Two New Records of Marine Hydromedusae (Cnidaria: Hvdrozoa) in Korea (한국 해산 히드라해파리 (자포동물문: 히드라충강) 2미기록종)

  • Some hydromedusae were collected from Shiwhaho (Kyonsgi-do), Korea Strait and Yeosu, Korea during years 2000-2002. They were identified into Sorsia tubulosa (M. Sars, 1835) of the order Anthomedusae, and Liriope tetraphylla (Chamisso and Eysenhardt, 1821) of the order Trachymedusae respectively. They are new to the Korean fauna. The order Trachymedusae is first recorded in Korea. The long tube-like manubrium and four marginal tentacles are characteristics of S. tubuloso. In L. tetraphylla, the four triangle-shaped gonads and their positions upon half of surface of subumbrella, the seven blind centripetal canals in a quadrant and the long fat manubrium are its distinct characteristics. Resulting from this work, the Korean hydromedusae identified so far consist of 13 species of 11 families in five orders.

  • The Mixed germ cell tumors of the mediastinum are very quite rare. The Prognosis is generally dominated by the most aggressive component, which is represented by a choriocarcinoma, an endodermal sinus tumor, an embryonal carcinoma, and a seminoma, in descending order of in the degree of malignancy. We experienced one a case of a mixed germ cell tumor at the anterior mediastinum. The patient was 27-year-old male, who complained of hemoptysis and cough. The Chest X-ray showed a well-defined lobulated mediastinal mass in the left upper lung field. The operation was done and The mass was excised surgically. A Biopsy showed elements of mature tissues, immature neuronal components, and seminoma components.

Studies on the Migratory Ability of Primordial Germ Cells from Embryonic Gonads at Different Developmental Stages in Quail (메추리의 발달 중 배자 생식선에서 분리한 원시생식세포의 이동능에 대한 연구)

  • Avian primordial germ cells (PGCs) originate from the epiblast and appear in the germinal crescent. These PGCs enter the developing blood vessels during stage 10∼12 (H&H), circulate in the blood stream, migrate into the developing gonadal anlage and differentiate into germ cells. However, it is not clear until when the migratory ability of PGC is maintained. This study was conducted to examine whether migratory ability is present in PGCs from the gonad at later embryonic developmental stages. In the present study, gonads were dissected from 5-, 6- and 10-day old quail embryos and treated with trypsin-EDTA. Gonadal PGCs (gPGCs) were purified by Ficoll-density-gradient-centrifugation and labeled with PKH26 fluorescent dye. The PKH26-labeled gPGCs were microinjected into the blood vessel of the recipient quail embryo. Manipulated recipients were incubated for 3 days, embedded in paraffin and sdctioned. The foreign gPGCs were detected by fluorescent and confocal laser microscopy. As a result, quail gPGCs, from 10, 6 and 5 day old embryos could migrate through the recipient blood stream at early stage and settle in the gonads. Thus, results suggest that gPGCs from upto 10-day old embryos keep properties seen in circulating PGC. Therefore, the PGCs of 10-day old embryonic gonads can be used for the tools of genetic manipulation.

  • Male sexual differentiation involves a cascade of events initiated by the presence on the Y chromosome of the of the SRY (sex determining region of Y chromosome) gene, which causes the indifferent gonad to develop into a testis. Hormonal products of the testis, predominantly testosterone and Mullerian inhibiting subtance (MIS), then control the sexual differentiation of the developing fetus. SRY is a transcription factor; however, target genes for its action have yet to be identified, because the DNA recognition sequence for SRY is found in many genes. Therefore the study of intersex disorders is being used to identify other genes active in the pathway of sexual differentiation. Patients with 46,XY gonadal dysgenesis, or Swyer's syndrome, have streak gonads, normal stature, and a sexually infantile phenotype with Mullerian structures present. The inheritance is usually sporadic but can be autosomal dominant or X-linked recessive. Unlike 45,X patients, stigmata of Turner syndrome are rare. As many as 20 to 30% of patients are at risk for malignant gonadal tumor formation and should undergo gonadectomy soon after the diagnosis is made. We have experienced a case of Swyer syndrome which showed a positive SRY gene in peripheral blood and gonad. So we report this case with a brief review of literatures.

