Bacteriocidal Effect of Chamical Sanitizers on Natural Microflora and Pathogenic Bacteria in Fish (화학적 살균소독제를 활용한 생선중 존재하는 자연균총 및 주요 식중독균 제어)

  • In this study, we evaluated bacteriocidal effect of CaO (scallop shell powder) for the reduction of microorganism in Scomber japonicus and Pseudosciaena ciean manchurica, and compared with main chemical sanitizers such as chlorine, ethanol, hydrogen peroxide. As a result, the effectiveness of CaO showed dramatic reduction rate for total aerobic bacteria, Escherichia coli and Vibrio parahaemolyticus. The reduction rates of total aerobic bacteria for Scomber japonicus and Pseudosciaena ciean manchurica showed $2.6{\times}10^3\;and\;3.2{\times}10^4$ respectively; Those of Escherichia Coli were $6.3{\times}10^3\;and\;1.2{\times}10^4$, those of Vibrio parahaemolyticus were $5.4{\times}10^4\;and\;5.6{\times}\2.6{\times}10^310^4$, respectively. According to this result, bacteriocidal effect of CaO was about $10^4$ and was similar to bacteriocidal effects of other three chemical sanitizers. According to these results, CaO can alternate the currently used chemical sanitizers due to its natural origin as well as the effectiveness for sterilization.

Evaluation of Efficacy and Development of Predictive Model of Sanitizers and Disinfectants on Reduction of Microorganisms on Food Contact Surfaces (스테인리스 스틸 식품기구 표면에 사용되는 주요 살균소독제의 살균력 평가 및 살균예측모델 개발)

  • This study was to evaluate the efficacy of sanitizer concentrations and treatment time against two major toad-borne pathogenic microorganisms such as Escherichia coli and Staphylococcus aureus on a stainless steel surface. As a result, stainless steel, treated with 100 ppm of chlorine showed reduction of E. coli(1.56, 1.49, 1.95 log cfu/25 $cm^2$) and S. aureus(0.49, 0.88, 1.27 log cfu/25 $cm^2$) after 0, 5 and 10 min, but none was not detected in treatment with 200 ppm. The population of E. coli(0.73, 0.90, 1.55 log cfu/25 $cm^2$) and S. aureus(0.37, 1.00, 1.45 log cfu/25 $cm^2$) reduced in 35.5% ethanol treated group, but none was not detected in treatment with 70%. The population was reduced E coli(0.28, 0.64, 1.07 cfu/25 $cm^2$) and S. aureus(0.53, 0.87, 0.99 log cfu/25 $cm^2$) by treatment with 45.5 ppm of hydrogen peroxide, but none was not detected in treatment with 91 ppm. Quarternary ammonium compound with 100 ppm was reduced E. coli(0.82, 1.62, 1.71 log cfu/25 $cm^2$) and S. aureus(0.46, 0.93, 1.38 log cfu/25 $cm^2$), but none was not detected in treatment with 200 ppm. Predictive models of sterilization for all 4 disinfectants were suitable to use with $r^2$ value of higher than 0.94. These models may be of use to food services and manufacture of safe products by controlling E. coli and S. aureus without the need for further detection of the organisms.

Effects of Fungicides on Inhibition of in Vitro Strawberry Pollen Germination (In Vitro에서 살균제의 딸기 화분발아 억제 효과)

  • Fungicide applications are required to prevent the strawberry from Botrytis fruit rot and powdery mildew that infect open strawberry flowers, however, their effects of fungicides on pollen germination of strawberry have been rarely documented, particularly those from recently developed active fungicidal ingredients. In this study we have evaluated the effects of 24 commercial fungicidal formulations and 6 organic materials on pollen germination in 3 strawberry cultivars using in vitro assays. Pollens from strawberry had higher germination rates on agar with sucrose of 18% and $25^{\circ}C$ than other tested conditions. Pollen germination rates of cvs. Seolhyang, Maehyang, and Kumhyang at 18% sucrose and $25^{\circ}C$ were 15.3, 18.4 and 30.7%, respectively. Pyraclostrobin, azoxystrobin, kresoxim-methyl, dichlofluanid, iminoctadine tris, and sulfur showed the strongest inhibitory efficacy with the germination rates of more than 93.8% compared to the no-fungicide control. Germination was not significantly affected by simeconazole and procymidone. This in vitro germination study may provide information useful for selecting fungicides in flowering stage to strawberry farmers.

