Plants of Vitex negundo are known to develop numerous trichomes throughout their body, where certain trichome types have been believed to be one of the plausible structures for the unique scents. In the current study. structural aspects of the trichomes have been examined in leaves and stems of Vitex negundo using TEM and SEM. Trichome types as well as structural changes that occurred in certain trichomes during secretion have been mainly focused. Three type of glandular trichomes and two types of non-glandular trichomes were developed in the epidermis of young and mature Vitex negundo plants. The glandular trichomes included the peltate type (Type 1), the capitate type (Type 2), and degraded capitate type (Type 3), whereas the non-glandular warty trichomes contained the multicellular (Types 4) and unicellular type (Type 5). Type 1 and 2 consisted of head and stalk cells, but their number and size were different. One secretory cavity was formed from the four head cells in the former, but only two head cells were involved in the latter. The cytoplasmic density in the head cell was quite high and in particular, sER and Golgi bodies were well developed. At initiation of their development, the cuticle layer of the head cells separated from the outer tangential wall to form a secretory cavity. Subsequently the cavity expanded acropetally and a large number of secretory vesicles continuously produced from the head cells until they filled the entire cavity. The cavity contained materials that would be soon discharged into intercellular spaces and/or into the air. The cavity began to decrease the volume by contracting at initial secretion but degrade rapidly within short time. It has been suggested that the mode of secretion in V. negundo is probably the eccrine secretion, since no break or rupture of the cavity has been observed during examination. Contrastingly Type 3 exhibited deterioration of the head cell at early stage. Type 4 was about $110{\sim}190{\mu}m$ long, consisting of $2{\sim}3$ cells, and distributed more in the adaxial epidermis compared to the abaxial surface. However, $20{\sim}30{\mu}m$ long Type 5 was extremely dense in both epidermis. Among several trichome types, Type 1 and 2 probably play an important role in discharging unique aromatic scents in plants of V. negundo.

This study is to produce multiple layers of liposomes in a supercritical state and encapsulates active ingredients in order to stably encapsulate thermodynamically unstable active ingredients. In order to form a liposome in a supercritical state, a mixed surfactant development including vegetable-derived hydrogenated phosphatidyl choline and their delivative, hydrogenated sucrose distearate was synthesized as high purity. It describes a manufacturing method of injecting liquid carbon dioxide into a reactor to create a supercritical state and stirring to produce a giant liposome, and adding and loading genistein and quercetin. The HLB of the mixed lipid complex (SC-Lipid Complex) was 12.50, and multiple layers of liposome vesicles were formed even at very low concentrations. This surfactant had a specific odor with a pale yellow flake, the specific gravity was 0.972, and the acid value was 0.12, indicating that it was synthesized with high purity. As a result of the emulsifying capacity experiment using 20 wt% capric/capric triglyceride and triethylhexanoin using SC-Lipid Complex, it was found to have 96.2% emulsifying power. SC LIPOSOME GENISTEIN was confirmed that a multi-layer liposome vesicle was formed through a transmission electron microscope (Cryo-TEM) for the supercritical liposome encapsulated with genistein. The primary liposome particle size in which genistein was encapsulated was 253.9 nm, and the secondary capsule size was 18.2 ㎛. Using genistein as the standard substance, the encapsulation efficiency of supercritical liposomes was 99.5%, and general liposomes were found to have an efficiency of 93.6%. In addition, the antioxidant activity experiment in which quercetin was sealed was confirmed by the DPPH method, and it was found that the supercritical liposome significantly maintained excellent antioxidant activity. In this study, thermodynamically unstable raw materials were sealed into liposomes without organic solvents in a supercritical state. Based on these results, it is expected that it can be applied to various forms such as highly functional skincare cosmetics, makeup cosmetics, and scalp protection cosmetics.

