Growth Characteristics of Nitrite Oxidizing Bacteria Isolated from Anaerobic Digestion Liquor (혐기소화액에서 분리한 아질산 산화세균의 생장특성)

  • Two nitrite oxidizing bacteria, NOB1 and NOB2, were isolated from anaerobic digester liquer of food wastewater and analyzed for their growth characteristics and the ability to oxidize nitrite under different temperature, pH, and DO( dissolved oxygen) concentrations. Both of the isolated strains have shown the best growth at pH 7.0 and at $35^{\circ}C$, and also shown higher growth rate with the increasing dissolved oxygen concentrations. As the factors to restrict the growth of these strains, parameters such as pH and DO were found to be effective ones, by increasing (up to 9.0) or decreasing pH (up to 5.0), or lowing DO below 1.0 ppm. Especially, the ability to oxidize nitrite in both strains was about 50% lower in below 1.0 ppm of DO than above of 1.0 ppm. NOB2 was found to be two times greater in both the growth rate and the nitrite-oxidizing ability than NOB1.

Induced of Systemic Resistance against Gray Leaf Spot in Pepper by Enterobacter Species Isolated from Family Gramineae Plants in Dok-do (독도의 벼과식물로부터 분리된 Enterobacter spp.에 의한 고추의 흰별무늬병에 대한 전신유도저항성)

  • This study's aim is to isolate and characterize plant growth promoting Enterobacter species for the biological control of gray leaf spot in pepper. Screening was carried out from the rhizosphere of Agropyron tsukushiensi var. transiens (Hack.) Ohwi in Dok-do. Rhizobacterial isolates were partially identified by 16S rDNA sequencing and Enterobacter species were tested for plant growth promoting capabilities and the induction of systemic resistance in pepper against gray leaf spot caused by Stemphylium solani. Isolates were tested for production of indole-acetic acid and siderophore, and for phosphate solubilization. The application of isolates was effective in controlling gray leaf spot in pepper with E. asburiae (KNUC5007) and E. cancerogenes (KNUC5008 and KNUC5010) having the highest efficacy in reducing gray leaf spot severity. This is the first report of the biological control of gray leaf spot in pepper using rhizobacteria and it is hoped that this study will increase the utilization of Enterobacter species as plant growth promoters and biocontrol agents.

A Comparison of Radical Scavenging Activity and Cyanobacteria Growth Inhibition of Aquatic Vascular Plants (수생관속식물의 라디칼 소거능과 남세균 생장에 대한 억제활성 비교)

  • Methanol extracts of aquatic plants were analyzed for allelopathic activities against Escherichia coli JM109 and $Microcystis$ $aeruginosa$ UTEX2385 which were compared to its 2,2-Diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activities. The radical scavenging activities were detected from the extracts of $Persicaria$ $thunbergii$, $Persicaria$ $amphibia$, $Trapa$ $japonica$, $Myriophyllum$ $spicatum$, and $Brasenia$ $schreberi$. Also, the inhibitory activities against cyanobacteria were analyzed according to the order of $B.$ $schreberi$, $T.$ $japonica$, $P.$ $amphibia$, and $M.$ $spicatum$. Most of the extracts from aquatic plants did not show any inhibition against $E.$ $coli$ except $B.$ $schreberi$. We found a positive correlation between the antioxidental activities of methanol extract of aquatic plants and the growth inhibitory activities for cyanobacteria in terms of the DPPH radical scavenging activities ($R^2$=0.381, $P$ <0.0001). The inhibitory activities of methanol extract against $E.$ $coli$ growth was not correlated with the other activities of aquatic plants ($P$ >0.04). We suggest from this study that the allelopathic effects of aquatic plants against cyanobacteria could be screened by using the bioassay based on DPPH.

