This study was carried out for the dietry safely based on the level of pesticide residues In 13 kinds of agricultural products consumed in Kyonggi-Do. Korea. From June to October 2000, sixteen organochlorine pesticides in 397 samples were analyzed by using GC/ECD and GC/MSD. According to the results, endosulfan were detected in 27(7.8%) samples and chlorothalonil were detected in 4(1.0%) samples. Detection ranges of endosulfan were 3.7437∼ 0.0488ppm for lettuce, 2.1902 ∼0.1423ppm for spinach, 2.4909 ∼0.0786ppm for mallow and 3.2333∼0.3997ppm for mustard leaf, respectively Chlorothalonil were 5.8097 for lettuce and 0.8962ppm for spinach. Consequently, six agricutural products were detected more than the maximum residue limits in Korea Endosulfan were detected In 5(1.8%) samples and cholrothalonil were detected in 1(0.3%) sample. Detection rates of endosulfan sulfate(45.9%) of agricultural products were more than $\alpha$-endosulfan(28.5% ) and $\beta$-endosulfan (23.9% ) .
Agrobacterium tumefaciens LBA 4404 harboring binary vector pBI 121 was used for genetic transformation of lettuce(Lactuca Sativa L.). Optimal shoot regeneration from cotyledon explants was obtained in MS medium supplemented with 0.1mg/L NAA and 1.0 mg/L 2ip. In this condition, cotyledon explants were cocultivated with A, tumefaciens for 2 days, and then transferred to selection medium supplemented with 50 mg/L kanamycin and 500 mg/L carbenicillin. These explants were subsequently subcultured every 2 weeks on shoot induction medium. PCR analysis indicated that the GUS gene was stably integrated into the nuclear genome of lettuce. Histochemical analysis based on the enzymatic activity of the CUS protein showed that GUS activity was associated with vascular tissue in leaves and roots. Progenies of Ro plants demonstrated a linked monogenic segregation for GUS gene.

Copper and Zinc are essential trace elements for all living organisms. When presenting in excess amount in soils, however they can be toxic to plants. In order to examine the transfer of Cu and Zn from soils to plants and to predict their contents in plants using soil factors, we investigated total and mobile contents of Cu and Zn in soils and their uptake by lettuce (Lactuca sativa L.) in plastic film houses. Total Cu and Zn contents in soils were $17.5{\sim}65.9mg\;kg^{-1}$ (mean: $39.3mg\;kg^{-1}$) and $63.2{\sim}200mg\;kg^{-1}$ (mean: $137mg\;kg^{-1}$), respectively. Mobile Cu and Zn contents in soils were $(0.04){\sim}0.55mg\;kg^{-1}$ (mean: $0.18mg\;kg^{-1}$) and $(0.05){\sim}2.62mg\;kg^{-1}$ (mean: $0.47mg\;kg^{-1}$), respectively. Soil pH ranged from 5.4 to 7.3 and OM from 24.1 to $59.9g\;kg^{-1}$. Mean Cu contents in leaves and roots of lettuce were 9.20 and $17.2mg\;kg^{-1}$, respectively which showed that Cu was accumulated mainly in root parts of lettuce and not easily transported to leaves. In contrast, Zn was fairly evenly distributed in leaves and roots with mean values of 54.5 and $56.7mg\;kg^{-1}$, indicating relative high mobility of Zn in lettuce. Transfer factors of Cu and Zn from soil total contents to roots and leaves of lettuce ($TFS_tR$ and $TFS_tL$) were between 0.1 and 1, while transfer factors from soil mobile contents to roots and leaves ($TFS_mR$ and $TFS_mL$) were between 10 and 1000. Transfer factors of Zn were higher than those of Cu, showing Zn was more easily absorbed by plants than Cu. Cu and Zn uptake was stronger influenced by soil pH and mobile contents than total contents and OM and could be significantly described by multiple regression equations including soil pH and soil mobile contents as variables.

