Occurrence of Syzygites Megalocarpus on Wild Mushrooms and Its Cultural Characteristics (야생(野生)버섯의 기생균(寄生菌)인 이분지털곰팡이의 출현(出現)과 배양(培養) 특성(特性))

  • Syzygites megalocarpus occurred on the fruit bodies of Tricholoma matsutake, Leccinum rugosiceps, Pulveroboletus ravenelii, Russula emetica, and Amanita pseudoporphyria in the field. We could select Malt Extract Agar, Mueller Hinton Medium and Potato Dextrose Agar as optimal media among eight media. Syzygites megalocarpus showed an optimal temperature around $23^{\circ}C$ with optimal pH 6.0. Growth of S. megalocarpus on PDA was inhibited by 36.5% at 5% NaCl compared with without NaCl and did not grow at more than 8% NaCl in 2 days after inoculation. However, it grew 1.1 cm in 10% NaCl in 5 days after inoculation. Growth of Tricholoma matsutake on PDA was inhibited by increasing contration of NaCl and did not nearly grow at 2.5% NaCl in 60 days after inoculation. Because S. megalocarpus grew at high concentration of NaCl, we concluded that NaCl should not use for controlling S. megalocarpus on the fruit body of T. matsutake.

Production of Phytoncide from Korean Pine Cone Waste by Steam Distillation (잣송이 부산물로부터 수증기 증류법에 의한 피톤치드의 추출)

  • Extraction of phytoncide oil from korea pine cone waste without damaging the pine cone tree itself was investigated using a steam distillation method. Also various components in the extracted phytoncide oil were separated using a column chromatography method. The extraction of phytoncide oil was effectively proceeded, and the maximum production yield of phytoncide oil could be obtained under $100^{\circ}C$ of distillation temperature and within 30 minute of distillation time. According to chemical analysis, it was found that the phytoncide oil from korea pine cone waste was consisted of more than 12 components such as ${\alpha}$-pinene, ${\beta}$-pinene, D-limonene, as main components. In addition, the aqueous hydrogel containing other components such as verbenone, ${\alpha}$-terpinieol, fenchol, different from components of phytoncide oil itself could be obtained through the steam distillation.

  • (1) 급액천의 규격별 양액공급량 $\bigcirc$ 합섬(비스코스+레이온) : 시간당 소(12.6ml), 중(21.2ml), 대(57.5ml)의 순으로 유입되었으며 중과 대의 급액천의 유입은 거의 같아 급액천의 규격간에 공급량은 비례하지 않았다. $\bigcirc$ 합섬(폴리에스텔) : 소(4.3ml), 중(20ml), 대(57.5ml)순으로 양액이 유출되었으며, $\bigcirc$ 합섬(인견) : 소(2.9ml), 중(23.7ml), 대(32.3ml)순으로 유입되었으나 소(급액천)의 유출량은 적어 급액천 재질로서 부적합하다고 생각되었으며, $\bigcirc$ 실(cotton) : 소(0.2ml), 중(3.0ml), 대(2.7ml)순으로 규격간에 유입은 차이가 없었으며 전체적으로 유출이 적어 급액천으로서는 부적합하였다. (2) 배지의 규격(종류)별 양액의 유입량 $\bigcirc$ 송이(Ф6.0mm~12.0mm이상) : 배지의 입자가 굵은 배지일수록 급액초기 1~2시간이 유입이 급격히 증가하였으나 시간이 경과할수록 일정한 량을 보였으며 유입량은 다른 배지에 비하여 많았고 급액천의 규격이 클수록 유입되는 량이 많았으나 시간이 경과함에 따라 감소하는 경향을 보였다. $\bigcirc$ 송이(Ф1~5mm) : 합섬A(비스코스+레이온)과 합섬B(폴리에스텔)에서는 시간당 15.0~50.1ml가 유입되었으나 합섬C(인견)과 실에서는 시간당 10ml이하로 토출량이 적었다. $\bigcirc$ 송이(Ф1~12mm) : 합섬A와 합섬B 급액천에서는 17.0~53.8ml가 유입되었으나 합섬C와 실에서는 시간당 평균 4.0~21.2ml가 흘러나와 배지의 공극이 일정하지 않아 시간당 통과하는 양이 일정하지 않았다고 생각되었다. $\bigcirc$ 펄라이트 : 합섬A(비스코스+레이온)급액천의 유입은 소(1$\times$60cm)에서 21.8ml, 중(2$\times$60cm) 33.5ml, 대(3$\times$60cm) 43.4ml가 통과되었고 합섬(폴리에스텔)에서는 19.0~30.7ml로서 급액천의 규격에 따라 통과되는 차이가 있었다. 배지가 규격화되어 있어 급액천의 규격별로 일정하게 유입되었으며 급액천의 재질이 유입에 영향을 미친 것으로 사료되었다. (2) 급액관과 베드상과의 높이에 따른 유출양 : 급액과 베드상과의 낙차가 클수록 유출이 증가함을 알수 있었으나 합섬C(인견)실험구에서는 낙차가 유출에 영향을 미치지 않았다. (4) 급액된 양액의 EC 및 pH조사 : 급액된 양액의 EC 및 pH에 전혀 변화가 없어 재배 적응에 문제가 없을것으로 사료되었다.

