understand damage patterns to photosynthetic apparatus under drought stress. Root sprout saplings of P. davildiana were treated with $0\%,\;2\%,\;5\%,\;and\;10\%$ of 300ml polyethylene glycol (PEG) once a weer far one month. After one month, we measured photosynthetic parameters and analyzed the photochemical and $CO_2$ fixation systems. Photosynthetic rate, stomatal conductance, and respiration rate in the leaves of P. davildiana decreased according to increasing stress strength. In the photochemical system, quantum yield of PSII was reduced by the increment of PEG concentration, The decrease of apparent quantum yield was related to reduction of electron transport. Respiration rate decreased with an increase in PEG concentration, whereas photorespiration rate in the $CO_2$ fixation system increased. In conclusion, photosynthesis of P. davidiana responded sensitively under drought stress, and the sensitivity depended upon the strength of water stress. P. davidiana exhibited an increase of water use efficiency under water stress.

Expressional Analysis of Superoxide Dismutase in Olive Flounder (Paralichthys olivaceus) against Viral Hemorrhagic Septicemia Virus Infection (Viral hemorrhagic septicemia virus (VHSV) 감염에 대한 넙치 superoxide dismutase(Of-SOD)의 발현분석)

  • Superoxide dismutase is a family of important antioxidant metalloenzymes and catalyzes the dismutation of toxic superoxide anions into dioxygen and hydrogen peroxide. A recent study identified the partial superoxide dismutase (SOD) gene in olive flounder (Paralichthys olivaceus). The same study reported that it strongly induced benzo[a]pyrene and that it was an indicator of aquatic oxidative stress responses. However, its transcriptional response against viral infection has not been investigated. In the present study, the spatial and temporal expression profiles were analyzed to investigate the function of Of-SOD in the antiviral response. The Of-SOD transcripts were ubiquitously detected at various levels in diverse tissues in a real-time PCR. The expression of Of-SOD was significantly higher in the muscles, liver, and brain but extremely low in the stomach and spleen. Following a VHSV challenge, the expression of Of-SOD increased within 3 h in the kidneys and decreased to the original level 2 days postchallenge. In muscle, liver, and brain, Of-SOD mRNA was similarly up-regulated at 3-6 h postchallenge and then decreased to the basal level. Although the expression pattern and induction time differed slightly depending on the tissue, the transcript of Of-SOD consistently increased in the acute infection response, but the expression was low in the chronic response. The expression of Of-SOD was induced after the VHSV infection, and Of-SOD was probably involved in the immune response against the viral challenge. These results suggest that SOD may play important roles in the immune defense system of P. olivaceus and perhaps contribute to the protective effects against oxidative stress in olive flounder.

Changes of Vascular Contraction and Relaxation of Rat aorta under Arsenic Stress (비소 스트레스에 의한 흰쥐 대동맥의 수축과 이완반응의 변화양상)

  • In order to examine whether arsenic, one of environmental stress, contribute to augumentation and relaxation of rat aorta, this study was performed in vivo and in vitro, using intacted or denuded rats aorta ring preparation, respectively. The carotid arterial pressure was recorded on an ink-writing physiograph(Grass Co. 79E) connected to strain gauge. The contractile response of vascular ring with or without endothelium preparation isolated from rat were determined in organ bath and was recorded on physiograph connected to isometric transducer. Vasopressin-,and phenylephrine- induced increase in arterial pressure significantly enhanced in arsenic-treated rats; increase of 19.1%, and 46.6%, respectively. Vascular contractile response was measured in vitro preparations exposed to 0, 0.5, 1, 2 and 4 mM of arsenic for 1, 3, 5 and 8 hours. The dose-vascular responses of phenylephrine augmented by increasing dose of arsenic in the strips exposed to arsenic for 8 hours, and did not augmented for 1, 3, 5 hours. The phenomenon was not affected by strips denuded endothelium. And the response of relaxation of rat aorta induced by nitroprusside was not influenced by arsenic stress, but acetylcholine was a little increased. compared to that of control. There were no significant difference in relaxation between control and arsenic treated rings with endothelium or denuded. All of the results, phenyleprine-induced vascular contraction was significantly enhanced in 4 mM arsenic-treated rat aortic rings compared with control, whether endothelium was present or denuded at 8 hours after arsenic treatment. It may be a mechanism by which long-term arsenic stresses play a role in development of hypertension.

