Development a glucose-FIA system with a fiber optic oxygen sensor

  • Flow injection analysis (FIA) system was developed to monitor glucose concentrations in biotechnological processes. A fiber optic oxygen sensor was used to determine consumption of oxygen concentration by reaction of immobilized glucose oxidase (GOD). The GOD was immobilized on VA-Epoxy carrier and integrated into FIA system. A calibration curve for glucose was obtained in the range of 0.5 $g/L{\sim}3.0$ g/L.

Production and characterization of anti-Salmonella polyclonal antibodies as bio-recognition element for developing a microbial monitoring method (미생물학적 모니터링 분석방법 개발을 위한 생물학적 수용체로서 살모넬라에 특이적인 다중클론 항체의 생산 및 특성 검토)

  • For the construction of the microbial monitoring method, anti-Salmonella polyclonal antibodies (pAbs) were produced from a rabbit and purified by saturated ammonium sulfate precipitation and protein A affinity column. The reactivity of anti-Salmonella pAbs was compared to that of commercial ones by using an indirect ELISA. The specificity of anti-Salmonella pAbs was investigated using 20 Salmonella serotypes and 20 non-Salmonella strains. A capturing ability of anti-Salmonella pAbs was investigated by exposing antibody-immobilized gold biosensor to different concentration of Salmonella mixture. Anti-Salmonella pAbs were successfully produced and purified with an antibody concentration of 2.0 mg/mL The reactivity of purified anti-Salmonella pAbs was greater than that of commercial one at all tested concentrations. All Salmonella serotypes, except S. Diarizonae, showed excellent binding efficiency with purified anti-Salmonella pAbs. Moreover, the purified anti-Salmonella pAbs showed excellent specificity against all non-Salmonella strains. The anti-Salmonella pAbs immobilized on the gold biosensor demonstrated the successful capturing capability against Salmonella with a dose-response manner. Therefore, the anti-Salmonella pAbs exhibited sufficient reactivity, specificity, as well as capturing capability against Salmonella to be considered as a bio-recognition element.

  • 본 연구의 목적은 한반도 연안해역에서 적조로부터 양식장을 보호하기 위한 연안환경모니터링시스템을 구축하는데 있다. 1995년 이후 적조의 발생은 한반도 남부 연안해역에서 빈번하게 발생하고 넓게 확산되었다. 따라서 적조연구의 가장 중요한 점은 유해성적조의 발생 초기단계에 적조를 탐지하는 것이다. 지리정보시스템 기술은 적조발생의 공간적, 시간적 변화를 분석하는데 필요한 적조모니터링 시스템을 만드는데 적합하다. 유해성 적조 분석시스템 프로그램은 매일 매일의 적조 분포범위, 적조 생물 종, 적조 밀도 등을 제공해준다. 우리는 유해성 적조분석프로그램을 이용하여 적조로부터 양식장의 보호지역을 찾을 수 있다. 적조의 공간분석은 현장조사에 의한 참조자료를 바탕으로 수행되었다.

Development of On-line Monitoring Techniques for Fumaric Acid and Succinic Acid by Flow Injection Analysis (흐름주입분석기술에 의한 푸마르산과 숙신산의 모니터링 기술 개발)

  • On-line monitoring techniques for fumaric acid and succinic acid were developed by flow injection analysis (FIA). For the determination of fumaric acid, two enzymes, fumarase and malic dehydrogenase were immobilized on VA-epoxy Biosynth E3-carrier and integrated into a FIA-system with a fluorescence detector. For the analysis of succinic acid, isocitrate lyase and isocitrate dehydrogenase were also immobilized on VA-epoxy polymer support and used in a FIA system. The immobilized enzymes in two FIA systems were characterized systematically, e.g. optimum pH and temperature, inhibitory effects etc. Two FIA systems were also used to on-line monitor the concentrations of fumaric acid and succinic acid in biotechnological processes. Good agreement between on-line monitored data and off-line data measured by HPLC showed extensive application of the FIA systems in bioprocesses.

