Physiological Activities of Fermented Garlic Broth during Fermentation (발효기간에 따른 마늘 발효액의 기능성)

  • The purpose of this research is to develop functional food with garlics(Allium sativum var. pekinense) as a healthy food. Fermented garlic broth(FGB)s were prepared with whole bulb of garlics preserved in sugar and sugar syrup, then fermented and aged at room temperature for 36 months. Biological activities of FGBs were tested antibacterial, antioxidative, fibrinolytic activities and analyzed for polyphenol contents. The total polyphenol contents of FGBs in 12~36 month fermented broth(870~885 mg/100 mL of broth) had significantly higher than those of 1~6 months fermented broths(p<0.001). The electron donating abilities(EDAs) and SOD-like activities of 24~36 month fermented broth had significantly higher than those of 1~6 months fermented broths(p<0.05). FGBs had shown strong antibacterial activities against four kinds of pathogenic bacteria(L. monocytogenes, S. aureus, E. coli O157:H7, and Sal. typhimurium). The fibrinolytic activities of 24~36 months fermented broth had more than twice of the fibrinolytic activity of plasmin. FGBs had increasing activities in antibacterial, antioxidative and fibrinolytic activity as the progress of fermentation period. FGBs can be used as natural antioxidant to prevent oxidative damage on normal cells probably because of their antibacterial, antioxidative and fibrinolytic activities.

Quality characteristics of smoked duck using natural curing agent (천연 염지제을 이용한 훈제오리의 품질특성)

  • The objective of this study was to examine quality characteristics of a smoked duck marinated in natural curing agents containing extracts of pear by sugar, celery powder, and vitamin C. We evaluated the composition food additives residue and total plate count, chromaticity, sensory properties, total polyphenol contents, 2,2-Diphenyl-1-picrylhydrazyl (DPPH) radical scavenging effect of general smoked duck (GSD) and natural smoked duck (NSD). The nitrite ion, antioxidant and sodium glutamate of NSD were not detected. The lightness, yellowness and redness of the two smoked duck decreased significantly as growing longer storage days. The total plate count (CFU/g) of coliform bacteria of two smoked duck were not grown. The sensory properties showed that NSD was higher than GSD in the aspect of taste, texture and overall preferences. The total polyphenol contents and DPPH radical scavenging effect of NSD were higher than those of GSD. These results suggested that it was possible to manufacture a smoked duck marinated in natural curing agents including extracts of pear by sugar, celery powder, and vitamin C for the improvement of safety and quality.

Antioxidative Activities and Antiproliferation Effects on Oral Carcinoma KB Cell of the Brazilin from Caesalpinia sappan L. (소목(Caesalpinia sappan L.)에서 분리한 Brazilin의 항산화 활성과 구강상피암 KB 세포주의 증식억제효과)

  • Caesalpinia sappan L. has long been commonly used in oriental folk medicines and as dyes materials. To investigate the antioxidative activities and antiproliferation effects of brazilin from C. sappan heart wood, the MeOH soluble extract was successively fractionated by using hexane, $CHCl_{3}$, EtOAc, BuOH, MeOH, and $H_{2}O$. In these fractions, we were purified brazilin from EtOAc fraction which partitioned to 3.94% of the highest yields. The effects of brazilin and the extracts on human oral carcinoma cells (KB) by MTT assay and their antioxidant activities by DPPH, TCA assay and Fenton reaction were tested. The results showed that the brazilin could inhibits the proliferation of KB cells and obviously decreased the production of nitric oxide of the cells. When the concentration of the brazilin reached to $100\;{\mu}g/ml$, the inhibition percentage of the cell growth was about 60%. In assay on antioxidant activities, The results showed that brazilin exhibit the highest capacity of DPPH free radical scavenging effects among tested extracts. When the concentration of brazilin reached to 1 mg/ml, the lipid peroxide inhibition and radical inhibition activities were determined to be 65.0% and 85.8%, respectively. These results are suggest that brazilin have stronger antiproliferation effect on KB cell and antioxidant properties.