Isolation and Expression of Aluminum Induced Protein(ClAIP) Gene from Codonopsis lanceolata (더덕에서 Aluminum Induced Protein (ClAIP) 유전자의 분리 및 발현분석)

  • A cDNA clone (GenBank accession no. CF924621) homologous to aluminum induced protein gene was isolated and characterized from Codonopsis lanceolata (ClAIP). The ClAIP is 906 nucleotides long and has an open reading frame of 711 bp with a deduced amino acid sequence of 236 residues. The ClAIP shows high homology to A. marina (84%), G. hirsutum(83%), V. radiata (83%), A. thaliana (80%), B. nap us (78%) and T. aestivum (68%). The deduced amino acid sequence of ClAIP also has homology to the N-terminal end of plant Asn synthetase. This region does not contain the active sites of the enzyme and the significance of this conservation is currently not clear. To investigate the expression of ClAIP against several heavy metal stresses, we treated the sliced tap root of C. lanceolata with various heavy metals. The expression of ClAIP was increased by 25 uM $Al_2$(SO$_3$)$_4$ in proportion to incubation time and also increased by 50 uM CdCl$_2$.

The Relationship of the Expressions of Stress-related Markers and Their Production Performances in Korean Domestic Chicken Breed (닭의 스트레스 연관 표지인자들의 발현도와 생산능력 간의 상관 분석)

  • This study was conducted to verify the relationships between the expression values of stress-related markers and their production performances in 25 strains of Korean domestic chicken breeds. For stress response markers, the amount of telomeric DNA; expression levels of heat shock protein (HSP)-70, $HSP-90{\alpha}$, and $HSP-90{\beta}$; and comet scores were analyzed. Production performances were measured by the survival rate, body weights, days at first egg laying, egg weight and hen housed egg production. The results showed that the production traits and values of stress-related markers showed significant differences between strains. In general, the stress response of pure bred chickens with heavy weights was relatively high, while that of hybrid chickens with light weights was relatively low. The correlation coefficients between telomere contents and body weights showed that there were weak negative relationships. However, the correlations of telomere content with the survival rate and egg production were weakly positive after 20 weeks old. The expression levels of HSP genes and DNA damage rate (comet scores) were positively correlated to body weight, but were negatively correlated to the survival rate and egg production. The results implied that increasing body weight was associated with increasing HSPs expression and the DNA damage rate was associated with decreasing telomere content. In addition, increasing HSPs expression and the DNA damage rate decreased the survival rate and egg production, but the relationships with the telomere content was the reverse. Correlations among the stress-related markers showed that there were significant correlation coefficients between all of the marker values. HSPs expression was negatively correlated to the telomere content, while it was positively correlated to the DNA damage rate. There was a highly negative correlation between the telomere content and DNA damage rate. In conclusion, increasing the HSP values and DNA damage rate can promote telomere reduction, which led to a decrease in disease resistance and robustness of the chicken. Thus, increasing the stress response was verified to adversely affect the laying performance and viability of chickens.

The Effects of Repeated Restraint Stress on the Rat Parotid Glands, Ultramicroscopical and Histochemical Study (구속 스트레스에 대한 백서 타액선 조직의 미세구조적, 조직화학적 변화)