Selection and Mechanisms of Indigenous Antagonistic Microorganisms against Sheath Rot and Dry Rot Disease of Garlic (마늘 잎집썩음병과 마른썩음병을 길항하는 토착길항미생물의 선발 및 기작)

  • Sheath rot and dry rot disease caused by Pseudomonas marginalis and Fusarium oxysporum were serious problems in garlic farmland. In this study, total of 160 indigenous antagonistic bacteria were isolated from 16 farmlands in Yeongcheon, Korea. Among these, 15 strains were able to inhibited P. marginalis and F. oxysporum. The 16s rDNA genes of the selected 15 strains were amplified and sequenced. The strains has strong antagonistic ability against garlic pathogens was achieved Bacillus subtilis YC82, B. vallismortis YC84, B. amyloliquefaciens YC240. The selected 3 strains tested for investigation of antifungal mechanisms further analyses; 3 strains of these validated for production of siderophore, ${\beta}$-glucanase and chitinase using CAS (chrome azurol S) blue agar, CMC-congo red agar and DNS method. The 3 strains were able to utilized insoluble phosphate as dertermined by vanado-molybdate method. The 3 strains verified for production of auxin and gibberellic acid using Salkowski test and holdbrook test. Also, 3 strains showed stimulation germination, stem growth promoting activity on the in vivo test. The 3 strains were able to effectively suppress P. marginalis and F. oxysporum causing sheath rot and dry rot diseases on the in vivo pot test.

Bacteriological Control of Cyanobacterial Bloom. (시안세균 수화의 세균학적 조절)

  • A Gram (-), rod-shaped bacterium in size of 1.3∼$1.8{\times}0.35{\mu}m$ inhibiting the growth of cyanobacterium (Ana-baena cylindrica) was isolated and designated NG-2 in this manuscript. This isolate showed positive reactions for catalase and oxidase, and optimal growth conditions of 35∼TEX>$40<^{\circ}C$ and pH 9.0. In a mixed-culture of A. cylindrica and the isolate, each microorganism grew inverse-proportionally, and the cyanobacterial vegetative cells almost completely disappeared within 24 hours. NG-2 lysed A. cylindrica only under light, which means that lytic activity of NG-2 was dependent on the photosynthetic activity of host. When observed under phase contrast microscope, the isolate lysed vegetative cells of A. cylindrica in scattered state in a liquid medium, whereas het-erocysts have not been lysed. When cyanobacterial cell walls have been lysed partly, NG-2 attatched around A. cylindrica filament and formed colony, then encouraged complete lysis of cyanobacterial cells. The isolate showed similar lytic activity in natural water as in an artificial medium. And lytic activity of NG-2 was enhanced when attached on expandable polystyrene bead.

Effect of Plant (Salvia sp.) Growth Using Mixed Microorganisms (혼합 미생물이 식물(Salvia)의 생장에 미치는 영향)

  • Effect of effective microorganisms on the growth of plant (salvia sp.) was investigated. Microorganisms used were photosynthetic bacteria, lactic acid bacteria and yeasts. When photosynthetic bacteria were inoculated to soil by 100 dilution, treated plants showed 160% growth by length compared to control. When photosynthetic bacteria, lactic acid bacteria and yeasts were mixed, diluted by 10 and inoculated to soil, the plants showed 212% growth compared to control. Microbial populations were increased in the treated soil.

  • The effluent sediment from the land-based seawater fish farms of Jeju consists of proteins, fats, ash and moisture. An evaluation of the effluent sediment as substrate for growth of phototrophic or lactic acid bacteria revealed that the sediment supported the growth of phototrophic bacteria but could support lactic acid bacteria only on supplementation with sugar. The possibility of using phototrophic bacteria for recycling the land-based seawater fish farm effluent is shown.

Growth kinetics and chlorine resistance of heterotrophic bacteria isolated from young biofilms formed on a model drinking water distribution system (모델 상수관망에 형성된 초기 생물막에서 분리한 종속영양세균의 생장 동역학 및 염소 내성)

