  • 중금속은 해수에서보다 해수중의 생물계에 그 농도가 더 높은 것으로 알려져 있으며 수은도 무기형으로 생물계에 존재하다가 생물활동으로 인해 알킬형으로 변한다. food chain이 미량금속 축적의 주통로로 알려져 있으나 그외 몇가지 방법이 제시되고 잇다. 특히 연체동물은 미량금속의 축적현상이 온도, 폭로시간, 시간, 생리적 활동에 다라 상당히 달라진다. 또한 유기알킬 수은이 무기수은보다 비교적 낮은 독성을 나타내지만 후자의 피해도 무시할 수는 없다. 굴(crassostrea virginica)은 염화수은농도가 비교적 낮은 상태에서도 상\ulcorner량의 축적현상을 보여주었다. 두개의 compartment system을 연계적인 상태에서 보면 첫 compartment에서의 초기축적은 가역적이었으나 둘째 Compartment에서 측정된 원실율로 보아 비가역적 축적이상을 확인해 주었다. 뿐만 아니라 미량금속이 아가미와 외부 근육에서 가장 높은 축적률을 보인것은 flow system의 regression moder과 매우 흡사함을 암시하며 농도가 낮은 상태에서의 축적이상에 중요한 의미를 부여하였다.

Measurement of Mass Flow of Water in the Stem of Musk Melon by Sap Flow Gauge (열목지 경유센서에 의한 멜론 경유양의 측정)

  • The mass flow of water in the stem of melon measured by Sap Flow Gauge was compared with the actual flow calculated by the difference between supply and drainage nutrient water to investigate the possibility and accuracy of estimation of melon's transpiration in rockwool culture. The Sap Flow Gauge which was made with copper-constantan theromocouple and nichrome fiber by our research team, was attached to the 3rd node of melon. The outdoor temperature, room temperature, solar radiation and relative humidity were continually measured. The amount of supply and drainage nutrient water were simultaneously measured for calculation of practical consumption of nutrient water to compare with mass flow of sap. The measuring errors of Sap Flow Gauge were 0.3 to 31.8%, which were small at solar radiation of 20MJ.m$^{2}$.d$^{-1}$ . The mass flow of water was lower for the measured value by Sap Flow Gauge than the actual value at higher solar intensity, however it was higher at lower solar intensity The variation of error rate of each Sap Flow Gauge was 0.1 to 13.0%. The measuring error with Sap Flow Gauge was negatively related with solar intensity and temperature. Therefore, to measure more exactly the mass flow of sap for estimation of melon's transpiration, the compensation factor must be calculated.

  • 이상 방사면역측정(放射免疫測定)의 기본원리(基本原理), 순수(純粹)한 항원(抗原), 표지항원(標識抗原), 항체생성(抗體生成), 표준곡선(表準曲線)의 작성(作成), 항원항체복합체(抗原抗體複合體)의 분리방법(分離方法)에 대(對)하여 설명(說明)하였다. 생물학적방법(生物學的方法),화학적방법(化學的方法), 기계적(機械的)인 측정방법(測定方法)에 의(依)해 생체내(生體內)의 미량(微量)으로 존재(存在)하고 있는 물질(物質)들을 정확(正確)하게 측정(測定)할 수 없다. 그러나 방사면역측정법(放射免疫測定法)에 의(依)해 쉽게 측정(測定)되어 기관(器管)의 기능검사(機能檢査)는 물론(勿論) 각(各) 기관(器管)과의 상관관계(相關關係)를 분석(分析)하여 질병(疾病)의 진단(診斷), 치료경과(治療經過)을 평가(評價)하는데 매우 중요(重要)한 정보(情報)를 제공(提供)해주고 있다. 이러한 점(點)을 고려(考慮)하여 이 방법(方法)을 잘 습득(習得)하고 진료(診療)의 수단(手段)으로 널리 이용(利用)되어 의료기술발전(醫療技術發展)에 토대(土臺)가 되기를 바란다.

Development of the Robot Manipulator for Kinematies (기구학적 분석을 이용한 로봇 매니퓰레이터 개발)

  • This study is kinematics for the manipulator development of cucumber harvesting. A theory value was verified by repeated error measurement after the forward kinematics or inverse kinematics analysis of manipulator. Manipulator is consisted of one perpendicular link and two revolution link. The transformation of manipulator can be valued by kinematics using Denavit-Hartenberg parameter. The value of inverse kinematics which is solved by three angles faction shows two types. Repeated errors refered maximum 2.60 mm, 2.05mm and 1.55 mm according to X, Y, Z axis. In this study, the actual coordinates of maximum point and minimum point were agreement in the forward kinematics or inverse kinematics. The results of repeated error measurement were reflect to be smaller compared to a diameter of cucumber. measurement errors were determined by experimented errors during the test. For reducing errors of manipulator and improving work efficiency, the number of link should be reduced and breeding and cultural environment should be considered to reduce the weight and use the hard stuff. The velocity of motor for working should be considered, too.