Correlation Coefficients between Pine Mushroom Emergence and Meteorological Elements in Yangyang County, Korea (양양지역 송이 발생과 기상요소의 상관관계)

  • The relationships between pine mushroom emergence and meteorological factors were analyzed with three years (from 2003 to 2005) of measurement data at Yangyang site, in order to evaluate the effect of micrometeorological environment on pine mushroom production. fine mushroom was daily monitored and collected in the survey area during the its producing period (approximately one month). Pine mushroom production was highest in 2005 with the meteorological conditions of high temperature and frequent rainfalls in October. The production was lowest in 2004 due to dry conditions from mid September to late October, The meterological factors related to humidity (i.e., relative humidity, soil water content, and precipitation) were better correlated than those related to temperature (i.e., air and soil temperature, soil heat flux and solar radiation) with pine mushroom production. However, all of the correlation coefficients were statistically insignificant with values ranging from 0.15 to 0.46. Such poor correlations may be attributed to various other environmental conditions (e.g., topography, soil, vegetation, other fungi, the relationship between pine mushroom and pine forest) affecting pine mushroom production. We found that a mycelium requires a stimulation of low temperature (of three-day moving average) below $19.5^{\circ}C$, in order to farm a mushroom primordium which grows to pine mushroom after 16 days from the stimulation. We also found that the pine mushroom production ended when the soil temperature (of three-day moving average) fell below $14.0^{\circ}C$.

Aroma Characteristics of Tricholoma matsutake Mushrooms Collected from Eleven Major Sites in Korea (한국(韓國)의 11개(個) 주요(主要) 산지(産地)에서 채집(採集)한 송이(松栮)의 향기성분(香氣成分)에 관(關)한 연구(硏究))

  • The objectives of this study were to identify aroma characteristics of Tricholoma matsutake(S. Ito et Imai) Sing. growing in different geograghic ranges in South Korea. Mushrooms were collected from 11 major sites which included four sites(Bonghwa, Uljin, Goryung and Chungdo) in Kyongbuk Province, three sites(Changnyung, Hadong and Hamyang) in Kyongnam Province, two sites(Yangyang and Inje) in Kangwon Province, one site(Goisan) in Choongbuk Province, and one site(Namwon) in Chonbuk Province. One of three mushrooms from each site were used for measurements of aromatic characteristics. Aromatic compounds were identified using a dynamic headspace trap method at $40^{\circ}C$ and GC-MSD(gas chromatograph-mass spectrometric detector) method. A total of 25 aromatic compounds were identified. Large variations in the composition and amount of aromatic compounds were noticed. Major aroma compound was 1-octen-3-ol, while methyl cinnamate known as the major aroma compound in matsutake was not detected at all. The total amount of aromatic compounds was highest in Bonghwa, and decreased in the order of Chungdo, Inje, Hamyang, Uljin, and Yangyang. Based on the total amount, kinds and amounts of individual aromatic compounds, the mushrooms from 11 major sites were grouped into following four types : 1) Bongwha Type : the total amount of aromatic compounds was highest, with major compound being 3-methyl 1-butanol. 2) Hamyang Type including Chungdo, Yangyang and Inje : 1-octen-3-of comprised 2/3 of total aromatic compounds, with second major being 3-methyl butanal. 3) Uljin Type : 1-octen-3-ol comprised 94% of total aromatic compounds, with other compounds being almost nothing. 4) Goisan Type including Goryung, Namwon, Changnyung, and Hadong : the total amount of aromatic compounds was lower than other three types.