  • The present work quantified the growth of young biofilm in a model distribution system that was fed with chlorinated drinking water at a hydraulic retention time of 2 h. Bacterial biofilms grew on the surface of polyvinyl chloride (PVC) slides at a specific growth rate of $0.14{\pm}0.09day^{-1}$ for total bacteria and $0.16{\pm}0.08day^{-1}$ for heterotrophic bacteria, reaching $3.1{\times}10^4cells/cm^2$ and $6.6{\times}10^3CFU/cm^2$ after 10 days, respectively. The specific growth rates of biofilm-forming bacteria were found to be much higher than those of bulk-phase bacteria, suggesting that biofilm bacteria account for a major part of the bacterial production in this model system. Biofilm isolates exhibited characteristic kinetic properties, as determined by ${\mu}_{max}$ and $K_S$ values using the Monod model, in a defined growth medium containing various amounts of acetate. The lowest ${\mu}_{max}$ value was observed in bacterial species belonging to the genus Methylobacterium, and their slow growth seemed to confer high resistance to chlorine treatment (0.5 mg/L for 10 min). $K_S$ values (inversely related to substrate affinity) of Sphingomonas were two orders of magnitude lower for acetate carbon than those of other isolates. The Sphingomonas isolates may have obligate-oligotrophic characteristics, since the lower $K_S$ values allow them to thrive under nutrient-deficient conditions. These results provide a better understanding and control of multi-species bacterial biofilms that develop within days in a drinking water distribution system.

Seasonal Variations of Environmental Factors and Distribution of Anabaena cylindrica Growth-Inhibiting Bacteria in the Lower Daechung Reservoir (대청호 하류에서 환경요인과 Anabaena cylindrica 생장억제세균의 계절별 분포 변화)

  • The authors surveyed the seasonal variations of environmental factors, the distributions of heterotrophic bacteria and Anabaena cylindrica growth-inhibiting bacteria at each water layer in Daechung Reservoir to verify the role of bacteria during the extinction of bloom. Average water depth at site 1, 2, and 3 were 25.5 m, 15.0 m and 12.3 m, respectively. Water temperature showed a typical pattern seasonally. The variation of DO was relatively inverse proportional to that of water temperature, although it was irregular during summer time. DO decreased gradually to early May, fluctuated sharply after then, and followed by gradual increasement after middle of September. This variation pattern was notable at surface layer. There was remarkable difference in DO concentraion between surface layer and the other water layers during the period in which DO irregulary varied. The variation range of chlorophyll-a concentraion at surface layer in summer time was broad, and it was relatively high when DO was high. The population size of heterotrophic bacteria was high from Spring to Autumn, an declined after September when the water temperature droped rapidly. Especially this variation pattern was prominent at the surface layer. Bacteria that inhibit the growth of A. cylindrica was almost not detected by June, and its distribution increased in July. Afterward, it showed different variation pattern between each site. The distribution of A. cylindrica growth-inhibiting bacteria was higher at the middle and bottom layer than the surface layer in July and October, when it was larger at all sites for the study period. This result suggests that the antagonistic bacteria exhibit higher activity when host activity drops. These results also suggest that natural water bacteria control the distirbution of cyanobacteria, especially its activity as controller is remarkable when cyanobacterial growth declines.

Bactericidal Effects of Nano-silver Liquid Against Various Plant Pathogenic Bacteria (은 나노 용액의 식물병원성 세균에 대한 살균활성)

  • We have conducted in vitro experiments with nano-silver liquid for their effect against various plant pathogenic bacteria. Different types of nano-silver liquid WA-CV-WA13B, WA-AT-WB13R and WA-PR-WB13R were used. These are classified based on different manufacturing processes. The tested bacteria were provided by KACC. We experimented ten bacterial isolates in Clavibacter, Erwinia, Pseudomonas, Ralstonia, and Xanthomonas genera. In order to determine the level of concentrations of control effects, different concentrations (10, 25, 50, and 100 ppm) of each different nano-silver liquid were added in the culture media. As a result, WA-CV-WA13B showed high inhibition effect against C-1 at 10 ppm, and showed minor inhibition effects against P-6, X-1, and X-2. WA-AT-WB13R showed bactericidal effect against P-6 at 10 ppm. At 10 ppm, WA-AT-WB13R showed relatively high inhibition effects against C-1, X-1, and X-2. WA-PR-WB13R showed bactericidal effects against P-5, P-6 and X-2 at 10 ppm or higher concentrations. All the tested three nano-silver liquid showed bactericidal effects against all the tested plant pathogenic bacteria at concentrations of 25 ppm or higher. These results indicated the possible use of nano-silver liquid for the control of plant pathogenic bacteria.
