Effect of the Mechanochemical Pretreatment on Antioxidant Material Extraction from Pu'er Tea Leaf (Camellia sinensis var. assalnlca) (보이차잎 항산화 물질추출에 메카노케미스트리 전처리 효과분석)

  • 보이차의 주요산지는 중국 운남성이며 특히 란창강 유역이 그 중심지이다. 천연 사포닌과 미네랄류를 풍부하게 포함하고, 지방의 용해, 다이어트 효과, 소화 촉진, 정장 작용, 숙취 해소, 위가 더부룩할 증상 개선, 혈당치 상승 억제, 혈액 순환 촉진에 효과가 있는 것으로 알려져 있다.또한, 면역력 강화 효과와 노화예방, 암 예방효과도 있는 것으로 알려져 있다. 보이차의 기능성 물질은 일반적으로 메탄올 또는 에탄올 용매에 의하여 추출한다. 용매에 의하여 추출하기 전에 메카노케미스트리 분쇄기술을 적용하여 전처리하면 재료의 표면적을 증가시키고 부분적으로 화학적 성분을 변화시켜 기능성 물질의 추출효율을 증가시킬 수 있다. 본 연구에서는 메카노케미스트리 분쇄기술을 적용하여 보이차를 전처리한 다음 메탄올과 에탄올 용매에 의하여 기능성 물질을 추출하여 메카노케미스트리 전처리가 항산화 물질의 추출효율에 미친 효과를 분석하였다. 보이차는 메탄올과 에탄올 추출 전에 유성밀에 의하여 분쇄되었으며 분쇄 후의 형상을 SEM 현미경으로 분석하였다. 아질산성질소 소거능은 pH 1.2에서 메탄올 용매추출을 했을 때 전처리하지 않는 경우 63.0-83.0%로부터 메카노케미스트리 분쇄기술을 적용한 전처리를 한 경우에 74.0-92.0%로 증가하였다. 전자공여능은 DPPH(1,1-diphenyl-2-pycrylhydrazyl)를 이용한 방법으로 측정하였으며, 메탄올 용매추출의 경우에 13.12-49.29%로부터 메카노케미스트리 전처리 후에 15.12-64.29%로 증가하였다. DPPH radical 50% 소거능을 나타내는 $IC_{50}$은 전처리하지 않는 경우 164 ug/mL로부터 전처리한 경우에 151ug/mL로 감소하였다.

Study on the Alleviation of an Alcohol Induced Hangover and the Antioxidant Activity by Mulberry Fruit (오디 추출물의 알코올 분해능 및 항산화 효과)

  • This study investigated the effects of mulberry fruit extract on antioxidant activities and the alleviation of an alcoholinduced hangover, which was measured with alcohol dehydrogenase(ADH) enzyme activity. The antioxidative capacity of each mulberry extract was measured by total phenolic compounds, electron donating ability, superoxide dismutase-like ability, thiobarbituric acid reactive substances, and nitrite scavenging ability. The 60% methanol extract yielded the highest total phenolic compounds. The electron donating ability of the 40% ethanol and 40% methanol extracts were 68% and 67%, respectively. The superoxide dismutase-like abilities of the 40% methanol and 40% ethanol extracts were 30% and 28%, respectively, when extracts were assayed at 2.5 mg/$m\ell$. The nitrite scavenging ability of the 60% ethanol and 40% methanol extracts were 98% and 97%, respectively. The 60% ethanol extract yielded the highest ADH.

Anti-Diabetic, Alcohol-Metabolizing, and Hepatoprotective Activities of Moringa (Moringa oleifera Lam.) Leaf Extracts (모링가 잎 추출물의 항당뇨, 알코올 대사 및 간 보호 활성)

  • This study was carried out to investigate anti-inflammatory, anti-diabetic, alcohol metabolizing, and hepatoprotective effects of hot water (MOW) and 80% ethanol (MOE) extracts from moringa (Moringa oleifera Lam.) leaf. The total phenol content of MOW and MOE were 45.49 and 63.06 mg tannic acid equivalents/g, respectively. 1,1-Diphenyl-2-picrylhydrazyl radical scavenging activities of MOW and MOE were remarkably elevated in a dose-dependent manner, and about 60.8% and 71.3% at 1 mg/mL, respectively (P<0.01). Superoxide dismutase-like activities of MOW and MOE were 2.8% and 7.4% at 5 mg/mL, respectively (P<0.05). ${\alpha}-Glucosidase$ inhibitory activity also increased in a dose-dependent manner in both extracts, and MOE was higher about two times than MOW at 5 mg/mL (P<0.001). The effects of MOW and MOE on alcohol metabolizing activity were determined by measuring generation of reduced nicotinamide adenine dinucleotide (NADH) by alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase (ALDH). ADH and ALDH activities significantly increased upon addition of MOW and MOE (P<0.05). Anti-inflammatory activity was examined in lipopolysaccharide-stimulated RAW 264.7 cells. Nitric oxide production was reduced to 32.1% and 81.2% by addition of MOW and MOE at 1 mg/mL, respectively (P<0.05). MOW and MOE showed significant protective effects against tacrine-induced cytotoxicity in Hep3B cells at $100{\mu}g/mL$. These results suggest that moringa leaf extracts have great potential as natural health products.