Antioxidant Properties of Cultured Wild Ginseng Roots Extracts (산삼배양근 추출물의 항산화 특성)

  • We obtained hot-water extracts (HWE) and 70% (v/v) ethanol extracts (EE) from cultured wild ginseng roots (CWGR) and determined the saponin and total polyphenol contents, and antioxidant activities. The yields of freeze-dried powder from the HWE and EE were 27.86% and 18.33% (both w/w), respectively. The total polyphenol content of the EE (22.63 mg/g) was higher than that of the HWE (17.90 mg/g). Ginsenoside-Rb1 and -Rg1 contents of hot-air-dried CWGR were 17.90 mg/g and 22.63 mg/g, respectively. The electron-donating ability of HWE and EE were 2.82-60.58% and 3.88?70.88%, respectively, and the reducing powers ($OD_{700}$) were 0.02-0.17 and 0.07-1.90, respectively, at concentrations of 1-20 mg/mL. Thus, the HWE reducing power was markedly lower than that of the EE, but the SOD-like activity of the EE was significantly higher than that of the HWE. The nitrite-scavenging activities of HWE and EE were 9.25-19.18% and 11.94-53.49%, respectively, at concentrations of 1-20 mg/mL. Additionally, the TBARS (Thiobarbituric acid reactive substances, % value) of the EE (1-20 mg/mL) was 9.18-66.59%, thus 1.9-2.8-fold greater than that of the HWE (4.74-24.88%). In conclusion, we provide experimental evidence that extracts of CWGR may be natural antioxidants.

Effect of collection time on the chemical composition and levels of thiobarbituric acid reactive substance of Godulbaegi (Youngia sonchifolia M.) (채취시기에 따른 고들빼기의 성분 조성과 산화방지활성)

  • This study analyzes the chemical composition and thiobarbituric acid reactive substance levels of Godulbaegi (Youngia sonchifolia M.) depending on collection time. The moisture and crude fat content in leaf and root decreased, while crude fiber, crude protein, carbohydrate, and ash increased with increases in collection time. The mineral elements tended to increase in each sample with increases in collection time. The content of vitamin B increased as collection time increased. Vitamin C content was approximately five times higher in the leaves than that in the roots. Total amino acids in leaf and root increased considerably as collection time increased content of phenolic compounds in root were higher than that in the leaf and these contents increased. Antioxidant activity of Godulbaegi was higher in the root than in the leaf and increased as collection time increased.

Effect of Solvent Extracts from Sargassum hemiphyllum on Inhibition of Growth of Human Cancer Cell Lines and Antioxidant Activity (짝잎모자반(Sargassum hemiphyllum)의 암세포주 증식 억제 및 항산화 효과)

  • This study was carried out to determine the inhibitory effects of solvent extracts from Sargassum hemiphyllum on growth of cancer cell lines (AGS human gastric adenocarcinoma and HT-29 human colon cancer cells) and production of lipid peroxides. Inhibitory effects of acetone with methylene chloride extract from S. hemiphyllum on the growth of AGS and HT-29 cancer cells were increased as dose dependent patterns (p<0.05). The methanol extract was more effective on inhibition of growth of AGS. The treatments of hexane, 85% aq. methanol, butanol and water fractions significantly inhibited the growth of cancer cells (p<0.05) and the inhibitory effect was stronger in HT-29. In DCFH-DA (dichlorodihydrofluorescin diacetate) assay, acetone with methylene chloride and methanol extracts showed a stronger inhibitory effect on the production of cellular lipid peroxides (p<0.05) compared with the butanol and hexane fractions. These results indicate that the consumption of S. hemiphyllum may be recommended as a potent functional food for preventing cellular oxidation and cancer.

