Volatile flavor components of Korean ginger(Zingiber officinale Roscoe)extracted with liquid carbon dioxide (액체 이산화탄소 추출법에 의한 생강 (Zingiber officinale Roscoe)의 향기성분)

  • The essential oil of Korean ginger(Zingiber officinale Roscoe) was isolated by liquid carbon dioxide extraction method and fractionated into one hydrocarbon fraction and two oxygenated hydrocarbon fractions by using silica gel column chromatography. The compositions of the resulting oils were investigated by GC and GC-MS spectrometry. Out of 102 identified compounds, 44 were identified by comparing GC retention time and mass spectral data with authentic samples and 58 were tentatively identified according to mass spectral data only. The major compounds of hydrocarhon fraction were $zingiberene,\;{\beta}-sesquiphellandrene,\;{\gamma}-bisabolene,\;{\gamma}-cardinene,\;ar-curcumene$, and those of oxygenated hydrocarbon fractions wee geranal, sesquisabinene hydrate, borneol and zingiberenol. The major compounds of ginger oil were zingiberene, $citronellol+{\beta}-sesquiphellandrene,\;geranial,\;{\gamma}-bisabolene\;and\;ar-curcumene+geranyl\;acetate$, and ginger oil contained higher amounts of sesquiterpene hydrocarbons. The yield of extract was 6.96%.

  • For the synthesis of polyurethane memory foam stabilizer with fine cells, hydrosilylation reaction with various polyalkyleneoxides and hydrogen functional group of polymethylhydrogensiloxane (D = 75, D' = 15) was conducted. Polyalkyleneoxides (PAO) used in this research were ethylene oxides or ethylene-co-propylene oxides with terminal groups of hydroxides or methyl groups. To analyze the molecular structures and molecular weights as well as the reaction yields (98%), NMR and GPC analysis were executed. Synthesized siloxane surfactants modified with polyalkylene (EO = 12 units) were applied to producing flexible polyurethane fine memory foams from 0.6 pphp to 2.0 pphp. By controlling the amount of the surfactant, physical characteristics, the polyurethane memory foam with cell size (minimum $0.868{\mu}m$), air flows (-78 KPa), and recovery times (8 sec) were achieved.

  • Propylene epoxidation by $H_2O_2$ (30% aqueous) as oxidant was studied in a semi-batch reactor using TS-1 catalyst: Effects of reaction temperature, time, pressure, solvent, catalyst and $H_2O_2$ concentration on $H_2O_2$ conversion (limiting reagent) and product distribution were investigated. Potential inhibition by propylene oxide on the epoxidation rate was also examined. Ti-MCM-22 with MWW zeolytic structure was found to exhibit better performance than TS-1 with MFI structure, provide that a proper choice of solvent(acetonitrile) is made.

  • LFG (Land-Fill Gas) includes components of $CH_4$, $CO_2$, $O_2$, $N_2$, and water. The preparation of synthesis gas from LFG as a DME (Dimethyl Ether) feedstock was studied by methane reforming of $CO_2$, $O_2$ and steam over $NiO-MgO-CeO_2/Al_2O_3$ catalyst. Our experiments were performed to investigate the effects of methane conversion and syngas yield on the amount of LFG components over $NiO-MgO-CeO_2/Al_2O_3$ catalyst. Results were obtained through the methan reforming experiments at the temperature of $900^{\circ}C$ and GHSV of 8,800. The results were as following; it has generally shown that syngas yield increase with the increase of oxygen and steam amounts and then decrease. Highly methane conversion of above 98% and syngas yield of approximately 60% were obtained in the feed of gas composition flow-rate of 243ml/min of $CH_4$, 241ml/min of $CO_2$, 195ml/min of $O_2$, 48ml/min of $N_2$, and 450ml/min of steam, respectively, under reactor pressure of 1 bar for 200 hrs of reaction time. Also, it was shown that catalyst deactivation by coke formation was reduced by excessively adding oxygen and steam as an oxidizer of the methane reforming.

  • The hydrocarbons containing more than 10 carbons(0.1% of total volume, C10+), residue of aromatization from aliphatic hydrocarbons, were condensated in the presence of catalyst aluminumchloride and cocatalyst nitrobenzene(NB) to be pitchs with desirable properties. The properties of pitch were affected by the concentration of cocatalyst chosen 20 and 30wt.%. The pitch with 30wt.% NB showed higher carbon yield and lower crystallinity than that with 20wt.% NB. The two pitches were heat treated at 1000C and measured of charge/discharge capacity of the carbon as an anode. The carbon prepared at 20wt.% NB exhibited excellent cyclic stability with a capacity of 218mAh/.g and that at 30wt.% exhibited rather low cyclic stability with higher capacity of 235mAh/g.

  • Poly-${\beta}$-hydroxybutyrates(PHB) are well known intracellular biopolymer which are completely degraded to carbon dioxide and water in the landfill. The pressurized fermentation method was used to increase the cell growth of Alcaligenes eutrophus and productivity of PHB. The experimental data were analyzed in terms of carbon source and gas composition effects at various elevated pressures and temperatures. The results reveal that the flow rate of hydrogen gas of 0.0075vvm for the culture is better PHB production than the no hydrogen flow when fructose was used as a sole carbon source. The higher yields and productivities of PHB biosynthesized by A. eutrophus were obtained when the oxygen composition was changed from 2% to 8% at 6atm and $30^{\circ}C$.