Inhibitory Effects of Angiotensin Converting Enzyme and α-Glucosidase, and Alcohol Metabolizing Activity of Fermented Omija (Schizandra chinensis Baillon) Beverage (오미자 발효음료의 알코올 분해능과 Angiotensin Converting Enzyme 및 α-Glucosidase 저해효과)

  • The nutraceutical role of fermented omija (Schizandra chinensis) beverage (FOB) was determined through the analysis of fibrinolytic and alcohol metabolizing activities, nitrite scavenging activity, and angiotensin converting enzyme and $\alpha$-glucosidase inhibitory effects. Firstly, FOB increased fibrinolytic activity in a dose-dependent manner and indicated angiotensin converting enzyme inhibitory activity of 94.8% at 20% FOB (0.6 mg/mL). In addition, the inhibitory activities of FOB on $\alpha$-amylase and $\alpha$-glucosidase were determined to be 100% at 100% FOB (3 mg/mL) and 49% at 60% FOB (1.8 mg/mL), respectively. Nitrite scavenging activity of FOB was about 96.1%, 72.3%, and 68.3% on pH 1.2, 3.0, and 6.0 at 100% FOB, respectively. To determine influence of FOB on alcohol metabolism, the generating activities of reduced-nicotinamide adenine dinucleotide (NADH) by alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase (ALDH) were measured. Facilitating rate of ADH activity was 70.3% at 50% FOB, but ALDH activity was not affected. These results revealed that FOB has a strong alcohol metabolizing activity, and fibrinolytic and nitrite scavenging activities and exhibits the angiotensin converting enzyme, $\alpha$-amylase, and $\alpha$-glucosidase inhibitory activities.

Comparison of Hepatic Detoxification activity and reducing Serum Alcohol concentration of Hovenia dulsis $T_{HUNB}$ and Alnus japonica Steud. (헛개나무와 오리나무 추출물의 간 해독작용 및 체내 알콜 분해능 비교)

  • It was found that the level of alcohol concentration in both mouse and human sera can be significantly decreased up to 42% by oral administration of the mixtures of the extracts of Hovenia dulcis THUNB and Alnus japonica Steud. A single treatment of extract from Hovenia dulcis reduced the serum alcohol concentration to 32%, compared to 13% in treating the extract of Alnus japonica. Similar patterns were observed in enhancing alcohol dehydrogenase (ADH) and glutathion-S-transferase (GST) activity in the liver. The inhibition of cathepsin activity was also greatly reduced by administrating the mixture of both extracts : however, the extract of Alnus japonica did not affect the acitivity of cathepsin. It was concluded that the mixture of both extracts had synergic effect on reducing serum alcohol concentration and improving the detoxification process due to alcohol administration in the liver.

Effect of Hot Water Extracts of Blue Mussel and Several Plants on Alcohol Metabolizing Enzymatic, Antioxidant, and Angiotensin Converting Enzyme Inhibitory Activities (홍합과 여러 식물의 열수 추출물의 알코올분해효소에 미치는 영향과 항산화 및 ACE 저해 활성)

  • For the development of a hangover soup containing blue mussel, 11 kinds of hot water extracts were prepared - A (mistletoe); B (shepherd's purse); C (arrowroot); D (bean sprout); E (oriental raisin); F (blue mussel); G (blue mussel and mistletoe); H (blue mussel and shepherd's purse); I (blue mussel and arrowroot); J (blue mussel and bean sprout); and K (blue mussel and oriental raisin). Extract C showed the highest effect for increasing the activities of alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase (ALDH); however, the addition of blue mussel did not provide a synergy effect on extract C. Other than the arrowroot-containing extracts (C and I), extract H showed relatively higher ADH ($237.4{\pm}1.7%$) and ALDH ($136.5{\pm}2.1%$) activities. Moreover, extract H showed the highest DPPH radical scavenging activity ($93.9{\pm}0.1%$) and angiotensin I converting enzyme (ACE) inhibitory activity ($42.7{\pm}1.6%$). The combination of blue mussel with shepherd's purse had a synergic effect on its ADH and ACE inhibitory activities.

Quality Characteristics and Storage Properties of Sikhe Prepared with Extracts from Hovenia dulcis THUNB (헛개나무열매 추출물을 첨가한 식혜의 품질특성과 저장성)

  • Sikhe, a traditional Korean drink, was prepared with Hovenia dulcis extract (HDE), and its quality and storage characteristics were evaluated through sensory and microbial analyses. In the color analysis, the lightness of the Sikhe showed a decreasing tendency according to the addition of HDE, while redness and yellowness increased. The Sikhe prepared with 20% HDE had the highest sugar concentration at 12days of storage ($4^{\circ}C$). Interms of sensory quality, both the 20% and 40% HDE-added Sikhes had the highest overall preference. The microbial cell counts presented no distinct differences early in the storage. period, but the general bacteria counts of the Sikhe made with the increasing concentrations of extracts decreased with a longer storage period. The coliform counts of the Sikhe prepared without HDE (control) was 10.5 CFU/g at 10 days of storage, but the 100% HDE-added Sikhe had a level of 7.9 CFU/g at 16 days of storage. These results indicates that Hovenia dulcis may inhibit normal microbial growth and extend the shelf-life on Sikhe.
