Lipase를 이용한 곤충 pheromone 합성 응용 - 생물 전환 기술

  • 본고에서는 화학적수법으로 다양한 특이한 기질을 제조하여 여기에 입체특이적 성질(stereospecificity)과 입체선택적 성질(steroselectivity)이 높은 생물학적 방법들 중에서 주로 lipase를 이용한 부제가수분해반응(asymmetric hydrolysis)과 그 역반응인 에스테르화반응(esterification)등을 도입한 chiral building block의 제조와 이를 이용한 곤충 생리활성물질인 pheromone의 합성에 대한 최근의 많은 예들중 상품화를 시도하고 있는 매미나방의 pheromne인 (+)-disparlure에 대하여 간단히 소개하고자 한다.

  • 생물화학에서 취급되고 있는 대부분의 효소는 metabolic map에 나타난 대사의 각각의 단계를 촉매하는 효소로써 생물학적 혹은 생리학적인 의의가 해명되어 있다. 그러나 현재로서는 metabolic map과는 관계없는 효소로 취급되는 것도, 생물이 생명유지를 위해서는 필수적인 어떤 화학반응을 촉매하고 있는 것으로 생각되고 있다. 생체내에 존재하는 무수한 효소 중에서 개개의 특이적인 효소의 성질을 해명하기 위해서는 촉매성의 기반이 되고 있는 효소단백질의 구조라든가 성질을 검토하지 않으면 안 된다. 그렇게 하기 위해서는 될 수 있는 한 목적효소를 생체내에 존재할 때와 같은 상태로 순수하게 정제할 필요가 있다. 일반적으로 효소류는 생체내에 존재할 때는 상당히 안정하게 유지되지만, 일단 세포 외에 추출되면 생체 내에서의 생리적 조건으로부터 벗어나기 때문에 서서히 실활되는 경우가 많다. 따라서 효소의 취급에는 안정성을 고려한 세심한 주의가 필요하다.

A Study on the Degradation of PAH in Organic and Aqueous Phases by Ozone (다환방향족탄화수소에 대한 오존처리의 방법에 관한 연구)

  • Pyrene of natural and anthropogenic sources is one of the toxic, mutagenic polycyclic aromatic hydrocarbons (PAHs) listed as priority pollutants. The objectives of this research are to break down pyrene by using ozonation, identify the intermediates and byproducts of pyrene, and test the biodegradability of intermediates and byproducts of pyrene in the aqueous phase. Since pyrene is non-polar, hexane was chosen as a solvent to effectively dissolve pyrene. Pyrene solutions were treated with ozone, as it has high oxidation capacity and electrophilic characteristic. After different ozonation pretreatment times (2, 3, and 10 minutes), intermediates of pyrene in the form of yellowish solid were collected from the hexane solution using a centrifuge. They were identified by gas chromatography/mass spectrometer (GC/MS). $BOD_5$, COD, and E-coli toxicity tests have been performed to assess the ozonation products.

  • This experiment was carried out to investigate the effect of sewage sludge compost amended soils on the growth and accumulation patterns of heavy metals in plant parts of Orchardgrass seedlings, changes in physical properties and chemical composition, and heavy metal residue in soils. Mixture ratios of sewage sludge compost and soil(loam) were 100:0, 80:20, 60:40, 40:60, 20:80 and 0:100(control), respectively. The results obtained were as follows; 1. The physical properties and chemical compostion of soils were improved by increase in mixture ratios of sewage sludge compost. 2. The biological yield of Orchardgrass seedlings was increased with mixture ratios of sewage sludge compost. 3. The dry weight of shoot(SH) was increased with both of yield components(NT and WT) and biological yield of Orchardgrass seedlings. 4. The total nitrogen concentrations(TN) of plants was increased with quadratically up to the biological yield of 100% mixture ratio of sewage sludge compost. 5. Lead(Pb) concentration of soil in over the 60% mixture ratios of sewage sludge compost were in excess of limiting level(50ppm) of organic fertilizers.

