The Effect of the UV irridation on the Cornea (UV조사가 각막에 미치는 영향)

  • This study was performed on the mouse to estimate the effect of UV radiation on the cornea in UV clean bench by LM & SEM. The results are as follows In the control groups, The cornea tissue have relatively compact and each layer have well identify, and the thick of cornea have constant. In the increasing experimental time, the experimental result have very different. The early experimental groups results have not severely degeneration. But, some substrate layer have a swelling and some epithelial tissue have not normal shape. The middle experimental groups results have very swelling of the stroma, the vacoule of some region the condensation of the epithelium, and the irregular arrangement of the endothelium. The last experimental groups results have shirinking of cornea tissue, the swelling and vacoule of the end endothelium, the partially disruption of epithelium, the irregular thick of the corneal tissue.

  • The venomous apparatus of the black widow spider, Latrodectus mactans, is composed of chelicera and paired venom glands in the cephalothorax. Each glands is surrounded by a thin adventitia and striated muscular bundles resting on a basal membrane. Along the musculature neuromuscular synaptic contacts are formed by a motor axon and the muscle fibers. The secretory epithelium, which made up of simple and long columnar cells with extensive finger-like processes, creates a simple acinar gland. The secretory surfac is increased by a sort of fringes extended from the basal membrane into the gland lumen, and the luminal surface of the epithelium is marked by the presence of closely spaced microvilli. The venoms of the black widow spider are produced from two types of secretory granules within the epithelial cells. During the secretory phase, these granules are transformed into droplets and suffering a condensation. Finally the secretory products are released by the apocrine secretion. After the gland is emptied, the basal epithelial cells present a high proliferative process and regenerate the columnar epithlial cells.

Abnormality of Reproduction and Organ Structure of the Oily Bitterling, Acheilognathus koreensis from Isa Stream (이사천에서 채집된 칼납자루, Acheilognathus koreensis의 생식과 기관계 구조 이상)

  • Reproduction and organ system of the oily bitterling, Acheilognathus koreensis in Isa stream were investigated by means of histological methods. Sex ratio was observed 1 : 1 in female to male. The gonadosomatic index (GSI) of the female showed two peak in December and April. The ovipositor length index (OLI) represented irregular change. From the histological analysis of the organ system, epidermal atrophy and hyperplasia of pigment cell were observed in the skin. Epithelial layer lifting of the lamella and bifurcation of the filament were observed in the gill. Also histological changes as congestion, cytoplasmic degeneration of hepatic cell, glomerular dilatation, hyaline droplet degeneration of renal tubule epithelium and pycnosis of interstitial cell were identified in the liver and kidney, respectively.

Expression of Id-1 Gene in Mouse Uterus (생쥐 자궁에서의 Id-1 유전자의 발현)

  • 연구 목적: Microarray data에 의해 밝혀진 생쥐자궁에서의 Id 유전자의 hormonal effect와 implantation process 동안의 관계를 조사하고자 실험을 수행하였다. 연구재료 및 방법: 난소절제한 생쥐에 estrogen을 주사하고 6시간, 12시간이 지난 후 자궁을 적출하여 두 가지 방법으로 sample을 준비하였다. 먼저 자궁전체의 RNA를 추출하여 실험하거나 laser capture microdissection (LCM) 방법으로 자궁내막상피세포, 자궁기질세포, 자궁근육층으로 분리하여 RNA를 추출하고 semi-quantative RT-PCR을 수행하여 Id 유전자의 발현을 조사하였다. 임신 4.5일째 생쥐에 Chicago blue dye를 주사하여 착상부위와 비착상부위를 분리하고 RNA를 추출하여 Id 유전자의 발현을 semiquantitative RT-PCR 방법으로 실험하였다. 결 과: Estrogen을 처리한 난소절제된 생쥐 자궁에서의 cDNA microarray 자료에서 Id-1 mRNA는 점진적으로 두 배 이상 증가하였고 Id-2 mNRA는 반대로 시간이 지날수록 두 배 이상 감소하였다. Microarray 자료를 재확인하기 위해 semi-quantitative RT-PCR을 이용하여 실험하였고, 그 결과 Id-1 유전자는 estrogen 처리 6시간까지는 큰 변화가 없었으나 12시간에서는 4배 이상의 높은 발현을 보였으며, Id-2 mRNA의 발현은 estrogen 처리 6시간과 12시간 모두에서 대조군에 비해 4배 가량 감소하였다. 이 실험군을 LCM을 이용하여 자궁내막상피세포, 자궁기질세포, 자궁근육층을 각각 분리하여 실험한 결과 estrogen 처리군에서 Id-1의 발현은 자궁내막상피세포에서만 높은 발현을 보였으며, estrogen 처리 6시간과 12시간에는 큰 차이를 보이지 않았다. 그러나, Id-2 mRNA는 자궁내막상피세포에서 estrogen 처리 6, 12시간 모두에서 높은 발현을 보였고, 근육세포층에서는 estrogen 처리 6시간에서는 변화가 거의 없었으나 12시간에는 현저하게 증가하였다. 단, 자궁기질세포에서는 대조군에 비해 estrogen 6, 12시간에서 Id-2 mRNA의 발현이 감소하였다. 임신한 생쥐 자궁의 착상부위에서는 Id-1 mNRA의 발현은 비착상부 위보다 월등하게 높은 증가를 보였다. 결 론: 난소절제 생쥐를 이용한 실험에서 Id-1, -3는 estrogen에 의해 발현이 증가하고, Id-2는 발현이 감소하였다. LCM을 이용한 실험에서는 Id-2는 이와는 달리 부위별 발현양상은 다르게 나타났지만 이는 넓은 부위를 차지하는 자궁기질에서의 발현감소가 전체적인 Id-2의 발현양상으로 나타난 것으로 추측된다. 착상부위에서의 Id의 발현은 Id-1만이 유일하게 월등한 증가를 보였다. 위의 결과를 종합해 볼 때 생쥐 자궁에서 Id 유전자는 estrogen에 의해 조절되며 직, 간접적으로 착상시기에 다양한 작용을 할 것으로 사료된다.

