Aspiration Cytology of Insular Carcinoma of Thyroid - A Case Report - (갑상선 미분화 "도암종"의 세침흡인 세포학적 소견 - 1례 보고 -)

  • Fine needle aspiration cytologic features of a case of insular carcinoma of the thyroid in a 23-year-old woman who presented a palpable neck mass is described. The aspirate showed cellular smear arranged in trabeculae, solid or loose clusters, and microfillicles in necrotic background. The tumor cells had uniform, small round, hyperchromatic nuclei. The chromatin was finely granular, and nuclear membrane was smooth. Nucleoli were not discernible. Nuclear pleomorphism was minimal. The cytoplasm was usually scanty, pale, poorly outlined, and almostly amphophilic. Sometimes paranuclear cytoplasmic vacuoles were noted. final diagnosis was confirmed by total thyroidectomy as insular carcinoma.

Recognition fo Cervical Cancer Cells Using Wavlelet Coefficient (Wavelet 계수를 이용한 자궁 경부 세포진 인식)

  • 본 논문에서는 세포 영상에 대해 Wavelet 계수을 이용한 인식 방법을 제안하고 있다. 자궁 경부세포진은 핵과 세포질을 분할하기 힘들기 때문에 영역분할을 통해 얻은 핵특징이 잘못 계산될 수 있어 인식율이 떨어진다. 따라서 핵의 세포만을 나타낼 수 있도록 핵의 정보를 포함하고 있는 고대역 부밴드에서는 20$\times$20 영상을 사용하였고, 세포질에 정보를 포함하고 있는 저대역 부밴드에서는 50$\times$50의 영상을 사용하였다. 영상 인식을 위한 특징 추출은 2단계 Wavelet 변환후 생성된 변환 영역에 대해서 Wavelet 계수 평균값 표준편차와 Energy를 사용하였다. 실험 결과 Wavelet 계수를 이용한 방법이 영역분할을 이용한 방법과 비교하여 더 높은 인식율 보였다.

  • A transmission electron microscopic study was performed to observe the ultrastructures of the male germinal cells and spermatozoa of Fibricola seoulensis. Spermatogonia were found in the periphery of the testis and characterized by large nuclei and comparatively little cytoplasms. Spermatocytes contained an oval to spherical nucleus. Their nuclear volume was little larger in comparative to that of cytoplasm, and the chromatin was comparatively little. The early spermatids were characterized by a great amount of cytoplasm, and numerous mitochondria encircled the nucleus. In a more advanced spermatids the electron-dense strands of chromatin appeared in the nucleus, and a pair of rootlet of the axoneme and a microtubule-organizing center (MTOC) were observed near the nucleus. The sectioned spermatozoa were found in the testis and the seminal vesicle. Their cross sectional views were divided into 6 types when they were distinguished on the basis of the morphology and components. The spermatozoa of F. seoulensis showed two flagella of 9+1 type axoneme.

  • The ultrastructures of spermatogenesis and sperm in Xiphophorus maculatus, ovoviviparous fish were investigated by electronmicroscopy The testis of Xiphophorus maculatus contained numerous testicular sacs, and spermatogenesis was synchronized in these testicular sac. In the case of spermatogonium, the nucleus was comparatively large ellipsoidal, and the nucleolus and mitochondria showed a marked development. The size of primary spermatocyte was smaller than that of spermatogonia, and that of secondary spermatocyte was smaller than that of primary spermatocyte. The chromatin of spermatocyte was highly condensed according to their development. The nucleus with electron-dense was round shape. In spermiogenesis, flagella started to be formed and chromatin was more condensed. The mitochondria were rearranged along the tail. The sperm was formed by loss of cytoplasm. The head of mature sperm was long cone shape and had not acrosome. The microtubules of flagella were arranged 9+2 structure. Also, the sperm has a loop-like structure at the end of a tail.

Effects of Donor Somatic Cell Conditions on In Vitro Development of Nuclear Transplanted Porcine Embryos (돼지 공여세포의 조건이 핵이식 수정란의 체외발달에 미치는 영향)