Analysis of Volatile Organic Compounds Produced from Incineration of Papers at 600°C (600°C에서 제지류 소각시 발생하는 휘발성 유기화합물 농도분석 연구)

  • This study analyzed concentrations of volatile organic compounds (VOCs) produced from incineration of papers at $600^{\circ}C$. The papers used in this study included A4 papers (new, printed with ink-jet, printed with carbon), newspapers (printed with bean oil, printed with a general newspaper ink), packaging box, document envelope, single-use paper cup, and cosmetic tissue. Papers were heated from room temperature upto $600^{\circ}C$ providing air inside of the electric furnace and then they were oxidized for 80 minutes at $600^{\circ}C$ maintaining the same air supply. VOCs emitted from the incineration process were sampled using an air sampling pump and bags for 160 minutes and then the components and concentrations of the VOCs were analyzed by a CC-MS. The most prominent chemical structure of the Vous identified from incineration of the papers was furans and then furans were followed by aromatics and aliphatic alkenes. About 40% of the identified VOCs contained double bonds, which have relatively a high ozone (ground level) formation potential, within their molecular structure. Also, some cancer suspecting compounds like benzene, dichlorormethane and chloroform were identified.

Basic characteristics of an indented cylinder broken rice separator (원통형 홈 선별기의 쇄미선별 특성)

  • 본 연구는 상업용 규모의 쇄미선별공정에 사용할 수 있는 파이로트 규모 원통형 홈 선별기 시작기의 설계 및 제작에 필요한 기초 연구로서, 실험실용 원통형 홈 선별기를 이용하여 쇄미선별 실험을 실시하였다. 원통 회전속도, trough각도, indent 크기와 형상, 공급량을 요인으로 하여 완전립의 수거율과 순도, 쇄립의 수거율과 순도 및 선별효율을 조사하였다. 수행한 연구결과를 요약하면 다음과 같다. 1. 선별효율에 대한 원통 회전속도와 trough각도의 단독효과 및 교호작용은 1% 수준에서 유의성이 인정되었다. 원통 회전속도가 커지면trough각도 역시 그에 따라 적절히 증가시켜야만 선별효율의 저하가 방지되는 것으로 나타났다. 2. 최고 선별효율 값은 홈의 모양과 크기, 그리고 공급량에 관계없이 낮은 회전속도 (17rpm)와 중간 trough각도 (37.5$^{\circ}$또는 60$^{\circ}$)가 조합된 처리에서 나타났으며, 60~70% 범위의 높은 값을 보였다. 선별효율에 관한 원통 회전속도와 trough각도의 최적 조합은 17rpm, 37.5$^{\circ}$라고 판단된다. 3. 말발굽형 홈과 반구형 홈 간 선별효율의 차이는 없었다. 따라서, 실제 상업용 규모의 원통형 홈 쇄미선별기 개발에 있어서는 제작이 쉽고 유지.보수가 간편한 반구형 홈을 채택하는 것이 바람직할 것으로 생각된다. 길이 2.5mm이하의 미립인 쇄미의 선별에 사용할 홈의 크기는 2.5mm 보다는 약간 큰 3.0mm 정도가 되어야만 할 것으로 판단된다. 4. 공급량에 따른 선별효율의 차이는 1% 수준에서 유의성이 인정되었으며, 공급량이 작았을 때 전반적으로 선별효율이 높았다.타리 시프터를 채택, 사용하고 있었으며, 로타리시프터 사용상의 문제는 회전몸체를 지지하는 rod spring의 파손 등 구조와 관련된 것이었다. 로타리 시프터에 의한 쇄립의 선별과 제거정도는 만족할 만한 수준은 아니었다. 4. 국내 유통백미 완전립의 길이, 폭, 두께는 각각 5.02mm, 2.93mm, 2.03mm이었으며, 산물밀도와 천립중은 각각 745.3kg/m3 및 20.46g이었다. 5. RPC 백미제품의 품질경쟁력 향상을 유도하고자 현행 쇄미의 정의와 기준을 보다 강화하여 다음과 같은 쇄미 기준과 계급을 설정, 제시하였다. "완전립" - 길이가 3.75mm이상인 미립 "준완전립" - 길이가 2.5∼3.75mm인 미립 "쇄미" - 길이가 1.75∼2.5 mm인 미립 "이물" - 길이가 1.75mm이하인 미립.볼 때 상토 종자혼합비 6 : 1, 성형롤 회전속도 약 7 rpm으로 판단되며, 이 때 제조능력은 시간당 약 65 Kg(펠렛종자 약 39,000 개), 성형률 약 87 %, 종자손실률은 약 30 %, 펠렛종자 내 평균 종자수는 약 5.5 개, 완전 벼 종자 3개 이상 포함 펠렛종자 비율은 약 100 %가 될 것으로 보인다. 세포의 Androgen 생성을 촉진시키는 역할이 있는 것으로 보여진다 따라서 옻나무 유래 F는 포유동물의 생식기능에 중요하게 작용하는 것으로 사료된다.된다.정량 분석한 결과이다. 시편의 조성은 33.6 at% U, 66.4 at% O의 결과를 얻었다. 산화물 핵연료의 표면 관찰 및 정량 분석 시험시 시편 표면을 전도성 물질로 증착시키지 않고, Silver Paint 에 시편을 접착하는 방법으로도 만족한 시험 결과를 얻을 수 있었다.째, 회복기 중에 일어나는 입자들의 유입은 자기폭풍의 지속시간