Survival and Histological Change of Integumentary System of the Juvenile Sea Cucumber, Apostichopus japonicus Exposed to Various Salinity Concentrations (염분 농도에 따른 어린 돌기해삼, Apostichopus japonicus의 생존 및 피부계의 조직학적 변화)

  • Juvenile sea cucumber, Apostichopus japonicus(wet weight $1.0{\pm}0.2g$, n=250) acclimated to environmental conditions($17^{\circ}C$, 33 psu) during 4 weeks for observation of survival and structural variation of integumentary system according to various salinity concentration. After that, it was exposed 0, 5, 10, 15, 20, 25, 30, 35, 40 psu in 96h, and recovered during 7 days. In this results, The 96-h lethal concentration(LC50) for sea cucumber was a salinity of 21.05 psu. Integumentary system of the sea cucumber exposed to below the salinity of 20 psu mainly observed thickness reduction of epidermal layer, nucleus condensation of epithelial cells, decreased of mucous cells and loose arrangement of connective tissue in dermal layer. Integumentary system of the sea cucumber exposed a salinity of 40 psu mainly observed nucleus hypertrophy of epithelial cells, increase of mucous cells and tight arrangement of connective tissue in dermal layer. Also, observed secretory cells showing the alcian blue(pH 2.5) positive. During 96-h exposure, this cells decreased after increase below the salinity of 20 psu but increased in the salinity of 40 psu compared with 25-35 psu.

Establishment of a Single Dose Radiation Model of Oral Mucositis in Mice (일회 방사선조사를 이용한 마우스 구강점막염 모델의 확립)

  • Purpose: Oral mucositis induced by radiotherapy to the head and neck area, is a common acute complication and is considered as the most severe symptom for cancer patients in the early stages of treatment. This study was proposed to establish the oral mucositis mouse model induced by a single dose of radiation for the facility of testing therapeutic candidates which can be used for the oral mucositis treatments. Materials and Methods: Fifty-five BALB/c mice were divided into four groups: control, 16 Gy, 18 Gy, and 20 Gy. Oral mucositis was induced by a single dose of radiation to the head and neck using 6 MV x-Ray from linear accelerator. After irradiation, body weight and physical abnormalities were checked daily. Tongue tissues from all groups were taken on days 1, 2, 3, 5, 7, 9, and 14, respectively and H&E staining was conducted to examine morphological changes. Results: Body weight dramatically decreased after day 5 in all irradiated mice. In the 16 Gy treatment group, body weight was recovered on day 14. The histology data showed that the thickness of the epithelial cell layer was decreased by the accumulated time after radiation treatment, up to day 9. Severe ulceration was revealed on day 9. Conclusion: A single dose of 16 Gy is sufficient dose to induce oral mucositis in Balb/C mice. Significant changes were observed in the Balb/C mice on days 7 and 9 after radiation. It is suggested that this mouse model might be a useful standard tool for studying oral mucositis induced by radiation.

