We have investigated the optimal conditions for the growth and ginsenoside production in adventitious root of “Chunpoong” (Panax ginseng C.A. Meyer). Ginseng adventitous roots induced from the embryo of “Chunpoong” were cultured in various plant media supplemented with several growth hormones. The best growth rate and high ginsenoside contents were obtained in B5 medium supplemented with IBA (2 mg/L) and kinetin (0.1 mg/L). The supplement of 2.5 mM KH$_2$PO$_4$ was good for high growth rate of the adventitious roots, but the accumulation of ginsenosides was increased by reducing the KH$_2$PO$_4$ concentration to 1.25 mM. We have established the effective liquid culture system for the optimal growth and ginsenoside production of “Chunpoong” adventitious roots.
This study was carried out to establish a mass production of normal somatic embryos of Oenanthe iavanica (BL.) DC. including examination of nitrogen and sucrose sources, and ABA concentration. Embryogenic cell clumps and embryos were formed on the MS medium devoid of growth regulators. Proliferation of embryogenic cells and clumps was enhanced by 2, 4-D. Meanwhile embryo growth and development occurred on the media containing NAA and IBA. Growth of embryos was generally good in the media containing both 20 mM $KNO_3$ and 20 mM $NH_4NO_3$. The rate of shoot forming embryos was higher on the media containing on1y 20mM $NH_4NO_3$ than on the former. Addition of sucrose at 3-6% enhanced the embryo development, and normal embryos with short hypocotyl was observed on the medium containing $10\mu\textrm{M}$ ABA. Embryogenic cell clumps or globular embryos, when transferred to MS solid media devoid of growth regulators, developed into mature embryos and then into plantlets which had entire primary leaves like zygotic seedlings.
To study the effects of temperature, day length, and various plant growth regulator treatments on the tuber formation of Irish cobbler, this experiment was carried out with 2 combinations of day-length and temperature and 11 kinds of growth regulator, including GA, and their combinations. For the tuber formation, low temperature-short day condition played decisive role, and exceeded the effects of growth regulators. 4 times foliar application of 10 ppm GA resulted marked elongation of stolon but did not inhibit the tuber formation even under high temperature longday condition.
The leaf and petiole segments of Farfugium japonica were cultured to investigate the influence of growth regulators on their callus induction and plant regeneration. The callus induction and growth showed a good response both leaf and petiole on MS media supplemented with 1∼2 mg/L 2,4-D and 1∼2 mg/L BA. Callus induction and growth were more effective in petiole segments than leaf one. The highest percentage of plant regeneration was obtained from 60-day-old calli on MS medium supplemented with 1 mg/L NAA and 2 mg/L BA. When subcultured to the same medium for about 60 days, multiple shoots were developed from regenerating callus. The shoots produced roots after transferring to rooting medium containing 0.5 mg/L IAA. The plantlets over 50 mm in height were successfully acclimatized in vermiculite, and the survival rate was over 95%.
This experiment was carried out in order to determine proper plant growth regulators (PGR) and their concentrations for direct shoot induction and somatic embryogenesis from leaf tissue cultures of Muscari armeniacum 'Early Giant'. Direct shoot formation from the leaf explant culture was effective only on a half-strength MS medium containing $0.1mg{\cdot}L^{-1}$ 2,4-D, while embryogenesis was occurred on a half-strength MS medium containing $0.1{\sim}1.0mg{\cdot}L^{-1}$ IPA or without PGR. The regenerated bulblets derived from embryos or shoots were harvested and transplanted into a greenhouse. The sprouting percentage of bulblets obtained from different culture media ranged from 80 to 100% and growth of quality bulblets was enhanced when the bulblets were harvested from the medium containing $0.1mg{\cdot}L^{-1}$ NAA and $1.0{\sim}3.0mg{\cdot}L^{-1}$ IPA.
The experiment was implemented to introduce the drainage electrode irrigation system as early as possible after transplanting in order to save the nutrient solution in a convenient way. Drainage electrode irrigation method was introduced 15, 19 or 22 days after transplanting after irrigation was firstly controlled by time clock. Time clock method was also treated as a control plot. Drainage electrode method could be adopted from 15 days after transplanting, 15 days earlier than the present introducing time. The growth and yield was better in treatments with drainage electrode method. Water and fertilizer use efficiency were the highest in the treatment of 15 days, the lowest in time clock treatment.