Perilla is an oilseed crop cultivated in Korea since ancient times. Due to the high ${\alpha}-linolenic$ acid content in perilla, perilla seed oil can easily become rancid. ${\alpha}-Linolenic$ acid is synthesized by two enzymes, endoplasmic reticulum-localized ${\Delta}15$ desaturase (FAD3) and chloroplast-localized ${\Delta}15$ desaturase (FAD7) in vivo. In order to lower the ${\alpha}-linolenic$ acid content of the seed oil without disturbing plant growth, we tried to suppress the expression of only the FAD3 gene using RNA interference, whilst maintaining the expression of the FAD7 gene. Seventeen transgenic plants with herbicide ($Basta^{TM}$) resistance were obtained by Agrobacterium-mediated transformation using hypocotyls of perilla plants. The transgenic plants were firstly confirmed by treatment with 0.3% (v/v) $Basta^{TM}$ herbicide, and the expression of FAD3 was measured by Northern blot analysis. The ${\alpha}-linolenic$ acid content was 10-20%, 30-40%, and 60% in two, seven, and three of the twelve $T_1$ transgenic perilla plants which had enough seeds to be analyzed for fatty acid composition, respectively. Analysis of the fatty acid composition of $T_2$ progeny seeds from $T_1$ plants with the lowest ${\alpha}-linolenic$ acid content showed that the homozygous lines had 6-10% ${\alpha}-linolenic$ acid content and the heterozygous lines had 20-26% ${\alpha}-linolenic$ acid content. It is expected that the reduction in ${\alpha}-linolenic$ acid content in perilla seed oil will prevent rancidity and can be utilized for the production of high-value functional ingredients such as high ${\gamma}-linolenic$ acid.

Mushroom production in Korea is concentrated on five major mushroom types. To create a new source of income for farmers, it is necessary to establish new mushroom production and marketing systems. This study was conducted to evaluate the marketability of and establish a marketing strategy for Pleurotus nebrodensis variety 'Uram'. The evaluation of distributors showed that it was necessary to cultivate mushrooms of uniform shape and size, to compensate for their low storability, and to sell them in small packages. The consumer evaluation showed that the texture of P. nebrodensis had the highest level of satisfaction for quality, but the levels of satisfaction for size and shape were low. In the consumer evaluation, as in the distributor evaluation, improvements in cultivation were found to be necessary, as the storage time is short due to a high moisture content. An evaluation of the market gave the following results regarding the marketing strategy. It was found to be necessary to consider the production of mushrooms of a uniform shape and size and sell them in small packages in the range of 150 to 300 g. The price of the mushrooms should be set using a high-end strategy for high-end sales. The mushrooms should be introduced to local food and eco-friendly stores in the early stages of production. Subsequently, if farmhouse production increases, shipments should be made to wholesale markets through a regular contract. Finally, considering that P. nebrodensis is an unfamiliar mushroom to consumers, it is necessary to promote it by increasing the accessibility of consumers through tasting events and experience groups.

This study aims to contribute to the research on resource recovery for the rapidly declining population of Liparis tanakae by observing the larval development process and the morphology of juveniles based on their growth. Natural spawning eggs collected in Yeosu were used for observing the process of egg development and larval morphology. The water temperature during the rearing process was maintained at 12.3~13.5℃ (average 12.7℃). The fertilized eggs had an egg diameter ranging from 1.57 to 1.79 mm (average 1.71 mm) and were spherical and adhesive. Within 4 hours 35 minutes after fertilization, they reached the two-cell stage, and after 74 hours 10 minutes, the formation of the yolk sac began. At 106 hours post-fertilization, a caudal fin appeared at the tail tip. Hatching began at 526 hours, and the larvae developed with the yolk sac positioned just behind the eyes. The newly hatched larvae had both the mouth and anus open. Melanophores appeared inside the lower jaw and around the tail on the third day after hatching. By the 16th day after hatching, most of the yolk was absorbed, and melanophores were visible in the head region. Finally, on the 63rd day after hatching, the head region significantly developed, and the body shape and mouth were similar to those of an adult fish, signifying the transition to the juvenile stage. This study will serve as valuable data for aquaculture techniques related to the conservation and restoration of fish species based on the hatching and juvenile morphology of Liparis tanakae.
The distribution of the secretory ducts, fine structures of the secretory epithelial cells, and the ingredients of the metaplasmic inclusions were studied at light and electron microscopical levels in seeds, stems, leaves, and roots of ginseng. The secretory ducts occurred in the hypocotyl of the embryo, in the cortex of the roots, and also both inside and outside of each vascular bundle in the stems and leaves. Especially, it is considered that the circular layers of the secretory ducts in roots may represent their ages. The epithelial cell has well developed nucleolus, mitochondria and smooth endoplasmic reticulum. Sudanophyl and osmiophilic inclusions were found in the epithelial cytoplasm and duct lumen. But these inclusions were not observed when extracted with pyridin or alcohol. In contrast to the lumen with red color, the epithelial cells were blue in color as stained with nile blue, suggesting that the former inclusions are neutral lipid while the latter are acidic lipid. The electron density of the cell inclusions was quite high as fixed with osmium tetroxide, indicating that most of these secretory materials seem to be unsaturated lipid. Therefore, since ginseng secretory ducts are closely associated with the lipid metabolism, it should be called lipid canal or lipid duct.
The object of this study is to prepare microcapsules containing a diisocyanate compound, apply them to self-healing protective coating, and evaluate the self-healing capability of the coating by atmospheric moisture. Isophorone diisocyanate (IPDI) polymerized under humid atmosphere, indicating that IPDI can be used as a healing agent. Microencapsulations of IPDI were conducted via interfacial polymerization of a polyurethane prepolymer with diol compounds. The formation of microcapsules was confirmed by Fourier-transform infrared (FTIR) spectroscopy and nuclear magnetic resonance (NMR) spectroscopy. The mean diameter, size distribution, morphology and shell wall thickness of microcapsules were investigated by optical microscopy and scanning electron microscopy (SEM). The properties of microcapsules were studied by varying agitation rates and diol structure. The self-healing coatings were prepared on test pieces of CRC board. When scratch was generated in the coatings, the core material flew out of the microcapsules and filled the scratch. The self-healing coatings were damaged and healed under atmosphere with 68~89% relative humidity for 48 h, and SEM and impermeability test for the specimens showed that the scratch could be healed by atmospheric moisture.