Growth Response according to Indoor Light Levels of Two Evergreen Conifer Species Native to Korea (자생 상록 침엽수 개비자나무와 비자나무의 실내 광량에 따른 생육반응)

  • 개비자나무[Cephalotaxus harringtonia (Knight ex Forbes) K. Koch]와 비자나무[Torreya nucifera (L.) Siebold & Zucc.]의 3년생 실생묘를 유리온실에 재배하면서 실험재료로 사용하였다. 실내의 광량을 측정하여 10, 50, 100 및 200 PPFD (${\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$)로 설정하였다. 광량을 제외한 광주기(12/12 h), 온도($25{\pm}1^{\circ}C$), 습도($55{\pm}3%$) 및 관수(1회/3일)주기를 일정하게 유지시킨 밀실에서 8주간 재배하였다. 유리온실에서 재배중인 식물과 광량별 처리구의 초장, 줄기직경, 엽수, 엽장, 엽폭, 엽록소함량(SPAD) 및 광화학반응을 평가하였다. 개비자나무의 생육결과, 모든 광량조건에서 줄기직경과 엽록소함량이 증가하는 경향을 보였으며, 초장, 엽폭의 경우 대조구와 유의한 차이가 없었다. 스트레스지수(Fm/Fo), 최대양자수율(Fv/Fm) 및 광계II 성능지수(Pi_Abs)는 대조구에 비해 모든 광량에서 증가하는 경향을 나타냈다. 10 PPFD의 광량을 제외한 처리구에서 재배된 비자나무의 초장, 줄기직경, 엽장, 엽폭은 대조구와 유사한 경향을 나타냈다. 한편, 엽수와 엽록소함량은 대조구에 비해 우수하였다. Fm/Fo, Fv/Fm, Pi_Abs 및 비광학적 에너지의 손실(DIo/RC)은 모든 광량처리구에서 대조구에 비해 유의적으로 높은 측정값을 나타냈다.

A Study of Skin Biophysical Parameters and Biomarkers related to the Anatomical Site and Age in Korean Women (한국 여성의 피부 부위 및 연령에 따른 피부 측정 인자와 생물 인자 연구)

  • The skin is one of the largest organs in our body and participates in many of the human organism's physiological and pathological events. Skin function were known for self-maintenance and self-repair, mechanical and chemical stress protection, protection against UV and environmental pathogenic micro-organisms, production of vitamin D, and social and psychological function through the physical aspect. The aim of this study was to evaluate the variation of biophysical parameters and to find relation with skin biomarkers in different anatomical site and age in Korean women. About 70 healthy volunteers in age range 20 to 49 were participated in this test. Test areas were the forearms and the cheek. Investigation to determine biophysical parameters on human skin, was carried out using various non-invasive methods. For analysis to skin biomarkers, we studied to examine various biomarkers for the quantitative determination of cortisol, fibronectin, keratin-1, 10, and 11, involucrin, and keratin-6 in human face and forearm. And we measured to skin biophysical parameters for skin anatomical site and age difference with non-invasive methods. As results of measuring site, some parameters were have following significant difference, stratum corneum hydration, trans epidermal water loss and skin color (L and a value). As results of age difference, skin colors were had only significant difference with age. For cortisol, keratin-6, fibronectin, keratin-1, 10, 11 contents, there were no significant difference in age and site. However, involucrin level in the cheeks were the highest for age group 30 ~ 39 compared to other age groups. These results suggest that in individual skin condition may explain detailed skin state variation.

Comparison of Oligosaccharyltransferase Assay Methods Using a Fluorescent Peptide (형광펩타이드를 이용한 Oligosaccharyltransferase Assay 방법 연구)

  • Oligosaccharyltransferase (OTase) catalyzes the transfer of a lipid-linked oligosaccharide (LLO) to the nascent polypeptide. Most eukaryotes have an OTase composed of a multisubunit protein complex. However, the kinetoplastid Leishmania major and the bacterium Campylobacter jejuni have only a single subunit for OTase activity, Stt3p and PglB, respectively. In this study, a new in vitro assay for OTase was developed by using a fluorescent peptide containing N-glycosylation sequon, Asn-Xaa-Thr/Ser, where Xaa can be any amino acid residue except Pro. L. major Stt3p and C. jejuni PglB as a model OTase enzyme demonstrated the formation of glycopeptides from a fluorescent peptide through OTase activities. For separation and measurement of the glycopeptides produced by the OTases, Tricine-SDS-PAGE, a lectin column and fluorospectrophotometer, and HPLC were applied. Comparison of these assay methods for analyzing a fluorescent glycopeptide showed HPLC analysis is the best method for separation of glycopeptides and nonglycosylated peptides as well as for quantify the peptides than other methods.

