Serratia marcescens, an enterobacterium of gram-negative bacteria, is characterized by resistance of the admium. Cadmium sensitive PM strain did not grow in the medium at cadmium concentration of 50 ppm. PA strain was induced to accommodate to cadmium by cultivating the mother strain (PC strain) in the medium with 50 ppm cadmium. As compared with PC and PM strains, PA strain revealed the excellent growth in cadmium media and accumulated four to five times higher cadmium concentration in cell than other strains. PA strain accumulated 23% of cadmium in cells when cultured in medium treated with 100 ppm cadmium and this cadmium was more accumulated in cytosol fractions than membrane fractions. Analysis by TEM indicated that cadmium was concentrated as a form of granule in cytosol. In protein patterns of cell after the treatment of cadmium, two inducible proteins (28 KDa and 64 KDa) and one reducible protein (45 KDa) were detected by SDS-PAGE. By Atomic Absorption Spectrophotometer, the amounts of cadmium attached to inducible proteins of 28 KDa and 64 KDa were 318.28 ㎍ and 325.37 ㎍ per gram of protein, respectively. It is assumed that these inducible proteins play an important role in the mechanism of cadmium accumulation in cells. A plasmid of 23Kbp was found in S. marcescens. The ability of resistance to cadmium in plasmid was confirmed by curing experiments.

This study was performed to investigate the characteristics of nutrient removal of municipal wastewater in the membrane bioreactor system with the different types of membrane. Membrane bioreactor consists of three reactors such as two intermittent anaerobic and the submerged membrane aerobic reactor with flat sheet and hollow fiber membrane, respectively. The removal efficiencies of $COD_{cr}$, BOD, SS, TN and TP on the flat sheet membrane bioreactor were 94.3%, 99.0%, 99.9%, 70.3% and 63.1%, respectively. In addition, The removal efficiencies of $COD_{cr}$, BOD, SS, TN and TP on the hollow fiber membrane bioreactor were 94.0%, 99.3%, 99.9%, 69.9% and 66.9%, respectively. The estimated true biomass yield, specific denitrification rate (SDNR), specific nitrification rate (SNR) and phosphorus removal content on the flat sheet membrane bioreactor were $0.33kgVSS/kgBOD{\cdot}d$, $0.043mgNO_3-N/mgVSS{\cdot}d$, $0.031mgNH_4-N/mgVSS{\cdot}d$, and 0.144 kgP/d, respectively. In addition, the estimated true biomass yield, specific denitrification rate (SDNR), specific nitrification rate (SNR) and phosphorus removal content on the hollow fiber membrane bioreactor were $0.30kgVSS/kgBOD{\cdot}d$, $0.067mgNO_3-N/mgVSS{\cdot}d$, $0.028mgNH_4-N/mgVSS{\cdot}d$, and 0.121 kgP/d, respectively. There was little difference between the flat sheet and hollow fiber on the nutrient removal efficiencies except SNR and SDNR. These differences between them were caused by the air demand to prevent the membrane fouling. The flux and oxygen demand for air scouring were $19.0L/m^2/hr$ and $2.28m^3/min$ for the flat sheet membrane, and $20.7L/m^2/hr$ and $1.77m^3/min$ for the hollow fiber membrane on an average.
Glutathione S-transferases (GSTs) are a superfamily of detoxification enzymes that primarily catalyze the nucleophilic addition of reduced glutathione to both endogenous and exogenous electrophiles. In this study, we isolated and characterized a full-length of alpha class GST cDNA from the abalone (Haliotis discus hannai). The abalone GST cDNA encodes a 223-amino acid polypeptide with a calculated molecular mass of 25.8 kDa and isoelectric point of 5.69. Multiple alignments and phylogenetic analysis with the deduced abalone GST protein revealed that it belongs to the alpha class GSTs and showed strong homology with disk abalone (Haliotis discus discus) putative alpha class GST. Abalone GST mRNA was ubiquitously detected in all tested tissues. GST mRNA expression was comparatively high in the mantle, gill, liver, and digestive duct, however, lowest in the hemocytes. Expression level of abalone GST mRNA in the mantle, gill, liver, and digestive duct was 182.7-fold, 114.8-fold, 4675.8-fold, 406.1-fold higher than in the hemocytes, respectively. Expression level of abalone GST mRNA in the liver was peaked at 6 h post-infection with Vibrio parahemolyticus and decreased at 12 h post-infection. While the expression level of abalone GST mRNA in the hemocytes was drastically increased at 3 h post-infection with Vibrio parahemolyticus. These results suggest that abalone GST is conserved through evolution and may play roles similar to its mammalian counterparts.