Pathogenicity of Rhizoctonia Isolates from Southern Horticultural Area in Korea (남부지방에 발생하는 Rhizoctonia solani의 병원성)

  • Pathogenicity of nine Rhizoctonia solani isolates of different anastomosis groups (AG) on seed and hypocotyls of red pepper, cucumber, Chinese cabbage and radish varied considerably from nonvirulent to highly virulent. Rhizoctonia solani AG 1 was highly virulent on the above four plant species. AG 2 type 1 was highly virulent on radish and Chinese cabbage, moderately virulent on red pepper, and AG 2 type 2 was avirulent or weakly virulent except red pepper. R. solani AG 5 was moderately virulent on hosts tested. In general, virulence of the R. solani isolates to a given host varied among anastomosis groups, but not within anastomosis groups. Anastomosis groups lacked host specificity. The pathogenicity was stronger in steam-sterilized soil than in non-sterilized field soil, if the inoculated plants were closely related with orginal host from which the pathogen was isolated. On the other hand, pathogen was more virulent in non-sterilized field soil than in steam-sterilized soil, if the inoculated ones were not closely related. Generally, contrary to other soil-brone plant pathogenic fungi, Rhizoctonia isolates tended to be more virulent in non-sterilized field soil than in the same soil which had been steamed. A potential danger of building up propagules of R. solani in southern horticultural area are discussed in terms of cropping system.

대구 수정란의 질병 제어에 의한 부화율 향상 연구

  • 대구 인공종묘 생산 기술 개발의 일환으로 수정란에 기생하여 폐사를 유발하는 세균 및 세균 감염 경로를 파악함으로써 수정란의 생존율과 부화율 향상을 위하여 본 실험을 실시하였다. 세균 감염 경로를 파악하기 위하여 난소와 정소 자체 세균 감염 여부를 조사하였다. 일반배지 (BHIA)와 비브리오 선택배지 (TCBS)를 사용하여 세균검사를 실시하였다. 난소의 세균 검사는 생식소 내부를 일부 절개한 후 멸균 roop를 찔러 세균을 검사하였고, 정소는 채정하기 전 복부를 절개하여 멸균 roop로서 적출하여 배지에 도말한 후 배양하였다. 수정란 내 세균 수는 수정란을 각각 20개씩 샘플하여 난 외부에 기생하는 세균을 제거하기 위하여 난소독제로 이용되고 있는 benzalkonium 0.1%로 1분간 소독한 후 멸균 생리식염수로 3회 세척하여 호모게나이즈하여 검사하였다. 호모게나이즈한 액 중 100${\mu}\ell$를 pipetting하여 일반배지(BHIA)에 도말하여 인큐베이터에서 배양하였다. 난소와 정소 내에서 세균이 검출됨에 따라 수정 시 세균 감염을 억제하기 위하여 자외선 살균해수와 일반해수를 수정액으로 사용하여 발생율을 비교 시험하였다. 수정 시간은 1분으로 동일하게 적용하였으며, 수정 용기는 멸균 처리된 일회용 100$m\ell$ 플라스틱 용기를 사용하였다. 수정 후 과다한 정자를 제거하기 위한 세란은 1$\ell$ 멸균 비이커에서 5회 30분 가량 실시하여 정자를 제거하였고 멸균 봉으로 저어주면서 수정란의 점착력을 제거하였다. 수정란은 100$\mu\textrm{m}$ 그물망으로 수정란 유실을 방지한 플라스틱 용기에 수용하여 유효수량 270$\ell$ FRP 수조에 수용하여 3$\ell$/min 환수하였고, 수온은 자연수온 1$^{\circ}C$로 유지하였다. 발생율은 만능투영기로 3회 측정하였다. 수정 후 세균과 기생충에 의한 수정란의 폐사를 억제하기 위하여 수정 직후, 수정 후 1일, 2일째 oxytetracycline과 iodine 처리에 따른 발생율 변화를 조사하였다. 발생율은 만능투영기로 조사하였고, 시험구별로 3회 측정하였다. 경과 일수별로 약제 처리는 약제 미처리 수정란 중 정상적인 발생이 이루어지고 있는 것을 선별하여 조사하였다. 약제 처리에 따른 배체 발생 단계는 수정 후 1일째는 상실기, 2일째는 포배기였다. 수정란 및 대구 자어에서 분리된 V. splendidus 에 의한 폐사를 예방하기 위해 in vivo에서 oxytetracycline 외 5종의 항생제를 대상으로 96well plate에서 최고 농도 250ppm부터 2 fold로 단계 희석하여 $25^{\circ}C$에서 48시간 배양하여 MIC를 조사하였다. 세균 감염경로 파악을 위하여 난소, 정소 및 정자에서 세균을 분리한 결과 일반배지 및 비브리오 선택배지에서 모두 균이 검출되었고, 균 동정 결과 터봇 자어에서 검출된 것으로 보고된(Gatesoupe et al., 1999) V. splendidus로 나타났다. 수정액과 정자 및 미수정란의 세균 분리 결과 일반배지에서 3$\times$10/ml ~ 7$\times$10/ml로 균이 검출되었다. 수정액을 일반해수와 자외선 살균 해수를 사용하여 발생율을 비교한 결과 수정 후 3일째 발생율은 자외선 살균해수 72.3%, 일반해수 52.7%였으며, 수정 후 7일째 40.9%와 25.1%로 자외선 살균해수가 유의적으로 높게 나타났다. 수정 후 경과 일수별로 oxytetracycline과 Iodine을 처리한 결과 수정 직후 처리한 시험구는 7일째 19.8%와 18.9%로 대조구 23.1%와 유의적인 차이가 없는 것으로 나타났다. 수정 후 1일째 처리한 시험구는 54.5%와 56.8%, 수정후 2일째 처리구는 47.9%와 50.6%로 두 시험구 모두 대조구와 수정 직후 처리구보다 유의적으로 높게 나타났다. 수정란 및 대구 자어에서 분리된 V. splendidus 에 의한 폐사를 예방하기 위해 in vivo에서 항생제 종류별 MIC 조사 결과 oxytetracycline이 0.48ppm으로 가장 효과가 높은 것으로 나타났다.