Recovery rate, growth and development of Heterophyopsis continua in experimental chicks (병아리에서 Heterophuopsis continua의 충체 회수율 및 성장 발육)

  • The growth and developmental pattern of H. continua was observed after experimental infection of their metacercariae to chicks. The recovery rate of worms from the chicks at 1 to 28 days post-infection(PI) was 12.8% in average. The rate remained fairly high for early 4 days of infection but decreased thereafter rapidly till 28 days PI. Most of the nukes, 91.9%, were recovered from the ileum of the chicks. In metacercariae, genital organs such as the ovary, testes, seminal vesicle, seminal receptacle and genital sucker were recognizable. At one day PI Mehlis'gland appeared, and at 2 days follicular vitellaria were observed. At 3 days PI, eggs were formed in the uterine tubule and increased in number as the worm grew old. The worms reached $2,990{\;}{\mu\textrm{m}}$ in length and $525{\;}{\mu\textrm{m}}$ in width at 28 days PI. Genital organs developed rapidly in early stages of infection but slowly thereafter to 28 days Pl, whereas non-genital organs developed steadily through the infection period. It was proved by this experiment that chicks should be a moderately suitable final host of H. continua.

Comparison of Vitrification and Slow Freezing for the Cryopreservation of Chicken Primordial Germ Cell (Ogye) (한국재래닭 (오계) 원시생식세포의 완만동결과 급속동결의 비교)

  • We sought to provide a method for freezing and preserving primordial germ cells, or an avian germ cell of a bird, as a material for developmental engineering or species preservation. The aim of this study was to compare the efficacy of slow freezing with a vitrification method for the cryopreservation of chicken primordial germ cells (PGCs). PGCs obtained from the germinal gonad of day 5.5-6 day (stage 28) cultured chick embryos, using the MACS method, were classified into two groups: slow freezing and vitrification. We examined the viability of PGCs after Cryopreservation. Four freezing methods were compared with each other, including the following: Method 1: The PGCs were frozen by a programmed freezer in a plastic straw, including 2.0 M ethylene glycol (EG) as cryoprotective additive (slow freezing) Method 2: The PGCs were vitrified in a plastic straw, including 8.0 M EG, plus 7% polyvinylpyrrolidone (PVP) (rapid freezing). Method 3: The slow freezing was induced with a cryotube including 2.0 M EG Method 4: The PGCs were frozen in a cryotube including 10% dimethyl suloxide (DMSO) (rapid freezing). After freezing and thawing, survival rates of the frozen-thawed PGCs from Method 1 to 4were 76.4%, 70.6%, 80.5% and 78.1% (p<0.05), respectively. The slow freezing ($-80^{\circ}C$ programmed freezer) method may provide better survival rates of frozen-thawed PGCs than the vitrification method for the cryopreservation of PGCs. Therefore, these systems may contribute to the cryopreservation of a rare avian species.

A Study on Sexual Maturation of Hen Clam Mactra chinensis Philippi (개량조개, Mactra chinensis Philippi의 성성숙에 관한 연구)

  • Gonadal development, the annual reproductive cycle and the first sexual maturity of hen clam, Mactra chinensis were studied histologically. Sexuality of the species was dioecious. The gonads were irregularly arranged from the subregion of mid-intestinal 91an4 in visceral cavity to the reticular connective tissue of the foot. The ripe eggs were about $50-60\;{\mu}m$ in diameter, and they were surrounded by gelatinous membrane. The spawning period was from May to September when the water temperature ranged $18.5-27.0^{\circ}C$, with the peak in June and July. The annual reproductive cycle of Mactra chinensis could be classified into five successive stages; multiplicative, growing, mature, spent, and degenerative and resting. The monthly changes of the fatness coefficient closely correlated with the annual reproductive cycle. Percentages of the first sexual maturity of female and male clams were over $50\%$ among those individuals ranging from 3.5 to 3.9cm, and $100\%$ in those over 5.0cm in shell length.