This study was carried out to investigate the effect of plant growth retardants (daminozide, chlormequat, uniconazole, paclobutrazole) and fungicides of triazole chemical (hexaconazole, microbutanil, difenoconazole, terbuconazole, bitertanol) on the growh of 'New Guinea' impatiens. Plant height and canopy were decreased by all kinds of plant growth retardants tested in both 'Anguilla' and 'Papete'. Especially, uniconazole and paclobutrazole were most effective in inhibition of top growth. However, the content of chlorophyll increased at all treatments of plant growth retardants and stem diameter tended to be increased at the highest concentration of all kinds of plant growth retardants tested. The results in all cultivars tested, with the experiment of triazole fungicides, were similar to the results of experiment with plant growth retardants. These results suggest that fungicides of triazole chemical such as hexaconazole, microbutanil, difenoconazole, terbuconazole and bitertanol can be used for the promotion of quality in potted 'New Guinea' impatiens.
This study was carried out to establish the in vitro culture system of potato germplasms for minimizing the occurrence of variation and maximizing the culture period. We used osmoticum such as sorbitol or mannitol with sucrose in the absence of plant growth regulators. The growth of potato germplasms in the medium containing osmoticum was increased when the growth temperature was lowered. After six months storage in low temperature, plant heights of tetraploid was somewhat higher than those of diploid with the exception of stn-16 and the difference due to media was not observed. But after twelve months storage, survival rates of plants cultured in LSM 1(sucrose and sorbitol) was higher than those of plants cultured in LSM 2(sucrose and mannitol). The survival rate of stn-16, diploid wild species, was approximately 75% and it was considerably high. In Atlantic, tetraploid cultivated variety, every individual was survived.

시설하에서 재배되는 채소류가 생육이 순조롭지 못하다던지 경엽은 생육하고 있더라도 착과되지 않거나 착과되더라도 모양이나 품질면에서 상품성이 낮은 것이 수확되는 경우가 많은데 그 원인으로는 병충해와 생리장해가 있다. 채소류가 생리장해를 일으키는 원인은 뿌리의 손상, 지상부가 생육이 불량하게되는 토양환경 및 비료조건의 불량, 지상부에 영향이 크게 나타나는 기상환경의 불량, 접목장해, 살균제, 살충제, 제초제, 생장조절 등의 살포에 의한 직접, 간접적 약해 등으로 나눌 수 있다. 이들 원인은 실제로는 서로 영향을 주어서 장해를 크게 하는 것이 많다. 또 장해는 주로 경엽이나 뿌리에 발생하고 장해가 심해지면 꽃이나 과실에 미치는 것과 꽃과 과실에만 나타내는 것이 있다. 우리나라에서 시설재배면적과 경제성이 크고 생리장해가 심한 오이와 토마토의 과실에 나타나는 생리장해를 중심으로 그 원인과 대책에 관해서 기술코저 한다.

최근 소득작물로 재배면적이 늘고있는 시설하우스의 채소재배는 시설의 대형화와 고정화로 인한 연작장해와 주년재배에 의한 포장환경의 오염, 시설내의 고온다습, 무리한 약제살포 등으로 노지재배에서는 별로 문제가 되지않던 병들이 하우스내의 온도보존을 위한 2중, 3중의 밀폐로인한 높은 습도 때문에 거의 모든 작물에서 잿빛곰팡이병, 균핵병등이 발생되고 있다. 또한 연작으로 인한 토양전염성 병균의 밀도가 높아져서 역병, 풋마름병, 덩굴마름병등이 발생될 뿐아니라 지력증진을 위한 미숙 유기물이나 생육촉진을 위한 제4종 복합비료, 생장조절제등의 남용으로 전에는 없었던 새로운 장해가 증가되고 있어 이에 대한 방제대책의 일환으로 주산단지를 이루고 있는 농가의 병해충 발생및 토양환경등을 중심으로 문제점을 분석하고 대책을 소개하기로 한다.