Performance assessment of Magnesium Bipolar Plates for Light Weight PEM Fuel Cell (PEM 연료전지 경량화를 위한 마그네슘 분리판의 성능평가)

  • In present paper, we used magnesium alloy having a lower density and higher electrical conductivity for bipolar plate to reduce the weight of PEM fuel cell. The silver was coated to prevent corrosion and form passivation film on the metal surface with sputtering. In acid proof evaluation for setting optimal coating conditions, the homogeneity of coating thickness was improved by coating with the thickness of 3 ${\mu}m$ which not indicated any micro cracks and the temperature $180^{\circ}C$. The performance test and evaluation based on the clamping pressure and channel depth to determine the configuration of bipolar plate for assembling single cell was implemented. And then we assembled single cell with this bipolar plate and implemented the performance test to ensure and compare the current-voltage performance followed as several factors such as coating or non-coating, the change of clamping pressure, the change of channel depth, etc. As these results, the maximum power density of single cell with the coated bipolar plate was 192 $mW/cm^2$ and it was confirmed that the power density per unit mass was better than existing metal bipolar plate.

Anti-oxidant and Anti-inflammatory activities of Zanthoxylum schinifolium Essential Oil (산초 종자 정유의 항산화 및 항염 효능)

  • The purpose of this study was to investigate anti-inflammatory and antioxidant effects of essential oil from seed of Zanthoxylum schinifolium on cultured RAW 264.7 cell line. Antioxidant activity of essential oil was evaluated by two different assays as 2,2-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging and super oxide dismutase (SOD) like activities. This essential oil was tested for cell viability on RAW 264.7 cell line by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-tetrazolium bromide (MTT) assay. The effects of anti-inflammatory on LPS-induced RAW 264.7 cell line was studied by the content of nitric oxide (NO) and prostagladin $E_2$ ($PGE_2$) in cells. The essential oil of seed from Zanthoxylum schinifolium obtained dose-dependently increased the scavenging activity on DPPH radical scavenging activity and SOD like activity. The essential oil showed low cytotoxicity as more than 98% cell viability in $40{\mu}g/mL^{-1}$ concentration. The essential oil of seed extracted from Zanthoxylum schinifolium presented obvious effect on inflammation. These results suggest that essential oil of seed from Zanthoxylum schinifolium may have value as the potential anti-inflammatory effects by decreasing the action of NO and $PGE_2$ and preventing the activation of oxidative.