Quantitative Evaluation of Foodborne Pathogenic Bacteria in Commercial Sangshik (시판 생식에서 식중독균의 정량적 평가)

  • This study was carried out to survey the prevalence of foodborne pathogens in Sangshik products and their raw materials far the purpose of ensuring safety of those products in market, and establishing microbial regulatory standard. From 2002 to 2004, a total of 191 Sangshik products were purchased from market or mail-order sales, and major foodborne pathogens; E. coli, Salmonella, Staphylococcus aureus, Bacillus cereus, Clostridium perfringens, Listeria monocytogenes, Campylobacter jejuni, Yersinia enterocolitica, E. coli O157:H7, Vibrio parahaemolyticus were tested. B. cereus, C. perfringens and E. coli were detected from 29 samples (15.2%), 21 samples (11.0%) and 1 sample (0.5%), respectively. But other tested bacteria were not detected. For the identification of contamination source, 53 Sangshik ingredients were collected from 9 different manufacture factories. The results were similar with the Sangshik products. Aerobic plate counts were ranging from $1.0X10^3cfu/g\;to\;1.5X10^8cfu/g$. B. cereus was detected from 13 samples (24.5%), and counted as less than 100 cfu/g. C. perfringens were detected from 2 samples (3.8%), and counted as less than 100 cfu/g. Other foodborne pathogens were not detected except for B. cereus and C. perfringens. From the results, it was revealed that potential of microbial hazard by Sangshik was relatively low. However, it would be suggested that hygienic management and controling be needed for the prevention of growing contaminated pathogens and cross contamination during process and sale due to improper storage and management.

Potential Reproductive Toxicity Study of p53 Expressing Adenoviral Vector in Mice (아데노바이러스 유전자치료벡터의 생식독성 연구)

  • The possibility of inadvertent introduction of therapeutic gene expressing viral vectors has raised safety concerns about germ-line infection. Particularly, for indications such as prostate cancer and ovarian cancer, the proximity of the point of viral administration to organs of the reproductive system raises concerns regarding inadvertent germ-line transmission of genes carried by the virus vector. To evaluate the safety of in vivo adenovirus mediated gene transfer, we explored the biodistribution, persistance and potential germ-line transmission of p53-expressing adenovirus (Ad-CMV-p53). Both male and female Balb/c mice were injected with $1{\times}10^9$ PFU of Ad-CMV-p53. The PCR analysis showed that there were detectable vector sequences in liver, kidney, spleen, seminal vesicle, epididymis, prostate, ovary, and uterus. The RT-PCR analysis for detecting inserted gene, p53 showed that Ad-CMV-p53 viral RNA were present in spleen, prostate and ovary. Direct injected male and female mice of adenovirus vector into testis and ovary were mated and their of offspring were evaluated for germ-line transmission of the adenoviral vector. The PCR and RT-PCR analysis showed no evidence of germline transmission, although vector sequences were detected in DNA extracted from gonadal tissues. Real-time PCR result confirmed a significant decrease of adenovirus in gonad tissues 1 week after injection. We have also analysed the cell specific localization of viral DNA in gonad tissues by using in-situ PCR. Positive signals were detected in interstitial tissue but not in seminiferous tubule in sperm. In the case of ovary, adenovirus signal were localized to the stromal tissue, but no follicular signals were observed. Together, these data provide strong evidence that the risk of the Inadvertent germ-line transmission of vector sequences following intraperitoneal or direct injection into genito-urinary system of adenovirus is extremely low.

Growth and EVelopment of Pygidiopsis summa in Rats and Mice with a Supplementary Note on Its Morphological Characters (Pygidiopsis summa의 흰쥐와 마우스내에서의 성장발육 및 충체 형태학적 특징에 대한 검토)

  • The growth and development of Pygidiopsis summa were studied in experimental rats and mice, and a special reference was given to its morphological characters differed from the type species, P. genata. The metacercariae were obtained from young mullets (Mugil sp.), and total 21 rats and mice infected each with 1,000 metacercariae. Worms of various ages of infection, from 3 to 28 days, were subjected to a microscopic observation. The worms grew rapidly and remarkably in size up to 10 days of infection, to become 0.53~0.82mm long and 0.31~0.39mm wide, but nearly stopped the growth thereafter. Their genital organs developed more rapidly and fully matured within 3~5 days. At 3 days several eggs were found in uterus. The presence of two groups of small spines, 5~6 on the right and 7~9 on the left side of the genital apparatus, was a new finding in this study and considered a distinct character of P. summa. The morphology of ventral sucker and intestinal ceca was also different from P. genata. This study confirms the validity of the species, P. summa.