  • It has been known that saliva may affect the most of oral diseases. On the contrary, several systemic conditions may affect salivary flow and cause oral dryness and psychosocial stress especially may a crucial role in the etiology of hyposalivation and oral dryness. Many studies have focused on macroscopic effects of the stress on the salivary glands by autonomic response, but on the other hand it has hardly been reported on cellular microscopic effects of the stress on the salivary glands. Therefore, this study was performed to examine clusterin, a antiapoptotic and cytoprotective protein, in the parotid glands under restraint stress condition. For this study, 18 rats were divided into 3 groups; 1) 2 rats of group I were selected as a normal control. 2) 2 rats of group II, as a experimental control were placed in the restraint cone for 2 hours 3) 14 rats of group III were placed in the restraint cone for 2 hours once a day. The rats were sacrificed immediately(group II, as a experimental control), 1, 2, 3, 4, 5, 6 and 7 days after application of the stress and the both parotid glands were excised. Immunohistochemistry and electron microscopy were performed. The finding were as follows: 1. In parotid glands, repeated stress denaturalize the acinar cells, interacinous tissues and interacinous connective tissues were separated to individual acinar cells. After 4 days of experiment, there were lots of vacuoles and intercalated ducts. 2. In parotid glands, repeated stress make the rER which is in acinar cells swollen after 3 days of experiment and it was intensified to 4 days. After 5 days of experiment the edema got worse and degenerated. 3. In parotid glands, clusterin was reduced in ductal cell cytoplasm but in intercalated duct clusterin was slightly stained until 3 days prominently increased until 4 days and then decreased again after 5 days of experiment.

Evaluation of Biological Effects of Low Concentrations of Mercury Chloride (II) and Ionizing Radiation in the Prepubertal Male Rats (미성숙 웅성 흰쥐를 이용한 이온화 방사선 조사 및 저농도 염화수은(II)의 음용에 따른 위해성 비교 평가)

  • Mercury, one of the most diffused and hazardous organ-specific environmental contaminants, exists in a wide variety of physical and chemical states. The murcury with the nature which evaporates easily can cause an acute or chronic mercury poisoning to workers at mercury-handling workplaces. Although many studies indicate that mercury induces a deleterious damage, little has been reported from the investigations of mercury effects at surrounding levels in living things. The purpose of this study was to evaluate the biological effects of mercury chloride and ionizing radiation. Prepubertal male F344 rats were administered mercury chloride in drinking water throughout the experimental period or were given wholebody irradiation with a dose of 6.5 Gy. The amount changed of body weight during the experimental period showed a 4.9% rise in the mercury-treated group and 14.4% decline in the irradiated group compared with the level of the control group. The results of hematological analysis (red blood cells, white blood cells, hemoglobin, and hematocrit) indicated the differential effects of mercury chloride and ionizing radiation. However the concentration of cortisol as assessed by radioimmunoassay increased in both of the groups. Relative expressions of mRNA related to mitochondrion-mediated apoptosis were investigated using semiquantitative reverse transcription polymerase chain reaction on gonad and urinary organs of the experimental groups. While the expression of Bcl-2 mRNA exhibited different patterns depending on the organs or the experimental groups, both of the experimental groups showed a conspicuous expressions of Bax mRNA. In conclusion, the target organ of mercury chloride seems to be a urinary organ and the pattern of damage induced by mercury chloride differs from that by ionizing radiation.

Miscanthus EST-originated Transcription Factor WRKY Expression in Response to Low Temperature in Warm-season Turfgrasses (억새 EST 정보 유래 전사요소 WRKY의 난지형 잔디의 저온 발현 반응성)

  • Whole genome transcriptomes from Miscanthus species were sequenced and analyzed, which provided 50 different types of transcription factor (TF) involving various developmental processes or environmental stresses. Among the explored TF, WRKY gene family was the major type and one of the WRKY genes, MSIR7180_WRKY4, induced under low temperature environment was selected to investigate how the Miscanthus-originated MSIR7180_WRKY4 TF responds when exposed to low temperature in four warm-season turfgrasses (Z. matrella 'Semil', bermudagrass, St. Augustinegrass, and seashore paspalum). The MSIR7180_WRKY4 was expressed higher during low temperature period in Bermudagrass, but the expression was enhanced in St. Augustinegrass. In contrast, the gene in 'Semil' cultivar was barely expressed and relatively less expressed, but repressed gradually in seashore paspalum, which seems to allow two turfgrasses stay-green longer in the fall season. The results indicate that bermudagrass and St. Augustinegrass adapt to low temperature quickly, but relative tolerance to low or cold temperature at the molecular level needs to be further investigated at different physiological stages and the corresponding genes systematically.