  • This study was conducted to examine in vitro developmental ability of porcine embryos after somatic cell nuclear transfer. The porcine ear fell was cultured in vitro for confluency in serum-starvation condition(TCM-199 + 0.5% FBS) far 3~6 days of cell confluency. The zona pellucida of IVM oocytes were partially drilled using laser system. Single somatic cell was individually transferred into enucleated oocytes. And the reconstructed embryos were electrically fused(single DC 1.9kv/cm, 30$\mu$ sec) with 0.3M mannitol. After electrofusion, embryos were activated(single AC 5v/mm, 5sec) and cultured in HCSU-23 medium containing 10% FBS at 39$^{\circ}C$, 5% $CO_2$ in air for 6 to 8 days. The fusion rate of donor cells was 45.6, 36.8 and 46.1% in 3~4, 5~6 days of serum starvation and non serum starvation(N-S), and were 52.7. 53.0 and 51.7% in 1~2. 5~6 and 13~14 passages of donor cell culture, respectively. No significant difference was found in the fusion rate of donor cells by the duration of serum starvation treatment or the number of donor cell passages. By the size of donor cells, however, the fusion rate was significantly higher(P<0.05) for reconstructed embryos derived from 25r $\mu$m $\geq$ site of donor cells (65.3%) than that of 25~30$\mu$ m(42.5%) or 30$\mu$ m(45.5%)$\leq$ cells. The cleavage rate was significantly (P<0.05) higher in 3~4 darts of serum starvation treatment(67.1%) than that in N-S (50.7%) or 5~6 days of starvation(57.1%). The activation rate by the size of donor cells in fused oocytes was 56.5, 68.8 and 58.5%, respectively, and was not significant.

A Study on Nucleus Extraction of Uterine Cervical Pap-Smears (자궁 경부진 핵 추출에 관한 연구)

  • If detected early enough, cervical cantor may have a good survival rate due to its preneoplastic state. However, the process is so time consuming that a medical expert can handle only a small amount of such examinations. In this paper, we propose a new nucleus extraction algorithm for uterine cervical pap smears in order to mitigate such burdens of medical experts. In the preneoplastic state cytodiagnosis images, it is important to differentiate three main areas - background, cytoplasm and nucleus. Thus, we apply lighting compensation and $3{\times}3$ mask of B channel in order to restore damaged image and remove noises respectively. The cell object is obtained from those clean binarized images with Grossfire algorithm. When there are clusters of cells, the target nucleus can be obtained with repetitive binarization of R channel brightness. In our experiment of using uterine cervical pap smears of 400 magnifications that is common in the diagnostic cytology, our method is able to extract 40 nucleus out of 45 population successfully.

  • 본 논문은 자궁경부암 검사를 위한 전처리 과정인 자궁경부암 세포 영상분할 문제 연구의 결과이다. 자궁경부암 세포 영상은 배경과 세포질 및 세포핵의 구별이 어렵다. 게다가 자궁경부암 검사 시스템은 짧은 시간동안 많은 영상을 처리해야 하기 때문에, 영상의 분석 속도가 빠르고 강력한 영상 분할 기법이 필요하다. 이를 위하여 우리는 thresholding 기법을 연구하였다. 먼저 세포 영상의 각 화소의 명암의 분포를 조사하여 히스토그램을 구하였다. 히스토그램은 0~255 사이에 존재하게 되는데, 0~255의 전 영역에 존재하기 보다는 그 중 일부분에만 존재한다. 우리는 히스토그램이 존재하는 영역을 백분율로 나누고 세포핵 및 세포질이 존재하는 영역의 분포를 구하여 global threshold를 찾았고, 이를 기준으로 각 점을 thresholding 할 때에 주위의 평균값을 보정값으로 두어 local thresholding을 수행하였다. 결과 영상은 핵의 영역을 탐색하기 위한 seed로 사용하기에 적합하다.

  • The present study was carried out light and electron microscopic analysis of the structures of the tritocerebral neurons in butterfly (Pieris rapae Linne) from Insecta. We have summarized our morphological analyses as follows: In insecta, tritocerebrum of butterfly shows tentatively classified 6 neuronal types without any arrangement of cell layers. Type I cells with long oval or spindle shape are the largest out of 6 kinds. The nucleus contains a small amount of chromatin. In the large volume of cytoplasm there contained rich cell organelles except granular endoplasmic reticulum. Type II cells are smaller than type I neurons, but their ultrastructural features are similar to type I neuron. They have well-developed granular endoplasmic reticulum. Type III cells are spherical or triangular and smaller than type I neuron with spherical nuclei. Cell organelles are mostly prominent, esp., mitochondria, ribosomes and fine fibrils which are arranged in periphery of the cytoplasm.

  • Single nuclei from two-, four- and eight-cell mouse embryos were transplanted into enucleated two-cell embryos by micromanipulation and Sendai virus mediated fusion. The developmental potential of these reconstituted embryos in vitro and in vivo was examined. It was found that the single nuclei which were transplanted to enucleated two-cell embryos were not only able to develop to the blastocyst stage in vitro(two-cell nuclei, 76.5%; four-cell nuclei, 68.4%; eight-cell nuclei, 48.3%), but also able to develop to full term in vivo after transfer to recipient mice(two-cell nuclei, 37.1%; four-cell nuclei, 29.6%; eight-cell nuclei, 16.3%). Although the proportion of live young produced after transfer of nucler of nuclear transplant embryos which received eight-cell nuclei was significantly (p<0.05) reduced, it would be suggested that the overall efficiency in producing identical offspring is greater when eight-cell embryos were selected for nuclear donor than two- or four-cell embryos were selected.