Development and Application of Acute Bioassay Method on Solid Samples using Bioluminescence Producing Bacteria (박테리아 발광 특성을 이용한 고형시료 급성 독성법 연구 및 적용)

  • A toxicity method using bioluminescence producing bacteria, Escherichia coli DH5 RB1436, was developed and applied on solid environmental samples. In the assay, 1 g soil sample was mixed with 4 ml RB1436 strain. Sets amended with p-buffer were employed for control in soil test, showing approximately 108% of sets amended with combusted soils. Measurable differences were observed between relatively more polluted groups (HP) and less polluted groups (LP) of soil samples, showing average toxicity 43 and 26%, respectively, in direct soil toxicity test. $EC_{50}$'s for all soil groups appeared in the range of $1.8{\sim}4.6\;g$, but those of sediments from dam reservoir and refuses were below 0.22 g. This developed bioassay should prove useful as a screening test for toxicity in various types of environmental solid samples.

Study on the Effects of the Healing Programs Using Emotional Insects - Bombyx mori (Linnaeus) Case - (정서곤충을 이용한 치유프로그램의 효과 분석 - 누에나방 사례 -)

  • The study aimed to investigate the psychological effect of the emotional insect-assisted healing program on the stress experienced by 43 children in a community child center. Silkworm (Bombyx mori [Linnaeus]) was used in the healing programs, and the stress was measured using salivary alpha-amylase levels. Saliva samples and a survey on insect preference were collected before and after the healing programs, respectively. After the program, the satisfaction score for participation was evaluated. Results indicate that the emotional insect-assisted healing programs led to statistically significant lowering of salivary alpha-amylase levels and increased the preference for insects. The children's satisfaction in the healing program activities using silkworms was 4.23 points out of 5.

Determination of the Bioconcentration Factor in Some Pesticides by Flow-through Fish Test (Flow-through fish test를 이용한 일부 농약의 생물농축예수의 측정)

  • Zebrafish(Brachdanio rerio)를 실험어류로 하여 methidathion 과 phosalone의 생물농축계수(bioconcentration factor : BCF)와 배설속도상수 (depuration rate constant) 및 LC$_{50}$를 측정하였다. Methidathion의 24, 48, 72, 96시간 LC$_{50}$는 각각 28.34, 35.98, 24.43, 22.03 mg/$\ell$로 측정되었다. Methidathion 0.22 mg/$\ell$(고농도)와 0.022 mg/$\ell$(저농도)에서 어류 체내에서의 농축정도는 두 농도군에서 각각 12시간 이후에 정류상태에 도달하여 72시간동안 거의 일정하였고, BCF값도 12시간에서 72시간 사이에 고농도와 저농도에서 8.72(n=4)와 11.25(n=4)로 조사되었다. 배설속도상수는 고농도와 저농도에서 6시간 이내에 모두 배설되어 배설속도상수를 구할 수 없었다. Phosalone의 24, 48, 72, 96시간 LC$_{50}$는 각각 3.76, 2.43, 1.86, 1.05 mg/$\ell$로 측정되었다. Zebrafish 체내에서의 농축정도와 BCF값은 고농도(0.01 mg/$\ell$)에서 12시간 이후에 정류상태에 도달하여 72시간동안 거의 일정하였고, BCF값은 12시간에서 72시간 사이에 48.88(n=4)로 측정되었다. 저농도(0.001 mg/$\ell$)에서는 실험 전기간동안 zebrafish 체내에서 phosalone이 검출되지 않아 BCF값을 산출할 수 없었다. Zebrafish 체내에서 phosalone(고농도)의 배설속도상수와 반감기를 구하기 위하여 6,12시간의 배설실험 결과 각각 0.17$hr^{-1}$과 4.01 시간이었다. Methidathion과 phosalone의 BCF값은 phosalone이 methidathion 보다 약 5배 정도 높게 나타났으며, 농약의 배설속도는 phosalone이 methidathion보다 빨랐다.
