THE EFFECTS OF DRYING AGENTS AND BONDING AGENTS ON THE SHEAR BOND STRENGTH OF SEALANTS TO ENAMEL (치면건조제와 접착제의 사용에 따른 치면열구전색재의 전단결합강도에 관한 연구)

  • The application of sealants is a highly technique-sensitive procedure, requiring an extremely dry field prior to placement. Moisture contamination of the etched enamel surface before sealant placement is cited as the main reason for sealant failure. The purpose of this study was to evaluate the effects of different methods of sealant application on the shear bond strength of sealants to enamel. In groups 1, 2, 3, 4 Teethmate(unfilled sealant) was used, while Ultraseal XTplus(filled sealant) was used in groups 5, 6, 7, 8. Groups 1 and 5(control) were acid etched for 15 seconds using 35% phosphoric acid, washed and then dried. In groups 2, 6 drying agents were applied, and in groups 3, 7 bonding agents were applied and light cured. In groups 4 and 8 both drying agent and bonding agent were applied. Then sealant was cured to the specimen using molds 3mm in diameter and 2mm in height. Thermocycling was performed and shear bond strength was finally measured. The following results were obtained : 1. Groups using filled sealant(groups 5, 6, 7, 8) showed higher shear bond strengths compared to groups using unfilled sealant(groups 1, 2, 3, 4). 2. Among groups using unfilled sealant(groups 1, 2, 3, 4), groups 2, 3, 4 showed significantly higher shear bond strength compared to group 1(p<0.05). There were no significant differences among groups 2, 3 and 4. 3. There were no significant differences(p>0.05) among groups using filled sealant(groups 5, 6, 7, 8). 4. When modes of fracture were examined, cohesive failure was observed in groups 2, 3 and 4.

Dental Management in a Patient with Infantile Osteopetrosis : A Case Report with a 7-Year follow-up (유아기형 골화석증 환자의 치과적 관리 : 7년간의 경과 관찰)

  • Osteopetrosis is characterized by impaired osteoclast function and increased bone density. Infantile osteopetrosis is a severe form of the disease and has characteristics such as diffusely sclerotic skeleton, pancytopenia, cranial nerve entrapment, infection susceptibility, and abnormal craniofacial appearance. Patients with infantile osteopetrosis often experience developmental delay, and may have a short life span. A 14-month-old girl with osteopetrosis presented to the department of pediatric dentistry. Incipient caries on deciduous incisors were observed. The patient revisited 4 years of age. Besides medical problems, oral complications such as growth retardation, narrow upper arch, crowding, dental caries, and abnormal tooth development were observed. After consultation with her pediatrician, dental treatments were performed on the deciduous molars under sedation after a prophylactic antibiotic injection. At a periodic follow-up, multiple deciduous teeth were treated and extracted, and oralrehabilitation with a removable partial denture was initiated. Patient with osteopetrosis are highly susceptible to infection because of their compromised immune system and problems associated with wound healing that lead to osteomyelitis or sepsis development. Active participation in dental care for sugar intake management and proper oral hygiene are obligatory.

Pathologic Change and Prognosis after Combination Therapy in Advanced Neuroblastoma (소아 신경모세포종의 치료 후 조직소견의 변화와 예후와의 관계)

  • Purpose : Neuroblastoma is a malignant neoplasm which arises from primitive sympathetic neuroblasts, and occasionally can matured from a malignant neuroblastoma into a benign ganglioneuroma. It has the highest rate of spontaneous regression of any pediatric tumor. We performed a retrospective study of pathologic features after combination therapy in advanced neuroblastoma. Prognostic effects of the individual morphologic feature and prognostic groupings according to modified Shimada classification systems were analyzed. Methods : The treatment results for six patients with neuroblastoma seen at Keimyung University from Jan. 1991 to June 2000 were analyzed. Patients were treated with a combination of chemotherapy, radiation therapy, and surgery, and classified by two major prognostic criteria based on morphological features of neuroblastoma, such as modified Shimada classification and histologic grading. Results : Three cases were classified to a good histologic group; among them, two cases survived, but one case was lost in follow-up. There were three cases classified in a poor histologic group. All of these patients expired due to sepsis and hemorrhagic pancreatitis. Conclusion : Prognostic classification due to pathologic findings had significant value in evaluating the survival rate of neuroblastoma patients.