  • The catalyst carriers of the mesoporous layer compounds were prepared to carry out the partial oxidation of methane(POM) to hydrogen. The catalytic activities of POM to hydrogen were investigated over Ru(3)/SPK and Ru(3)/SPM catalyst in a fixed bed flow reactor under atmosphere. In addition, the catalysts and carriers were characterized by BET, TEM, TPR. The BET surface areas of the silica-pillared $H^+-kenyaite$(SPK) and the silica-pillared $H^+-magadite$(SPM) were $760m^2/g$ and $810m^2/g$, repectively, and the average pore sizes were 3.0 nm and 2.6 nm, repectively. The nitrogen adsorption isotherms were type IV with developed hysteresis. The TEM showed that the mesoporous layer compounds were formed well. The Ru(3)/SPK and the Ru(3)/SPM catalyst were obtained high hydrogen yields(90%, 87%), and were kept constant high hydrogen yields even about 60 hours at 973 K, $CH_4/O_2=2$, $1.25{\times}10^{-5}g-Cat.hr/ml$. The TPR peaks of Ru(3)/SPK and the Ru(3)/SPM catalyst showed the similar reducibilities around 453 K and 413 K. It could be suggested that SPK and SPM had the physicochemical properties as oxidation catalyst carries from these analysis data.

  • In order to obtain basic information on the production of single cell protein from petroleum, more than 400 yeast strains were isolated from various soil samples in Korea utilizing petroleum hydrocarbon as the sole carbon source. A yeast strain showing the highest cell yield among the isolated strains was selected and identified. The optimal culture condition was searched in the flasks shaken throughout the procedure. And the growing characteristics for the selected yeast strain and chemical analysis of the yeast cell component were carried out. The results obtained were as follows: 1. The selected yeast strain was identified as Candida curvata and we named it Candida curvata-SNU 70. 2. The composition of the medium proposed for the present yeast strain is: Light Gas Oil 30ml, Urea 400mg, Ammonium sulfate 100mg, Potasium phosphate (monobasic) 670mg, Sodium phosphate (dibasic) 330mg, Magnesium sulfate 500mg, Calcium carbonate 3g, Yeast extract 50mg, Tween 20 0.05ml, Tap water 1,000ml. 3. Other culture conditions employed for the yeast were pH 5.5-7.0, temp. $30^{\circ}C$ under an affluent aerobic state. 4. Addition of light gas oil in portions to the culture media as the growth proceeded was more effective, especially in the cultivation on the higher oil concentration media. 5. Studies on the propagation of the yeast cells in the light gas oil medium revealed that the yeast has the lag phase lasted 16 hours and the logarithmic growth phase covered 16 to 28 hours. The specific growth rate was about $0.22\;hr^{-1}$ and doubling time was 3.2 hrs. during the logarithmic growth phase. 6. Under the cultural condition employed, the cell yield against the amount of light gas oil (wt%) was 16.1% and the protein content of the dried yeast cells was 48.4%.

Inhibitory Effects of Copper on the Anaerobic Degradation of Propionate (프로피온산의 혐기성 분해시 구리의 저해 효과)

  • The effects of copper on the anaerobic degradation of propionate were studied using anaerobic batch reactors. The apparent inhibitory effects of copper on the anaerobic degradation of propionate could be observed from behaviors of intermediates, ultimate methane yield(UMY) and specific methanogenic activity(SMA) There was little inhibition at the concentration of $2.5mg\;Cu^{2+}/L$. Beyond this concentration, the inhibitory effects increased with increasing dose of coppers. The 50% inhibition of UMY and SMA occurred at copper dosage of 33.8 and $24.1mg\;Cu^{2+}/gVSS$, respectively. The inhibitory effect based on the UMY was gradually reduced with the operation time dueprobably to the acclimation of microorganisms and/or binding of the added copper by ligands(and possibly ion exchange sites)contained on the cell membrane and extracellular polymer matrix whereas it based on the SMA might exclude the this phenomena. Therefore, the methodology for interpretation of inhibition data based on the SMA was more accurated than the UMY. There was no inhibitory effect in batch reactors supplemented with sulfate due to an antagonistic action of the sulfate reducing bacteria. Propionate degradation was initially retarded for copper inhibited samples but it gradually degraded afterward. Based on the mass removal considering take into account the propionate to acetate conversion, propionate degradation may appeal more affected than acetate. This result revealed that the hydrogenotrophic methanogens were the most affected by copper.

Molecular Analysis of the Microorganisms in a Thermophilic CSTR used for Continuous Biohydrogen Production (연속수소생성에 사용되는 고온 CSTR 내의 미생물의 분자적 분석)

  • Molecular methods were employed to investigate microorganisms in a thermophilic continuous stirred tank reactor(CSTR) used for continuous $H_2$ production. The reactor was inoculated with heat-treated anaerobic sludge and fed with a glucose-based medium. Denaturing gradient gel electrophoresis showed dynamic changes of bacterial populations in the reactor during 43 days of operation. Gas composition was constant from approximately 14 days but population shift still occurred. Populations affiliated with Fervidobactrium gondwanens and Thermoanaerobacterium thermosaccharolyticum were dominant on 21 and 41 days, respectively. Keeping pH of the medium at 5.0 could suppress methanogenic activity that was detected during initial operation period. $CH_4$ and mcrA detected in the samples obtained from the reactor or inoculum suggested the heat treatment condition employed in this study is not enough to remove methanogens in the inoculum. PCR using primer sets specific to 4 main orders of methanogens suggested that major $H_2$-consuming methanogens in the CSTR belong to the order Methanobacteriales.