Pine needle(Pinus densiflora sieb, et zucc) extract has been used to improve cardiovascular disorders, detoxification of nicotine, the infirmities of age and curing diseases of unidentified symptoms. It has various useful components including amino acids, vitamin C, terpenoids and chlorophyll. In this study we have identified 8 different yeast strains that are developed spontaneously causing self fermentation in the extract. The self-fermented pine extract(SFPE) inhibited the growth of some bacterial strains like E. coli, Bacillus subtilis and Staphylococcus aureus. The SFPE($0.2{\mu}{\ell}/ml{\sim}0.3{\mu}{\ell}/ml$) showed 90% NBT superoxide scavenging activities which is similar for all tested samples of different ages. The 7 years old SFPE(0.15 mg/ml and 0.3 mg/ml) caused relaxation of spontaneous contraction and relaxation rhythm of thoracic arterial tissues from rat. Therefore, SFPE has useful effects such as antibacterial, antioxidant and improved blood circulation and could be a good source of functional food development.
Peroxiredoxins (Prxs) are a ubiquitous family of antioxidant enzymes that participate in a variety of biological processes, including $H_2O_2$-mediated signal transduction, molecular chaperoning, and mitochondrial function. In this study, we isolated and characterized a Prx 2 cDNA from abalone (Haliotis discus hannai). The abalone Prx 2 cDNA encoded a 199-amino acid polypeptide that belongs to a class of typical 2-Cys Prxs that contain peroxidatic and resolving cysteines. The deduced abalone Prx 2 protein showed strong homology (64-99%) with Prx 2 proteins from other species, including mollusk, fish, amphibians, and mammals, and it was most closely related to disk abalone (H. discus discus) Prx 2. Abalone Prx 2 mRNA was ubiquitously detected in tested tissues, and its expression was comparatively high in the mantle, gills, liver, foot, and digestive duct. The expression level of abalone Prx 2 mRNA was 106.7-fold, 51.9-fold, and 437.8-fold higher, respectively, in the gills, digestive duct, and liver than in the muscles. The expression level of abalone Prx 2 mRNA in the liver peaked at 6 hr postinfection with Vibrio parahemolyticus and decreased at 12 hr postinfection. The expression level of abalone Prx 2 mRNA in hemocytes was drastically increased at 1 hr postinfection with V. parahemolyticus. These results suggest that abalone Prx 2 is conserved through evolution and that it may play a role similar to that of its mammalian counterpart.
The present study was carried out to investigate the effect of microwave fixation in comparison with that of chemical fixation in preparing the microscopic samples. The microwave fixator was equipped with infrared-temperature sensor, and that was designed to compensate air temperature in the microwave fixator. In the microwave fixation, rat tongue was well preserved in terms of muscular fasciculus and pancreas stained by Feulgen reagents showed clear reaction products in the nucleus. Reaction products by PAS method in duodenal villi appeared specifically at the goblet cells. In electron microscopy, pancreatic cellular components such as secretory granules and collagen bundles were well preserved in both fixations. In aspect of histochemical reaction and electron microscopy, high quality was due to the protein content of microwave fixed specimen. The microwave fixation method saved total duration engaging microscopic preparation.
Insects represent the largest class within the animal kingdom in terms of species number. Humans had been utilized insect in the broad area, including food, agriculture, industry, pharmaceuticals and so on. At present, insects are emerging as a leading group for identifying and extracting novel bioactive substances due to enormous number and a high nutritional value. Insects rely on a suite of systemic response to resist infection such as immune cells, hemocytes, activation of enzymes cascades, and antimicrobial peptide/protein. Among the substances, antimicrobial peptides (AMPs) are main components of potent antimircrobial innate defense system into the insect hemolymph. AMPs raise influential candidate as avenue to resolve the development of antibiotic-resistant microbial organism. Insect AMPs are classified into four main classes: cecropins, insect defensins, glycine/proline-rich peptides. Insect AMPs have been purified, over 150. In this review, AMPs derived from several insects were summarized including honey bee, dung beetle, butterfly and longicorn beetle. These peptides almost exhibited potent antimicrobial activities against human microbial pathogens without causing remarkable hemolysis to erythrocytes excluding melittin, and their mode of action(s) are based on disruption of the plasma membrane or fungal apoptosis. Therefore, study of insect AMPs is expected to be useful for designing novel therapeutic antimicrobial applications.