Chemical Preservation Methods of Urine Sediment for Quality Control (정도관리를 위한 요침사의 화학적 보존방법)

  • There is greater standardization of quality control for microscopic examination of urine than for physicochemical test. In this study, we investigated whether it is possible to control the sediment accuracy by microscopic examination through the real thing by preserving the essential sediment with glutaraldehyde, which is required for the rationality of sediment quality control. A urine specimen was prepared using 2.5% glutaraldehyde as a preservation solution. Samples treated with urine preservatives confirmed the morphological deformation of the cells for four weeks at intervals of one week and confirmed whether they should be preserved for 4 weeks thereafter. After preparing the required sediment slide, two more slides were produced; one was stored in a refrigerator for, and the other was stored at room temperature. The morphological deformation of the specimen was confirmed. Glutaraldehyde has the effect of preserving the refrigerated essential sediments and storing them for up to 8 weeks, refrigerated storage after slide production, stabilized by 3 days. Moreover, after treatment with preservatives, the production of the slide and comparison between the measured values between the laboratories and examiners showed a low consistency. In conclusion, we showed that the urine sediment components can be preserved, and it can be used for quality control and education through real objects.

The effect of Swd2's binding to Set1 on the dual functions of Swd2 in Saccharomyces cerevisiae (Saccharomyces cerevisiae의 Swd2와 Set1의 결합이 Swd2의 이중적인 기능에 미치는 영향)

  • In eukaryotic cells, histone modification is an important mechanism to regulate the chromatin structure. The methylation of the fourth lysine on histone H3 (H3K4) by Set1 complex is one of the various well-known histone modifications. Set1 complex has seven subunits including Swd2, which is known to be important for H2B ubiquitination dependent on H3K4 methylation. Swd2 was reported to regulate Set1's methyltransferase activity by binding to near RNA recognition motif (RRM) domain of Set1 and to act as a component of CPF (Cleavage and Polyadenylation Factors) complex involved in RNA 3' end processing. According to the recent reports, two functions of Swd2 work independently of each other and the lethality of Swd2 knockout strain was known to be caused by its function as a component of CPF complex. In this study, we found that Swd2 could influence the Set1's stability as well as histone methyltransferase activity through the association with RRM domain of Set1. Also, we found that ${\Delta}swd2$ mutant bearing truncated-Set1, which cannot interact with Swd2, lost its lethality and grew normally. These results suggest that the dual functions of Swd2 in H3K4 methylation and RNA 3' end processing are not independent in Saccharomyces cerevisiae.

A STUDY ON THE EFFECT OF THE ND:YAG LASER IRRADIATION ON THE MECHANICALLY EXPOSED PULP (기계적 노출치수의 Nd:YAG 레이저 조사효과에 대한 연구)

  • The purpose of this study was to investigate the effects of Nd:YAG laser on mechanically exposed pulp of dog by the observation of pulpal inflammatory change and heal process including dentinal bridge, structural changes of fibroblasts of thr remaining vital pulp tissue. In experimental group 1, the exposed pulps were irradiated with Nd:YAG laser(3W, 30Hz, 0.2sec) for $2{\sim}3$ second followed by capped with aluminium tin foil. In group 2 and group 3, the exposed pulps were irradiated with Nd:YAG laser via contact(Group 2) and non-contact(Group 3) methods followed by capped with calciumhydroxide paste. The animal were sacrificed at the intervals of 3, 7, 14 and 30days for histologic evaluation. The results were as follows : 1. The dentinal bridges were formwd more fast and broadly in the experimental group 1 and 2 than other groups, but there were no histologic differences in the degree of their formation among control group, experimental group 1. 2. Odontoblastic activities at amputated pulp was increased in the experimental group 2, 3 than other group but there no histologic difference in the odontblastic activitiy among control group, experimental group 1. 3. The infalmmation was severe at the postoperative 1 week of all groups, but its condition subsideed with time elapsed. At the postoperative 3, 7 days, its condition in experimental group 2, 3 were less severe than in the group 1. 4. There were no histologic differences between the experimental group 2 and 3 according to the degree of dentin bridge formation.