This study was conducted to compare the quality of baby leaves grown under several temperature conditions and the storage properties of MA storage for romaine lettuce. It was grown for 5 weeks under an artificial light source (200 µmol·m-2·s-1) in a chamber at 21℃, 28℃, and 35℃. The growth and quality of red romaine lettuce that grown in different temperatures were investigated at the end of cultivation, and the oxygen, carbon dioxide, and ethylene concentrations in the 20,000 cc OTR film and perforated film packed with lettuces were measured for 36 and 12 days, respectively. The red romaine lettuce baby leaf was examined for color, chlorophyll, and visual quality at the end of storage. The maximum quantum yield of baby leaf grown in different temperatures at 7days before the harvest was higher at 21℃ and 28℃ growth temperature treatments. On harvest day, the leaf length measured was longest at 28℃, and the leaf width was wider at 21℃ and 28℃, and the number of leaves was similar to 5-6 at all cultivation temperatures. Leaf weight, root weight, and dry weight were found to be higher at 21℃, and tended to decrease as the cultivation temperature increased. The concentration of ethylene in the film of the MA storage treatments was maintained at 1~2 µL·L-1 until the end of storage in all treatments regardless of the cultivation temperature. Oxygen concentration in the MA treatment used 20,000 OTR film was maintained at around 19.5%, and carbon dioxide concentration around 1% that was satisfied the CA conditions. Both Hunter a* and b* values were generally higher in the MA storage treatment at the end of storage day. The chlorophyll content was decreased as the cultivation temperature increased, and was lower in the MA storage treatment than in the perforated film treatment. Visual quality was 3 points or higher in the MA storage treatment at 21℃ growth treatment, and it was maintained marketability. As the above results, the growth of baby leaves of romaine lettuce was the best at 21℃ treatment, and the lower the cultivation temperature, the longer the shelf life. And it was possible to extend the shelf life by 3 times by showing excellent visual quality at the MA storage treatment that satisfies the carbon dioxide concentration of CA condition until the end of storage day.

This study was evaluated the effect of attractant or repellent plants for establishing push-pull strategy for insect pest management of organic Chinese cabbage cultivation. The attractant or repellent effect of Ten plants, marigold, rye, Chinese chive, lettuce, chicory, Nongwoo-chicory, crown daisy, Treviso, green leaf mustard, and red leaf mustard were evaluated against diamondback moth, striped flea beetle, aphids, brown-winged stink bug and cabbage stink bug in organic Chinese cabbage field in Seosan in 2012. Of the ten repellent or attractant plants, rye and chicory attracted significantly brown-winged stink bug and cabbage stink bug, respectively. Aphids ate attracted significantly to the rye rather than the other plants. Leaf mustard was attracted cabbage stink bug and brown-winged stink bug in order. Cabbage stink bug was attracted by only a combination of lettuce and Chinese chive. Diamondback moth, striped flea beetle, aphid, brown-winged stink bug and cabbage stink bug were significantly repelled by single or combined cultivation of crown daisies, Treviso, and leaf mustard in organic Chinese cabbage cultivation. The results above-mentioned indicate that selected insect-attractant and -repellent plants can be used as tools for integrated pest managements of Chinese cabbage.
This study was investigate the occurrence of bottom rot caused by Rhizoctonia solani at the crisphead lettuce fields in Uiryeong-gun, Gyeongsangnam-do from November to December in 2003. Incidence of bottom rot on crisphead lettuce was up to 5.3% at the six plastic houses. A total of 30 isolates of R. solani were obtained from diseased leaves of plants and were tested by artificial inoculation to the host. Among them, PY-1 isolate was selected showing highly virulent on the whole plant and was identified as R. solani AG1 (IB) based on the anastomosis test, morphological and cultural characteristics. Symptoms of bottom rot by PY-1 isolate produced small dark brown, depressed and elliptical spots on the lower part of leaves in the early stage as same as at the fields, were enlarged onto the upper part of leaves later, and the infected plant wilted and ultimately died in the end. For the pathogenicity test, triturated mycelia-inoculum (A$_{550}$=1.0) of PY-1 isolate was selected the most effective inoculum showing disease incidence of 51.1% for the mycelial inoculation at pot assay. Otherwise, WSRP media-inoculum (wheat brane : sawdust : rice brane : PDB media=30 g : 10 g : 10 g : 100 ml, w/w/w/v) of PY-1 isolate was effectual inoculum showing disease incidence of 61.6% for soil inoculation at the plastic house. Also, in selection of density and amount of inoculum, most suitable density of triturated mycelia-inoculum and amount of WSRP media- inoculum were determined as $A_{550}$=1.0 and 40 ml, respectively. This is the first report on the pathogenicity test using by WSRP media-inoculum of R. solani PY-1 isolate for the bottom rot of crisphead lettuce.

