Application of Neutral Red Staining Method to Distinguishing Live and Dead Marine Plankton for the Investigation of Efficacy of Ship's Ballast Water Treatment System (선박평형수 처리 시스템 효율 검증을 위한 해양 플랑크톤 생사판별시 Neutral red 염색법 적용 가능성 연구)

  • In order to prevent the spread of non-indigenous aquatic species through the ballast water in commercial ships, International Maritime Organization (IMO) adopted in 2004 the International Convention for Control and Management of Ship's Ballast Water and Sediments. The Convention mandates treatment of ballast water for most transoceanic voyages and its confirmation of treatment is made with plankton live/dead assay. Fluorescein diacetate assay (FDA), which produces bright green light for live phytoplankton, has been a de facto standard method to determine the survival of marine plankton, but its staining efficacy has been in dispute. In the present study, we examined the limitation of FDA, and compared its efficacy with Neutral red (NR) staining, another promising assay and widely used especially for zooplankton mortality. For all phytoplankton species studied in the present study, except Ditylum brightwellii, the staining efficiency was <50% with FDA. The green FDA fluorescence interfered with phytoplankton autofluorescence in most samples. In contrast, NR assay stained over 90% of both phytoplankton and zooplankton species tested in this study. FDA assay also showed that green FDA fluorescence rapidly faded when phytoplankton cells were exposed to microscope light. Both FDA and NR assay were negative on formalin-killed individuals of both phytoplankton and zooplankton species. Our results suggest that NR assay is more effective for determining the survival of marine plankton and can be applied to test the efficacy of ballast water treatment.

Characteristics of environmental condition and planktonic organisms in ship's ballast water originating from international ports of Japan (우리나라 주요 국제항에 입항하는 일본 기원 선박의 평형수내 환경 및 부유생물 특성)

  • This study investigated the environmental conditions and planktonic organisms in the ballast waters (BW) of 22 vessels originating from the international ports of Japan for the purpose of negotiating exemptions from the Ballast Water Management Convention (BWM Convention). The shortest duration of the BW was $3.33{\pm}1.87days$ in area "A", which included Kyushu and Suo Nada at Seto Inland. The total suspended solids, dissolved organic carbon, and particulate organic carbon ranged from 4.60 to 60.9 mg L-1, from 0.97 to 2.69 mg L-1, and from 0.24 to 4.51 mg L-1, respectively. A low average concentration of nutrients was measured in the BW from area "A", but that in the BW from area "C" (around central Honshu) was high, which may be related to the ballasting periods. High chlorophyll-a concentrations (>$1{\mu}g\;L-1$) were measured in four vessels, three of which carried the BW in area "A". High abundances of phytoplankton (> 50,000 cells L-1) were measured in four vessels, three of which carried the BW in area "A". The two vessels originating from Tokyuyama Bay in area "A" showed high densities of dinoflagellates, which are known to be harmful algae. Our results suggest that the negotiations for an exemption from the BWM Convention for Japan should proceed with caution.

  • Potassium iodide (KI) TRO sensor for measuring total residual oxidant which is an index for disinfection and neutralization in ballast water has been constructed. The results of UV absorption wavelength and absorbance test in various TRO concentrations show high linear correlation ($r^2=0.9825$) at 350 nm wavelengths and it can be possible to visualize numerical value of TRO concentration (0.1~7.0 mg/L). The result of comparison test according to TRO concentration between the KI based TRO sensor system and the DPD based TRO sensor system showed identical trend. Overall results indicated that the KI TRO sensing system could be applied for continuous TRO concentration measurement in ballast water treatment system.

A Study of Characteristics on Water Quality and Phytoplankton in Ship's Ballast Water Originating from International Ports of China (우리나라 주요 국제항에 입항하는 중국 기원 선박의 평형수내 수질 및 식물플랑크톤 특성 연구)

  • The water quality and phytoplankton presence in the ballast water (BW) of 37 vessels originating from the international ports of China were investigated to facilitate negotiations for exemptions to the Ballast Water Management Convention (BWM Convention). The shortest duration given BW spent in a vessel was $3.91{\pm}4.61days$ in area "A", which included the Bohai Sea. Total suspended solids, dissolved organic carbon, and particulate organic carbon ranged from 1.80 to $266mg\;L^{-1}$, from 1.09 to $5.79mg\;L^{-1}$, and from 0.17 to $3.65mg\;L^{-1}$, respectively. A low average concentration of nutrients was measured in BW from area "C", but the concentration of nutrients in BW from area "B" (around the Changjiang estuary) was high, which may be related to the relevant supply of freshwater. A high chlorophyll-a concentration (> $1{\mu}g\;L^{-1}$) was measured in six vessels, three of which carried BW in the area "A". High abundance of phytoplankton (> $10,000cells\;L^{-1}$) was measured in four vessels, two of which carried BW in the area "C". Vessel No. 37, originating from Hong Kong Bay in area "C", showed a high density of dinoflagellates. The results suggest that BWM Convention exemption negotiations with China should be performed cautiously.

