An Analysis Microbiological Contamination on the Facilities and Utilities of Japanese Restaurants (일식 레스토랑 설비와 조리기구의 미생물 오염 실태 분석)

  • The microbiological examinations was conducted for the hygienic evaluation on ten Japanese restaurants during summer season in Gwangju, Korea. Total two hundreds swabbed samples using sponge were collected from the surface of facilities and utensils at restaurants. The number of total microorganism, coliform, E. coli, Staphylococcus, Salmonella, and Listeria were measured. The results demonstrated that most swabbed samples were contaminated with microorganisms and coliforms. The degree of contamination depended on the sample sites. The severely contaminated sites were floor, trench, and working table for the fish and total counts of those samples were over $10^4$ CFU/100 $cm^2$. The coliforms were detected on the floor, trench, and wood cutting board over $10^3$ CFU/100 $cm^2$. Moreover, coliforms was detected on the towel, dish, and working place for fish and vegetables. The E. coli was detected on the floor and trench of several restaurants. The Staphylococcus spp. was detected on the cutting board, towel, floor and trench of several restaurants. The Salmonella spp. and Listeria spp. were not detected all over the samples. From those results showed that sanitation management and disinfection were required on the Japanese restaurants.

Analysis of Microbiological Contamination in Ready-to-eat Foods (즉석 섭취 식품에 대한 미생물 오염 분석)

  • This study was carried out to examine microbiological contamination of ready-to-eat foods (kimbab, sushi, salad, sandwich, sashimi) and to prove hazard of ready-to-eat foods by microbiological analysis. Collection of 440 samples of ready- to-eat foods were obtained from department stores, discount stores, super-markets in Seoul, Gyeonggi, Chungcheong, Gyeongsang, Honam areas. Sushi showed the highest detection rate of micro organism with value of20.4%, and then detection rates of kimbab, sashimi and salad were 13.0%, 12.5% and 6.9%, respectively. S. aureus was the most detected microorganism with value of 6.6%, and then Coliform and L. monocytogenes were detected 2.7% and 2.3%.

Antibiotic Resistance Among Salmonella spp. Isolated from Feces of Patients with Acute Diarrhea in Gwangju Area, Korea, during 2000-2009 (2000년-2009년 광주 광역시 지역의 급성설사환자의 분변에서 분리한 살모넬라 균주에서의 항생제 내성률 조사)

  • Antibiotic susceptibility was examined for 596 Salmonella isolates from patients with acute gastroenteritis during 2000-2009 in Gwangju area in South Korea. Of 16 antibiotics tested, ampicillin resistance (43%) was the most commonly observed resistance among the 596 Salmonella sp. isolates, followed by tetracycline (35.9%), nalidixic acid (31.5%), and chloramphenicol (26.2%). Antibiotic resistance varied among serotypes: The highest resistance of S. Enteritidis and S. Typhimurium was to ampicillin (51.1%) and tetracycline (77.9%), respectively. A total of 89 resistance patterns were observed, and 26% (155/596) of Salmonella isolates were susceptible to all antibiotics tested in this study. About 21% (127/596) and 15% (87/596) of the isolates were resistant to one and two antibiotics, respectively. The rest of Salmonella isolates (227/596, 38%) were resistant to three or more antibiotic agents. The highest multi-drug resistance (MDR) was observed in serotype S. Paratyphi B (76.5%), followed by S. Typhimurium (58.2%), and S. Enteritidis (40.2%). The most common resistance pattern of MDR isolates was ampicillin-chloramphenicol-nalidixic acid-ticarcillin (36/227, 15.8%), most of which (35/36, 97.2%) were S. Enteritidis.

  • A very rare case of mycotic aneurysm in the descending thoracic aorta due to salmonellosis was treated in our hospital The patient was a 62 year-old male who 48 days before the operation was admitted to the depart,cent of internal medicine complaining of fever. nausla. vomiting, and loose stool. He was treated for 35 days and discharged. Three days after discharge, however, the patient was readmitted to the hospital comE긴ainin프 of $\ulcorner$i포ht uppe$\ulcorner$ 킥uadrant abdominal pain. Fever developed on the third hospiti린 day, and on the eighth hospital day, the patient complAined of back pain and epigastric pale. A simple chest x-ray showed evidence of hemothorax in the left plueral space, and therefore, computed tomography of the chest was done. The patient was diagnosed as a ruptured mycotic aneurysm of the descending thoracic aorta, and was transferred to our Department of Thoracic and CArdiovascular Surgery. The aneurysm and infected tissues was widely debrided, and the site was then patched with a Dacron graft. Salmonella choleraesuis was identified in the blood and aneurysm cultures, and antibiotics were administered for weeks according to the sensitivity of the organism. The patient experienced no complication thereafter and for the last three months since the operation, he has been leading a healthy and normal social life.