Pasteurization Efficiency and Physico-chemical Changes of Soymilk HTST Pasteurized Using Microwaves (두유의 마이크로파 고온단시간 살균시 살균효과 및 이화학적 성분 변화)

  • This work was to determine the microbial and physico-chemical changes of HTST-pasteurized soymilk using microwave energy. Soymilk was HTST pasteurized$(at\;90^{\circ}C\;for\;20\;sec)$ by three methods: by heating in a stainless steel tube immersed in a hot water bath(MP0), by heating in a microwave cavity to a defiled temperature and then holding in a hot water bath(MP1), and by both heating and holding in a microwave cavity(MP2). The microbial quality based on the total plate count was in the order of MP0, MP2 and MP1. The three samples pasteurized by different methods showed the similar microbial quality with respect to the coliform count, psychrotrophic bacterial count and phosphatase activity. The destruction of trypsin inhibitor was in the order of MP0, MP1 and MP2. There were no significant differences in pH, titratable acidity, viscosity and vitamin $B_2$ content before and after pasteurization and among the different pasteurization methods. The similar or higher quality retention of the MP1 or MP2 supports the possibility of using microwave energy for the HTST pasteurization of soymilk and other fluid food products.

Efficacy of Listeria Innocua Reduction on Enoki Mushrooms by Utilization of an Air Sterilization Device (공기 살균 장치 적용 팽이버섯 재배사의 Listeria Innocua 저감 효과)

  • For sterilization of microorganisms of the Listeria genus contaminating enoki mushroom, pilot mushroom grower equipped with air sterilization devices were developed. Sterilization experiments were performed using physical and chemical treatments. Internal temperature and humidity were controlled, maintaining 6.62℃±0.30 in the upper shelves, 6.46℃±0.24 in the middle shelves, and 6.48℃±0.25 in the lower shelves. Humidities were 79.97%±4.42, 79.43%±4.06, and 79.94±4.30%, respectively, with a temperature setting of 6.5℃, and a relative humidity of 75%. A suitable enoki mushroom cultivation stage for air sterilizer application was during the growth stage, with temperature in the 6.5~8.5℃ range, and humidity of 70~80%. At these same internal conditions, the ozone concentration in the mushroom cultivator was found to be 160 ppb during ion-cluster generator operation. After physical sterilization, the Listeria innocua survival rate was 0.1 to 0.9% using ion cluster sterilization, and 9.3 to 10.6% using UV air sterilization. The Listeria innocua survival rates on different materials were 9.3~10.6% on the metal specimen, and 9.9~16.2% on the plastic wrapper. The survival rate was particularly high on the rough side of the plastic wrapper. Ion cluster air sterilization is a labor-saving and effective method for suppressing the occurrence of Listeria bacteria on mushroom growers walls and shelves. For the plastic wrapper, chemical sterilization is more effective than physical sterilization.

환경변이원 및 항변이원에 관한 연구: 살균제 Captan 및 유기인계 살충제 (Diazinon, MEP, Malathion 및 EPN)의 혼합처리가 E. coli $WP_2 S$ (uvrA-, trp-) 균주의 돌연변이와 생잔율로 미치는 영향

  • 살균제 Captan, 살충제 Diazinon, MEP, Malathion 및 EPN 의 단독 처리 그리고 Captan 반복처리와 이들 살충제와 서로 다른 조합으로의 동시적, 시간적 복합처리가 E. coli WP2S(uvrA-, trp-) 균주의 돌연변이유발 빈도 및 생잔율에 미치는 영향을 검토하여 다음과 같은 결과를 얻었다. 1. Captan, Diazinon, MEP, Malathion, EPN 및 UV 의 동시적 복합처리는 이들 농약의 단독 처리에서 보다 E. coli WP2S (uvrA-, trp-)균주의 돌연변이유발빈도가 높았고 이들 농약의 단독처리효과를 합한 가산적 영향으로 나타났으며, 동일조건하에서 이들 농약의 처리농도를 배로 늘렸을 때는 배가효과를 보였으나 이 균주의 증가된 돌연변이 유발빈도는 cinnamaldehyde 30ng 및 60ng/ml 그리고 tannic acid 50ng/ml 배지로 각각 첨가했을 때 감소되었고 생잔율은 증가되었다.