A Comparison Study on the Sterilization Effect of Ballast Water with Cylinder Type and Groove Type (선박평형수 처리장치의 cylinder type과 groove type의 살균효과 비교 연구)

  • Current ballast water treatment technologies are applying chemical or electrical treatment technology which are not free from secondary environmental pollution. The purpose of this study is to treat the ballast water by shear stress without an additional environmental pollution and to find out the optimal treatment apparatus. We tried to treat ballast water by applying shear stress with two different type of combination of inner and outer cylinder, such as non-pattern type and groove type. In the case of non-pattern type of inner and outer cylinder, sterilization effect was comparatively low because of a slip between inner and outer cylinder. But in the case of groove type of inner and outer cylinder, sterilization effect was superior to the non-pattern type. With a same revolutional speed of 8000rpm, an extinction effect was acquired in the gap of 1 mm of inner and outer cylinder at non-pattern type, but 3mm of that of groove type.

  • 수중방전은 다양한 라디칼을 직접 물 속에서 발생시키기 때문에 수처리 공정에 다양한 응용이 가능하다. 특히, 최근에 선박평형수 등의 살균이 국제적인 이슈가 되고 있고, 2017년까지는 모든 선박에 살균을 위한 수처리 설비가 의무화된다. 본 연구에서는 염분이 있는 수체에서의 방전공정을 연구하고 이를 수처리공정에 적용할 수 있는 방법에 대해 연구하였다. 해수의 경우 전도도가 53mS로 자유로운 전하의 이동이 가능하기 때문에 일반적인 민물방전의 전원과 전극 등으로는 방전을 할 수 없다. 이에 세라믹과 금속의 이중구조로 되어 있는 모세관전극을 개발하여 전도성이 있는 수체에서의 방전을 이루었다. 전원장치로는 60 Hz, 380 V를 1차측에 인가하여 2차측에서 약 3 kV, 10 kW의 파워가 발생하는 12위상차 교류전원장치를 개발하여 사용하였다. 모세관 내부에 전압이 인가되면 전류가 발생하여 joule heating에 의하여 모세관 내부에 기포가 형성된다. 이 때, 전류의 단락이 이루어지면서 고전압쪽에 전하가 축적되며 기포내부의 E-field가 상승한다. 이후 기포 내에서 방전이 개시되며 각종 라디칼을 생성한다. 방전에 의해 생성되는 산화제로는 오존, OH라디칼, 과산화수소 등이 있으며, 해수에서는 Cl-의 결합에 의하여 Cl2 가스가 발생한다. 약 30,000 J/L의 체적에너지에 대하여 생성되는 총염소의 농도는 2.5 mg/L이다. 수중방전의 적용대상으로 선박평형수, 멤브레인과의 결합, 용존기포부상법을 선정하여 적용가능성을 연구하였다. 먼저 선박평형수 살균처리를 위해 해수의 처리유량을 20 lpm으로 유지하고 대장균, 바실러스, 조류(테트라셀미스) 등을 투입하여 전극 12개가 삽입된 12위상차 플라즈마 반응기를 통과시켰더니, 약 30,000 J/L의 체적에너지에 대하여 1일 후의 살균력이 각각 99.99, 99.99, 99.9%의 살균력을 나타내었다. 이는 국제해사기구에서 권장하는 살균수준인 99.9%를 초과하는 수치이다. 플라즈마를 이용한 해수살균공정의 안정적 운전을 위해 후단에 UF멤브레인을 추가하여 잔류생존 미생물을 제거할 수 있다. 이를 위해 플라즈마가 후단의 멤브레인 운전에 미치는 영향을 평가하였다. 카올린과 탄산칼슘을 오염원으로 각각 투입하여 멤브레인으로 처리를 하였을 때, 방전 직후 멤브레인에 걸리는 막간압력차가 약 30% 감소하였는데, 이는 막에 형성된 파울링이 방전에 의해 제거된 것으로 평가할 수 있다. 수중방전은 다양한 산화제를 생성함과 동시에 미세기포를 발생시키는데 이는 수중유기물의 부상분리에 적용될 수 있다. 방전모세관전극의 내부직경을 1 mm로 유지하고, 60 Hz, 교류전원으로 방전한 결과 평균입경 44 um의 기포를 발생시켰고, 이는 일반적으로 용존공기부상법에 사용되는 기포의 크기와 일치한다.