미토콘드리아의 $tRNA^{Asp}$ 유전자의 한 돌연변이와 그의 억제 유전자들

  • tRNA는 그 생화학적인 역할이 잘 알려져 있고 구조적으로 안정하며, 이용할 수 있는 분자 생물학적인 자료가 많아, 유전자 발현과 유전자 산물간의 조직적인 상호작용을 연구하는데 적합한 재료이다. 효모의 미토콘드리아에는 24개의 tRNA 유전자가 잇어, 단백질 합성에 필요한 tRNA를 자급하고 있으나, 유전자 발현과 processing에 관여하는 모든 정보가, tRNA의 5' 부위를 process하는데 관여하는 효소중 RNA subunit인 9S RNA를 산출하는 tsl 유전자를 제외하고, 핵 유전자에 존재한다. 효모의 대표적인 종인 Saccharomyces cerevisiae의 $tRNA^{Asp}$ 유전자에 결함이 생긴 한 돌연변이 균주의 성질을 조사하고, 억제현상(suppression)을 규명하므로써 tRNA의 구조적 특성을 파악하고, 나아가 미토콘드리아 생성에 관여하는 핵 유전자를 찾아보고자 한다.

  • Natural actomyosin was extracted from the dorsal muscle of carp (Cyprinus carpio), and some Physico-chemical properties were determined. The sedimentation constant ($s_{20},\;_\omega$) of the purified skeletal muscle actomyosin was about 24.76s and the ATP sensitivity 147-176. The specific viscosity was rapidly ascended with the increase of protein concentration as other vertebrate actomyosins. ATPase activating site under the effect of pH variation was showed on the both sides of acid and base resembling with the tendency of rabbit skeletal muscle actomyosin, and found at 6.0 on the acidic side and at 9.5 on the basic side.

  • A new extracellular biopolymer was produced by Methylobacterium organophilum from methanol as a sole carbon and energy source. The purified biopolymer was found to have a high molecular weight of about 4-5$\times$10$^6$ dalton and contained 66% (w/w) of carbohydrate but no polyhydro xybutyrate. Other organic constituents were consisted of protein, pyruvic acid, uronic acid, and acetic acid, whereas content of inorganic ash was 22%. Based on the chemical analysis of the biopolymer by TLC method, the polymer was consisted of glucose, galactose, and mannose with an approximate molar ratio of 2:3:2. The biopolymer solution showed a characteristics of pseudoplastic non-Newtonian fluid. The viscosity of the 1%-biopolymer solution was found to be 18,000 cp at a shear rate, 1 sec$^{-1}$, which was almost 10 times higher than that of a commercial xanthan gum.

Changes on Physical Property of Ilmenite due to Microorganism (미생물에 의한 티탄철석의 물리적 특성 변화)

  • Laboratory tests for measuring absorption, porosity, P-wave velocity and uniaxial compressive strength (UCS) were performed to examine weathering characteristics of ilmenite by microorganism. Physical property changes were quantitatively estimated with comparing culture period on the condition of abiotic oxidation without microorganism and biooxidation with microorganism. As a result, the measured pH during 45 days was distributed in the range from 3.82 to 4.26, on the other hand, biooxidation showed the range from 2.20 to 2.57. The measured absorption according to microorganism and culture period represented 0.052% at final stage in the case of abiotic oxidation and 0.073% in the case of biooxidation. Porosity showed 0.206% at final stage in the case of abiotic oxidation and 0.281% in the case of biooxidation. In general, the values by biooxidation showed higher than that by abiotic oxidation. Change range of P-wave velocity with culture period showed that the measured value as 1410 m/s at final stage in the case of biooxidation was lower than 1886 m/s of that in the case of abiotic oxidation. The UCS was decreased with increasing culture period in all specimens and represented 241.1 MPa at final stage in the case of abiotic oxidation and 140.0 MPa in the case of bioxidation. In conclusion, it implies that influence of physical property on ilmenite by biooxidation related with microorganism was larger than that by abiotic oxidation.

  • Land treatment를 이용한 폐수와 폐기물의 성공적이며 안전한 처리는 토양에서의 적절한 생물학적, 물리적, 화학적 반응들이 일어날때 이루어지며 이를 위해서는 처리해야할 폐기물과 토양의 성질에 따라 적절한 공정의 설계가 따라야할 것이다. Land treatment는 다른 물리화학적 처분방법에 비해 약간 느리고 불완전하다 할지라도 비용이 저렴하고 또한 landfill이나 deepwell injection등 토양을 이용한 다른 처분방법에 비해 지하수나 인근지역의 오염, 가스발생 등의 문제점이 훨씬 적다. 토지의 반복적인 처리장 부지로서의 사용 후에는 경우에 따라서는 농작물이나 가축사육용 초지로 이용할 수 있고 농경지로 이용 못할 경우에는 공원이나 건축용부지, 체육시설 등으로 이용이 가능하므로 우리나라에서도 정유, 식품공장 등지에서 폐기물의 처리및 처분방법으로 시도해 보는 것이 바람직하다고 생각된다.
