Physiological Functionality of Various Extracts from Danmemil and Legumes (단메밀과 콩 추출물들의 생리 기능성)

  • Physiological functionalities of various extracts from Danmemil and legumes were determined and its optimal extraction conditions were also investigated. Angiotensin I-converting enzyme (ACE) inhibitory activity and tyrosinase inhibitory activity of Danmemil were higher in water extracts (53%, 58%) than those of ethanol extracts. However, its electron-donating ability was the highest in ethanol extracts (72%). ACE inhibitory activity and electron-donating ability of Black bean No. 1 and Taekwangkong(one of bean) were higher in water extracts than those of ethanol extracts, whereas SOD-like activity was the highest in ethanol extracts. ACE inhibitor and tyrosinase inhibitor of Danmemil were maximally extracted when it were treated with 20 times of distilled water at 35$^{\circ}C$ for 24 h and 36 h, respectively. Its electron donating compound was maximally extracted by treatment of 50$^{\circ}C$ for 18 h. ACE inhibitor of Black bean No. 1 was extracted maximally when it was treated with distilled water (1 :20) at 20$^{\circ}C$ for 24 h, whereas the other functional compounds were maximally extracted at 20$^{\circ}C$ for 18 h.

Changes in Physiological Activities of Scutellariae baicalensis by Heating (열처리에 의한 황금(Scutellaria baicalensis) 추출물의 생리활성 변화)

  • To elucidate the changes in physiological activities of Scutellariae baicalensis by heating, composition analysis with HPLC and evaluation of physiological activities of extracts were conducted. HPLC analysis of CW(water extract at room temperature) and CHW(heat treated CW) indicated that two distinctive peaks in the HPLC chromatogram were disappeared after boiling and additional peak was newly observed. This observation suggested that certain chemical changed in the extract occurred by heat treatment. In the estimation of physiological activities, CW showed higher ACE inhibiting activity than CHW. In the case of cytotoxic activity against cancer cells, CW exhibited higher activity than CHW against CCL-185. On the other hand, CHW showed higher activity than CW against HeLa, SNU-601 and HepG2. In addition, CHW showed the inhibitory effect on melanogenesis (80% inhibition) while CW did not show any activity.

  • Water extracts from Polygonum aviculare and Salix koreansis markedly inhibited the germination of lettuce and rice seeds, indicating the presence of biologically active substances. The biochemical substances such as salicylic and+vanillic acid, tannic acid + gallic acid, p-coumaric acid, p-cressol, sinapic acid and catechol etc. belonging to phenolic compounds were detected in the cultured cells, suggesting that the secondary metabolites can be synthesized in plant cell and tissue culture. In addition, fatty acid like linolenic acid and organic acid such as oxalic acid were presented in the highest amount, 3.7 mg/g and 14.288 mg/g, respectively, which seem to be related to exhibiting phytotoxicity of P. aviculare. Petroleum ether extract exhibited another potential relating to inhibitory effect which needs further investigation. Calli from two plant sources were easily introduced by uses of 1.0 mg/l of 2.4-D and 0.1 to 0.2 mg/l of BAP in MS basal medium which can be implemented for a large scale production through cell culture.

  • We investigated the immunological function of cheese whey protein concentrate (CWPC), which is a by-product of cheese production, using mitogenic activity in murine splenocytes as an index. A fraction isolated by gel filtration and anion exchange chromatography of CWPC showed high mitogenic activity, comparable to the activity of lipopolysaccharide (LPS). The fraction was detected as a single band on SDS-PAGE. It contained calcium, inorganic phosphorus, and carbo-hydrate, indicating the active component to be a glycophosphopeptide (GPP) Since pronase digestion of GPP did not reduce its mitogenic activity, carbohydrate rather than peptide may be important in the activity, When applied on an anti-${\beta}$-caseinophosphopeptide (${\beta}$-CPP ) antibody affinity column, the GPP was separated into two components, one with affinity to ${\beta}$-CPP and the other without such affinity. Both the components contained N-linked oligosaccharide chains and had the mitogenic activity. These results demonstrate that cheese whey contains a GPP having strong mitogenic activity

Phenolic Compounds of Ligustrum japonicum Leaves (광나무 잎의 페놀성 화합물)

  • This study was designed to investigate the possible utilization of Ligustrum japonicum leaves as a source of functional ingredients. Contents of total phenolic compounds and condensed tannin were $0.89{\sim}1.53%$ and $0.10{\sim}0.13%$, respectively. The major flavonoid compounds in the leaves of L. japonicum were luteolin, apigenin and their glycosides. Tyrosol, t-cinnamic acid, p-hydroxybenzoic acid, vanillic acid, shikimic acid and protocatecuic acid were detected in free phenolic acid, while tyrosol, t-cinnamic acid, ferulic acid, esculetin, caffeic acid, p-coumaric acid and hydroxytyrosol were detected in esterified phenolic acid. The insoluble phenolic acid contained tyrosol, t-cinnamic and p-caoumaric acid.