  • The outbreaks of red tide were sporadic in the South Sea until 1994, but have become frequent and widespread in whole coastal waters of the South Sea and East Sea since 1995 For monitoring of red tide, many kinds of techniques such as remote sensing, GIS and fuzzy model system have been developed and applied. The purpose of this paper is to develop red tide monitoring system for collection of red tide data and biological-oceanography parameters using wireless sensor network. The wireless sensor network has been noticed as a core technology in order to realize ubiquitous computing. In this paper, we design red tide database using wireless sensor network and suggest red tide monitoring software and web-service for user and biological-oceanographer.

  • Now global climate change is changing environmental factors, such as temperature and precipitation, which have a great effect on crop yields. Accordingly, crop yield forecast is becoming more important to global food supplies and sustainable development of rural areas. Worldwide, many countries, such as USA, China, Canada, and institutions, such as FAO, USDA, NASA, maintain the cooperative relationship to operate the crop monitoring system at both the national and global scale. This paper aims to investigate the current developments of crop monitoring systems in terms of information level, remotely-sensed data, and biophysical parameters, and to propose the direction of the advanced corp monitoring system based on remote sensing.

  • Korean government has planned to increase the productivity of horticultural crops and to expand supply smart greenhouse for energy saving by modernization of horticultural facilities based on ICT in policy. However, the diversity and linkages of monitoring and control are significantly insufficient in the agricultural sector in the current situation. Therefore, development of a service system with smart-farm based on the internet of things(IoT) for intelligent systemization of all the process of agricultural production through remote control using complex algorithm for diverse monitoring and control is required. In this study, domestic and international research trend related to ICT-based horticultural facilities was briefly introduced and limits were analyzed in the domestic application of the advanced technology. Finally, future core technologies feasible to graft in agricultural field were reviewed.

Background Modeling for Object Detection from Tidal Flat Images (갯벌 영상에서 객체 검출을 위한 배경 모델링)

  • Tidal flats provide important indicators that inform the condition of the environment, so we need to monitor them systematically. Currently, the projects to monitor tidal flats by periodically observing the creatures in tidal flats are underway. Still, it is done in a way that people observe directly, so it is not systematic and efficient. In this paper, we propose a background modeling method for tidal flat images that can be applied to a system that automatically monitors creatures living in tidal flats using sensor network technology. The application of sensor network technology makes it difficult to collect enough images due to the limitation of transmission capacity. Therefore, in this paper, we propose a method to effectively model the background and generate foreground maps by reflecting the characteristics of tidal flat images in the situation where the number of images to be used for analysis is small. Experimental results show that the proposed method models the background of a tidal flat image easily and accurately.

In this study we developed recombinant E. coli system for the high productivity of ALA and for the on-line monitoring of fusion protein (EGFP::ALAS) by a fluorescence sensor.

  • A recycling reactor system operated under sequential anoxic and oxic conditions for the swine wastewater has been developed, in which piggery slurry is fermentatively and aerobically treated and then part of the effluent recycled to the pigsty. This system significantly removes offensive smells (at both pigsty and treatment plant), BOD and other loads, and appears to be costeffective for the small-scale farms. The most dominant heterotrophs were Alcaligenes faecalis, Brevundimonas diminuta and Streptococcus sp. in order while lactic acid bacteria were dominantly observed in the anoxic tank. We propose a novel monitoring system for a recycling piggery slurry treatment system through neural networks. Here we tried to model treatment process for each tank(influent, fermentation, aeration, first sedimentation and fourth sedimentation tanks) in the system based on population densities of heterotrophic and lactic acid bacteria. Principle component analysis(PCA) was first applied to identify a relation between input(microbial densities and parameters for the treatment such as population densities of heterotrophic and lactic acid bacteria, suspended solids (SS), COD, $NH_3-N$, ortho-P, and total-P) and output, and then multilayer neural networks were employed to model the treatment process for each tank. PCA filtration of input data as microbial densities was found to facilitate the modeling procedure for the system monitoring even with a relatively lower number of input. Neural networks independently trained for each treatment tank and their subsequent combinatorial data analysis allowed a successful prediction of the treatment system for at least two days.