Functional expression of CalB in E.coli (대장균에서의 Candida antarctica lipase B 최적 발현)

  • Candida antarctica lipase B (CalB) is an efficient biocatalyst for many organic synthesis reactions. To make full use of CalB, we need effective expression system. Previously recombinant CalB was successfully expressed in the methylotropic yeast Pichia pastoris. In addition, we succeed in the functional expression of CalB in the Escherichia coli cytoplasm. This CalB expression system in E.coli has many considerable advantages in comparison with other expression systems and enables high-throughput screening of gene libraries as those derived from directed evolution experiments. To optimize E.coli system, we investigate comparing between OrigamiB (DE3) and BL21 (DE3) and observing effect of IPTG amount.

Comparison of Oligosaccharyltransferase Assay Methods Using a Fluorescent Peptide (형광펩타이드를 이용한 Oligosaccharyltransferase Assay 방법 연구)

  • Oligosaccharyltransferase (OTase) catalyzes the transfer of a lipid-linked oligosaccharide (LLO) to the nascent polypeptide. Most eukaryotes have an OTase composed of a multisubunit protein complex. However, the kinetoplastid Leishmania major and the bacterium Campylobacter jejuni have only a single subunit for OTase activity, Stt3p and PglB, respectively. In this study, a new in vitro assay for OTase was developed by using a fluorescent peptide containing N-glycosylation sequon, Asn-Xaa-Thr/Ser, where Xaa can be any amino acid residue except Pro. L. major Stt3p and C. jejuni PglB as a model OTase enzyme demonstrated the formation of glycopeptides from a fluorescent peptide through OTase activities. For separation and measurement of the glycopeptides produced by the OTases, Tricine-SDS-PAGE, a lectin column and fluorospectrophotometer, and HPLC were applied. Comparison of these assay methods for analyzing a fluorescent glycopeptide showed HPLC analysis is the best method for separation of glycopeptides and nonglycosylated peptides as well as for quantify the peptides than other methods.

A case of Niemann-Pick disease type A (A형 Niemann-Pick 병 1례)

  • Niemann-Pick disease is a group of autosomal recessive disorders associated with hepatosplenomegaly, variable neurologic deficits, and the storage of sphingomyelin and other lipids. Seven cases have been reported in Korea. We report an additional case presenting with hypotonia, early neurodevelopmental delay, hepatosplenomegaly and death by persistent pneumonia and asphyxia at the age of 23 months. MRI of brain and fundoscopic findings of our case at 4 months of age were normal. However, abnormal intensity of the thalamus and atrophy of the right temporal lobe on the MRI and macular cherry red spots were noticed at the age of 17 months. A bone marrow biopsy showed large foamy cells, while hexosaminidase A and B levels were normal. Although biochemical or molecular workup was not done, these findings led to the diagnosis of infantile onset Niemann-Pick disease, probably type A. A brief review of the related literatures was made.

Anti-oxidative properties of ginseng (인삼의 항산화 작용)

  • Clinical and animal studies have shown that free radical overload is an important cause for a variety of diseases. Although ginseng has been recognized as antioxidant, how it modulates anti-oxidative process at the molecular level remains unknown. Free radical production is induced by tumor necrosis factor-$\alpha$ (TNF-$\alpha$) under the stress condition, and (TNF-$\alpha$) release is activated by TNF-$\alpha$-converting enzyme (TACE). Since TACE inhibitor is also well known for anti-inflammatory agent, ginseng seems to show anti-oxidative activity by repressing TACE pathway. Further studies on signal transduction would be warranted to elucidate molecular action mechanisms of ginseng on anti-oxidation and anti-inflammation.
