Adsorption Properties and Production of Biosorbent from Bacillus sp. SK31 (Bacillus sp. SK3l의 생물흡착제 흡착특성 및 생산)

  • A strain (designated SK3l) which produces an excellent adsorption substance was isolated from soil samples and identified as Bacillus specied. The major adsorption substance (biosorbent SK3l) produced by Bacillus sp. SK31 was purified by ethanol precipitation and cetylpyridinium (CPC) precipitation. The adsorption charactics of zinc and lead ions on bioadsorbent SK3l were investigated. The equilibrium isotherms showed that bioadsorbent SK3l took up zinc and lead from aqueous solutions to the extent of about 52 mg/g and 112 mg/g. respectively. The culture conditions at the flask level of Bacillus sp. SK3l were investigated for the production of polysaccharide bioadsorbent, SK3l. The optimum pH and temperature for sorbent production were 7.5 and $30^{\circ}C$, respectively. The important carbon and nitrogen sources for sorbent formation were glucose and ammonium nitrate, respectively. In the optimized medium, sorbent production was improved three folds in comparison with the basal medium. In the jar fermenter, the highest sorbent production was obtained at 60 h cu1tivation time and the amount of biosorbent SK3l at that time was 9.2 g/$m\ell$.

  • Many researchers have reported that many biopolymers making up cell walls of the microorganisms display an ion-exchange property and play a major role in the sorption of the metal ions. Such polymers derived from microbial biomass are potentially useful as biosorbent materials for recovery various metal ions in industrial applications. although synthetic polymers such as ion-exchange resins and chelating resins have been widely used as commercial sorbents. In this study, valuable and commercial biopolymers for metal removal will be introduced.

  • Immobilization of biosorbent is very important for application to real wastewater treatment process because biosorbent itself does not have enough tough structure. Therefore, resent research on heavy metal biosorption using biomass has been focused on its efficient immobilization method. To improve the mechanical strength of freely biosorbent, many immobilization methods have been suggested for applications to the biosorbent such as microorganisms or polysaccharides. In this study, various immobilization methods such as adsorption, covalent binding, entrapment, encapsulation, and crosslinking will be introduced.

Removal/Recovery of Heavy Metals Using Biopolymer (생물고분자를 이용한 중금속 제거/회수에 관한 연구)

  • Zoogloea ramigera 115, well known type of bacteria to produce slime in sewage plants, was selected for biopolymer production. The extracted biopolymer showed high uptake capacity of metals such as cadmium and zinc. Especially the fermentor broth itself showed high adsorption of metal and could be used a biosorbent without an additional separation process. Biopolymer was immobilized into beads of calcium alginate and used in a packed bed reactor for the purpose of valued metals recovery. The biopolymer showed high removal efficiencies of 80% or greater for Cu, Cd, Mn and Zn, and high stability in sorption-desorption-resorption experiments. The immobilized biopolymer systems were found to be comparable to other metal removal systems such as ion exchange resins and to be of potential industrial application value.

Effect of Pore Size of Mesoporous Spherical Silica for the Purification of Paclitaxel from Plant Cell Cultures (식물세포배양으로부터 Paclitaxel 정제를 위한 메조다공성 실리카의 기공크기 영향)

  • Four types of mesoporous spherical silica adsorbents with different physical properties were prepared by spray pyrolysis and were used for the purification of the anticancer agent paclitaxel from plant cell cultures. Pore size had a greater effect on the removal of plant-derived impurities during the pre-purification of paclitaxel compared with surface area and pore volume. An appropriate pore diameter (~9.07 nm) was required to achieve the highest purity (~46.1%) and yield (~82.3%) of paclitaxel. These results were confirmed by HPLC analysis of the absorbent after treatment and Thermogravimetric analysis of the organic substances bonded to the adsorbent.