Anatomy and Histology of the Olfactory Organ of Asian Swamp Eel Monopterus albus (드렁허리 Monopterus albus 후각기관의 해부 및 조직학적 특성 연구)

  • The anatomy and histology of the olfactory organ of Monopterus albus was investigated using stereo microscopy, light microscopy, and scanning electron microscopy. The external structure of the olfactory organ exhibited closed anterior and posterior nostrils parallel to the skin surface. The interior structure consisted of a pipe-like chamber, and lower and upper accessory nasal sacs. The olfactory chamber was composed of the sensory and non-sensory epithelium, and an unidentified organ. The sensory epithelium of the pseudostratified epithelial layer was composed of olfactory receptor neurons, supporting cells, basal cells, and lymphatic cells; and the non-sensory epithelium of the stratified squamous layer contained stratified epithelial cells and mucous cells with acidic mucopolysaccharides. The presence of intraepithelial blood capillaries and abundant dermal vascularization in the sensory epithelium of the olfactory chamber may provide strong histological evidence that respiration occurs through the olfactory epithelium.

A Study of Structure of the Sucker of Common Freshwater Goby (Rhinogobius brunneus) and Triden Goby (Tridentiger brevispinis) (밀어 (Rhinogobius brunneus)와 민물검정망둑 (Tridentiger brevispinis)의 흡반 구조에 관한 연구)

  • The structures of sucker of two Cobiidae; Common freshwater goby and Triden goby were observed by light and electron microscopy. Scanning electron microscopy revealed the characteristic narrow ridges and grooves on the apical portion of sucker of Common freshwater goby, and hexagonal structures similar to a honeycomb representing the intercellular junctional area on the middle and basal portions. Some ridges were present on the epithelial surface on the middle and basal portions. The openings of several mucus-secreting cells were present between main epithelial cells. Light and transmission electron microscopy revealed the core of the fin; soft rays with a surrounding dense collagen fiber layer. Some loosely arranged fibers (collagen fiber) radiated toward the surface epithelium. The surface epithelium was cuboidal or columnar in shape. Scanning electron microscopy revealed the coiled irregular ridges and grooves, which was less developed and had sparser distribution than in Common freshwater goby, on the apical portion of sucker of Triden goby. The middle and basal portions had honeycomb structures as in Common freshwater goby. Fewer mucoussecreting cells were present. Light and transmission electron microscopy showed the core of soft rays, dense collagen fiber layer, however, the radiating fibers observed in the Common freshwater goby was rarely present. The sucker was thinner because the epithelium is squamous or polygonal in shape and rare presence of the radiating fibers.

Histological Study of the Minute Tubercles on Larval Skin Surface of a Korean Endemic Bitterling, Acheilognathus koreensis (Pisces, Cyprinidae), with Its Larval Growth (칼납자루, Acheilognathus koreensis (어강, 잉어목) 자어의 표피돌기에 관한 조직학적 연구)

  • Morphology and distribution of the minute tubercles projected on the skin surface of larvae with its development was observed in the Korean bitterling, Acheilognathus koreensis, known as an endemic freshwater fish. The epidermis of the larvae consisted of a thin single layer, having smaller basophilic flat or round-flattened basal cells. In between the single cell layer, two or three layers were added and they consisted mainly of large epidermal cells just above basal cells. These large unicellular epidermal cells were mainly scale-shaped and rarely cone-shaped, and do not give any histochemical tests for mucosubstances. They were present in anterior region and most region of yolk sac. Whereas, vestigial epidermal cells were distributed in the body region and the caudal fin-fold region. These two kinds of epidermal cells, called minute tubercles, increased in number and height from Just to 8 days after hatching, but as the larvae develop gradually, they became to reduce. At 31days after hatching of free swimming stage and absolute absorption stage of the yolk sac, the minute tubercles did not exist on the whole skin of the larvae.

The Differentiation of the Olfactory Placode in Xenopus (Xenopus 후각원판의 분화)

  • Normal development of the olfactory placode was studied to describe the sequence of events involved in the development of the olfactory placode. It has been primarily concerned with the morphological differentiation of the sensory neurons, their initial growth, maturation patterns and the contacts of their axons with the primitive prosencephalic vesicle. The olfactory organ first appears at stage 23 as a paired thickening of the two ectodermal layers: the superficial non-nervous layer (NNL) and the inner nervous layer (NL). Receptor cells differentiate from the NL and the supporting cells develop from the NNL. After stage 26 the placodal cells begin to migrate toward the epithelial surface between the NNL cells and their apical processes reach the surface at stage 28. As the apical process reaches the epithelial surface, basal processes (presumptive axons) sprout from the base of the NL cells at stage 29/30. They penetrate the underlying telencephalon by stage 32. Sensory synaptic contacts first appear at stage 37/38. Some placodal cells remain at the olfactory epithelium as basal cells while other placodal cells differentiate into olfactory neurons. The results confirmed that neurons originate exclusively from the nervous layer of the ectoderm while supporting cells originate from the NNL layer. The results also indicate that the development of olfactory neuron is independent of information from the target ftssue.