Embryonic and larval developments on Pacific cod, Gadus macrocephalus are described from laboratory-reared specimens. The eggs (1.075~1.168 mm in diameter) were weakly adhesive, almost spherical in shape, transparent and unpigmented, showing a pale yellow yolk without oil globules. Hatching was occurred 220 hrs after fertilization at $9.8^{\circ}C$. The newly hatched larvae (3.72~4.40 mm in total length, TL) had 43~47 (11~13+32~34) myomeres, mouth and anus which was not yet opened. The yolk was completely absorbed at 4.58 mm TL, and transformed to postlarval stage. Locations of the anus and second dorsal fin origin, and distinctive growth of the larvae head were observed at 5.48 mm TL with melanophores in the gill cover and upper region of the occipital. In addition, melanophores were distinctive in the ventral region and caudal fin base at 6.88 mm TL. All fins were formed with a complete set of fin rays having the following counts (D. 3~7-12~14-13~14; A. 13-15) at 11.54 mm TL. Both lateral line and barbel of lower jaw were observed at 15.07 mm TL.

Morphological changes in eggs and larvae of spotted knifejaw, Oplegnathus punctatus were observed by cultivation of fertilized eggs to juvenile stage. Fertilized egg was demersal and transparent. The egg has one oil glouble and its size ranged 1.01~1.15 mm ($1.08{\pm}0.10mm$; $mean{\pm}SD$, n=20). Culture chambers were maintained $22.5{\sim}24.5^{\circ}C$ in water temperature and 33.5~34.5ppt in salinity. It took 25 mins from fertilization to the blastoderm stage, 55 mins to 2 cell stage and 20 hrs 50 mins to hatching. Newly hatched yolksac larvae ranged 2.59~3.02 mm ($2.81{\pm}0.25mm$, n=5) in total length, and mouth and anus were not still open. Yolk and oil globule were absorbed 2 days after hatching (DAH). Flexion larvae in 12 DAH ranged 3.02~3.07 mm TL ($3.04{\pm}0.04mm$), and the caudal fin grew to fan-shaped, melanophores appeared on top of digestive canal.
Phosphodiesterase (PDE) plays an important role in cAMP-mediated signaling within cells. We previously showed that the long-form of Aplysia PDE4 (ApPDE4) was localized in the plasma membrane and the presynaptic terminal in Aplysia sensory neurons, and the 16 N-terminal amino acid was sufficient for this targeting process. In this study, we characterized the cellular localization of various ApPDE4 mutants. We first identified the roles of each amino acid within the group of 16 N-terminal amino acids of long-form ApPDE4. As a result, we were able to identify various mutants that were localized to both the plasma membrane and the Golgi complex, Golgi only, or both the endoplasmic reticulum (ER) and the Golgi complex. To examine the role of palmitoylation on the cellular localization of ApPDE4 mutants, 2-bromo palmitate (2-BR) was used as a treatment. As a result, in the presence of 2-BR, the plasma membrane targeting of many mutants was impaired, indicating that palmitoylation was involved in the plasma membrane targeting of the mutants. We also found that PI4P play crucial roles in the Golgi targeting of (N16,C3S/VV/G)-mRFP, L(N16,C3S/LFS/R)-mRFP, and L(N16,EPL/R)-mRFP.