Adsorption Characteristics of Cadmium ions from Aqueous Solution using by-product of Brewing (주정오니를 활용한 수중의 카드뮴(Cd) 흡착 특성)

  • Biosorption is considered to be an alternative method to replace the present adsorbent systems for the treatment of metal contaminated wastewater. In this study, by-product which was abandoned from brewing factory was used to remove metal component in aqueous solution. The experimental results showed that the range of the removal efficiency is 60~91% and adsorption equilibrium was reached in about 3 hr. FT-IR and stereo microscope has been used to observe the surface conditions and changes in functional groups by calcination. At the end of elution, the amount of nitrogen and phosphorus in water was increased 11 and 7 times compare raw sample to calcinated samples. The Langmuir isotherm adequately described the adsorption of waste materials and the maximum adsorption capacity was 28.17 mg/g for Cd. The overall results suggested that waste material might can be used for biosorption of Cd.

Treatment of Nickel Ions in Water Phase Using Biochar Prepared from Liriodendron tulipifera L. (백합나무 유래 biochar를 이용한 수중에서 니켈 이온의 처리)

  • In this work, a new type of biosorbent was prepared from the biochar of Liriodendron tulipifera L. by adding an activation process using water vapor. By using the biosorbent, the removal characteristics of nikel ions in the water phase were investigated. When the equilibrium experiments to remove both 5 and 10 mg/L of nikel ions were performed, the adsorption amount of nickel ions was 4.2 and 5.4 mg/g, respectively. Also, the optimal initial pH was 6 to increase the removal efficiency with respect to two different nickel concentrations of 5 and 10 mg/L. To enhance the removal efficiency of 10 mg/L of nikel ions, a chemical treatment using critic acid was applied for the biosorbent. In addition, 100% removal efficiency was observed for 10 mg/L of nikel ions when the experiment was conducted for 2 h using the modified biosorbent treated by 4 M of critic acid. The results of desorption experiment to recover nikel ions indicated that 0.1 M of nitrilotriacetic acid (NTA) was selected as the optimal desorption agent. Consequently, these experimental results could be employed as an economical and environmentally friendly technology for the development of nickel removal processes.

The Effects of Surface Functional Groups to Protein Adsorption (단백질흡착에 있어서 표면작용기의 영향)

  • The adsorption characteristics of bovine serum albumin(BSA) on the modified carbon fiber and cellulose surfaces were investigated. In order to define the effects of solid surface characteristics on protein adsorption, surfaces of carbon fiber and cellulose were modified by physical and chemical treatment. The amounts of BSA adsorbed onto various solid surfaces were evaluated by batch method under various pH and ionic strength. The amount of adsorbed BSA was highly dependent on pH as well as surface functional groups.

  • The soluble paclitaxel was found in the supernatant of the plant cell cultures of Taxus chinensis, The percentage of soluble paclitaxel depends on paclitaxel concentration in bioreactor. As paclitaxel concentration decreases the percentage of soulbe paclitaxel increases. it is therefore important to develop a new process for the recovery of soluble paclitaxel. The use of hydrophobic resin HP20 gives nearly perfect recovery of paclitaxel in supernatant. The resin was more effective in treatment of th cell and debris free filtrate probably because of the reduced solids content In this case 3 g.l resin and 1 day reaction were enough for recovery the soluble paclitaxel in medium.

  • Three types of adsorbents were developed by immobilizing synthetic zeolite, Philipsite-Gismonine, in alginate, cellulose acetate and dialysis membrane for the in situ removal of ammonium ion which inhibits growth and productivity of animal cells such as CHO cells producing tPA. Ammonium ion removal efficiency and cell growth promoting effect with various immobilized adsorbents were evaluated and the membrane type was selected as an optimal immobilized adsorbent. The experiments were then simulated by adding 8mM ammonium chloride and immobilized adsorbent in order to validate the removal effect under high density cell cultures. The results showed increase in maximum cell density by three times, in cell viability, and in tPA productivity by 40%. And it was found that the promoting effects were more significant in case of high ammonium ion concentration system. It was also found that the optimum addition time for immobilized adsorbents was 48 hr in the absence of ammonium chloride addition and 72 hr in the presence of ammonium chloride addition.