Characteristics of new strain 'Jinbudlsongyi' in Agrocybe aegerita (버들송이 신품종 '진버들송이'의 특성)

  • Jinbudlsongyi mushroom (Agrocybe aegerita) was bred in Mushroom Research Institute, Gyonggi Province A.R.E.S in 2005. It was bred with mating between moookarytic strains isolated from Budlsongyi#1 and KME45202. The temperature of optimal mycelial growth was $24{\sim}26^{\circ}C$ on PDA and $18{\sim}20^{\circ}C$ in sawdust medium. To harvest fruiting body of Jiobudlsoogyi the period required for colonization after inoculation was 37days and the period of cultivation was 13days after scratching of inoculum. The major characteristics of the mushroom was shown a lot of primordia, light brown colored stipe and convex-shaped pileus. Compared with Budlsongyi#1, it was shown dark brown-colored pileus and straight stipe. The fruiting body yield was $115g{\pm}6/850cc$ bottle. It was demanded proper circulation at the time of primordia and put it around with cloth that is prevented to be bend the stipes.

Studies on the Host Selectivity and Pathogenicity of White Pine Blister Rust Fungus (Cronartium ribicola J. C. Fischer ex Rabenhorst) in Korea (우리나라 잣나무 털녹병균(病菌)의 기주선택성(寄主選擇性)과 병원성조사(病原性調査))

  • Pedicularis spp. are the alternate hosts of the white pine blister rust fungi in the forests of Korea, and Ribes spp. were found as other alternate hosts by artificial inoculation test. New biotypes (races) with different host selectivity and pathogenicity of the causal fungi were speculated at various places in Korea. The aeciospores of the fungi were collected from two isolated places, Pyeongchang and Namwoen, and artificially inoculated on Pedicularis resupinata and six species of the genus Ribes. Among the tested alternate host species, P. resupinata and R. montigenum were only susceptible to both the Pyeongchang population and the Namwoen population, showing similar pathogencity between the two fungai populations. Three white pine species were planted in fungus-infested stands to study the susceptibility of each tree species. Four years later, 17.7% of Pinus koraiensis and 26.3% of the P. strobus were infected, but P. parviflora was free from attack. From the above results, it was concluded that the white pine blister rust fungus populations in Korea are of the same biotype, with P. koraiensis and P. strobus as aecial hosts, and Pedicularis spp. and Ribes spp. as telial hosts.

Growth and Fruit Characteristics of Grape 'Campbell Early' by Securing Planting Distance (재식거리 확보에 의한 '캠벨얼리' 포도의 생육 및 과실 특성)

  • Shoot and the fruit growth characteristics of 'Campbell Early' grape cultivar were investigated after vine thinning using the main branch leader. Before vine thinning, branch diameter was 11.8 mm, but it decreased to 9.3 mm after vine thinning. The number of shoots per vine increased to 55.9-64.5 after vine thinning. The number of fruit clusters per shoot was higher (1.5-1.75) in the vine thinning plot than in the control (0.8). Consequently, the number of clusters per vine was 98.1-106.5 in the vine thinning plot, while that in the control was 24.4-28.0. However, soluble sugar content (SSC) and tartaric acidity showed no significant differences between the control and the vine thinning plot. The proportion of >351 g fruit cluster weight after vine thinning (81.0%) was significantly higher than that of the control (32.5%). These results imply that fruit quality based on the variation in cluster weight and fruit yield was enhanced by vine thinning using the main branch leader.

A Study on the Ground Reinforcement of Jeju Scoria Layer by Chemical Grouting (약액주입에 의한 제주도 송이지층의 지반보강에 관한 연구)

  • Recently, public works becoming bigger in Jeju are implemented various kinds of ground reinforcement method including the chemical grouting method. In this study, we have been investigated on the proper material and the injection condition for the excellent injection effect and the excellent strength of injection material and the permanent waterproof and reinforcement through the experiment. The kind of injection material has been selected through the uniaxial compression test and the endurance test of injection material as the chamber test. An experiment was performed with model ground made of scoria, the injection performance of selected material has been identified through the evaluation test of injection range using the decision test of injection amount and the calibration chamber test. As a result of test, it has been analyzed that MSG appeared to have the excellent strength, durability and injection performance all compared with the ordinary cement, this result is judged to be possible as the ancillary data of design at time of design and construction with the chemical grouting method in the future.