Mesenchymal stem cells (MSC) are multipotent and can be isolated from diverse human tissues including bone marrow, fat, placenta, dental pulp, synovium, tonsil, and the thymus. They function as regulators of tissue homeostasis. Because of their various advantages such as plasticity, easy isolation and manipulation, chemotaxis to cancer, and immune regulatory function, MSCs have been considered to be a potent cell source for regenerative medicine, cancer treatment and other cell based therapy such as GVHD. However, relating to its supportive feature for surrounding cell and tissue, it has been frequently reported that MSCs accelerate tumor growth by modulating cancer microenvironment through promoting angiogenesis, secreting growth factors, and suppressing anti-tumorigenic immune reaction. Thus, clinical application of MSCs has been limited. To understand the underlying mechanism which modulates MSCs to function as tumor supportive cells, we co-cultured human adipose tissue derived mesenchymal stem cells (ASC) with cancer cell lines H460 and U87MG. Then, expression data of ASCs co-cultured with cancer cells and cultured alone were obtained via microarray. Comparative expression analysis was carried out using DAVID (Database for Annotation, Visualization and Integrated Discovery) and PANTHER (Protein ANalysis THrough Evolutionary Relationships) in divers aspects including biological process, molecular function, cellular component, protein class, disease, tissue expression, and signal pathway. We found that cancer cells alter the expression profile of MSCs to cancer associated fibroblast like cells by modulating its energy metabolism, stemness, cell structure components, and paracrine effect in a variety of levels. These findings will improve the clinical efficacy and safety of MSCs based cell therapy.
Recent evidence has revealed that the intratesticular injection of hypertonic saline(20%) resulted in a chemically castrated state such as nadir testosterone levels in rats. To confirm the efficacy of this simple saline-injection method further, we investigated the changes in the gross and microscopic anatomy of testis. Our study comprised three groups; intact(control) group, orchidectomy group and saline-injection (experimental) group. Single dose of hypertonic saline (sterilized, $750{\mu}{\ell}/testis$) were directly administered into both testis of adult rats (about 300 g BW). Bilateral orchidectomy was performed at the same day of saline injection. Following 30 days post-injection, reproductive tissues were surgically removed, weighed and fixed for histological examination. The body weights were not changed in both orchidectomy group and saline-injection group when compared to those in intact group. The wet weights of testis were significantly decreased in saline-injection group when compared to those in intact group. The wet weights of epididymis and seminal vesicle and prostate were significantly decreased in orchidectomy group and saline-injection group when compared to those in intact group. Macroscopically, the testes exerted slight atrophy and the tunica albuginea seemed to be intact in saline injection group. Histologically, however, larger parts of testicular tissue underwent necrosis and were barely recognizable after hematoxylin-eosin staining. In the same section, only the opposite part of the injection site was stained showing abnormal state of cell layers mostly fibrosis and infiltrated leukocytes. Sloughing of immature germ cells from the basement membrane along with shedding cells in the intraluminal space was notable in most seminiferous tubules from the saline injected testis. The present study confirmed that the direct injection of hypertonic saline into testis can induce a castration-like, testosterone-depriving effects on accessory sex organs. Our findings suggest that the efficacy of this less expensive and minimally invasive method seems to be almost even with that of conventional orchidectomy and chemical castration, though more in-depth evaluation should be supported.
Liquiritigenin (LG) is a chiral flavonoid isolated from the roots of licorice. It exhibits multiple biological activities including anti-oxidant, anti-cancer, and anti-inflammatory effects. In particular though, the anti-cancer activity of LG in oral squamous cell carcinoma has yet to be elucidated, and LG-induced apoptosis in oral squamous cell carcinoma remains poorly understood. In the present study, we tested the role of LG in inducing apoptosis in oral squamous cell carcinoma cells. LG treatment of HN22 cells resulted in a dose-dependent inhibition of cell viability as detected by a 3-(4,5-dimethylthiazol-2-yl)-2,5 diphenyltetrazolium bromide assay. The induction of apoptosis in terms of Annexin V/7-Aminoactinomycin D staining, sub-G1 population, and multi-caspase activity were assessed with a $Muse^{TM}$ Cell Analyzer. Flow cytometric analysis revealed that LG treatment resulted in G2/M arrest in cell cycle progression and downregulation of cyclin B1 and CDC2 expression in a concentration-dependent manner. It also resulted in significant upregulation of p27. In addition, LG was seen to trigger the generation of reactive oxygen species and induce CCAAT/enhancer-binding protein homologous protein and 78-kDa glucose-regulated protein in concentration-dependent upregulation. The LG treatment of HN22 cells led to a loss of mitochondrial membrane potential (${\Delta}{\Psi}m$); it also reduced the levels of anti-apoptotic protein and increased the expression of apoptotic protease activating factor-1, cleaved poly (ADP-ribose)polymerase and Bax. Overall, our results indicate that the pro-apoptotic effects of LG in HN22 cells depend on the activation of both intrinsic and extrinsic signaling pathways. Thus, our results suggest that LG constitutes a natural compound with a potential role as an anti-tumor agent in oral squamous cell carcinoma.