A Study on Bioserotype and Drug Resistance of Salmonella and Escherichia coli Isolated from Feces in Zoological Animals (동물원의 야생동물 분변에서 분리한 살모넬라균과 대장균의 생물형, 혈청형 및 약제내성에 관한 연구)

  • This study was undertaken the bioserotype and drug resistance of Salmonella and Escherichia coli isolated from feces for the prevention and treatment of salmonellosis and colibacillosis in zoological animals. The results obtained from the research were as follows 1. Salmonella were isolated 19, or 4.7% from 408 samples and E. coli were isolated 12, or 40.0% from 30 diarrheal samples. 2. The biotypes in 19 Salmonella were Subspecies 1. 3. The serogroups of Salmonella isolated were 47.4% in B group, 31.6% in C, 5.3% in D and 15.8% in other, and serotype of E. coli was 100% in 0127a. 4. The antibiotic resistance of Salmonella and E. coli isolated were 13, or 68.4% and 7, or 58.3% strains, respectively 5. The multiple resistant patterns of antibiotics in Salmonella were 2drugs- and 3 drugs-resistance 30.8%, respectively, and those in E. coli were mono drug-, 2 drugs- and 7 drugs-resistance 28.6%, respectively. 6. The transferred rate of resistance to recipients (E. coli ML 1410 NA$^{r}$ ) in Salmonella was 38.5%, but that in E. coli was 71.4%.

  • 본 연구는 자돈 설사의 원인이 되는 대장균, 살모넬라 등 그람음성균을 억제할 수 있는 유산균의 이용을 위해 선발된 균주의 과산화수소 대사를 구명하기 위해 수행하였다. 대장균 억제유산균중 superoxide dismutase 활성이 우수한 균주가 관찰되었으며, 과산화수소 이외에 그람음성균 억제에 유효한 bacteriocin-like compounds의 존재 유무, 대사과정에서 과산화수소 생성과 관련된 다른 효소 활성, superoxide dismutase 유전자 클로닝에 관한 연구가 진행중이다.

Effects of Ginseng Saponin Metabolites and Intestinal Health Active Ingredients of Vegetables Extracts and Fermented Lactic Acid Bacteria (비지터블 추출물 및 유산균 발효물의 진생사포닌 대사산물과 장건강 활성성분 효과)

  • In this study, 8 kinds of fruits and vegetables such as apples, pears and radishes were cut and hot water extracts and Steamed hot water extract from fruits and vegetables were prepared and used as experimental substrates. As a result of fermenting with 1% (W/V) red ginseng extract (W/V) and 8 types of lactic acid bacteria mixed starter added to the lactic acid bacteria fermented extract, the pattern and content of ginsenosides were almost unchanged in the fruit and vegetable extract group and the steam treatment group. However, in the lactic acid bacteria fermented group, the TLC pattern was changed according to the fermentation process and treatment, and the content of ginsenosides converted into Rg3(S) and Rg5 increased. No change in the number of lactic acid bacteria (cfu) was observed in all four types of fruit and vegetable extracts. The number of lactic acid bacteria CFU was slightly decreased in the four fermented groups of fruit and vegetable extracts, but the growth inhibitory effect of beneficial bacteria was not significant. The growth inhibitory effect of the three harmful bacteria was not affected by the growth of E. coli and Pseudomonas in the four fruit and vegetable extracts. However, the proliferation of Salmonella was inhibited, which was confirmed as the growth inhibitory effect of the fruit and vegetable extract regardless of whether the steamed hot water extract or red ginseng extract was added.

간장질환 치료제 G009의 개발 - 급성 및 유전독성 연구

  • 3) 결과 및 고찰 : 급성독성시험 : 대조군 및 G009투여군(최저 312.5mg/kg, 최고 5000mg/kg) 5용량에서 모두 사망예가 관찰되지 않았다 체중변화에 있어서도 대조군과 투여군 사이에 유의성 있는 차이는 없었다. 육안적 소견은 생존동물 모두에 약물에 기인한 내부장기의 이상이 관찰되지 않았다. 유전독성시험 : 마우스 골수세포를 이용한 소핵시험에서 약물 투여에 의한 어떠한 독성의 징후도 관찰되지 않았다. 포유류 배양세포를 이용한 염색체이상 시험에서 모든 농도에서 염색체 이상을 가진 세포의 출현빈도가 3% 이하로서 G009는 CHL세포에 대하여 염색체 이상유발성이 없었다. 살모넬라균을 이용한 복귀돌연변이 시험에서 투여군은 음성대조와 같은 정도 또는 그 이하의 복귀변이 집락수를 나타내었다.

Primers for Typing Salmonella spp. using Random Amplified Polymorphic DNA (RAPD) Analysis (Salmonella spp.의 RAPD Typing을 위한 Primer의 분리력 비교)

  • Random amplified polymorphic DNA (RAPD) analysis is based on the amplification of random DNA segment using a single arbitratrary primer. For typing Salmonella spp., polymorphic DNA patterns identified by this method can be used. To select the primers for RAPD typing Salmonella spp., the performances of 20 primers were compared by analyzing 16 Salmonella spp. reference strains. Reproducible electrophoresis patterns were obtained. Among the 20 primers tested, 4 primers (A, OPG04, OPG10, OPL03) showed better differentiation than the others. At the time discrimination index, band clarity, band number and difficulty of band scoring were considered. These primers will be useful for typing Salmonella spp. in the future. Curretly, we are under investigation for the RAPD typing of contaminated Slmonella spp. for the risk analysis of pork processing plant using the primers.

  • In order to find a solution to protect pigs from bacterial diarrhea and the nasty smell in stalls which are the most trouble, we composed a bio-formula with Stretococcus thermophilus, Bacillus amyloliquifaciens and Bacillu subtilis. The antagonistic microbe Bacillus amyloliquifaciens can control the growth of Salmonella typhimurium KCTC 1926, Escherichia coli O-157, Listeria and Staphylococcus. S. thermophilus from pig's stomach can live in gastric juice so it also control germs. They worked in its living cell state and its culture fluid. As a result of feeding with piglings, it showed effects of preventing diarrhea and increasing the weight.