  • Ninty three species of domestic weeds were collected and screened for antimicrobial, antitumor, antioxidant and herbicidal activities. Among them, few showed antifungal activities. Cuscuta japonica showed inhibitory activity against Alternaria mali, Ambrosia artemisiifolia and Geranium sibiricum against Phytophthora capsici, Aster yomema and Aster pilosus against Phytophthora parasitica. Ambrosia artemisiifolia, Artemisia princeps, Artemisia capillaris, Ludwigia prostrata, Chrysanthemum zawadskii, Bidens frondosa, and Geranium sibiricum showed broad antibacterial activities. Carex chordorhiza, Artemisia capillaris, Persicaria nodosa, Senecio koreanus, Pariicum bisulcatum, Geranium sibiricum showed antiblebbing activity on human chronic leukemia K562 cell, among them, Persicaria nodosa was the strongist. Angelica decursiva, Equisetum arvense, Cimicifuga heracleifolia, Persicaria nodosa, Geranium sibiricum, Oenothera odorata, Cyperus sanguinolentus showed antioxidant activities. Ludwigia prostrata and Peucedanum terebinthaceum showed strong herbicidal activities.

Biological Activities of Antheraea yamamai Silk Fibroin Hydrolysates (천잠견단백질 가수분해 분말의 생리활성)

  • Antheraea yamamai silk fibroin powder was prepared by treatment with HCl. Amino acid contents of the prepared Antheraea yamamai fibroin hydrolysates were composed of Gly, Ala, Glu, and so on. Biological activities of Antheraea yamamai silk fiborin powder were examined. It is showed about 75% of antioxidation activity compared to the reference ascorbic acid. And it lowered blood glucose level down from 550 mg/dl to 300 mg/dl by serving with wild silk protein.

Isolation and Characterization of Bio-active Materials from Prickly Lettuce (Lactuca serriola) (가시상추 유래 생리활성물질의 분리 및 특성 규명)

  • The bio-active materials were isolated from prickly lettuce (Lactuca serriola) by using several extraction solvents. The contents of general compounds, vitamins, polyphenols and flavonoids of prickly lettuce were analysed. In addition, nitrite scavenging activity, anti-oxidative activity and anti-microbial activity of methanol extracts were measured. The ethylacetate (EtOAc) fraction of methanol extracts of prickly lettuce showed the best on the nitrite scavenging activity, anti-oxidant activity and anti-microbial activity. And also, the contents of polyphenols and flavonoids were the highest among all fractions. The subfraction 2 (EtOAc:MeOH=3:1) of EtOAc fraction showed the strongest anti-oxidative activity among 5 subfractions. The subfraction 2 was identified as ${\rho}$-coumaric acid and caffeic acid by GC-MS. The anti-microbial activity of each solvent fraction from prickly lettuce was measured. The EtOAc fraction showed the strongest anti-microbial activity. Overall, these results may provide the basic data to understand the biological activities of bio-active materials derived from prickly lettuce.

Screening of Biologically Active Compounds from various weeds (다양한 잡초로부터 생리활성물질의 탐색)

  • To search for bioactive compounds from plant resources, 80% methanol extracts of 46 species of weeds were screened for their activities of antimicrobial, antioxidative, antiblebing, antitumor and herbicidal. Among extracts tested, some showed activities at the concentration of $50\;to\;100\;{\mu}g/ml$. Phryma leptostachya var. asiatica, Aster ageratoides, Centipeda minima, Cirsium pendulum, Lythrum anceps showed antibacterial activity. Penthorum chinense, Lindernia procumbens, Aster ageratoides, Dianthus superbus var. longicalycinus showed antiblebing activity. Phyma leptostachya var. asiatica, Juncus effusus var. decipiens, Lindernia procumbens, Aster ageratoides, Dianthus superbus var. longicalycinus, Viscum album var. coloratum showed antitumor activity. Juncus effusus var. decipiens, Hypericum ascyron, Juncus papillosus, Inula britannicar var. chinensis, Scirpus wichurae, Hypericum laxum showed antioxidant activity.