A new red leaf lettuce (Lactuca sativa L.) cultivar, "Gopungjeokchukmyeon" which has late bolting and good red expression leaf color was developed from a cross between "Andongjeokchukmyeon" (red leaf color and late bolting) and "Dukseomjeokchukmyeon" (high yield). The cross and selection for advanced lines had been done by the pedigree method during 2000-2006. The advanced lines were evaluated for yield and adaptability at several locations in Korea (Gangwon, Kyunggi, Chungbuk, Kyungnam, Jeonnam, Jeonbuk, and Jeju) from 2007 to 2008. The "Gopungjeokchukmyeon" has gray seed color and traverse elliptic leaves. Compared to "Dukseomjeokchukmyeon", marketable yield of "Gopungjeokchukmyeon" was higher by 17% (at 330 g per plant) and Gopungjeokchukmyeon has particularly improved expression of red leaf color in high temperature cultivation in the field. The shelf-life of "Gopungjeokchukmyeon" was three weeks longer than "Dukseomjeokchukmyeon" at $4^{\circ}C$. The anthocyanin and sugar contents of "Gopungjeokchukmyeon" were higher than those of "Dukseomjeokchukmyeon" with 29.4 mg/100 g and 4.8 brix, respectively. The content anthocynin of "Gopungjeokchukmyeon" is about 10 times higher than those of "seomjeokchukmyeon". Furthermore, its taste is better, more crispy, and sweeter than those of "Dukseomjeokchukmyeon", This new cultivar "Gopungjeokchukmyeon" can be harvested all year around even summer cultivation.
This study developed predictive growth models of Salmonella enterica Serovar Typhimurium on lettuce washed with chlorine (100~300 ppm) and ultrasound (US, 37 kHz, 380 W) treatment and stored at different temperatures ($10{\sim}25^{\circ}C$) using a polynomial equation. The primary model of specific growth rate (SGR) and lag time (LT) showed a good fit ($R^2{\geq}0.92$) with a Gompertz equation. A secondary model was obtained using a quadratic polynomial equation. The appropriateness of the secondary SGR and LT model was verified by coefficient of determination ($R^2=0.98{\sim}0.99$ for internal validation, 0.97~0.98 for external validation), mean square error (MSE=-0.0071~0.0057 for internal validation, -0.0118~0.0176 for external validation), bias factor ($B_f=0.9918{\sim}1.0066$ for internal validation, 0.9865~1.0205 for external validation), and accuracy factor ($A_f=0.9935{\sim}1.0082$ for internal validation, 0.9799~1.0137 for external validation). The newly developed models for S. Typhimurium could be incorporated into a tertiary modeling program to predict the growth of S. Typhimurium as a function of combined chlorine and US during the storage. These new models may also be useful to predict potential S. Typhimurium growth on lettuce, which is important for food safety purposes during the overall supply chain of lettuce from farm to table. Finally, the models may offer reliable and useful information of growth kinetics for the quantification microbial risk assessment of S. Typhimurium on washed lettuce.
Absorption of Cadmium(Cd) and Arsenic(As) by lettuce following to the change of polyamine content in lettuce were examined to find the effect of these elements on lettuce growth and on the basic resistance mechanism. As the contents of Cd and As were increased in soil, the contents of these elements were significantly increased in lettuce but lettuce growth was decreased. Sequential fractionation experiment of the Cd and As treated soil was shown that exchangeable, dilute acid extractable, and organically bound form, which were more mobile and phyto-available, of Cd were increased by 5.6, 42.9, and 56.7% during a growing season whereas 17.6, 25.0, and 24.1% were increased in case of As, respectively. Specially, the Cd content of leaves and roots in lettuce was positively correlated with exchangeable, dilute acid extractable, and residual form of Cd in the treated soil. However, the As content of roots in lettuce was positively correlated with all chemical forms of As in the treated soil. The contents of putrescine and cadaverine, a kind of polyamine, were also increased in lettuce as both elements were increased in soil.
Heavy metals exist in soils in various chemical forms including free metal ions and organo-metal complexes. The ratio of free metal ions has been known to be highly associated with the plant absorption of heavy metals. This study aims to understand the effect of free ions and organo-metal complexes on the absorption of lead (Pb) and cadmium (Cd) by plants. For this, lettuce grown in a hydroponic system for 28 days was consequently grown another 48 hours using Pb and Cd solutions. The ratios of free ion to organo-metal complexes in the solutions were adjusted at 100:0, 90:10, 70:30, 60:40 by four different organic acids (citric, oxalic, acetic, and humic acid). After that, the concentration of Pb and Cd in lettuce were analyzed. The Pb and Cd absorption by lettuce was more relied on the types of organic acids treated and the type of metals rather than the ratio of free metal ions. For example, citric acid increased the Pb absorption while it decreased the Cd absorption by lettuce. There was no significant relationship between free metal ion ratios and both Pb and Cd uptake by lettuce. It could be explained that citric acid, a relatively higher molecular weight organic acid, has higher ion binding capacity, so it forms organo-Pb complex easily due to the higher affinity of Pb on the binding site in comparison with Cd. Consequently, this complexation would assist Pb uptake by lettuce.
